15份多花黑麦草优良引进种质的表型变异分析   总被引:1,自引:0,他引:1  
多花黑麦草(Lolium multiflorum Lam.)是世界栽培牧草中的优良禾本科牧草。为更好地利用多花黑麦草种质资源,本研究对引自国外的15份多花黑麦草种质的15个形态性状和农艺性状进行了变异系数、相关性分析、主成分分析和聚类分析。结果表明:15份材料间存在较大变异,除单株干重外,其余性状在供试材料间均表现出显著性差异,变异系数范围为10.28%~39.15%,变异系数平均值为19.49%,从小到大依次为小穗数小穗长株高分蘖数千粒重花序长倒二叶长小花数茎粗第一节间长倒二叶宽旗叶宽旗叶长单株干重单株鲜重。主成分结果表明,前5个主成分累积贡献率达到84.51%,第1主成分以株高为主要特征;第2主成分以旗叶和倒二叶长、宽为主要特征;第3主成分以花序长和小花数为主要特征;第4主成分以千粒重为主要特征;第5主成分以产草量为主要特征。15份种质材料经基于欧氏距离的UPGMA聚类分析被划分为3大类,类别间存在较显著的差异,其中第2类因其植株高大、叶片宽大的特点,具备选育高产种质的潜力。  相似文献

New rice lines, restorer line RB207 and maintainer line Yewei B, with better agronomic traits were separately developed from variant progeny of R207 (rice restorer line) and V20B (rice maintainer line) through transformation of genomic DNA ofEchinochloa crusgalli (C4 plant) andOryza minuta, respectively. The phenotypes of the variant lines were apparently different from those of the receptors. Yewei B had stronger tolerance to high temperature than did V20B. The number of spikelets per panicle and the 1000-grain weight of RB207 increased by 40% over those of R207. The results of amplified fragment length polymorphism (AFLP) analysis indicated that the polymorphism rates were both 4.4% between genomes of the variant lines and their receptors. Results demonstrated that special DNA segments fromE. crusgalli andO. minuta might integrate into the genome of cultivated rice and could be stably passed on. The study further shows that transformation of genomic DNA of distant relatives is an effective approach for creating new rice germ plasm. These authors contributed equally to this work.  相似文献

A high throughput protocol was established to preserve 140,000 mutants of a moss, Physcomitrella patens, a model plant for functional genomics studies, over liquid nitrogen. Regarding the reliable long-term storage of diverse mutant phenotypes, as well as time and cost effectiveness, each working step was optimized: 1) plant preparation, 2) freezing regime, cryogenic conditions, 3) regrowth after thawing. A prerequisite for maximum regrowth was a 1-week preculture of chopped plant material on a supplemented medium prior to freezing. Cryo vials as preculture vessels resulted in identical regrowth rates, compared to petri dishes. The cryo vial type had a significant influence on regrowth rates. A cooling rate of - 1 degrees C/min down to - 35 degrees C with a 10 min holding time before transferring plants to - 152 degrees C was the most suitable freezing regime. This protocol allows a cryopreservation of 1100 plants during a 5-day working week, practicable by one person. For more than 650 cryopreserved mutants a maximum regrowth rate of 100 % was obtained, independently of mutant phenotypes.  相似文献

Summary Christmas bush (Ceratopetalum gummiferum Sm) is a shrubby tree species of the east coast of New South Wales in Australia. It is much prized as a cut flower crop because of its bright, pinky red floral calyces. New varieties are being developed, the storage of which is an important issue. In this study, it was shown that shoot tips sampled from in vitro plantlets withstood cryopreservation using the encapsulation-dehydration technique. The protocol leading to optimal regrowth was the following: excised shoot tips were pretreated for 1 d in the dark on hormone-free Murashige and Skoog (MS) medium with 0.3 M sucrose, then encapsulated in 3% calcium alginate and precultured in liquid MS medium with 0.5 M sucrose for 3 d. Precultured beads were dehydrated for 6 h in the air current of the laminar flow cabinet to 24.3% moisture content (fresh weight basis) before rapid immersion in liquid nitrogen. Under these conditions, regrowth of shoot tips after cryopreservation reached 61.4%. Regrowth of cryopreserved shoot tips was not affected by the period of cold acclimation of in vitro mother plants.  相似文献

Plant genetic resource conservation strategies, informed by an understanding of the geographical distribution of genetic variation within species, are likely to result in a wider representation of conserved diversity in ex situ gene banks and in situ genetic reserves. The main objective of this study was to map the geographical distribution of genetic variation, as revealed by randomly amplified polymorphic DNA (RAPDs), in four wild relatives of the cultivated lentil, namely Lens culinaris ssp. orientalis, L. odemensis, L. ervoides and L. nigricans . Areas of high diversity and unique diversity were located for each taxon, and regions where further germplasm collection was most likely to yield novel genetic variation were identified. There were centres of diversity for L. culinaris ssp. orientalis in southeast Turkey and northwest Syria, and in south Syria and Jordan. A centre of diversity was found to exist in Sweida province, south Syria, for L. odemensis , and for L. ervoides along the coastal border region between Syria and Turkey stretching down along the Syrian coast. There was a centre of diversity for L. nigricans in west Turkey. Analytical techniques previously used at the species level were found to be useful at the genotypic level to objectively target areas for future collection missions, to increase diversity in ex situ collections and to target areas for in situ conservation.  相似文献

对武夷山茶区42份武夷名丛茶树资源的主要生化成分进行了评价鉴定和遗传多样性分析。结果发现,武夷山茶树资源的生化成分存在丰富的多样性和变异,平均遗传多样性指数达到2.06%,平均变异系数达到22.01%。通过多变量的主成分分析,前3个主成分代表了茶树生化成分多样性的89.02%的信息。基于生化成分,把42份武夷名丛聚类划分为3个类群。3个类群除了咖啡碱含量差异不显著外,其他各生化成分均存在显著差异。第Ⅰ、Ⅱ类群大部分为适制乌龙茶的资源,第Ⅲ类群大部分为适制绿茶的资源。并从中筛选出一批生化成分特异的资源。  相似文献

SCo T是一种新型的目的基因分子标记,该标记不仅能获得与性状联系紧密的目的基因,而且能对性状进行跟踪,已被广泛应用于多种植物的研究。本文概述了SCo T标记的原理、引物设计方法及特点,并从PCR反应体系建立与优化、种质资源遗传多样性与亲缘关系分析、种质鉴定与指纹图谱构建、基因差异表达与分子遗传连锁图谱构建等方面总结了SCo T标记的应用进展。同时,对其存在的问题进行了讨论,并展望了该标记的发展应用前景。  相似文献

采用针刺叶片接种法、温室地面水层保湿方式,对146份普通菜豆种质资源进行抗普通细菌性疫病鉴定和评价,结果表明,该接种方法简便实用,保湿方式效果良好,鉴定结果准确;从146份普通菜豆种质中筛选出抗病种质(R)2份、中抗(MR)种质50份、感病(S)种质81份、高感种质(HS)13份。本研究表明针刺叶片接种、温室地面水层自然蒸发保持湿度可以作为大规模菜豆抗普通细菌性疫病鉴定的适宜方法。  相似文献

We report here that, during periodical surveys of insects inhabiting diverse habitats for the collection of entomopathogenic fungi; a large number of isolates were recovered belonging to seven species, from various regions of Madhya Pradesh and Chhattisgarh forest areas and agricultural fields. The most common entomopathogenic fungi such as Beauveria bassiana, Nomuraea rileyi, Paecilomyces farinosus and Paecilomyces fumosoroseus were found to infect various insect hosts species naturally viz. Hyblaea puera, Eutectona machaeralis, Diachrysia orichalcea, Spodoptera litura, and few new insect hosts of these fungal pathogens among Indian insect population were collected for the first time from Central India, such as beetles of Agrilus species, hairy caterpillars of Lymantria species. The isolation, identification, maintenance and pathogenicity assay of these isolates was performed prior to deposition in culture collection center.  相似文献

采用离体叶片接种法,对43份马铃薯种质资源进行晚疫病抗性鉴定、比较和评价。以接种5 d后的感病品种‘Désirée’和抗病品种‘加湘1号’叶片症状为对照,鉴定出5份表现为高抗的种质资源,其中包含3个Solanum phurejia和2个S. tuberosum ssp. andigena材料。另外,还鉴定出14份中抗材料(No. 7~20)。结果表明,野生种和安第斯山栽培亚种马铃薯资源的抗性材料较为丰富,可作为晚疫病抗病育种的亲本。  相似文献
