Nanosecond pulsed electric fields (nsPEFs) are increasingly recognized as a novel and unique tool in various life science fields, including electroporation and cancer therapy, although their mode of action in cells remains largely unclear. Here, we show that nsPEFs induce strong and transient activation of a signaling pathway involving c-Jun N-terminal kinase (JNK). Application of nsPEFs to HeLa S3 cells rapidly induced phosphorylation of JNK1 and MKK4, which is located immediately upstream of JNK in this signaling pathway. nsPEF application also elicited increased phosphorylation of c-Jun protein and dramatically elevated c-jun and c-fos mRNA levels. nsPEF-inducible events downstream of JNK were markedly suppressed by the JNK inhibitor SP600125, which confirmed JNK-dependency of these events in this pathway. Our results provide novel mechanistic insights into the mode of nsPEF action in human cells.  相似文献

Electrical excitability and signaling, frequently associated with rapid responses to environmental stimuli, have been documented in both animals and higher plants. The presence of electrical potentials (EPs), such as action potentials (APs) and variation potentials (VPs), in plant cells suggests that plants make use of ion channels to transmit information over long distances. The reason why plants have developed pathways for electrical signal transmission is most probably the necessity to respond rapidly, for example, to environmental stress factors.We examined the nature and specific characteristics of the electrical response to wounding in the woody plant Persea americana (avocado). Under field conditions, wounds can be the result of insect activity, strong winds or handling injury during fruit harvest. Evidence for extracellular EP signaling in avocado trees after mechanical injury was expressed in the form of variation potentials. For tipping and pruning, signal velocities of 8.7 and 20.9 cm/s, respectively, were calculated, based on data measured with Ag/AgCl microelectrodes inserted at different positions of the trunk. EP signal intensity decreased with increasing distance between the tipping and pruning point and the electrode. Recovery time to pre-tipping or pre-pruning EP values was also affected by the distance and signal intensity from the tipping or pruning point to the specific electrode position. Real time detection of remote EP signaling can provide an efficient tool for the early detection of insect attacks, strong wind damage or handling injury during fruit harvest.Our results indicate that electrical signaling in avocado, resulting from microenvironment modifications, can be quantitatively related to the intensity and duration of the stimuli, as well as to the distance between the stimuli site and the location of EP detection. These results may be indicative of the existence of a specific kind of proto-nervous system in plants.  相似文献

While electric fields at intermediate frequencies are not widely utilized for industrial technologies, surprisingly, certain toys emit the highest electric fields found in our living environment. These toys, plasma balls, are devices that use high voltage to create ionized light discharges. In this study, we assessed exposure to electric fields and contact/induced current from a recreational plasma ball device. The electric field strength was measured as a function of distance from the device, and the contact/induced current was measured with a current clamp in different exposure situations with point or grasping contact. The characteristic spectra of the electric field and contact current were measured, and both the multiple frequency rule and weighting of the spectra were applied according to the International Commission on Non-Ionizing Radiation Protection (ICNIRP) 1998 and 2010 guidelines. The results indicate that the recommended reference levels for the general public are exceeded at distances <1.2 m, and that the contact currents in the hand may be twice higher than recommended by the general public guidelines.  相似文献

The biological effects of extremely low-frequency electric fields (ELF) on living organisms have been explored in many studies, but the results are controversial and only a few studies investigated the influence of the intensity of the applied field on seedling growth. Here we assess the effects of a 50 Hz sinusoidal electric field on the early growth of Vigna radiata seedlings while varying the field intensity. Experiments performed in a dark, constant-climate chamber on several thousands of seedlings show that the field produces an inhibitory effect at a low field intensity and an enhancing one at a higher intensity. The maximum negative effect occurs at about 450 V/m, which is an intensity much lower than the exposure limits currently in force in the safety regulations.  相似文献

Electrotherapy with direct current delivered through implanted electrodes is used for local control of solid tumors in both preclinical and clinical studies. The aim of this research is to develop a solution method for obtaining a three-dimensional analytical expression for potential and electric current density as functions of direct electric current intensity, differences in conductivities between the tumor and the surrounding healthy tissue, and length, number and polarity of electrodes. The influence of these parameters on electric current density in both media is analyzed. The results show that the electric current density in the tumor is higher than that in the surrounding healthy tissue for any value of these parameters. The conclusion is that the solution method presented in this study is of practical interest because it provides, in a few minutes, a convenient way to visualize in 3D the electric current densities generated by a radial electrode array by means of the adequate selection of direct current intensity, length, number, and polarity of electrodes, and the difference in conductivity between the solid tumor and its surrounding healthy tissue.  相似文献

Irreversible electroporation (IRE) is a novel technique that deals with killing undesirable cells, mainly cancer cells, directly without using any cytotoxic drugs. Commonly in this technique very high electric field up to 1000?V/cm is used but for very short exposure time (nanoseconds). Low electric fields (LEFs) are used before to internalize molecules and drugs inside the cells (electroendocytosis) but mainly not in killing the cells. The aim of this work is to determine the ability of using LEFs to kill cancer cells (Hela cells). The Physics idea is in making LEFs energy equivalent to IRE energy. Four IRE protocols were selected to represent very high, high, moderate and mild voltages IRE, then we make equivalent energy for each of these protocols using different LEFs’ parameters of different amplitudes (7, 10, 14 and 20?V), different pulse numbers (40, 80, 160 and 320 pulses), different frequencies from 0.5 to 106.86?Hz and different pulse widths from 9.38 to 2000?ms. Each of the calculated LEF equivalent to IRE was applied on Hela cell line. The results show complete destruction of the cancer cells for all the tested exposure protocols. This damage was not due to thermal effect because the measured temperature was not changed before and after the exposure. The possible effect mechanism is discussed. It was concluded that the lethal effect on the cancer cells can be achieved using LEFs if the same energy equivalent to IRE is used. This work will help in using low-risk drug-free techniques in cancer treatment.  相似文献

An innovative design is reported of a direct‐current triboelectric nanogenerator (DC‐TENG) based on a rotating disk design for harvesting rotational mechanical energy. The DC‐TENG consists of two disks and two pairs of flexible electric brushes that are made of carbon fiber and contact two electrodes, respectively. During the rotation, two disks have distinct triboelectric polarities for a cyclic in‐plane charge separation between them and an alternating current is generated between the two electrodes. Because of the sliding contact and automatically switch between the electric brushes and the two electrodes, the current is reversed in the second half of the cycle and a direct current is generated. The role that the rotating speed and the segmentation number have is thoroughly investigated and shows that there is direct current enhancement not only at higher speed but also with more segments. The DC‐TENG has been demonstrated as a constant current source for directly and continuously driving electronic devices and/or charging an energy storage unit without a rectifier bridge. This work presents a novel DC‐TENG technology and opens up more potential applications for harvesting rotational mechanical energy and powering electronics.  相似文献
