Environmental gradients have been shown to disrupt gene flow in marine species, yet their influence in structuring populations at depth remains poorly understood. The Cape hakes (Merluccius paradoxus and M. capensis) are demersal species co‐occurring in the Benguela Current system, where decades of intense fishing resulted in severely depleted stocks in the past. Previous studies identified conflicting mtDNA genetic substructuring patterns and thus contrasting evolutionary trajectories for both species. Using 10 microsatellite loci, the control region of mtDNA and employing a seascape genetics approach, we investigated genetic connectivity and the impact of prolonged exploitation in the two species, which are characterized by different patterns of fishing pressure. Three consecutive years were sampled covering the entire distribution (N = 2100 fishes). Despite large estimated population sizes, both species exhibited low levels of contemporary genetic diversity (0.581 < HE < 0.692), implying that fishing has had a significant impact on their genetic composition and evolutionary trajectories. Further, for M. paradoxus, significant temporal, but not spatial, divergence points to the presence of genetic chaotic patchiness. In contrast, M. capensis exhibited a clear latitudinal cline in genetic differentiation between Namibia and South Africa (FST = 0.063, P < 0.05), with low (0.2% per generation) estimates of contemporary gene flow. Seascape analyses reveal an association with bathymetry and upwelling events, suggesting that adaptation to local environmental conditions may drive genetic differentiation in M. capensis. Importantly, our results highlight the need for temporal sampling in disentangling the complex factors that impact population divergence in marine fishes. 相似文献

The tropospheric level of the phytotoxic air pollutant ozone has increased considerably during the last century, and is expected to continue to rise. Long-term exposure of higher plants to low ozone concentrations affects biochemical processes prior to any visible symptoms of injury. The current critical level of ozone used to determine the threshold for damaging plants (biomass loss) is still based on the seasonal sum of the external concentration above 40 nl·l−1 (AOT40). Taking into account stomatal conductance and the internal capacity of leaf defences, a more relevant concept should be based upon the 'effective ozone flux', the balance between the stomatal flux and the intensity of cellular detoxification. The large decrease in the Rubisco/PEPc ratio reflects photosynthetic damage from ozone, and a large increase in activity of cytosolic PEPc, which allows increased malate production. Although the direct detoxification of ozone (and ROS produced from its decomposition) is carried out primarily by cell wall ascorbate, the existing level of this antioxidant is not sufficient to indicate the degree of cell sensitivity. In order to regenerate ascorbate, NAD(P)H is needed as the primary supplier of reducing power. It is hypothesised that increased activity of the catabolic pathways and associated shunts (glucose-6-phosphate dehydrogenase, NADP-dependent glyceraldehyde-3-phosphate dehydrogenase, isocitrate dehydrogenase and malic enzyme) can provide sufficient NAD(P)H to maintain intracellular detoxification. Thus, measurement of the level of redox power would contribute to determination of the 'effective ozone dose', serving ultimately to improve the ozone risk index for higher plants. 相似文献

Chronic wounds usually remain in the inflammatory phase of the healing process during several months or even years. Hence, a continuous research has been resulting in the development of wound dressings with improved performance. Herein, we report a delivery system for cutaneous wound healing, consisting of a textile material (non-woven gauzes) covered with lipidic vesicles containing diclofenac, a non-steroidal anti-inflammatory drug (NSAID). This study also aims to compare the entrapment efficiency data with previous works and confirm that this parameter and drug amount are not directly correlated. A method of dehydration–rehydration of the liposomes presenting different sizes and lamellarities was used to assess the best conditions to attain the highest drug entrapment efficiency. Optimum conditions for the NSAID release were achieved with high phospholipid concentrations and dried-rehydrated vesicles (DRVs) prepared from multilamellar liposomes (MLVs). A chemical activation of the gauzes was performed to enhance the vesicles attachment, also contributing to a higher drug amount in the surrounding media. In spite of the entrapment efficiency being lower comparatively with other values presented by us previously, the diclofenac concentration was considerably higher in this formulation. Entrapment efficiency is, therefore, not sufficient per se to define the real amount of drug contained in the formulation. The cytocompatibility assessment in human skin fibroblasts showed that DRVs from MLVs and DRVs from large unilamellar liposomes (LUVs) with less than 750?μM of egg-yolk phosphatidylcholine (EPC), containing diclofenac, were not cytotoxic after 72?h of contact, greatly implying potential for their application in the chronic wounds healing. 相似文献

The Kennedy Krieger lead paint study is a landmark case in human experimentation and a classic case in research ethics. In
this paper I use the lead paint study to assist in the analysis of the ethics of research on less expensive, less effective
interventions. I critically evaluate an argument by Buchanan and Miller who defend both the Kennedy Krieger lead paint study
and public health research on less expensive, less effective interventions. I conclude that Buchanan and Miller’s argument
is flawed but that does not mean that research designed to find less effective interventions cannot be justified in some situations.
Based on my analysis, I suggest questions to ask when considering such research and I offer some principles to guide us. In
the process, light is shed on the various debates and issues raised by the lead paint study; e.g. standards of care, researchers’
responsibilities to research subjects, the distinction between treatment and research and the question of what it is that
legitimizes public health research.

Radiological dispersion devices (RDDs), commonly called “dirty bombs,” utilize a conventional explosive to deliberately disperse non-fissile material as an aerosol. This analysis models total effective dose equivalent (Sv) at various locations down-wind from the detonation site subsequent to terrorists detonating a 241Am, 137Cs, 60Co, 192Ir, or90Sr RDD. A source term for each isotope equaling 3.7 × 1013 Bq with an instantaneous release by either high explosives or low explosives at street level is assumed in order to evaluate total effective dose equivalent (TEDE) under various meteorological scenarios for intentional releases of non-fissile materials by terrorists. The inhalation pathway on average contributes most to TEDE. The inhalation pathway accounts for 96% (0.22 Sv) of the mean exposure estimate of 0.2321 Sv and occurs over an extremely short time frame (i.e., a few minutes). Ground shine, on average, contributes the second most to TEDE estimates accounting for approximately 4% (0.009 Sv) of the estimate. A cautionary note with regard to ground shine is warranted, however, because Hotspot estimates for this pathway are based on the assumption that a person is exposed for 4 days (96 hours). The TEDE for submersion (i.e., passing through the plume without inhaling particles) is negligible for the scenarios evaluated contributing less than 1% (5.2 × 10?6 Sv) to the TEDE estimate averaged across all 140 model runs (5 nuclides × 2 rainfall scenarios × 2 explosive scenarios × 7 wind and atmospheric stability scenarios). The TEDE value for 241Am from inhalation is much greater, on average, than the inhalation TEDE value for 60Co, 137Cs, 192Ir, or 90Sr. This underscores the potentially high risk to human health posed by exposure to 241Am. Ground shine is the primary exposure pathway for 60Co and 137Cs due to the energetic and penetrating gamma rays those radionuclides emit. 192Ir and 90Sr have relatively low mean TEDE values for all of the pathways examined. 相似文献

Brown fat mitochondria obtained from a hibernator, the golden hamster, were investigated in order to elucidate the significance of membrane permeability for metabolic functioning at different temperatures. The mitochondria were shown to have active permeases for phosphate and pyruvate, but very poorly developed permeases for di- and tricarboxylate substrate anions. This was shown with both osmotic swelling techniques and respiration-driven uptake studies. It was shown that the very limited malate permeation observed was compatible with it being a non-carrier-mediated diffusion process. The role of malate transport in supporting fatty-acid oxidation in vitro as a function of temperature was studied in detail. The results support our earlier suggestion that physiologically pyruvate carboxylase probably functions to generate oxaloacetate when high concentrations of condensing partner are needed during thermogenesis. They may also explain earlier observations that acetate was produced from palmitoyl-carnitine at low temperatures even when malate was present; this is here shown to be due to the limited malate permeability at these low temperatures. Thus, even at the body temperature of the hibernating hamster (4–5°C), brown fat is probably able to combust fatty acids totally. 相似文献

The power of single contrast tests strongly depends on the a priori unknown shape of the dose response relationship. Two approaches can be used to overcome this major disadvantage: the likelihood ratio test for order restriction or multiple contrast tests. Within the closure principle, the shape for the different hyptheses varies. Therefore multiple contrast tests are used as trend tests. The Piétrain pig originates from the Belgian village Piétrain some time between 1920 and 1950. Owing to its superior conformation, the Piétrain has spread worldwide since the 1960s. As initial population sizes were limited and close inbreeding was commonplace, the breed’s genetic diversity has been questioned. Therefore, this study examines Piétrain breed substructure, diversity and selection signatures using SNP data in comparison with Duroc, Landrace and Large White populations. Principal component analysis indicated three subpopulations, and FST analysis showed that US Piétrains differ most from European Piétrains. Average inbreeding based on runs of homozygosity (ROH) segments larger than 4 Mb ranged between 16.7 and 20.9%. The highest chromosomal inbreeding levels were found on SSC8 (42.7%). ROH islands were found on SSC8, SSC15 and SSC18 in all Piétrain populations, but numerous population-specific ROH islands were also detected. Moreover, a large ROH island on SSC8 (34–126 Mb) appears nearly fixed in all Piétrain populations, with a unique genotype. Chromosomal ROH patterns were similar between Piétrain populations. This study shows that Piétrain populations are genetically diverging, with at least three genetically distinct populations worldwide. Increasing genetic diversity in local Piétrain populations by introgression from other Piétrain populations seems to be only limited. Moreover, a unique 90 Mb region on SSC8 appeared largely fixed in the Piétrain breed, indicating that fixation was already present before the 1960s. We believe that strong selection and inbreeding during breed formation fixed these genomic regions in Piétrains. Finally, we hypothesize that independent coat color selection may have led to large ROH pattern similarities on SSC8 between unrelated pig breeds. 相似文献
