In the present study the cold acclimation potential of two accessions of Arabidopsis thaliana was investigated. Significant variation was found for basic tolerance as well as the capacity to acclimate to freezing temperatures. During cold acclimation, levels of soluble sugars increased in both genotypes, but raffinose accumulation discriminated the more tolerant accession Col‐0 from C24. Concentrations of other compatible solutes such as proline and glutamine were also higher in cold‐acclimated Col‐0 than C24 plants. Changes of invertase activity during cold exposure corresponded to changes in sucrose and fructose, but not glucose concentrations and were consistent with an initial chilling response and a later decline in hexose metabolization. When vacuolar invertase was suppressed by siRNA expression, reduced sucrolytic activity resulted in elevated leaf sucrose concentration, whereas the fructose content was strongly reduced. This led to elevated freezing tolerance in the cold‐tolerant genotype Col‐0, but not in C24. The most pronounced metabolic changes in invertase‐inhibited Col‐0 plants occurred for proline and glutamine concentrations, indicating indirect metabolic effects of altered sugar concentrations.  相似文献

Although plant adaptation to serpentine soils has been studied for several decades, the mechanisms of plant adaptation to edaphic extremes are still poorly understood. Arbuscular mycorrhizal fungi (AMF) are common root symbionts that can increase the plant hosts' establishment and growth in stressful environments. However, little is known about the role plant-AMF interactions play in plant adaptation to serpentine. As a first step towards understanding this role, we examined the AMF assemblages associated with field populations of serpentine and non-serpentine ecotypes of California native plant Collinsia sparsiflora. We sampled roots of C. sparsiflora from three serpentine and three non-serpentine sites in close proximity (110 m to 1.94 km between sites) and analysed the small subunit ribosomal DNA gene amplified from root DNA extracts using AMF-specific primers. A total of 1952 clones from 24 root samples (four from each site) were sequenced. We used sequence similarity and phylogenetic analysis to determine operational taxonomic units (OTU) resulting in 19 OTUs representing taxa from six AMF genera, including one serpentine-specific OTU. We used Bray-Curtis similarity, multidimensional scaling and analysis of similarity to compare root sample AMF assemblages. These analyses clearly showed that plant ecotypes associated with distinct AMF assemblages; an Acaulospora OTU-dominated serpentine, and a Glomus OTU-dominated non-serpentine assemblages. Species diversity and evenness were significantly higher in serpentine assemblages. Finally, relate analysis showed a relationship between ecotype AMF assemblages and soil nutrients. This study reveals a strong relationship between AMF associates and plant adaptation to edaphic extremes.  相似文献

Worldwide degradation of coral reef communities has prompted a surge in restoration efforts. They proceed largely without considering genetic factors because traditionally, coral populations have been regarded as open over large areas with little potential for local adaptation. Since, biophysical and molecular studies indicated that most populations are closed over shorter time and smaller spatial scales. Thus, it is justified to re-examine the potential for site adaptation in corals. There is ample evidence for differentiated populations, inbreeding, asexual reproduction and the occurrence of ecotypes, factors that may facilitate local adaptation. Discovery of widespread local adaptation would influence coral restoration projects mainly with regard to the physical and evolutionary distance from the source wild and/or captive bred propagules may be moved without causing a loss of fitness in the restored population. Proposed causes for loss of fitness as a result of (plant) restoration efforts include founder effects, genetic swamping, inbreeding and/or outbreeding depression. Direct evidence for any of these processes is scarce in reef corals due to a lack of model species that allow for testing over multiple generations and the separation of the relative contributions of algal symbionts and their coral hosts to the overall performance of the coral colony. This gap in our knowledge may be closed by employing novel population genetic and genomics approaches. The use of molecular tools may aid managers in the selection of appropriate propagule sources, guide spatial arrangement of transplants, and help in assessing the success of coral restoration projects by tracking the performance of transplants, thereby generating important data for future coral reef conservation and restoration projects.  相似文献

Kokanee are the nonanadromous (freshwater resident) form of sockeyesalmon (Oncorhynchus nerka) found in lake ecosystems throughoutthe North Pacific region. Kokanee commonly exhibit two reproductiveecotypes; `stream-spawners' that reproduce in streams tributary tolakes, and `beach-spawners' that reproduce on submerged lakeshore gravelbeaches. Okanagan Lake, in the southcentral interior of BritishColumbia, Canada, contains beach- and stream-spawning kokanee and bothecotypes have declined dramatically in abundance over the last 20 years.We examined developmental biology (developmental rate to hatching andemergence) and genetic divergence at eight microsatellite loci toinvestigate phenotypic and genetic differentiation between ecotypes tounderstand selective and demographic factors that might influence therecovery of depressed populations. Beach-spawning female kokanee weresmaller and produced smaller eggs than females from stream-spawningpopulations. There was no striking difference in time to 50%hatching between ecotypes, but beach-spawning kokanee developed fasterfrom hatching to emergence. Microsatellite loci were highly polymorphicin kokanee (between 5 and 23 alleles per locus) and showed significantdifferentiation among populations (average = 0.018). There was,however, no significant variation attributable to spawning ecotype afteraccounting for variation within ecotypes. Simulated population-mixtureanalyses indicated good potential for genetic classification of kokaneeas beach- or stream-spawners; estimated mixture proportions were within11% of actual proportions averaged over 50 replications. Our datasuggest that Okanagan Lake kokanee constitute at least two managementunits within a single watershed; the ecotypes appear adapted to distinctthermal reproductive environments and show modest moleculardifferentiation from one another. Persistence of kokanee within OkanaganLake may depend, in part, on management plans that recognize thedistinctions between the sympatric reproductive ecotypes.  相似文献

Adjustment of hydraulic architecture in response to environmental conditions was studied in two warm-desert sub-shrubs, Hymenoclea salsola and Ambrosia dumosa, both at the level of genetic adaptation along a climatic gradient and plastic response to immediate growth conditions. Individuals of both species originating from southern populations developed higher leaf-specific hydraulic conductance in the common greenhouse than individuals from northern populations. Hydraulic conductance was higher in plants grown at high temperature, but did not vary as a function of growth relative humidity. Hydraulic conductance was not correlated within species with individual variation in vessel diameter, cavitation vulnerability, or root:shoot ratio, but was strongly, negatively correlated with the fraction of total plant biomass allocated to leaves. For both species, stomatal conductance (g s) at high leaf-to-air vapor pressure difference (ν) was tightly correlated with variability in hydraulic conductance, as was the sensitivity of stomatal closure to increasing ν. Experimentally increasing shoot water potential by soil pressurization, under conditions where high ν had already caused stomatal closure, led to substantial stomatal reopening in both species, but recovery was significantly higher in H. salsola. Hydraulic conductance was higher in H. salsola than A. dumosa. H.salsola also differed from A. dumosa by being a representative of a highly specialised group of desert shrubs which use the twigs as a major photosynthetic organ. The southern population of H. salsola produced far fewer leaves and relied much more heavily on twig photosynthesis than the northern population. At the whole-plant level, increased reliance on twig photosynthesis was associated with higher leaf-specific hydraulic conductance, but equivalent whole-plant photosynthesis on either a dry weight (μmol CO2 g–1) or nitrogen basis (μmol CO2 g–1)). This suggests that twig photosynthesis might be one way of increasing hydraulic conductance per unit photosynthetic canopy by increasing allocation to an organ which simultaneously performs photosynthetic, support, and transport functions.   • Crop wild relatives can be a useful source of genotypes that maximise crop survival and yield in specific habitats. Lupinus angustifolius is an annual forb with crop varieties derived from a narrow genetic basis but that are cultivated worldwide. Its seeds have high nutritional value, but they naturally contain alkaloids with anti‐nutritive factors. The study of its wild populations can be useful to find genotypes that contribute to higher climate resilience and greater yield under stressing environmental conditions.
  • Using a common garden, we evaluated ecotypic differentiation in four natural populations from two contrasting latitudes in terms of plant biomass, seed mass and number, alkaloid content in seeds for the three main alkaloids present in the plant and seed colour, including its possible influence on post‐dispersal predation. Correlations among traits were also assessed.
  • We found differences among populations for all traits except final biomass. Northern populations had lighter seeds and a tendency to yield more seeds when they produced white seeds, compared to southern populations and variegated seeds. Regardless of latitude, populations showed differences in alkaloid concentration, with all three alkaloids found generally in high or low concentrations in each population. Proportion of white seed morphs varied in each population. Seed colour did not influence predator preference. In addition, white seed colour was related to a low alkaloid content.
  • Our results evidence the existence of natural ecotypic differentiation in L. angustifolius not only due to latitudinal range, but also to local environmental factors. White seed coat colour could be used as a visual clue for identification of low‐alkaloid genotypes, a priority trait in L. angustifolius breeding programmes.

Geographic variation in floral morphology is often assumed to reflect geographic variation in pollinator communities and associated divergence in selective pressures. We studied populations of Nerine humilis (Amaryllidaceae) to assess whether geographic variation in floral form is the result of local adaptation to different pollinator communities. We first tested for associations between floral traits and visitor communities, and found that populations with similar floral morphologies were visited by similar insect communities. Mean style length in each population was also closely associated with the mean body length of the local visitor community. A reciprocal translocation experiment demonstrated that native phenotypes set more seed than translocated phenotypes. Single visitation experiments showed that native flowers received more pollen, and set more seed per visit, than introduced phenotypes in both populations. This suggests that the effectiveness of pollinator visits is determined by the degree of mechanical fit between flowers and visitors. We provide strong evidence that the observed among‐population variation in floral traits is an adaptive response to geographic variation in the pollinator community.  相似文献

Freshwater populations of three-spined sticklebacks (Gasterosteus aculeatus) in northern Germany are found as distinct lake and river ecotypes. Adaptation to habitat-specific parasites might influence immune capabilities of stickleback ecotypes. Here, naive laboratory-bred sticklebacks from lake and river populations were exposed reciprocally to parasite environments in a lake and a river habitat. Sticklebacks exposed to lake conditions were infected with higher numbers of parasite species when compared with the river. River sticklebacks in the lake had higher parasite loads than lake sticklebacks in the same habitat. Respiratory burst, granulocyte counts and lymphocyte proliferation of head kidney leucocytes were increased in river sticklebacks exposed to lake when compared with river conditions. Although river sticklebacks exposed to lake conditions showed elevated activation of their immune system, parasites could not be diminished as effectively as by lake sticklebacks in their native habitat. River sticklebacks seem to have reduced their immune-competence potential due to lower parasite diversity in rivers.  相似文献
