A nearly complete skeleton of a juvenile sauropod from the Lower Morrison Formation (Late Jurassic, Kimmeridgian) of the Howe Ranch in Bighorn County, Wyoming is described. The specimen consists of articulated mid-cervical to mid-caudal vertebrae and most appendicular bones, but cranial and mandibular elements are missing. The shoulder height is approximately 67 cm, and the total body length is estimated to be less than 200 cm. Besides the body size, the following morphological features indicate that this specimen is an early juvenile; (1) unfused centra and neural arches in presacral, sacral and first to ninth caudal vertebrae, (2) unfused coracoid and scapula, (3) open coracoid foramen, and (4) relatively smooth articular surfaces on the limb, wrist, and ankle bones. A large scapula, short neck and tail and elongate forelimb bones relative to overall body size demonstrate relative growth. A thin-section of the mid-shaft of a femur shows a lack of annual growth lines, indicating an early juvenile individual possibly younger than a few years old. Pneumatic structures in the vertebral column of the specimen SMA 0009 show that pneumatisation of the postcranial skeleton had already started in this individual, giving new insights in the early ontogenetic development of vertebral pneumaticity in sauropods. The specimen exhibits a number of diplodocid features (e.g., very elongate slender scapular blade with a gradually dorsoventrally expanded distal end, a total of nine dorsal vertebrae, presence of the posterior centroparapophyseal lamina in the posterior dorsal vertebrae). Although a few diplodocid taxa, Diplodocus, cf. Apatosaurus, and cf. Barosaurus, are known from several fossil sites near the Howe Ranch, identification of this specimen, even at a generic level, is difficult due to a large degree of ontogenetic variation. 相似文献

Some experiments were carried out with purified neutral proteinases I and II of Aspergillus sojae in relation to their characteristics as metalloenzyme.The both enzymes contained one gram atom of zinc and about two gram atoms of calcium per mole (molecular weights of 41,700 for I and 19,800 for II were estimated by gel filtration) of enzyme protein, and the zinc was essential for the activity. Some metal-chelating agents, such as ethylenediaminetetraacetic acid (EDTA), o-phenanthroline, 8-hydroxyquinoline and α,α′-dipyridyl, inhibited the activity of the both enzymes. In the inactivation of neutral proteinase II by EDTA a distinct pH-dependency was observed. The EDTA-inactivated enzymes were reactivated fully or partially by the addition of some metal ions such as Zn2+, Co2+, Mn2+, Cu2+ (only neutral proteinase II) and Ni2+. Zinc-free apo-enzymes were prepared from the native enzymes by the dialysis against EDTA solution. The apo-enzyme of neutral proteinase I still contained calcium, while that of neutral proteinase II did not. The apo-enzymes restored their activity for the most part either by the addition of excess amount of zinc or by mixing with a stoichiometric amount of zinc in the presence of calcium at an alkaline condition. 相似文献

Establishing CYFRA 21-1 detection for noninvasive differential diagnosis of urothelial carcinoma (UC) of bladder would help to improve assessment and follow-up of patients, as well as to improve screening of high-risk groups. The study group comprised of 147 subjects including 72 patients with UC of bladder, 75 controls and 17 follow-up cases. The levels of CYFRA 21-1 in serum, urine and urinary cell lysate were estimated by high sensitivity ELISA. Our results indicate that urinary CYFRA 21-1 provides a high value of overall sensitivity for UC of bladder and is also useful even for detection of low grade tumors that might indicate possible earlier detection and treatment administration. 相似文献

Atherosclerosis is a complicated and multifactorial disease, induced not only by genotype, but also, even more importantly, by environmental factors. Study on the metabolic perturbation of endogenous compounds may offer deeper insight into development of atherosclerosis. Gas chromatography/mass spectrometry (GC/MS)-based metabonomics was used to profile a metabolic fingerprint of serum obtained from hamsters with induced cholesterol. The deconvoluted GC/MS data were processed by multivariate statistical analysis tools, such as principal component analysis (PCA) and partial least squares projection to latent structure and discriminant analysis (PLS-DA). For the first time we showed a time-dependent development of the model animal from normal to hypercholesterolaemia, and further to early atherosclerosis. Twenty-one compounds were identified as markers involved in the development to atherosclerosis. Identification of the compounds suggests that amino acid metabolism and fatty acid oxidation are significantly perturbed following cholesterol overloading. The data provide novel information to approach the pathophysiological processes of the hypercholesterolaemia and atherosclerosis disease continuum. 相似文献

AbstractPrevention and early detection are considered to be the most effective means of managing non-native invasive species that have serious ecological and economic impacts. Consequently, the study of those taxa that are still in an early stage of invasion but are highly likely to spread and to have a significant adverse impact in future is becoming an increasingly important issue in both the research on and management of invasive species. In Italy, one such species is Arctotheca calendula (L.) Levyns, a herbaceous plant of the Asteraceae family that is native to South Africa. Here, we present an overview of current knowledge on the distribution and potential impacts of this species in Italy, and we outline some features that are crucial to understanding and preventing its invasive spread. 相似文献

Next, a brief history of music teaching and learning is followed by a discussion of Brazilian children's current musical practices. Intestinal celiac disease (CD) is triggered by peptic-tryptic digest of gluten, known as Frazer's Fraction (FF), in genetically predisposed individuals. Here, we investigate the immediate effects of FF on the actin cytoskeleton and the subsequent trafficking of actin-dependent and actin-independent proteins in COS-1 cells. Morphological alterations in the actin filaments were revealed concomitant with a drastic reduction in immunoprecipitated actin from cells incubated with FF. These alterations elicit impaired protein trafficking of intestinal sucrase–isomaltase, a glycoprotein that follows an actin-dependent vesicular transport to the cell surface. However, the actin-independent transport of intestinal lactase phlorizin hydrolase remains unaffected. Moreover, the morphological alteration in actin is induced by direct interaction of this protein with gliadin peptides carrying the QQQPFP epitope revealed by co-immunoprecipitation utilizing a monoclonal anti-gliadin antibody. Finally, stimulation of cells with FF directly influences the binding of actin to Arp2. Altogether, our data demonstrate that FF directly interacts with actin and alters the integrity of the actin cytoskeleton thus leading to an impaired trafficking of intestinal proteins that depend on an intact actin network. This direct interaction could be related to the endocytic segregation of gliadin peptides as well as the delayed endocytic vesicle trafficking and maturation in gliadin-positive intestinal epithelial cells and opens new insights into the pathogenesis of CD. 相似文献

Trafficking kinesin proteins (TRAKs) 1 and 2 are kinesin-associated proteins proposed to function in excitable tissues as adaptors in anterograde trafficking of cargoes including mitochondria. They are known to associate with N-acetylglucosamine transferase and the mitochondrial rho GTPase, Miro. We used confocal imaging, Förster resonance energy transfer and immunoprecipitations to investigate association between TRAKs1/2, N-acetylglucosamine transferase, the prototypic kinesin-1, KIF5C, and Miro. We demonstrate that in COS-7 cells, N-acetylglucosamine transferase, KIF5C and TRAKs1/2 co-distribute. Förster resonance energy transfer was observed between N-acetylglucosamine transferase and TRAKs1/2. Despite co-distributing with KIF5C and immunoprecipitations demonstrating a TRAK1/2, N-acetylglucosamine transferase and KIF5C ternary complex, no Förster resonance energy transfer was detected between N-acetylglucosamine transferase and KIF5C. KIF5C, N-acetylglucosamine transferase, TRAKs1/2 and Miro formed a quaternary complex. The presence of N-acteylglucosamine transferase partially prevented redistribution of mitochondria induced by trafficking proteins 1/2 and KIF5C. TRAK2 was a substrate for N-acetylglucosamine transferase with TRAK2 (S562) identified as a site of O-N-acetylglucosamine modification. These findings substantiate trafficking kinesin proteins as scaffolds for the formation of a multi-component complex involved in anterograde trafficking of mitochondria. They further suggest that O-glycosylation may regulate complex formation. 相似文献

A study on the feeding ecology of juvenile cod Gadus morhua, haddock Melanogrammus aeglefinus and whiting Merlangius merlangus during the pelagic to demersal transition was carried out on fishes sampled throughout their settlement season at a local nursery ground in the north-western North Sea, off the Scottish east coast. A comprehensive quantitative taxonomic analysis of the diets, as described in the paper, showed the emergence of distinctive feeding niches, minimizing the potential for competition between species and size categories. The diet of the juveniles changed with fish size, water depth, time of year and distance offshore. Small G. morhua were present in the study area earlier in the season, settled further inshore and ate a higher proportion of pelagic prey (copepods) and as size increased they moved into deeper waters and targeted larger, more benthic prey. As M. aeglefinus grew larger and moved into deeper waters, a diet of largely copepods, amphipods, pelagic Ammodytes spp., cyprids and pelagic gastropods evolved to one dominated predominantly by fishes and benthic invertebrates. In the case of M. merlangus, widespread ages and sizes throughout the sampling season, a consequence of their more protracted spawning season, resulted in dietary changes which were more likely to be influenced by seasonal changes in the prey field, in addition to developmental (size) changes, than the diets of the other two species. 相似文献
