Growth autocorrelation and animal size variation   总被引:2,自引:1,他引:1  
It has long been recognized that variability in animal size is affected by how individual growth rate is autocorrelated in time. Earlier studies have attributed the mechanism generating the autocorrelation primarily to size‐dependent growth rate and autocorrelation in resource abundance. All of these studies have shown that positive autocorrelation in individual growth rate always translates into increased variability in size. We show that energy reserves in individuals induce growth autocorrelation by acting like a low pass filter between the resource and the internal energy that is available for metabolism, growth and reproduction. However, the reserve also reduces the variance in growth rate. Consequently, reserve‐induced growth autocorrelation has relatively little effect on size variability in the population, contradicting existing ideas about the relationship between the growth autocorrelation and size variability.  相似文献

Dialysis is a well-known technique for laboratory separation. However, its efficiency is commonly restricted by the dialyzer volume and its passive diffusion manner. In addition, the sample is likely to be precipitated and inactive during a long dialysis process. To overcome these drawbacks, a dynamic dialysis method was described and evaluated. The dynamic dialysis was performed by two peristaltic pumps working in reverse directions, in order to drive countercurrent parallel flow of sample and buffer, respectively. The efficiency and capacity of this dynamic dialysis method was evaluated by recording and statistically comparing the variation of conductance from retentate under different conditions. The dynamic method was proven to be effective in dialyzing a large-volume sample, and its efficiency changes proportionally to the flow rate of sample. To sum up, circulating the sample and the buffer creates the highest possible concentration gradient to significantly improve dialysis capacity and shorten dialysis time.  相似文献

Ganglion cells are the output neurons of the retina and their activity reflects the integration of multiple synaptic inputs arising from specific neural circuits. Patch clamp techniques, in voltage clamp and current clamp configurations, are commonly used to study the physiological properties of neurons and to characterize their synaptic inputs. Although the application of these techniques is highly informative, they pose various limitations. For example, it is difficult to quantify how the precise interactions of excitatory and inhibitory inputs determine response output. To address this issue, we used a modified current clamp technique, dynamic clamp, also called conductance clamp 1, 2, 3 and examined the impact of excitatory and inhibitory synaptic inputs on neuronal excitability. This technique requires the injection of current into the cell and is dependent on the real-time feedback of its membrane potential at that time. The injected current is calculated from predetermined excitatory and inhibitory synaptic conductances, their reversal potentials and the cell''s instantaneous membrane potential. Details on the experimental procedures, patch clamping cells to achieve a whole-cell configuration and employment of the dynamic clamp technique are illustrated in this video article. Here, we show the responses of mouse retinal ganglion cells to various conductance waveforms obtained from physiological experiments in control conditions or in the presence of drugs. Furthermore, we show the use of artificial excitatory and inhibitory conductances generated using alpha functions to investigate the responses of the cells.  相似文献

Historic land‐cover/use change is important for studies on climate change, soil carbon, and biodiversity assessments. Available reconstructions focus on the net area difference between two time steps (net changes) instead of accounting for all area gains and losses (gross changes). This leads to a serious underestimation of land‐cover/use dynamics with impacts on the biogeochemical and environmental assessments based on these reconstructions. In this study, we quantified to what extent land‐cover/use reconstructions underestimate land‐cover/use changes in Europe for the 1900–2010 period by accounting for net changes only. We empirically analyzed available historic land‐change data, quantified their uncertainty, corrected for spatial‐temporal effects and identified underlying processes causing differences between gross and net changes. Gross changes varied for different land classes (largest for forest and grassland) and led to two to four times the amount of net changes. We applied the empirical results of gross change quantities in a spatially explicit reconstruction of historic land change to reconstruct gross changes for the EU27 plus Switzerland at 1 km spatial resolution between 1950 and 2010. In addition, the reconstruction was extended back to 1900 to explore the effects of accounting for gross changes on longer time scales. We created a land‐change reconstruction that only accounted for net changes for comparison. Our two model outputs were compared with five commonly used global reconstructions for the same period and area. In our reconstruction, gross changes led in total to a 56% area change (ca. 0.5% yr?1) between 1900 and 2010 and cover twice the area of net changes. All global reconstructions used for comparison estimated fewer changes than our gross change reconstruction. Main land‐change processes were cropland/grassland dynamics and afforestation, and also deforestation and urbanization.  相似文献

Kinetic information during human gait can be estimated with inverse dynamics, which is based on anthropometric, kinematic, and ground reaction data. While collecting ground reaction data with a force plate is useful, it is costly and requires regulated space. The goal of this study was to propose a new, accurate methodology for predicting ground reaction forces (GRFs) during level walking without the help of a force plate. To predict GRFs without a force plate, the traditional method of Newtonian mechanics was used for the single support phase. In addition, an artificial neural network (ANN) model was applied for the double support phase to solve statically indeterminate structure problems. The input variables of the ANN model, which were selected to have both dependency and independency, were limited to the trajectory, velocity, and acceleration of the whole segment's mass centre to minimise errors. The predicted GRFs were validated with actual GRFs through a ten-fold cross-validation method, and the correlation coefficients (R) for the ground forces were 0.918 in the medial–lateral axis, 0.985 in the anterior–posterior axis, and 0.991 in the vertical axis during gait. The ground moments were 0.987 in the sagittal plane, 0.841 in the frontal plane, and 0.868 in the transverse plane during gait. The high correlation coefficients(R) are due to the improvement of the prediction rate in the double support phase. This study also proved the possibility of calculating joint forces and moments based on the GRFs predicted with the proposed new hybrid method. Data generated with the proposed method may thus be used instead of raw GRF data in gait analysis and in calculating joint dynamic data using inverse dynamics.  相似文献

In this work the effect of betaine on the structure and aggregation of the GST-GFP fluorescent fusion protein was studied by different complementary techniques, including electron microscopy, dynamic light scattering, circular dichroism, and FTIR spectroscopy. Although osmolytes are known to be protein stabilizers in vivo, the effect of betaine on the structure and aggregation of our model protein was found to be strictly concentration dependent. We demonstrated that, by changing betaine concentration, it was possible to tune the formation of protein soluble assemblies and insoluble aggregates, as well as to disaggregate preformed aggregates. In particular, at a critical concentration of betaine between 5 and 7.5 mM, the protein precipitated into macroscopic prefibrillar structures, rich in intermolecular beta-sheets, which were found to bind thioflavine T and to be inaccessible to protease. Instead, at higher betaine concentration (10-20 mM) the misfolded protein lost its fluorescence, but formed soluble assemblies with hydrodynamic radius of about 16 nm. These structures displayed a reduced propensity to further aggregate under thermal treatment. In addition, betaine at this high concentration was also found to disrupt large preformed aggregates, obtained under different conditions, into protein soluble assemblies. It is the first time that a disaggregation process has been described for a chemical chaperone. A mechanism for the betaine concentration-dependent effect on protein misfolding, aggregation, and disaggregation is proposed and its possible physiological implications are discussed.  相似文献

The effects of a recombinant small heat shock protein with an apparent molecular weight of 27 kDa (Hsp27) and both wild type (Hsp27-wt) and S15D, S78D, S82D mutants (Hsp27-3D), which mimic the naturally occurring phosphorylation of this protein, on the thermal denaturation and aggregation of F-actin were studied. It has been shown that at a Hsp27/actin weight ration of 1/4, both Hsp27-wt and Hsp27-3D do not affect the thermal denaturation of F-actin, but efficiently prevent its further aggregation by forming soluble complexes with denatured actin. Formation of these complexes occurs upon heating and accompanies the F-actin thermal denaturation. Hsp27-wt is known to exist as a high-molecular-weight oligomer, whereas Hsp27-3D forms only small dimers or tetramers. However, the complexes formed by Hsp27-wt and Hsp27-3D with denatured actin did not differ in their size, as measured by dynamic light scattering, and demonstrated the same hydrodynamic radius of 17–18 nm. On the other hand, the sedimentation coefficients of these complexes differed: they ranged 10–45 S in the case of Hsp27-3D and 18–60 S in case of Hsp27-wt. Thus, the initial oligomeric state of Hsp27 does not significantly affect its capacity to form small soluble complexes with denatured actin.  相似文献

采用HPLC法测定不同采收期雷公藤根、茎、叶的主要活性成分雷公藤甲素含量,掌握雷公藤甲素的动态积累。结果表明,雷公藤甲素浓度在7.75~250 μg·mL-1范围内线性关系良好(R2=0.9997),雷公藤加样回收率为99.21%,RSD值为1.578%,该方法回收率较好。雷公藤甲素的含量在不同生长期、不同部位存在一定差异,不同部位甲素含量由高到低依次为叶、根、茎,雷公藤叶片具有资源开发利用潜力。7~8月采收的雷公藤叶片甲素含量最高,11~12月雷公藤根部甲素含量达最高。雷公藤不同器官均具有较高的药用价值,研究结果为雷公藤人工采收、质量评价和资源开发提供一定的科学依据。  相似文献

The spatial-temporal variation of understory light availability has important influences on species diversity and community assembly. However, the distribution characteristics and influencing factors of understory light availability have not been fully elucidated, especially in temperate deciduous, broad-leaved forests. In this study, the understory light availability was monitored monthly (May–October) in a temperate deciduous, broad-leaved forest in Henan Province, China. Differences in the light availability among different months and habitat types were statistically analyzed using Kruskal–Wallis method, respectively. Partial least squares path modeling (PLS-PM) was used to explore the direct and/or indirect effects of stand structure, dominant species and topographic factors on the light environment. Results showed that there were differences in light environments among the four habitat types and during the studied six months. The PLS-PM results showed that the stand structure and the dominant species were negatively correlated with the light environment, and the path coefficient values were −0.089 (P = 0.042) and −0.130 (P = 0.004), respectively. Our result indicated that the understory light availability exhibit a distinct spatial and temporal heterogeneity in temperate deciduous, broad-leaved forest of north China. The characteristics of woody plant community, especially the abundance of one of the dominant plant species, were the important factors affecting the understory light availability.  相似文献

The alarming increase in multi- and extensively drug-resistant (MDR and XDR) strains of Mycobacterium tuberculosis (MTB) has triggered the scientific community to search for novel, effective, and safer therapeutics. To this end, a series of 3,5-disubstituted-1,2,4-oxadiazole derivatives (3a–3i) were tested against H37Rv, MDR and XDR strains of MTB. Of which, compound 3a with para-trifluorophenyl substituted oxadiazole showed excellent activity against the susceptible H37Rv and MDR-MTB strain with a MIC values of 8 and 16 µg/ml, respectively.To understand the mechanism of action of these compounds (3a–3i) and identify their putative drug target, molecular docking and dynamics studies were employed against a panel of 20 mycobacterial enzymes reported to be essential for mycobacterial growth and survival. These computational studies revealed polyketide synthase (Pks13) enzyme as the putative target. Moreover, in silico ADMET predictions showed satisfactory properties for these compounds, collectively, making them, particularly compound 3a, promising leads worthy of further optimisation.  相似文献
