以浙江省的水稻黑条矮缩病和河北省的玉米粗缩病的病毒分离物为材料,对我国南北方两种病毒分离物进行了比较研究。两种病毒分离物的粒子大小形态相似,且在血清学上密切相关;二者的介体、寄主相同,并引起相同的症状,提纯两种病毒分离物的基因组片段凝胶电泳显示它们相应的基因组片段之间大小极相似。根据水稻黑条矮缩病毒(Rice black strecaked dwarf virus),RBSDV的S7和S8设计引物,利用PR-PCR技术,在两种病毒分离物中均可特异扩增到预期大小的片段,序列测定比较分析表明:它们的同源性达97.0%-98.9%,与日本RBSDV的同源性(92.2%-95.5%)高于与意大利MRDV的同源性(76.6%-88.4%)。从而认为我国南方的水稻黑条矮缩病和北方和玉米粗缩病都是同一种病毒-RBSDV引起的,也就是说我国北方的玉米粗缩病病原实际上是水稻黑条矮缩病毒,而不玉米粗缩病毒。  相似文献

Further work on the subcellular localization of two lipid-degrading enzymes, lipolytic acyl hydrolase (LAH) and lipoxygenase (LOX) has been carried out on brassica florets, potato shoots and pea roots. In all cases, the LAH profile on sucrose and Ficoll density gradients was coincident with ‘lysosomal’ acid phosphatase activity. However, the localization of LOX activity was different for each tissue. In pea roots the activity of LOX was localized in the ‘lysosomal’ fraction, whereas with brassica florets (cauliflower and calabrese) it was present in a heavy body with a similar density to plastids and in potato shoots LOX gave only low particulate recoveries.  相似文献

Summary A field experiment on N turnover in rotations of groundnut, sorghum and pigeonpea crops was conducted in an Indian Alfisol during 1978–80.15N-labelled urea N was applied @ 40 kg ha–1 in 1978. In the first year, the groundnut utilized 19.5% of the applied labelled N, sorghum 25.5%, and pigeon pea 10.3%. More fertilizer N was removed through weeding than by crop uptake in sorghum and pigeon pea. The fertilizer N left behind in soil upto 40 cm depth was 44.4% in groundnut plots, 35.1% in sorghum plots and 40.1% in pigeon pea plots.The uptake in 1979 of the residual fertilizer N in the soil was 8.9% in sorghum, 8.3% in groundnut and 2.8% in pigeon pea. In 1980, it declined to less than 2% for pigeon pea and groundnut and to about 4% for sorghum.A balance sheet drawn at the end of each rotation showed that about 51.3% of the applied labelled N was accounted for in groundnut-sorghum-pigeon pea rotation, 70.9% in sorghumpigeon pea-groundnut, and 43.5% in pigeon pea-groundnut-sorghum.  相似文献

Abstract A new experimental system was devised for the simultaneous measurement of elongation rate and the activity of the spatially separate electrogenic ion pumps of a hypocotyl segment excised from a seedling of Vigna unguiculata L. Walp. under enforced intra-organ perfusion by artificial solutions. The pathway of the perfusion medium was apoplastic space, including xylem vessels as main routes. The elongation rate of the segment was highly dependent on the perfusion pressure applied. It was possible to increase the growth rate under pressurized perfusion by 10-30 times as much as that without perfusion. Elongation rate was also dependent on respiration under perfusion, being retarded reversibly by anoxia a few minutes after the activities of the electrogenic ion pumps were stopped. Perfusion pressure had a little influence on the membrane potential (Vpx) below a breakdown level (c. 130 kPa). Perfusion of mannitol or sorbitol solution of appropriate concentration reduced the elongation rate reversibly.  相似文献

采用聚合酶链式反应技术,扩增了水稻矮缩病毒(RDV)基因组第10号片段的编码序列,该片段编码病毒的非结构蛋白。对扩增产物进行了克隆和限制性内切酶分析,并绘制了物理图谱。克隆片段大小为1150 bp,含Sac I、Hind III、Nde I、BamH I、Sai I 等酶切位点,引物设计时还在该片段两侧增加了Bgl II和 EcoR I 切点,以便克隆到植物中间载体质粒。利用上述酶切位点对该片段进行了亚克隆和序列分析,结果表明,本研究克隆的RDV中国流行林基因组第10号片段的编码区与日本流行株的相应区域比较,核酸的同源率为96.03%,编码的氨基酸的同源率为97.17%。  相似文献

Abscisic acid (ABA) plays a multifaceted role in plant immunity and can either increase resistance or increase susceptibility to some bacterial and fungal pathogens depending on the pathosystem. ABA is also known to mediate plant defence to some viruses. In this study, the relationship between the ABA pathway and rice black‐streaked dwarf virus (RBSDV) was investigated in rice. The expression of ABA pathway genes was significantly reduced upon RBSDV infection. Application of exogenous hormones and various ABA pathway mutants revealed that the ABA pathway plays a negative role in rice defence against RBSDV. Exogenous hormone treatment and virus inoculation showed that ABA inhibits the jasmonate‐mediated resistance to RBSDV. ABA treatment also suppressed accumulation of reactive oxygen species by inducing the expression of superoxidase dismutases and catalases. Thus, ABA modulates the rice–RBSDV interaction by suppressing the jasmonate pathway and regulating reactive oxygen species levels. This is the first example of ABA increasing susceptibility to a plant virus.  相似文献

Field experiments were conducted in 2004 and 2005 to determine the effects of seed treatment with Rhizobium leguminosarum bv. viceae on damping‐off, seedling height, root nodule mass, root biomass, shoot biomass and seed yield of pea and lentil in a field naturally infested with Pythium spp. Compared with the untreated controls, treatment of pea seeds with R. leguminosarum bv. viceae strains R12, R20 or R21 significantly (P < 0.05) reduced incidence of damping‐off, promoted seedling growth and increased root nodule mass, root biomass and shoot biomass. Seed treatments with R12 or R21 also resulted in a significant (P < 0.05) increase in seed yield of pea. The strain R21 was most effective among the four strains of R. leguminosarum bv. viceae tested in peas. Although, the level of disease control by strain R21 was similar to seed treatment with the fungicide ThiramTM, R21 was more effective in enhancing root nodule production and promoting plant growth. For lentil, treatment of seeds with R. leguminosarum bv. viceae strains R12 or R21 significantly (P < 0.05) reduced incidence of damping‐off compared with the untreated control. All of the four strains of R. leguminosarum bv. viceae tested increased lentil seedling height, root nodule mass and shoot biomass, and all except R20 increased root biomass. Seed yield was higher for the treatments of R12 and R21. The strain R12 was most effective among the four strains of R. leguminosarum bv. viceae tested in lentil. Although, strain R12 was as effective as ThiramTM for control of damping‐off of lentil, it was more effective than ThiramTM for the production of root nodules and promotion of plant growth. The study concludes that seed treatment with R. leguminosarum bv. viceae is effective in control of Pythium damping‐off of pea and lentil and that the efficacy of control is strain specific, strain R21 for control of the disease on pea and strain R12 for control of the disease on lentil.  相似文献

The respiratory uptake or photosynthetic evolution of oxygen by mesophyll protoplasts of pea ( Pisum sativum L. cv. Arkel) were monitored during successive short. (3–5 min) cycles of darkness and illumination. The rate of respiration was nearly doubled after 3–4 short periods of illumination while there was a 15–20% enhancement in photosynthesis with cycles of illumination and darkness preceding illumination. Such interaction between photosynthesis and respiration was statistically significant when bicarbonate was present in the reaction medium. The inhibitors of photosynthesis [3(3,4–dichlorophenyl)-l,l-dimethylurea (DCMU), glyceraldehyde] decreased respiration after periods of illumination, whereas inhibitors of respiratory electron transport (Rotenone, antimycin A, NaN3) suppressed photosynthesis, as well. We suggest that a rapid beneficial interaction exists between photosynthesis and respiration in protoplasts, even during short cycles of light and darkness.  相似文献

The gibberellin (GA) economy of young pea (Pisum sativum L.) fruits was investigated using a range of mutants with altered GA biosynthesis or deactivation. The synthesis mutation lh-2 substantially reduced the content of both GA4 and GA1 in young seeds. Among the other synthesis mutations, ls-1, le-1 and le-3, the largest reduction in seed GA1 content was only 1.7-fold (le-1), while GA4 was not reduced in these mutants, and in fact accumulated in some experiments (compared with the wild type). Mutation sln appeared to block the step GA20 to GA29 in young pods and seeds, but not as strongly as in older seeds. Mutations ls-1, le-1 and le-3 markedly reduced pod GA1 levels, but pod elongation was not affected. After feeds of [13C,3H]GA20 to leaves, the pods contained 13C,3H-labelled GA20, GA1, GA29 and GA81, and the seeds, [13C,3H]GA20 and [13C,3H]GA29.
