Bacillus thuringiensis (Bt) bacteria produce Cry toxins that are able to kill insect pests. Different models explaining the mode of action of these toxins have been proposed. The pore formation model proposes that the toxin creates pores in the membrane of the larval midgut cells after interaction with different receptors such as cadherin, aminopeptidase N and alkaline phosphatase and that this pore formation activity is responsible for the toxicity of these proteins. The alternative model proposes that interaction with cadherin receptor triggers an intracellular cascade response involving protein G, adenylate cyclase (AC) and protein kinase A (PKA). In addition, it was shown that Cry toxins induce a defense response in the larvae involving the activation of mitogen-activated kinases such as MAPK p38 in different insect orders. Here we analyzed the mechanism of action of Cry1Ab and Cry1Ac toxins and a collection of mutants from these toxins in the insect cell line CF1 from Choristoneura fumiferana, that is naturally sensitive to these toxins. Our results show that both toxins induced permeability of K+ ions into the cells. The initial response after intoxication with Cry1Ab and Cry1Ac toxins involves the activation of a defense response that involves the phosphorylation of MAPK p38. Analysis of activation of PKA and AC activities indicated that the signal transduction involving PKA, AC and cAMP was not activated during Cry1Ab or Cry1Ac intoxication. In contrast we show that Cry1Ab and Cry1Ac activate apoptosis. These data indicate that Cry toxins can induce an apoptotic death response not related with AC/PKA activation. Since Cry1Ab and Cry1Ac toxins affected K+ ion permeability into the cells, and that mutant toxins affected in pore formation are not toxic to CF1, we propose that pore formation activity of the toxins is responsible of triggering cell death response in CF1cells. 相似文献

水氮供应对夏棉产量、水氮利用及土壤硝态氮累积的影响 总被引:6,自引:0,他引:6
通过田间试验,研究了黄淮地区水氮供应对夏棉生长、产量及水氮利用效率的影响,探索在保证产量的同时提高水氮利用效率、减少农田水氮排放的管理模式.试验设置5个氮素水平(0、60、120、180、240 kg·hm-2,分别记为N0、N1、N2、N3、N4)和3个灌水水平(滴灌,灌水定额30、22.5、15 mm,分别记为I1、I2、I3),使用裂区设计,主区为氮用量,裂区为灌水水平,共15个处理,3次重复.结果表明: 氮素和水分施用对夏棉生长和产量都有明显促进作用,但氮素影响更显著,是该地区调控夏棉生长和籽棉产量的主要因素.随着施氮量和灌水量的增加,花铃期生殖器官积累量、地上部干物质积累量和籽棉产量在开始阶段都逐步增加,当施氮量超过180 kg·hm-2时,进一步增施氮肥会导致生殖器官积累量、地上部干物质积累量和籽棉产量减小.籽棉产量在N3I1处理达到最大,为4016 kg·hm-2.增加施氮量能显著提高地上部总吸氮量和茎叶含氮量,但会降低氮肥偏生产力.灌溉水利用效率和田间水分利用效率分别在N3I3和N3I1处理最大,分别为5.40和1.24 kg·m-3.随着施氮量的增加,土壤硝态氮含量明显增加,且硝态氮累积区域有下移趋势.综合考虑对地上部干物质积累、产量、水氮吸收利用及土壤硝态氮累积等的影响,N3I1处理可作为试验区夏季棉花生产的最优水氮管理方案. 相似文献

大气CO2浓度升高影响植物光合作用过程和生物量积累,改变植物地上和地下生物量的动态分配.土壤有机质的形成和周转依赖于植物组分的输入,因此,CO2浓度升高所造成的植物生理和代谢的变化对土壤碳库收支平衡具有重要影响.采用稳定碳同位素(13C)技术研究土壤-植物系统的碳循环可阐明大气CO2浓度升高条件下光合碳在植物各器官的分配特征和时间动态,明确光合碳在土壤中的积累、分解与迁移转化过程以及对土壤有机碳库周转的影响.本文综述了基于13C自然丰度法或13C示踪技术研究大气CO2浓度升高对土壤-植物系统碳循环的影响,主要包括:1)对植物光合作用的同位素分馏的影响;2)对植物光合碳(新碳)分配动态的影响;3)对土壤有机碳新老碳库动态以及微生物转化过程的影响.明确上述过程及其调控机制可为预测CO2浓度升高对陆地生态系统碳循环及源汇效应的长期影响奠定基础. 相似文献

为了分析保水剂与有机肥配合施加对铁尾矿理化性质的影响,确定适宜尾矿改良的保水剂与有机肥最优配比,在迁安首钢所辖的铁尾矿区内,布设保水剂与有机肥2因素4水平的野外小区试验,并测定试验小区铁尾矿容重、孔隙度及持水量和pH、有机质、氮、磷、钾含量等理化性质,分析有机肥和保水剂对尾矿改良的作用;同时在小区内播种紫苜蓿和紫穗槐两种植物,测其生物量,验证改良效果.保水剂水平为(L·m-3):0(B0)、10(B1)、50(B2)、100(B3);有机肥水平为(kg·m-2):0(N0)、2.25(N1)、11.24(N2)、22.49(N3).结果表明:不同配比处理对尾矿理化性质的改良作用主要体现在0~20 cm表层尾矿,各指标均在0~20 cm层与对照(CK)差异显著.有机肥对尾矿理化性质的改良作用优于保水剂.在0~20 cm层,保水剂单因素的所有处理尾矿容重、非毛管孔隙度、有机质、速效磷和速效钾含量均与CK无显著差异,而单独施加有机肥B0N2、B0N3处理的上述指标均与CK差异显著.保水剂与有机肥二者配合施加优于保水剂或有机肥单独添加的改良效果.0~20 cm层各项指标均在配施处理B3N3下达到最佳,且均与CK达到显著差异.综合考虑,B3N3可以作为尾矿改良的保水剂与有机肥最优配比. 相似文献

More than 90% of US Navy Ships are coated with copper ablative paint. These ships may spend long periods of time pier-side, which makes them vulnerable to fouling. Hull grooming has been proposed as a means of maintaining the coatings in an operational condition. This study investigated the effect of grooming on a copper ablative coating exposed statically for six years. Grooming was performed weekly or monthly with controls left ungroomed. The fouling community was visually assessed, dry film thickness measurements were taken to monitor coating loss, and the copper leaching rates were measured. It was found that weekly and monthly groomed surfaces reduced fouling, and the ungroomed surfaces became fully fouled. Coating loss was similar for weekly, monthly and ungroomed surfaces. The results suggest that grooming is a viable method for maintaining copper ablative coatings in a fouling-free condition without adverse increases in the total copper output. 相似文献

In this study, the fractionation and distribution of phosphorus (P) in the core sediments of the Shanmei reservoir were investigated by using the chemical extraction method in directions for the first time in order to understand its bio-availability, adsorption characteristics, potential release and environmental significance. The results of the study showed that P in the sediments mainly consisted of inorganic phosphorus (IP) and that IP mainly consisted of non-apatite phosphorus (NAIP). The horizontal and temporal distributions of the P fractions were different from each other, but the vertical distribution was similar, which indicated a trend of stabilization after falling. The content of total phosphorus (TP), IP, organic phosphorus (OP), NAIP, apatite phosphorus (AP), and bio-available phosphorus (BAP) in the sediments during the three seasons ranged from 193.85 to 1664.05 mg·kg?1, 126.90 to 1127.70 mg·kg?1, 43.74 to 669.29 mg·kg?1, 57.62 to 937.07 mg·kg?1, 32.58 to 250.71 mg·kg?1, and 41.06 to 871.82 mg·kg?1, respectively. NAIP contents in the sediments accounted for more than 50% of TP. Using an analysis from three aspects, the eutrophication risk index (ERI) could be used to assess the potential release of P in the sediments, and there was a high release risk of P in the sediments in the Shanmei reservoir. 相似文献

The paucity of late Paleogene and Neogene floras from Antarctica limits our ability to understand the interplay between Antarctic climate evolution and the impact that glaciation had on the vegetation, in particular, how the vegetation changed from temperate Eocene forests, to today’s sparse vegetation. Fluvial and lacustrine strata deposited in a wet-based glacial sequence (Friis Hills, McMurdo Dry Valley sector, Transantarctic Mountains) have yielded abundant megaspores. These strata are early Miocene based on correlation with a volcanic ash dated at 19.76 ± 0.11 Ma. The megaspores are up to 736 µm in diameter with well-developed wing-like laesurae and equatorial zona. The morphology is consistent with extant Isoetes, and demonstrates the presence of Lycopsida and the Isoetaceae within Antarctic Miocene floras. Today, Isoetes is widespread from the Tropics to the Arctic such as Greenland (I. echinospora, I. lacustris) and from marginal marine (I. ekmanii) to high altitudinal environments (I. lechleri), though commonly associated with lacustrine or aquatic environments. The fossil spores occur in fluvial and lacustrine beds, suggesting the parent plants were aquatics. The occurrence together with mosses and Nothofagus leaves points to persistent vegetation in the early Miocene of Antarctica. 相似文献

The adaptive significance of avian egg shape in birds is poorly understood. The pyriform (pear‐like) shape of the Common Guillemot's Uria aalge egg has long been considered to be an adaptation to prevent eggs rolling off the bare cliff ledges on which this species breeds. Rolling was thought to be prevented either by the egg spinning like a top, which is not the case, or by rolling in an arc, which it does but with little influence on whether the egg will fall from a ledge. We therefore sought alternative explanations for the pyriform shape of the Common Guillemot's egg. This species breeds in extremely dense colonies, which makes their eggs vulnerable to mechanical damage from conspecifics, and to contamination by debris such as faeces and soil. We present evidence consistent with both these possible explanations. First, the pyriform shape of Common Guillemot eggs means that a higher proportion of the eggshell lies in contact with the substrate and this may minimize the effect of impacts. Resistance to impacts may be further enhanced because their eggshells are especially thick where they are in contact with the substrate. Secondly, Common Guillemot eggs are often heavily contaminated with faecal material and other debris during incubation. Most contamination is on the pointed end of the egg where it is in contact with the substrate; the pyriform shape thus keeps the blunt end of the egg, which has the highest porosity, relatively free of contamination, which in turn may facilitate both gas exchange during incubation and the hatching process, because the chick emerges from the blunt end of the egg. 相似文献

Slugs feed on some plant species but not on others for reasons that are not yet fully understood. We re‐examined the two largest studies on the preference of Arion lusitanicus for different plant species. Briner & Frank (1998) measured the consumption index of slugs (CIB) based on the dry leaf mass eaten per day per gram of slug mass. Kozlowski & Kozlowska (2009) also measured a consumption index of slugs (CIK) but on the basis of the area eaten per day per gram of slug mass. The two indices showed a moderate positive rank correlation. Briner & Frank (1998) found that slugs preferred annual to perennial plants and crops rather than wild species. On the other hand, Kozlowski & Kozlowska (2009) found no statistically significant difference in slug preference for annuals, biennials or perennials. Using botanical databases we selected plant traits related to slug herbivory and analysed 141 species. We first analysed the two data sets separately and found that CIB was higher for seedlings with a higher relative growth rate although this result was only marginally statistically significant. In addition, we found that annuals were consumed more than perennials and that cultivated plants were consumed more than wild plants. This is in agreement with the conclusions of Briner & Frank (1998). For CIK slugs consumed less from plants with high dry matter content (DMC). The use of classification and regression tree models solves the problem of analysing datasets with missing values. Regarding CIB, the tree models indicated that life cycle was the major branch. Within perennials, cultivated/wild origin of plants was the most important predictor of slug feeding. Within wild perennials slugs ate more from plants with low specific leaf area (SLA), i.e. relatively thick leaves. Within annuals we found the unexpected result that slugs ate more from plant species with either alkaloids or glucosinolates than from species without one of these compounds. For CIK, the results of tree models showed that slugs preferred shade‐intolerant plants to shade‐tolerant plant species. Within the shade‐tolerant group slugs fed more on leaves with low DMC. We only partially understand which factors make plants acceptable to slugs. We discuss this issue along with the pros and cons of the two methods used to calculate CI. 相似文献
