烟粉虱B生物型的若虫、皮蜕及其成虫的提取物作为一种利它素信息源,在室内对其在双斑蚜小蜂寻找寄主取食、寄生行为的影响进行了生物测定。烟粉虱的若虫、皮蜕及其成虫分别用正己烷、乙醇和无菌水进行粗提。研究结果发现,双斑蚜小蜂在处理区寻找寄主停留的时间高于对照区。在处理区,双斑蚜小蜂行动活泼,对利它素源表现出高的正趋向性和选择性。对于同一利它素源、同一提取介质的两种不同浓度,双斑蚜小蜂在若虫 水提取物的高浓度区停留的时间(111.23s)最长,与在低浓度区的停留时间差异显著;而在烟粉虱皮蜕及其成虫的水、正己烷和乙醇提取物处理区,不同浓度的提取物对蚜小蜂停留的时间影响差异不显著。本研究的结果表明,利它素可以增加蚜小蜂寻找寄主的效率,有利于蚜小蜂寻找到适宜的寄主。 相似文献

1. The distribution and location of the ciliated protozoan Lagenophrys aselli on the freshwater isopod Asellus aquaticus were studied in relation to water flow over the third pleopod surface. 2. At low densities L. aselli had a significant preference for the anterior centre of the pleopod; however, at the highest densities this preference was no longer significant. The distribution ranged from closer than random at low densities to further than random at the highest densities, and may have been a product of feeding and reproduction of L. aselli as well as the short intermoult period of the host. 3. An individual L. aselli has an effective area of both the lorica and the ciliated feeding disc. By application of immunocytochemical techniques at the electron microscope level, glucoamylase was localized to the cell periphery in Clostridium thermosaccharolyticum during and following growth on starch, sucrose or glucose. Levels of immunolabelling were found to be relatively independent of growth substrate and of phase of growth, whereas previous studies had demonstrated strong dependence of glucoamylase activity on growth conditions; previously high levels of glucoamylase activity had been detected after growth on starch (i.e. during the stationary phase after growth) and only very low activities detected during exponential growth and following growth on glucose. The results presented demonstrate that levels of the glucoamylase protein are independent of measurable enzyme activity, and imply that the protein is constitutive. This indicates that the protein can exist in active and inactive states in the cell. By analogy with similar systems, we consider it likely that maturation or activation of newly synthesized glucoamylase occurs during (or following) transport through the cytoplasmic membrane. Electron microscopy of individual protein molecules which had been subjected to negative staining revealed that the enzyme consists of two domains of approximately equal size which are linked by a hinge region. 相似文献

Field test of an automated radio‐telemetry system: tracking local space use of aerial insectivores 下载免费PDF全文
Documenting local space use of birds that move rapidly, but are too small to carry GPS tags, such as swallows and swifts, can be challenging. For these species, tracking methods such as manual radio‐telemetry and visual observation are either inadequate or labor‐ and time‐intensive. Another option is use of an automated telemetry system, but equipment for such systems can be costly when many receivers are used. Our objective, therefore, was to determine if an automated radio‐telemetry system, consisting of just two receivers, could provide an alternative to manual tracking for gathering data on local space use of six individuals of three species of aerial insectivores, including one Cliff Swallow (Petrochelidon pyrrhonota), one Eastern Phoebe (Sayornis phoebe), and four Barn Swallows (Hirundo rustica). We established automated radio‐telemetry systems at three sites near the city of Peterborough in eastern Ontario, Canada, from May to August 2015. We evaluated the location error of our two‐receiver system using data from moving and stationary test transmitters at known locations, and used telemetry data from the aerial insectivores as a test of the system's ability to track rapidly moving birds under field conditions. Median location error was ~250 m for automated telemetry test locations after filtering. More than 90% of estimated locations had large location errors and were removed from analysis, including all locations > 1 km from receiver stations. Our automated telemetry receivers recorded 17,634 detections of the six radio‐tagged birds. However, filtering removed an average of 89% of bird location estimates, leaving only the Cliff Swallow with enough locations for analysis of space use. Our results demonstrate that a minimal automated radio‐telemetry system can be used to assess local space use by small, highly mobile birds, but the resolution of the data collected using only two receiver stations was coarse and had a limited range. To improve both location accuracy and increase the percentage of usable location estimates collected, we suggest that, in future studies, investigators use receivers that simultaneously record signals detected by all antennas, and use of a minimum of three receiver stations with more antennas at each station. 相似文献

通过对大麦小孢子进行基因枪轰击获得4株转绿色荧光蛋白基因(gfp)的植株(A、C、D、E),以gfp基因为探针进行荧光原位杂交(FISH)研究转化植株中转基因插入位置和基因表达。4个株系在染色体7L(5HL)的不同位置都有一个插入点,而E株系在染色体5S(7HS)还有第2个插入点。所有的转基因T0代植株都是半合子并在T1、T2代发生分离。D株系GFP未表达,但FISH和PCR分析表明gfp基因已成功插入其染色体。各株系在根尖和花粉中的GFP表达水平不同:C株系在花粉表达强而在根尖表达中等;A株系在花粉中等表达而在根尖表达较淡;E株系则在根尖高表达,花粉中等表达。A和C株系在根尖和花粉的GFP分离都表现单位点特性,而E株系的根尖分离表现重叠作用(15:1)特征,但在花粉中表达GFP的频率低。PCR结果和3个分离株系的根尖表达结果一致。D和E株系的GFP表达不正常可能和加基因插入位置或基因的结构有关。 相似文献

Recent investigations concentrate on the correlation between the myocardial expression of the inducible 70-kDa heat shock protein (HSP70i) by different stress conditions and its possible protective effects. Only few studies have focused on the involvement of small heat shock proteins in this process. We analyzed the location of the small heat shock protein HSP25 in isolated cardiomyocytes as well as its location and induction in isolated perfused hearts of rats. By immunofluorescence microscopy HSP25 was found to colocalize with actin in the I-band of myofibrils in cardiomyocytes of isolated perfused hearts as well as in isolated neonatal and adult cardiomyocytes. Hyperthermic perfusion of isolated hearts for 45 min resulted in modulation of different parameters of heart function and in induction of HSP25 and HSP70i. Temperatures higher than 43°C (44–46°C) were lethal with respect to the contractile function of the hearts. Compared to control hearts perfused at 37°C, significant increases during hyperthermic perfusion at 42°C and 43°C were obtained for heart rate, contraction velocity and relaxation velocity. In response to hyperthermia at 43°C and after subsequent normothermic perfusion for 135 min at 37°C, left ventricular pressure, contraction velocity and relaxation velocity remained significantly elevated. However, heart rate returned to control values immediately after the period of heat treatment. HSP25 is constitutively expressed even in normothermic perfused hearts as shown by Western blotting. Hyperthermia increased the content of HSP25 only in the left ventricular tissue. In contrast, HSP70i was strongly induced in all analyzed parts of the myocardium (left ventricle, right ventricle, septum). Our findings suggest a differential regulation of HSP25 and HSP70i expression in response to hyperthermia in isolated perfused hearts. The constitutively expressed HSP25 seems to be located adjacent to the myofibrils which implies a specific role of this protein even under unstressed conditions for the contractile function of the myocardium. 相似文献

Chd64是含有钙结合蛋白同源(Calponin homology,CH)结构域的蛋白,在果蝇蜕皮激素与保幼激素信号通路中发挥重要作用。本研究以家蚕Bombyx mori为研究对象,利用PCR克隆了与果蝇Drosophila melanogaster DmChd64同源的BmChd64基因,与表达载体pET-28a连接后,成功获得体外原核表达的BmChd64蛋白,并对其进行了亚细胞定位分析。家蚕BmChd64基因开放阅读框(ORF)的序列为567 bp,编码188个氨基酸,预测分子量大小为20.9 kDa,理论等电点为8.41,编码的蛋白在第27~130个氨基酸处存在CH结构域。同源性比对与进化分析显示,BmChd64与赤拟谷盗Tribolium castaneum和果蝇的Chd亲缘关系较近。qRT-PCR结果显示,BmChd64在家蚕5龄游走期的不同组织中均有表达,且在翅原基中的表达趋势与家蚕体内蜕皮激素(20-hydroxyecdysone,20E)滴度变化规律一致。亚细胞定位结果显示,BmChd64在细胞核与细胞质中均有分布,细胞核内荧光信号较强,且在细胞核外围较弱,推测细胞质中BmChd64也可入核,最终定位在细胞核中。研究结果表明BmChd64可能主要通过20E信号通路调控翅原基等的变态发育,这为进一步完善20E调控昆虫变态发育的分子机制提供了实验基础。 相似文献

ABSTRACT Global Positioning System (GPS) collars are increasingly being used to study fine-scale patterns of animal behavior. Previous studies on GPS collars have tried to determine the causes of location error without attempting to investigate whether the accuracy of fixes provides a correspondingly accurate measure of the animal's natural behavior. When comparing 2 types of GPS collar, we found a significant effect of collar weight and fit on the rate of travel of plains zebra (Equus burchelli antiquorum) females in the Makgadikgadi, Botswana. Although both types of collar were well within accepted norms of collar weight, the slightly heavier collars (0.6% of total body mass [TBM]) reduced rate of travel by >50% when foraging compared with the collar that was 0.4% of TBM. Collar effect was activity specific, particularly interfering with grazing behavior; the effect was less noticeable when zebras crossed larger interpatch distances. We highlight that small differences in collar weight or fit can affect specific behaviors, limiting the extrapolation of fine-scaled GPS data. This has important implications for wildlife biologists, who hitherto have assumed that collars within accepted weight limits have little or no effect on animal movement parameters. 相似文献
