为了明确自然种群白背飞虱Sogatella furcifera中Wolbachia和Cardinium的感染情况以及Wolbachia与其特有的WO噬菌体之间的关系, 以采自中国7个省区9个地点的白背飞虱为研究材料, 运用PCR检测的方法调查了Wolbachia, Cardinium以及WO噬菌体在各飞虱种群中的感染率和组织分布特点。结果表明: 白背飞虱广泛双重感染Wolbachia和Cardinium, 并且都表现出很高的感染率。白背飞虱各种群Cardinium的感染率几乎均为100%; Wolbachia的感染率也较高, 但雌雄虫感染率差异较大, 雌虫的感染率几乎均为100%, 而雄虫的感染率从22.2%~95.0%不等。另外, 通过不同DNA聚合酶、 不同提取方法的对比, 揭示了DNA粗提样品在基于PCR技术的胞内共生菌检测中的不足之处。对白背飞虱头部、 胸部、 腹部、 足和翅5个不同部位组织的检测结果表明, 不仅在含有生殖组织的腹部有这两类共生菌的感染, 在其他非生殖组织中同样也感染了这两类共生菌; 虽然Wolbachia和Cardinium在寄主的各个组织中均有分布, 但是两者在白背飞虱成虫(尤其是雄虫)阶段的动态变化有明显的差异。进一步对Wolbachia宿主特异性WO噬菌体的检测结果表明, 自然种群雄虫中Wolbachia的感染率与不感染个体中WO噬菌体的比率呈明显的负相关。因此推测, 雄虫中Wolbachia感染率相对较低的原因可能是由于Wolbachia基因组中溶原性的WO噬菌体受到某种因素的诱导已转化为裂解性噬菌体。研究结果为进一步揭示Wolbachia和Cardinium双重感染条件下对寄主的生殖调控作用及其机制、 垂直传播规律、 两者之间的相互关系以及进一步的应用研究等方面提供了重要的理论基础。 相似文献

The aim of this study was to explore the action characters of total flavonoids from MDQ on cerebral ischemic tolerance with blood stasis. Fully understanding the mechanism of action of total flavonoids from MDQ is helpful for the development of new drugs and the utilization of resources. Male Wistar rat model of blood stasis was established by injecting dexamethasone into the intramuscular side of the thigh. Then they were given related drugs via an intragastric administration for a successive 10 days. After 7 days, the following occurred: firstly, the method of blocking the bilateral common carotid artery (CCA) was used for 10 min, followed by a restoration of perfusion. After 72 h, we performed a temporary occlusion of the rat’s middle cerebral artery for 2 h with an intraluminal thread method. This was followed by reperfusion for 24 h, respectively, to establish the rat model of cerebral ischemic tolerance with blood stasis. Viscosity of the whole blood was measured after the last administration was given blood. Brain was removed, and then the activity of ATP enzyme and T-SOD was determined. To observe the pathological changes of the hippocampus area by HE staining, and the expression of Bcl-2 and Bax were observed by immunohistochemical method. The rat model of cerebral ischemic tolerance with blood stasis was copied successfully. The whole blood viscosity, the activity of NOS, the content of Gluin in the ischemic brain in the IPC model group and the ischemia–reperfusion group were increased significantly. The activity of ATPase was decreased significantly. Compared with the ischemia–reperfusion model group, the activity of ATPase and the whole blood viscosity in the ischemic preconditioning (IPC) group were increased significantly. The activity of NOS and the content of Gluin were decreased significantly. The degree of pathological injury of the brain tissue was also relieved significantly. Total flavonoids of MDQ were used, improving blood circulation, improving energy metabolism, activating endogenous anti-oxidative capability, enhancing the antiapoptotic effect, and relieving the injury of the nerve cell. Hence, the use of MDQ flavonoids improves the tolerance ability of cerebral ischemia. 相似文献

The anatomical traits of xylem are the characteristics of tree rings at the cellular and subcellular scales, and are often reflection of environmental signals. Studying the relationships between anatomical traits of xylem and environmental change not only provide physiological explanations to the statistics in dendroclimatology, but can also provide a new vision for studying the adaptation process and response strategies of tree growth to climate change. In this paper, with the relationships between the anatomical characteristics of xylem in tree-rings (cell chronology) and climate change as a main thread, we first outline the basic principles and mechanisms of wood anatomical features to record environmental signals, and expounded the basic methods involved in the process of xylem anatomy. Secondly, we discuss the relationship between the anatomical features of xylem and climate factors. We then propose the following as possible directions of future research based on the existing knowledge gap in the topical area: (1) to explore the temporal and spatial variations in the anatomical characteristics of xylem in tree-rings along radial and tangential directions and the relationships with environmental changes; (2) to explore the threshold of tree growth response to environmental plasticity and adaptation processes; (3) to assess the synergistic and antagonistic effects as well as the formation mechanisms of climate response among different tree-ring proxies, and to determine the specific roles and contributions of major climatic factors during different periods of tree-ring formation. 相似文献

Aims Soil total organic carbon and labile organic carbon are important indicators in evaluating soil quality. Mulching is widely applied to promote the emergence of bamboo shoot in winter time through stand management. Yet the consequences of mulching on soil quality in Phyllostachys edulis have not been well studied. We aim at the quantitative effect of mulching duration on soil quality in P. edulis stands.
Methods Several P. edulis stands located in Huangyan District of Taizhou, Zhejiang Province of China, had been applied with mulching for 1-2 years and were used in this study to assess the mulching effects. We also selected stands without mulching treatment as the reference sites (or control, CK) for comparisons.||||Important findings Total soil organic carbon (TOC), light fraction organic matter (LFOM), and easily-oxidized carbon (EOC) contents at stands with 1-year and 2-year mulching treatments were significantly increased compared with those at the CK sites. The 1-year mulching increased TOC, LFOM and EOC by 11.2%-74.2%, 31.7%-196.9% and 5.0%-79.6%, respectively, than those of CK sites, while by 22.2%-90.8%, 36.7%-238.5%, and 21.9%-97.5% with 2-year treatment. However, the contents of water-soluble organic carbon (WSOC) changed insignificantly. Among the indicators, we found that WSOC:TOC in CK was higher than that with the mulching treatments, while EOC:TOC with 1-year treatment was higher than that with 2-year treatment, and EOC:TOC with 2-year treatment was higher than that of CK. Additionally, WSOC, EOC, and LFOM at all three treatments showed high correlations with TOC, with a higher correlation coefficient of WSOC with TOC of 0- 30 cm soil layers in CK than those with mulching treatments. The correlation coefficient of EOC and LFOM with TOC was the highest at the 2-year mulching sites. More importantly, TOC, WSOC, EOC, and LFOM were significantly (p < 0.05), or extremely significantly (p < 0.01), correlated with soil nutrient content, including total N, hydrolysis N, available P, available K, exchangeable Ca, and exchangeable Mg in all treatments. In sum, it appeared that mulching in short term can increase the contents of TOC, soil labile organic carbons and soil nutrients in bamboo soils, yielding an improved soil quality and thus can be promoted as a plausible practice for the sustainable management of P. edulis stands. 相似文献

Adaptation to different environments can promote population divergence via natural selection even in the presence of gene flow – a phenomenon that typically occurs during ecological speciation. To elucidate how natural selection promotes and maintains population divergence during speciation, we investigated the population genetic structure, degree of gene flow and heterogeneous genomic divergence in three closely related Japanese phytophagous ladybird beetles: Henosepilachna pustulosa, H. niponica and H. yasutomii. These species act as a generalist, a wild thistle (Cirsium spp.) specialist and a blue cohosh (Caulophyllum robustum) specialist, respectively, and their ranges differ accordingly. The two specialist species widely co‐occur but are reproductively isolated solely due to their high specialization to a particular host plant. Genomewide amplified fragment‐length polymorphism (AFLP) markers and mitochondrial cytochrome c oxidase subunit I (COI) gene sequences demonstrated obvious genomewide divergence associated with both geographic distance and ecological divergence. However, a hybridization assessment for both AFLP loci and the mitochondrial sequences revealed a certain degree of unidirectional gene flow between the two sympatric specialist species. Principal coordinates analysis (PCoA) based on all of the variable AFLP loci demonstrated that there are genetic similarities between populations from adjacent localities irrespective of the species (i.e. host range). However, a further comparative genome scan identified a few fractions of loci representing approximately 1% of all loci as different host‐associated outliers. These results suggest that these three species had a complex origin, which could be obscured by current gene flow, and that ecological divergence can be maintained with only a small fraction of the genome is related to different host use even when there is a certain degree of gene flow between sympatric species pairs. 相似文献

In this study, a cough cycle is reproduced using a computational methodology. The Eulerian wall film approach is proposed to simulate airway mucus flow during a cough. The reproduced airway domain is based on realistic geometry from the literature and captures the deformation of flexible tissue. To quantify the overall performance of this complex phenomenon, cough efficiency (CE) was calculated, which provided an easily reproducible measurement parameter for the cough clearance process. Moreover, the effect of mucus layer thickness was examined. The relationship between the CE and the mucus viscosity was quantified using reductions from 20 to 80%. Finally, predictions of CE values based on healthy person inputs were compared with values obtained from patients with different respiratory diseases, including chronic obstructive pulmonary disease (COPD) and respiratory muscle weakness (RMW). It was observed that CE was reduced by 50% in patients with COPD compared with that of a healthy person. On average, CE was reduced in patients with RMW to 10% of the average value of a healthy person. 相似文献

基于江西省地面气象资料和农业气象试验站数据,分析了江西省双季稻生育期的变化趋势,并利用生长度日(GDD)、低温度日(CDD)和高温度日(HDD)对1981-2007年江西省水稻各生育期热量资源的变化趋势进行分析.结果表明:气候变暖背景下,江西省水稻生长季平均气温、平均最低气温和平均最高气温均呈升高趋势,引起双季稻生长季缩短,其中,营养生长期日数减少最明显,而生殖生长期延长;生长度日和高温度日均增加,低温度日减少.研究期间,江西省双季稻有效热量资源增加,低温风险减少,但高温风险增多;江西省水稻有效热量资源的空间变化特征表现为东北部的增幅大于西南部,南部的低温风险大于北部,中部的高温风险最大. 相似文献
