Introduction: The introduction of the MALDI Biotyper in laboratories substantially changed microbiology practice, this has been called a revolution. The system accelerated diagnostic while costs were reduced and accuracy was increased. In just a few years MALDI-TOF MS became the first-line identification tool for microorganisms. Ten years after its introduction, more than 2000 MALDI Biotyper systems are installed in laboratories which are performing routine diagnostic, and the number is still increasing.
Areas covered: This article summarises changes in clinical microbiology introduced by the MALDI Biotyper and its effects, as it has been published in peer reviewed articles found in PubMed. Further, the potential of novel developments to increase the value of the system is described.
Expert commentary: The MALDI Biotyper has significantly improved clinical microbiology in the area of microorganism identification. Now new developments and applications, e.g. for typing and resistance testing, might further increase its value in clinical microbiology. Lithium (Li) metal is an ideal anode material for high energy density batteries. However, the low Coulombic efficiency (CE) and the formation of dendrites during repeated plating and stripping processes have hindered its applications in rechargeable Li metal batteries. The accurate measurement of Li CE is a critical factor to predict the cycle life of Li metal batteries, but the measurement of Li CE is affected by various factors that often lead to conflicting values reported in the literature. Here, several parameters that affect the measurement of Li CE are investigated and a more accurate method of determining Li CE is proposed. It is also found that the capacity used for cycling greatly affects the stabilization cycles and the average CE. A higher cycling capacity leads to faster stabilization of Li anode and a higher average CE. With a proper operating protocol, the average Li CE can be increased from 99.0% to 99.5% at a high capacity of 6 mA h cm?2 (which is suitable for practical applications) when a high‐concentration ether‐based electrolyte is used. 相似文献
An automated closed‐chamber system was developed to measure N2O fluxes in the field. It was deployed at two N‐fertilized grassland sites in two successive years, together with replicated manual chambers, to investigate the spatial and temporal variability in fluxes, and the likely impact of sampling frequency on cumulative flux values. The automated system provided flux data at 8‐h intervals, while manual sampling was conducted at intervals of 3–7 days. The autochambers showed fluctuations in emissions not detected by manual sampling. However, integrated flux values based on the more intensive measurements were on average no more than 14% greater than those based on data from the autochambers that were obtained at the same time as manual sampling. This difference was not significant and well within the spatial variability determined with manual chambers. If daily sampling intervals were used immediately after fertilization, the agreement was closer still, increasing the confidence that can be placed in manual procedures. Diurnal variations in temperature and flux were small, and results from sampling at mid‐day were not significantly different from those based on early morning or evening sampling. Where diurnal fluctuations in temperature and flux are likely to be much larger, the autochamber/sampler system could prove very useful to quantify the effect. 相似文献
Image processing using traditional photogrammetric methods is a labor-intensive process. The collection of photogrammetry images during aerial surveys is expanding rapidly, creating new challenges to analyze images promptly and efficiently, while reducing human error during processing. Computer vision-assisted photogrammetry, a field of artificial intelligence (AI), can automate image processing, greatly enhancing the efficiency of photogrammetry. Here, we present a practical and efficient program capable of automatically extracting the fine-scale photogrammetry of East Asian finless porpoises (Neophocaena asiaeorientalis sunameri). Our results indicated that computer vision-assisted photogrammetry could achieve the same accuracy as traditional photogrammetry, and the results of the comparisons were validated against the direct measurements. Three-dimensional (3D) models using computer vision-assisted photogrammetric morphometrics generated trustworthy body volume estimates. We also explored the one image-based 3D modeling technique, which is less accurate, but still useful when only one image of the animal is available. Although several limitations exist in the current program, improvements could be made to narrow the virtual-reality gap when more images are available for machine learning and training. In the current study, fifteen halophilic and halotolerant bacteria were isolated from salt-affected soil of ?anl?urfa, Turkey. The isolates were characterized by conventional and molecular techniques (16S rDNA sequence analyses) as belonging to seven different genus including Bacillus (5 isolates), Halobacillus (1 isolate), Oceanobacillus (2 isolates), Halomonas (3 isolate), Nesterenkonia (1 isolate), Chromohalobacter (2 isolates) and Jeotgalibacillus (2 isolates). According to the results obtained, the investigated bacterial strains have high salt tolerance and significant enzyme activities which can improve soil nutrient cycling and fertility. Furthermore, these bacterial strains have been investigated for their ability to dissolve common salts available in salt-affected soils. Salt dissolving experiments showed that two Chromohalobacter isolates were able to dissolve CaCO3 and one of the Halomonas isolate was able to dissolve both CaCO3 and MgCO3. As these bacterial isolates can dissolve CaCO3 and MgCO3, the availability of Ca2+ and Mg2+ ions may increase which can enhance the removal of the excess Na+ in soil profile. 相似文献
Data integration is needed in order to cope with the huge amounts of biological information now available and to perform data mining effectively. Current data integration systems have strict limitations, mainly due to the number of resources, their size and frequency of updates, their heterogeneity and distribution on the Internet. Integration must therefore be achieved by accessing network services through flexible and extensible data integration and analysis network tools. EXtensible Markup Language (XML), Web Services and Workflow Management Systems (WMS) can support the creation and deployment of such systems. Many XML languages and Web Services for bioinformatics have already been designed and implemented and some WMS have been proposed. In this article, we review a methodology for data integration in biomedical research that is based on these technologies. We also briefly describe some of the available WMS and discuss the current limitations of this methodology and the ways in which they can be overcome. 相似文献
Inclusion complexes between dexamethasone acetate (DMA), a poorly water soluble drug, and β-cyclodextrin (βCD) were obtained
to improve the solubility and dissolution rate of this drug. Phase-solubility profile indicated that the solubility of DMA
was significantly increased in the presence of βCD (33-fold) and was classified as AL-type, indicating the 1:1 stoichiometric inclusion complexes. Solid complexes prepared by different methods (kneading, coevaporation,
freeze drying) and physical mixture were characterized by differential scanning calorimetry, thermogravimetry, infrared absorption
and optical microscopy. Preparation methods influenced the physicochemical properties of the products. The dissolution profiles
of solid complexes were determined and compared with those DMA alone and their physical mixture, in three different mediums:
simulated gastric fluid (pH 1.2), simulated intestinal fluid (pH 7.4) and distilled water. The dissolution studies showed
that in all mediums DMA presented an incomplete dissolution even in four hours. In contrast, the complexes formed presented
a higher dissolution rate in simulated gastric fluid (SGF pH 1.2), which indicate that these have different ionization characteristics.
According to the results, the freeze–dried and kneaded products exhibited higher dissolution rates than the drug alone, in
