Investigation of the bone and the bone marrow is critical in many research fields including basic bone biology, immunology, hematology, cancer metastasis, biomechanics, and stem cell biology. Despite the importance of the bone in healthy and pathologic states, however, it is a largely under-researched organ due to lack of specialized knowledge of bone dissection and bone marrow isolation. Mice are a common model organism to study effects on bone and bone marrow, necessitating a standardized and efficient method for long bone dissection and bone marrow isolation for processing of large experimental cohorts. We describe a straightforward dissection procedure for the removal of the femur and tibia that is suitable for downstream applications, including but not limited to histomorphologic analysis and strength testing. In addition, we outline a rapid procedure for isolation of bone marrow from the long bones via centrifugation with limited handling time, ideal for cell sorting, primary cell culture, or DNA, RNA, and protein extraction. The protocol is streamlined for rapid processing of samples to limit experimental error, and is standardized to minimize user-to-user variability.  相似文献

BackgroundBreast cancer patients with positive sentinel lymph node biopsy (SLNB) may be spared axillary lymph node dissection (ALND) in favour of irradiation. The aim of the study was to estimate local control probability in the axilla (axLCP).Materials and methodsWe identified 1832 invasive breast cancer patients who had undergone SLNB at our centre. We measured maximal metastasis diameter (SLDmax) in the sentinel lymph nodes and lymph node metastasis volume (VALN) from ALND in 246 patients with one or two positive SLNs. We calculated axLCP after irradiation and systemic treatment for different molecular types.ResultsVALN values are higher for high grade tumours and larger metastases in SLNs (> 5 mm). It is smaller in luminal A tumours. axLCP is high, nearly 100%, in all molecular types in radiation sensitive tumours (SF2 Gy = 0.45), except luminal B. Expected axLCP is relatively low (67%) in luminal B radiation sensitive tumours with no chemotherapy and nearly 100% with chemotherapy.ConclusionVALN values differ among molecular tumour types. They depend on SLNDmax and tumour grade. New prognostic factors are needed for selected luminal B breast cancer patients (i.e. high grade tumours, large metastases in SLNs) after positive SLNB intended to be spared ALND and chemotherapy.  相似文献

Background: Multiple studies showed that long-chain noncoding RNA H19 (LncRNA H19) is high-expressed in human and mouse abdominal aortic aneurysms (AAAs). We speculated that it plays an important role in arterial disease, and therefore studied the role and mechanism of H19 in aortic dissection (AD).Methods: The expressions of related genes in human aortic smooth muscle cells (HASMCs) induced by platelet-derived growth factor BB (PDGF-BB) or in the aortic tissue of AD patients/mice were identified by Western blot and quantitative real-time polymerase chain reaction. The targeting relationship between H19 and miR-193b-3p was predicted and verified by bioinformatics analysis, dual luciferase assay, RNA pull-down assay, RNA immunoprecipitation (RIP), and Pearson correlation coefficient. The H19 and miR-193b-3p effects on the biological functions of tissues and cells were examined by MTT (3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide, thiazolyl blue tetrazolium bromide) assay, wound-healing assay, and Hematoxylin–Eosin (HE) staining.Results: LncRNA H19 was abnormally high-expressed in thoracic aorta tissues of AD patients, and it could competitively bind to and inhibit miR-193b-3p. In the PDGF-BB group, the expressions of H19, matrix metallopeptidase (MMP) 2 (MMP-2) and MMP-9 were up-regulated and the expressions of miR-193b-3p, α-SMA, and SM22α were down-regulated; moreover, the proliferation and migration rate of HASMCs were increased. However, H19 silencing reversed the regulation of PDGF-BB on HASMCs. More interestingly, miR-193b-3p inhibitor could partially reverse the effect of H19 silencing. In addition, the above results were verified by animal experiments, showing that shH19 and up-regulated miR-193b-3p could significantly reduce the thoracic aorta pathological damage in AD mice.Conclusion: LncRNA H19 regulated smooth muscle cell function by sponging miR-193b-3p and it participated in the development of AD.  相似文献

Several neutral glycosphingolipids were hydrogenated and subjected to trifluoroacetylation in trifluoroacetic acid/trifluoroacetic acid anhydride under conditions leading to complete exchange of the N-acetyl groups of GalNAc for N-trifluoroacetyl. The derivatized glycosphingolipids were analyzed for binding by P-fimbriated uropathogenic Escherichia coli, recognizing the globo series of glycolipids (carrying Gal1-4Gal). Using E. coli it was shown that a GalNCO-CF3 next to the minimum binding epitope Gal1-4Gal did not substantially influence the binding, as did not a trifluoro acetyl group on the ceramide. Exchange of N-acetyl of GalNAc in the receptor active gangliotetraosylceramide, Gal1-3GalNAc1-4Gal1-4Glc1-1Cer, for N-trifluoroacetyl, did not change the binding of two out of the three strains tested of the bacterium Neisseria subflava. Discussion concerning the binding epitopes of the bacterial adhesins to carbohydrates is based on these results.  相似文献

Although the bursting patterns with spike undershoot are involved with the achievement of physiological or cognitive functions of brain with synaptic noise, noise induced-coherence resonance (CR) from resting state or subthreshold oscillations instead of bursting has been widely identified to play positive roles in information process. Instead, in the present paper, CR characterized by the increase firstly and then decease of peak value of power spectrum of spike trains is evoked from a bursting pattern with spike undershoot, which means that the minimal membrane potential within burst is lower than that of the subthreshold oscillations between bursts, while CR cannot be evoked from the bursting pattern without spike undershoot. With bifurcations and fast-slow variable dissection method, the bursting patterns with and without spike undershoot are classified into “Sub-Hopf/Fold” bursting and “Fold/Homoclinic” bursting, respectively. For the bursting with spike undershoot, the trajectory of the subthreshold oscillations is very close to that of the spikes within burst. Therefore, noise can induce more spikes from the subthreshold oscillations and modulate the bursting regularity, which leads to the appearance of CR. For the bursting pattern without spike undershoot, the trajectory of the quiescent state is not close to that of the spikes within burst, and noise cannot induce spikes from the quiescent state between bursts, which is cause for non-CR. The result provides a novel case of CR phenomenon and extends the scopes of CR concept, presents that noise can enhance rather than suppress information of the bursting patterns with spike undershoot, which are helpful for understanding the dynamics and the potential physiological or cognitive functions of the nerve fiber or brain neurons with such bursting patterns.  相似文献

ABSTRACT

This study explored the periodic changes in the onset of aortic dissection in a single center in northern China, with the goal to improve predictability and prevention. We collected the clinical data of 1121 patients from Hebei Province treated for acute aortic dissection at Fuwai Hospital between January 2010 and December 2016. The patterns of aortic dissection during different periods of each day, each month, and each quarter of a year were analyzed. Variations in the number of cases were summarized for weekdays, weekends, and different periods of a day in each season. We compared the differences in gender, age, body mass index, Marfan syndrome, hypertension, and the type of aortic dissection during different time periods and different seasons. In the study, 774 patients (69.1%) with type A aortic dissection and 347 patients (30.9%) with type B aortic dissection were included. The average age of 1121 patients was 51.4 ± 12.0 years. Overall, the peak period for the onset of aortic dissection in a day was between 13:00 and 18:00. There was no statistically significant difference in the number of cases between weekdays and weekend (P = 0.94). Most cases occurred in winter (30.9%); the rate of onset was the lowest in summer (16.6%). No statistically significant difference was observed between male and female patients in terms of variations during different periods of a day (P = 0.45) and seasons (P = 0.12). In conclusion, aortic dissection displayed circadian and seasonal patterns in northern China. Onset of the disease peaked between 13:00 and 18:00 in a day. Winter was the peak season for the onset of acute aortic dissection, regardless of sex, type of dissection and age.  相似文献

In recent years, the simple picture of a viral fusion protein interacting with the cell and/or viral membranes by means of only two localized segments (i.e. the fusion peptide and the transmembrane domain) has given way to a more complex picture in which multiple regions from the viral proteins interact with membranes. Indeed, possible roles in membrane binding and/or destabilization have been postulated for the N-terminal heptad repeats, pre-transmembrane segments, and other internal regions of fusion proteins from distant viruses (such as orthomyxo-, retro-, paramyxo-, or flaviviruses). This review focuses on the experimental evidence and functional models postulated so far about the role of these regions in the process of virus-induced membrane fusion.  相似文献

The gaseous molecule hydrogen sulfide (H2S) has been proposed as an endogenous signal molecule and neuromodulator in mammals. Using a newly developed method, we report here for the first time the ability of intact and living brain and colonic tissue in the mouse to generate and release H2S. This production occurs through the activity of two enzymes, cystathionine-γ-lyase and cystathionine-β-synthase. The quantitative expression of messenger RNA and protein localization for both enzymes are described in the liver, brain, and colon. Expression levels of the enzymes vary between tissues and are differentially distributed. The observation that, tissues that respond to exogenously applied H2S can endogenously generate the gas, strongly supports its role as an endogenous signal molecule.  相似文献

Current-voltage (I/V) analysis and pharmacological dissection were applied to membranes of Lamprothamnium at the time of hypotonic stress. At least three types of process were found to be involved in the response to this stress.
  • 1 The first 10min of exposure to hypotonic medium resulted in a depolarization of about 50mV accompanied by a decrease or no change in conductance. This depolarization occurred with either K+ or Ca2+ (and consequently C? channels inactivated.
  • 2 The CI? channels opened mainly in the first 15min of the hypotonic stress, increasing the membrane conductance by about an order of magnitude.
  • 3 The K+ conductance rose as the Cl? conductance started to diminish and reached a maximum after about 40 min.
Both types of channel were strongly potential-dependent with a conductance peak between -150 and 0mV. An inactivation of K+or CI? channels resulted in moving the membrane potential away from the conductance maximum toward either EK or ECI, diminishing the ion efflux (and turgor regulation). The time courses of the conductance increases remained the same, suggesting that the conductance changes are not driven by feedback to some preset turgor level. The electrophysiology of the Lamprothamnium transporters is compared to that of salt-sensitive charophytes.  相似文献
