1. In this paper the results are presented of two experiments designed to examine the relationship between sward structure variables and the nature of patch depression experienced by a large grazing herbivore, Bos taurus, grazing from small patches of forage. In experiment 1 the spatial distribution of leaves within patches was varied. In experiment 2 the role of reproductive stems of different lengths as potential intake rate inhibitors was examined.
2. Both experiments were conducted by offering cattle hand-constructed swards of Orchardgrass ( Dactylis glomerata ) leaves attached to plywood boards. Each of three experimental animals was allowed to take 10, 20, 30, 45 or 60 bites from a patch and intake was estimated on the basis of pregrazing and postgrazing dry-matter. All trials were videotaped and the trial length in seconds was determined from playback of the tapes.
3. In both experiments patch depression (reduction in instantaneous intake rate) was due to decreases in bite mass concomitant with patch depletion. Handling time per bite was unaffected by depletion level. In experiment 1, different spatial configurations resulted in markedly different gain functions ranging from linear to sigmoid. In experiment 2, the presence in the sward of long (15 cm) reproductive stems slowed cumulative gains by restricting bite mass and by increasing the per-bite handling time relative to patches with shorter (5 cm) or no stems.
4. The study of patch depression mechanisms now needs to be generalized over a range of herbivore body sizes and feeding styles.  相似文献

Measurements of single neurone activity in the peripheral pheromone receptors of male Agrotis segetum (Denis & Schiffermüller) (Lepidoptera: Noctuidae) were performed in a wind tunnel using a portable electrophysiological recording unit. Filter paper and rubber septa loaded with synthetic sex pheromone, as well as individual conspecific female glands, were used as pheromone sources. Recordings, up to 3 h long, were analysed for temporal variation in spiking activity. The recordings were performed 2 m downwind of the source, where the pheromone plume had a width of approximately 12 cm, as could be measured with the single cell preparations. The system allowed reliable measurements of relative pheromone concentration with a 20-s time resolution. The release rate from rubber septa loaded with pheromone was more or less constant over time, whereas the release rate from filter paper loaded with pheromone decreased to one tenth of the initial value within 6 min from the application of the pheromone. The release of pheromone from female pheromone glands was pulsed with an interval of 2–10 min between bursts. This pulsing was not caused by retraction of the gland, as the glands were forcibly extruded during the entire experiment, but should reflect variation in transport of pheromone to the gland surface and subsequent release. The demonstrated stability of the preparations using tungsten electrodes, the reliable monitoring of female-produced pheromone plumes at several metres distance, and the time resolution obtained are important steps towards field monitoring of natural pheromone plumes, as well as pheromone concentration and distribution in applications for mating disruption.  相似文献

The final second of the landing approach of black bean aphids, Aphis fabae, was analysed in three dimensions using video techniques. A yellow landing platform was placed upwind or downwind from aphids aggregating under a ceiling light in a laboratory wind tunnel with 10, 20, 30, 40 or 50 cm s–1 wind speeds, and up-tunnel or down-tunnel in still air. As individual aphids flew to the platform, body orientation (assessed by direct observation) was predominantly into-wind whether the initial flight direction to the landing platform was upwind or downwind. A greater proportion showed into-wind body orientation as wind speed increased. Flight track parameters which differed significantly between wind speeds were the track length, linear start to finish distance, linearity index, horizontal ground speed, speed vertical to the ground, vertical turning rate, and horizontal turning rate. The position of the landing platform was important for track length, linear start to finish distance, horizontal ground speed, three-dimensional turning rate, horizontal turning rate, vertical turning rate, and sinuosity. As wind speed increased above 30 cm s–1 the ground speed became more consistent and indicated considerable variation in air speed to adjust for ground speed. For the majority of aphids there was a strong preference (88%) for into-wind landings with initial upwind directed flight, while for downwind flights a significant number (55%) of insects reversed initial flight direction and landed into-wind. Field recorded landings showed that 66% of aphids landed into-wind and there was a mean bearing to the wind of 71 ± 42°, a similar finding to wind-tunnel studies.  相似文献

Abstract.Sixth-stadium nymphs of the grasshopper Schistocerca americana (Drury) (Orthoptera: Acrididae) were observed in a series of experiments designed to measure feeding behaviour in response to suitable and unsuitable phytosterols. In the first experiment, grasshoppers were presented with artificial diet that contained either sitosterol, a suitable phytosterol, or a spinach lipid extract which contained only unsuitable sterols as well as other spinach lipids. The diet with the spinach lipid extract, but not the sitosterol diet, evoked a deterrent response. To determine if the spinach sterols were responsible for the deterrent response, a second experiment was performed where the spinach lipid extract was separated into three lipid classes, including desmethyl sterols, dimethyl sterols and the remaining spinach lipids. Grasshoppers presented with artificial diet containing the desmethyl sterols (the end-product sterols in spinach) exhibited deterrent responses. Finally, feeding behaviour to a suite of different sterols, including cholesterol (suitable), stigmasterol (unsuitable), and lathosterol (unsuitable), was observed; these sterols were selected because they show variation in the position of double bonds. Grasshoppers presented with diets containing unsuitable sterols again exhibited deterrent responses. Overall, the deterrent effect was strongest when sterols with a double bond at position 22 were in the diet.  相似文献

Abstract.The diel locomotor activity patterns of wandering larvae in the flesh fly, Sarcophaga crassipalpis Macquart (Diptera: Sarcophagidae), were examined using a novel apparatus and shown to be primarily diurnal, but with a minority (37%) showing nocturnal activity. In response to the environmental stress of heat shock, a significantly larger proportion (72%) of the larvae became nocturnal. In comparison, adult circadian activity also was predominantly diurnal, but not correlated with the larval activity patterns. In addition, adult patterns showed age-related changes in entrainment and free running period. Finally, the phase of circadian-gated adult eclosion was shown to be entrained by a 3-day exposure to light–dark cycles delivered prior to pupariation, with the phase maintained throughout pupal–adult metamorphosis under constant dark conditions. These results demonstrate that environmental changes may have profound effects on the expression of 24-h activity patterns and circadian rhythms during different life stages throughout development.  相似文献

The questing behaviour of ixodid ticks serves for identification and localisation of approaching hosts and is evoked by carbon dioxide, vibrations, visual and odour stimuli. In an olfactometer, we examined the specificity of the questing response of larvae of Boophilus microplus, a one-host tick which develops mainly on cattle, and Ixodes ricinus, a three-host tick with a broader host spectrum. While all mammalian odours tested were equally stimulatory for I. ricinus, B. microplus was clearly more activated by bovine odours. A phenolic fraction of bovine odour stimulated B. microplus only. Attractive components of the host odours were identified by exposing the ticks to single chemicals and mixtures. Single chemicals stimulated questing responses only at levels higher than the levels detected in the bovine odour. However, an artificial odour blend of 37 pure chemicals, diluted to concentrations at which the individual components were inactive, proved to be as effective as natural host odour for both tick species. Further fractionation of the blend revealed that the combinatory effect was achieved by only 7 compounds in both species. Although B. microplus responded to the same synergistic mixture of volatiles as I. ricinus, it showed significant higher sensitivity to the cattle-associated compounds 1-octen-3-ol and 2-nitrophenol and this might contribute to its host-specificity. 利用抗TNF的T5单抗作为筛选配基,对经DNA碱基组成分析证明具有良好随机性的15肽库进行亲和筛选.经过三轮筛选后,以硝酸纤维素膜斑点印迹法观察到良好的富集效果.由第三轮挑选出的31个克隆进行DNA测序,结果推出的优势克隆的短肽为CYRRPAGGLPGICSA等,竞争性ELISA实验证明带有以上短肽的噬菌体与TNF能竞争性地与T5单抗结合.该多肽可能是T5单抗所识别的模拟表位  相似文献

It is known that the braconid Asobara tabida, a parasitoid of Drosophila larvae, takes oviposition decisions in accordance with survival probabilities in several host species. Nevertheless, larvae of D. simulans, in which the survival probability is virtually zero, are readily accepted for oviposition by searching females. This even happens when they are offered together with D. melanogaster larvae, in which the parasitoid can develop. Here I show that A. tabida can act as a kleptoparasitoid in D. simulans larvae: it can develop in D. simulans larvae, once these larvae are parasitized by another parasitoid, the eucoilid Leptopilina boulardi. Analysis with an optimal foraging model suggests that the increase in survival probability and the occurrence of L. boulardi in the field are high enough to make this kleptoparasitic behaviour of A. tabida pay.  相似文献

Peptide display methods are a powerful tool for discovering new ligands of pharmacologically relevant targets. However, the selected ligands often suffer from low affinity. Using phage display, we identified a new bicyclic peptide binder of prostate-specific membrane antigen (PSMA), a metalloprotease frequently overexpressed in prostate cancer. We show that linking multiple copies of a selected low-affinity peptide to a biocompatible water-soluble N-(2-hydroxypropyl)methacrylamide copolymer carrier (iBody) improved binding of the conjugate by several orders of magnitude. Furthermore, using ELISA, enzyme kinetics, confocal microscopy, and other approaches, we demonstrate that the resulting iBody can distinguish between different conformations of the target protein. The possibility to develop stable, fully synthetic, conformation-selective antibody mimetics has potential applications for molecular recognition, diagnosis and treatment of many pathologies. This strategy could significantly contribute to more effective drug discovery and design.  相似文献
