Cell therapy has been intensely studied for over a decade as a potential treatment for ischaemic heart disease. While initial trials using skeletal myoblasts, bone marrow cells and peripheral blood stem cells showed promise in improving cardiac function, benefits were found to be short‐lived likely related to limited survival and engraftment of the delivered cells. The discovery of putative cardiac ‘progenitor’ cells as well as the creation of induced pluripotent stem cells has led to the delivery of cells potentially capable of electromechanical integration into existing tissue. An alternative strategy involving either direct reprogramming of endogenous cardiac fibroblasts or stimulation of resident cardiomyocytes to regenerate new myocytes can potentially overcome the limitations of exogenous cell delivery. Complimentary approaches utilizing combination cell therapy and bioengineering techniques may be necessary to provide the proper milieu for clinically significant regeneration. Clinical trials employing bone marrow cells, mesenchymal stem cells and cardiac progenitor cells have demonstrated safety of catheter based cell delivery, with suggestion of limited improvement in ventricular function and reduction in infarct size. Ongoing trials are investigating potential benefits to outcome such as morbidity and mortality. These and future trials will clarify the optimal cell types and delivery conditions for therapeutic effect.  相似文献

The present study highlights the importance of preculture time and concentration of TDZ (thidiazuron) for direct regeneration from in vitro leaves (attached to shoots) in Arnebia euchroma. Shoot buds proliferated to form multiple shoots on MS medium (Murashige and Skoog medium) with 5.0 μM Kn. Different additives viz. ascorbic acid, PVP (polyvinylpyrrolidone), PVPP (polyvinylpolypyrrolidone) or activated charcoal (50, 100 and 250 mg/l each) were used to check the phenolic exudations. Direct shoot regeneration was obtained when shoots were initially precultured for 40 days on medium with a higher concentration of TDZ (20.0 μM) and then transferred to a lower concentration (5.0 μM TDZ). The identity of shoot buds was confirmed by histological studies. Regenerated shoots were cultured for 30 days on medium containing Kn (5.0 μM) for proliferation and then transferred to IBA (0.25 μM)‐containing medium for rooting. Rooted plantlets were transferred to greenhouse with 45–50% survival.  相似文献

With the rapid growth of highway mileage and vehicles, the Chinese highway traffic system (HTS) has become one of the great resource consumers. This article attempts to evaluate the material metabolism of China's HTS during 2001–2005 using the approach of material flow analysis (MFA) and to explore possible measures to promote circular economy throughout HTS. We measured a set of indicators to illustrate the whole material metabolism of China's HTS. The results indicated that the direct material input (DMI) of China's HTS increased from 1181.26 million tonnes (Mt) in 2001 to 1,874.57 Mt in 2005, and about 80% of DMI was accumulated in the system as infrastructure and vehicles. The domestic processed output (DPO) increased by 59.0% from 2001 to 2005. Carbon dioxide and solid waste accounted for 80.5% and 10.4% of DPO, respectively. The increase of resource consumption and pollutant emissions kept pace with the growth of transportation turnover. All these suggest that China's HTS still followed an extensive linear developing pattern with large resource consumption and heavy pollution emissions during the study period, which brought great challenges to the resources and the environment. Therefore, it's high time for China to implement a circular economy throughout the HTS by instituting resource and energy savings, by reducing emissions in the field of infrastructure construction and maintenance, by reducing vehicles’ energy and materials consumption, and by recycling waste materials.  相似文献

The study focused on the dynamics of Malondialdehyde (MDA) contents and the activities of protective enzymes in the leaves of alfalfa varieties with various resistances to Aphis medicaginis Koch. The results showed that susceptible varieties always had higher MDA contents than resistant varieties, and the MDA contents tended to rise in both susceptible and resistant varieties in period of the varieties were pierced and sucked by aphids. Superoxide dismutase (SOD), peroxidase (POD) and phenylalanine ammonia-lyase (PAL) activities in susceptible varieties were lower than those in resistant varieties, and in both susceptible and resistant varieties the SOD and POD activities tended to rise at first and then decline, and the PAL activities rose to their peaks and then tended to remain stable. In the susceptible and resistant varieties the catalase (CAT) activities appeared to rise and decline alternatively; the PPO activities in resistant varieties were lower than those in susceptible varieties in early growth, but higher than those in susceptible varieties in later growth. It follows that infested by aphids, susceptible and resistant varieties had the MDA contents, variations of SOD, POD, PAL and PPO activities were closely correlated with their aphid resistances, hence these indexes could be used as physiological indexes for testing aphid resistance of alfalfa, whereas the relations of their CAT activities to their resistances needed to be further studied.  相似文献

Using immature embryos and cotyledons as explants, a successful immature embryo culture and efficient plant direct regeneration via organogenesis from cotyledons, which showed different patterns, was established for the “Xuemei” cultivar of Prunus mume. For immature embryo culture, high frequency plantlet forming (89.5%) from embryo axis was obtained on half-strength Murashige and Skoog (½MS) medium supplemented with 13.2 μM 6-benzyladenine (BA) and 2.7 μM 1-naphthaleneacetic (NAA). At the same time, shoots direct differentiation from cotyledons with the embryo axis development was also observed on ½MS medium containing 2.2 μM BA together with different combinations of NAA (2.7, 5.4 μM) and indole-3-butyric acid (IBA) (0, 2.5, 5.0 μM). Better results were achieved when embryo axes were removed from cotyledons and cultured on ½MS medium supplement with 13.2 μM BA, 2.7 μM NAA (72.9%) or 2.2 μM BA, 2.2 μM thidiazuron (TDZ), and 2.7 μM NAA (84.2%), respectively. Regenerated shoots were successfully rooted on ½MS or Woody Plant medium (WPM) supplemented with 2.5–5.0 μM IBA. The effect of embryo axes, BA and TDZ, on cotyledons’ regeneration were investigated in detail. The rooted plantlets were transferred to soil successfully with normal morphology.  相似文献

【目的】研究秸秆还田方式对东北黑土理化性质及微生物群落的影响。【方法】试验周期为2019年12月至2021年10月,秸秆还田采用2种方式: 秸秆直接还田+微生物菌剂WJ(strawdirect return+microbial agent WJ;MD),秸秆堆肥还田+微生物菌剂WJ(straw compost return +microbial agent WJ;MC)。分析土壤肥力、酶活和微生物群落。【结果】分析两种方式土壤有机质(SOM)、腐殖酸(HS)和富里酸有机碳(FA-C)含量,发现秸秆直接还田+微生物菌剂WJ比秸秆堆肥还田+微生物菌剂WJ分别增加2.28g/kg、7.82g/kg和5.26g/kg。土壤铵态氮(NH4+-N)、速效磷(AP)略高于秸秆堆肥还田+微生物菌剂WJ,均在6月份达到峰值。胡敏酸有机碳(HA-C)含量下降。此外,土壤脲酶、转化酶、纤维素酶活性和碱性磷酸酶活性对比发现,秸秆直接还田+微生物菌剂WJ比秸秆堆肥还田+微生物菌剂WJ分别高8.55%、15.46%、4.35%和6.19%。高通量测序结果显示,秸秆直接还田+微生物菌剂WJ中细菌和真菌的多样性均比秸秆堆肥还田+微生物菌剂WJ丰富。其中AnaerolineaBacteroidetesPseudomonas为优势细菌,TausoniaMrakiaMrakiella为优势真菌。【结论】秸秆直接还田+微生物菌剂WJ比秸秆堆肥还田+微生物菌剂WJ更有利于土壤有机质、腐殖酸、土壤酶活性和微生物多样性的增加,这说明秸秆添加WJ菌剂直接还田可以减少有机养分的流失,保持田间土壤肥力。  相似文献

Recent studies have highlighted the role of coupled side‐chain fluctuations alone in the allosteric behavior of proteins. Moreover, examination of X‐ray crystallography data has recently revealed new information about the prevalence of alternate side‐chain conformations (conformational polymorphism), and attempts have been made to uncover the hidden alternate conformations from X‐ray data. Hence, new computational approaches are required that consider the polymorphic nature of the side chains, and incorporate the effects of this phenomenon in the study of information transmission and functional interactions of residues in a molecule. These studies can provide a more accurate understanding of the allosteric behavior. In this article, we first present a novel approach to generate an ensemble of conformations and an efficient computational method to extract direct couplings of side chains in allosteric proteins, and provide sparse network representations of the couplings. We take the side‐chain conformational polymorphism into account, and show that by studying the intrinsic dynamics of an inactive structure, we are able to construct a network of functionally crucial residues. Second, we show that the proposed method is capable of providing a magnified view of the coupled and conformationally polymorphic residues. This model reveals couplings between the alternate conformations of a coupled residue pair. To the best of our knowledge, this is the first computational method for extracting networks of side chains' alternate conformations. Such networks help in providing a detailed image of side‐chain dynamics in functionally important and conformationally polymorphic sites, such as binding and/or allosteric sites.
