By capturing time-lapse images of primary stromal-vascular cells (SVCs) derived from rat mesenteric adipose tissue, we revealed temporal and spatial variations of lipid droplets (LDs) in individual SVCs during adipocyte differentiation. Numerous small LDs (a few micrometers in diameter) appeared in the perinuclear region at an early stage of differentiation; subsequently, several LDs grew to more than 10 microm in diameter and occupied the cytoplasm. We have developed a method for the fluorescence staining of LDs in living adipocytes. Time-lapse observation of the stained cells at higher magnification showed that nascent LDs (several 100 nm in diameter) grew into small LDs while moving from lamellipodia to the perinuclear region. We also found that adipocytes are capable of division and that they evenly distribute the LDs between two daughter cells. Immunofluorescence observations of LD-associated proteins revealed that such cell divisions of SVCs occurred even after LDs were coated with perilipin, suggesting that the "final" cell division during adipocyte differentiation occurs considerably later than that characterized in 3T3-L1 cells. Our time-lapse observations have provided a detailed account of the morphological changes that SVCs undergo during adipocyte division and differentiation.  相似文献

目的:探讨彩色多普勒超声(CDFI)对乳腺癌新辅助化疗(NCT)疗效评价的临床应用价值。方法:选取2013年1月~2013年12月在我院接受新辅助化疗后行手术治疗的女性乳腺癌患者55例,以病理学评价为金标准,化疗后根据化疗效果分为有效组和无效组,利用CDFI观察患者NCT前后病灶超声指标、病灶内血流分级及阻力指数(RI)值变化。结果:55例患者中,临床触诊疗效评价符合率为36.4%,敏感度为60.7%;CDFI评价符合率为70.9%,敏感度为85.7%。CDFI检查显示,乳腺癌NCT后原发肿瘤病灶大小显著缩小,边界多清晰可见,内部回声及后方回声倾向正常。有效组NCT前后病灶内的血流类型和RI值变化有统计学意义(P0.05),无效组NCT前后病灶内的血流类型和RI值无明显变化(P0.05)。结论:CDFI技术可对乳腺癌NCT前后病灶大小及病灶内部血流动力学变化提供客观参数,是评价乳腺癌NCT疗效的有效方法。  相似文献

The system comprising bacteriophage (phage) lambda and the bacterium E. coli has long served as a paradigm for cell-fate determination. Following the simultaneous infection of the cell by a number of phages, one of two pathways is chosen: lytic (virulent) or lysogenic (dormant). We recently developed a method for fluorescently labeling individual phages, and were able to examine the post-infection decision in real-time under the microscope, at the level of individual phages and cells. Here, we describe the full procedure for performing the infection experiments described in our earlier work. This includes the creation of fluorescent phages, infection of the cells, imaging under the microscope and data analysis. The fluorescent phage is a "hybrid", co-expressing wild- type and YFP-fusion versions of the capsid gpD protein. A crude phage lysate is first obtained by inducing a lysogen of the gpD-EYFP (Enhanced Yellow Fluorescent Protein) phage, harboring a plasmid expressing wild type gpD. A series of purification steps are then performed, followed by DAPI-labeling and imaging under the microscope. This is done in order to verify the uniformity, DNA packaging efficiency, fluorescence signal and structural stability of the phage stock. The initial adsorption of phages to bacteria is performed on ice, then followed by a short incubation at 35°C to trigger viral DNA injection. The phage/bacteria mixture is then moved to the surface of a thin nutrient agar slab, covered with a coverslip and imaged under an epifluorescence microscope. The post-infection process is followed for 4 hr, at 10 min interval. Multiple stage positions are tracked such that ~100 cell infections can be traced in a single experiment. At each position and time point, images are acquired in the phase-contrast and red and green fluorescent channels. The phase-contrast image is used later for automated cell recognition while the fluorescent channels are used to characterize the infection outcome: production of new fluorescent phages (green) followed by cell lysis, or expression of lysogeny factors (red) followed by resumed cell growth and division. The acquired time-lapse movies are processed using a combination of manual and automated methods. Data analysis results in the identification of infection parameters for each infection event (e.g. number and positions of infecting phages) as well as infection outcome (lysis/lysogeny). A study on subjective perception has been carried out in order to gain further insight into subjective discomfort and sensations experienced during 7 T magnetic resonance imaging (MRI). This study provides information about subjective acceptance, which is essential if 7 T MRI is to become a clinical diagnostic tool. Of 573 subjects who underwent 7 T MRI, 166 were also examined at 1.5 T, providing a means of discriminating field‐dependent discomfort. All subjects judged sources of discomfort and physiological sensations on an 11‐point scale (0 = no side effects, 10 = intolerable side effects) and scores were analyzed separately for exam phases, with and without table movement at each field strength. Results revealed that 7 T MRI was, in general, judged more uncomfortable than 1.5 T; however, most subjects rated the effects as being non‐critical (mean scores between 0.5 and 3.5). Significant differences were detected regarding vertigo and sweating between subjects positioned “head‐first” and “feet‐first” at 7 T (worse in “head‐first”) and between 7 and 1.5 T (worse at 7 T), with the effects being more pronounced in the moving compared to the stationary table position. The most unpleasant factor at 7 T was the extensive examination duration, while potentially field‐dependent sensations were rated less bothersome. In summary, our study indicates that although certain sensations increase at 7 T compared to 1.5 T, they are unlikely to hinder the use of 7 T MRI as a clinical diagnostic tool. Bioelectromagnetics. A series of tridentate ligands consisting of mixed aromatic and aliphatic amine derivatives of single amino acid chelates and phenylpiperazine have been developed, and their reactions with [NEt4]2[ReBr3(CO)3] have been investigated. The compounds [Re(CO)3{(NC5H4CH2)NCH3(C2H4)NHCH3}]Br (4), [Re(CO)3{(NC5H4CH2)NCH3(C2H4)NCH3(CH2)xCOOC2H5}]Br (x = 1, 5; x = 4, 6) [Re(CO)3{(NC5H4CH2)NH(C2H4)N(CH3)2}]Br (7), [Re(CO)3{(NC5H4CH2)N(CH 2COOC2H5)(C2H4)N(CH3)2}]Br (8) and [Re(CO)3(NC5H4CH2)(C2H4NH2)N(CH2)3-CH3Ophenpip]Br (9) (phenpip: phenylpiperazine, -C6H4-(CH2CH2)2N-) were prepared and characterized by elemental analysis, NMR, IR, HSMS and X-ray crystallography. All complexes exhibit fac-{Re(CO)3N3} coordination geometry in the cationic molecular unit. 目的:建立荧光素酶标记的人鼻咽癌细胞裸鼠模型,活体成像系统监测肿瘤的生长并与肿瘤的体积进行对比。方法:构建表达荧光素酶基因2(1uc2)的慢病毒载体,与辅助质粒共转染293T细胞以制备慢病毒,感染人鼻咽癌SUNEl细胞后经嘌呤霉素筛选获得表达luc2的细胞株。活体成像设备体外检测不同数量细胞的发光强度,最后以5×10 6个细胞皮下接种BALB/cnu/nu裸鼠,活体成像系统动态记录接种后肿瘤的信号并与肿瘤的体积对比。结果:成功构建慢病毒表达质粒pLenti.1uc2并包装出慢病毒颗粒,病毒感染后嘌呤霉素筛选6天得到鼻咽癌细胞株SUNEl一luc2。细胞株传代后有稳定的发光强度,且经活体检测的每秒光子数与细胞数成正相关(R2=0.96);活体成像观察发现裸鼠接种第2天接种部位的发光强度就达到3-2×10^8,而且成瘤过程中发光强度的变化与肿瘤大小一致。结论:成功构建适用于活体成像的人鼻咽癌SUNEl细胞的裸鼠成瘤模型,该模型从细胞接种开始即可有效动态监测鼻咽癌皮下瘤的生长及转移,从而为鼻咽癌的成瘤机制及药物干预研究提供一个新的手段。  相似文献
