Abstract. In an experimental arena, Melanoplus bivittatus (Say) (Orthoptera: Acrididae) nymphs of four stadia were observed to determine whether they learn to associate food with positional or light-related cues. Nymphs were observed for 4 h in the arena containing one yellow box and one green box, then they were trained by placing food in the green box overnight. Following training, the responses of nymphs to green and yellow boxes were observed for a second 4 h period.
Before training, nymphs of all stadia visited the yellow box more frequently than the green box. Training did not affect the behaviour of second-stadium nymphs, but training increased the relative frequency of visits to the green box by third, fourth and fifth instars. Effects of training on duration of visits to boxes were not evident. Investigations of the mechanism of learning showed that positional cues were used by third-stadium nymphs, but that fourth and fifth instars responded to light-related cues. The progression from no learning in second instars, through use of positional cues by third instars, to use of light-related cues by fourth and fifth instars is probably associated with the greater mobility, greater food consumption, and more varied diet of older nymphs.  相似文献

Many microorganisms are capable of producing and secreting exopolysaccharides (EPS), which have important implications in medical fields, food applications or in the replacement of petro-based chemicals. We describe an analytical platform to be automated on a liquid handling system that allows the fast and reliable analysis of the type and the amount of EPS produced by microorganisms. It enables the user to identify novel natural microbial exopolysaccharide producers and to analyze the carbohydrate fingerprint of the corresponding polymers within one day in high-throughput (HT). Using this platform, strain collections as well as libraries of strain variants that might be obtained in engineering approaches can be screened. The platform has a modular setup, which allows a separation of the protocol into two major parts. First, there is an automated screening system, which combines different polysaccharide detection modules: a semi-quantitative analysis of viscosity formation via a centrifugation step, an analysis of polymer formation via alcohol precipitation and the determination of the total carbohydrate content via a phenol-sulfuric-acid transformation. Here, it is possible to screen up to 384 strains per run. The second part provides a detailed monosaccharide analysis for all the selected EPS producers identified in the first part by combining two essential modules: the analysis of the complete monomer composition via ultra-high performance liquid chromatography coupled with ultra violet and electrospray ionization ion trap detection (UHPLC-UV-ESI-MS) and the determination of pyruvate as a polymer substituent (presence of pyruvate ketal) via enzymatic oxidation that is coupled to a color formation. All the analytical modules of this screening platform can be combined in different ways and adjusted to individual requirements. Additionally, they can all be handled manually or performed with a liquid handling system. Thereby, the screening platform enables a huge flexibility in order to identify various EPS.  相似文献

A delayed matching-to-position (DMP) T-maze task was used to examine the effects of estrogen replacement on spatial learning and memory, as well as the ability of estrogen replacement to reduce performance deficits produced by acute systemic and intrahippocampal muscarinic cholinergic inhibition. Two experiments were performed. In Experiment 1, ovariectomized animals were trained to criterion on the DMP task and then tested with increased intertrial delays and following systemic scopolamine administration. The animals then received either continuous estrogen replacement or sham surgery and were retested beginning 10 days later. In Experiment 2, ovariectomized animals received guide cannulae implanted bilaterally into the hippocampus. Half of these animals also began receiving continuous estrogen replacement. Two months later, the animals were trained on the DMP task and then tested with increased intertrial delays and following systemic as well as intrahippocampal scopolamine administration. Animals received the same test battery 8 months later and were then immediately trained on a reversal task. The results indicate that estrogen-treated animals acquired the DMP task at a significantly faster rate than the ovariectomized, non-estrogen-treated controls. In addition, estrogen replacement significantly reduced deficits in DMP performance produced by intrahippocampal, but not systemic, scopolamine administration. This occurred when animals were tested after 3.5 months, as well as after 12 months, of continuous estrogen replacement. No evidence for an effect of estrogen replacement on spatial working memory or reversal learning was detected. These findings demonstrate that estrogen replacement can enhance acquisition of a spatial memory task and reduce performance deficits associated with hippocampal cholinergic impairment.  相似文献

Several groups of parasitic protozoa, as represented by Giardia, Trichomonas, Entamoeba and Microsporida, were once widely considered to be the most primitive extant eukaryotic group―Archezoa. The main evidence for this is their 'lacking mitochondria' and possessing some other primitive features between prokaryotes and eukaryotes, and being basal to all eukaryotes with mitochondria in phylogenies inferred from many molecules. Some authors even proposed that these organisms diverged before the endosymbiotic origin of mitochondria within eukaryotes. This view was once considered to be very significant to the study of origin and evolution of eukaryotic cells (eukaryotes). However, in recent years this has been challenged by accumulating evidence from new studies. Here the sequences of DNA topoisomerase II in G. lamblia, T. vaginalis and E. histolytica were identified first by PCR and sequencing, then combining with the sequence data of the microsporidia Encephalitozoon cunicul and other eukaryotic groups of different evolutionary positions from GenBank, phylogenetic trees were constructed by various methods to investigate the evolutionary positions of these amitochondriate protozoa. Our results showed that since the characteristics of DNA topoisomerase II make it avoid the defect of 'long-branch attraction' appearing in the previous phylogenetic analyses, our trees can not only reflect effectively the relationship of different major eukaryotic groups, which is widely accepted, but also reveal phylogenetic positions for these amitochondriate protozoa, which is different from the previous phylogenetic trees. They are not the earliest-branching eukaryotes, but diverged after some mitochondriate organisms such as kinetoplastids and mycetozoan; they are not a united group but occupy different phylogenetic positions. Combining with the recent cytological findings of mitochondria-like organelles in them, we think that though some of them (e.g. diplomonads, as represented by Giardia) may occupy a very low evolutionary position, generally these organisms are not as extremely primitive as was thought before; they should be polyphyletic groups diverging after the endosymbiotic origin of mitochondrion to adapt themselves to anaerobic parasitic life.  相似文献

Three elements of wetland biodiversity (aquatic invertebrates, waterbirds and overstorey vegetation of the wetland edge) have been monitored since 1998 at Lake Eganu and Paperbark Swamp in the Western Australian Wheatbelt to provide information about the changes occurring in wetland biodiversity in a landscape that is severely affected by dryland salinization. Changes in extent of wetland vegetation since the 1960s were examined using historical aerial photographs and waterbird use of Lake Eganu during the early 1980s was compared with recent waterbird survey results. Lake Eganu, which is within a major drainage line, started to become salinized in the mid-1960s, about 70 years after land clearing began in the catchment, and its salinity has increased an order of magnitude. The extent of wetland overstorey vegetation and the richness of freshwater aquatic invertebrates have both declined about 80%. Waterbird richness has also declined over the past 20 years, with changes in species composition. Salinization has not occurred at Paperbark Swamp, which is in a small catchment off the main drainage line, and there has been no consistent change in the biodiversity elements monitored.  相似文献

于2004~2005年对江西庐山地区蜻蜓资源进行调查,通过对春、夏、秋三季蜻蜓多样性的研究,共收集蜻蜓1145只,隶属于10科、35属、52种。种类个体数量均表现为夏季最多,春季次之,秋季最少;属数和科数也是以夏季最多,春季次之,秋季最少。三季多样性指数(H′)的排序为春季(2.7603)>夏季(2.6420)>秋季(2.5601)。  相似文献

Searches using position specific scoring matrices (PSSMs) have been commonly used in remote homology detection procedures such as PSI-BLAST and RPS-BLAST. A PSSM is generated typically using one of the sequences of a family as the reference sequence. In the case of PSI-BLAST searches the reference sequence is same as the query. Recently we have shown that searches against the database of multiple family-profiles, with each one of the members of the family used as a reference sequence, are more effective than searches against the classical database of single family-profiles. Despite relatively a better overall performance when compared with common sequence-profile matching procedures, searches against the multiple family-profiles database result in a few false positives and false negatives. Here we show that profile length and divergence of sequences used in the construction of a PSSM have major influence on the performance of multiple profile based search approach. We also identify that a simple parameter defined by the number of PSSMs corresponding to a family that is hit, for a query, divided by the total number of PSSMs in the family can distinguish effectively the true positives from the false positives in the multiple profiles search approach.  相似文献

采用分光光度法,研究了不同生长期香椿叶片的抗氧化活性,并对其抗氧化作用进行了分析。结果表明,阴干处理对香椿总抗氧化力无显著影响,不同生长期的香椿总抗氧化力无显著差异;香椿嫩芽、嫩叶和老叶提取物均有清除.OH的作用,老叶、嫩叶对.OH的清除率均显著大于嫩芽,且阴干香椿对.OH的清除率均显著高于同一生长期的新鲜香椿,尤以阴干老叶的清除率最高;不同生长期的香椿提取物均有抑制亚油酸过氧化的作用,在低浓度时,嫩芽提取物对亚油酸过氧化的抑制率显著高于嫩叶或老叶,而在高浓度时,不同生长期香椿叶提取物对亚油酸过氧化的抑制率无显著差异。  相似文献

Based on a linear mixed-effect model, we propose here a non-destructive, rapid and reliable way for estimating leaf area, leaf mass and specific leaf area (SLA) at leaf scale for broadleaf species. For the construction of the model, the product of leaf length by width (LW) was the optimum variable to predict the leaf area of five deciduous broadleaf species in northeast China. In contrast, for species with leaf thickness (T) lower than 0.10 mm, the surface metric of a leaf (e.g., LW or width) was more suitable for predicting leaf mass; and for species with leaf thickness larger than 0.10 mm, the volume metric of a leaf (e.g., the product of length, width and thickness together, LWT) was a better predictor. The linear mixed-effect model was reasonable and accurate in predicting the leaf area and leaf mass of leaves in different seasons and positions within the canopy. The mean MAE% (mean absolute error percent) values were 6.9% (with a scope of 4.1–13.0%) for leaf area and 13.8% (9.9–20.7%) for leaf mass for the five broadleaf species. Furthermore, these models can also be used to effectively estimate SLA at leaf scale, with a mean MAE% value of 11.9% (8.2–14.1%) for the five broadleaf species. We also propose that for the SLA estimation of the five broadleaf species examined, the optimum number of sample leaves necessary for good accuracy and reasonable error was 40–60. The use of the provided method would enable researchers or managers to rapidly and effectively detect the seasonal dynamic of leaf traits (e.g., leaf area, leaf mass or SLA) of the same sample leaves in the future.  相似文献
