Zeaxanthin, an important component in protection against overexcitation in higher plants, is formed from violaxanthin by the enzyme violaxanthin de-epoxidase. We have investigated factors that may control the maximal degree of conversion in the violaxanthin cycle. The conversion of violaxanthin to zeaxanthin in isolated spinach thylakoids was followed at different temperatures and in the presence of lipid packing modifiers. The maximum degree of conversion was found to be 35%, 70% and 80% at 4 °C, 25 °C and 37 °C respectively. In the presence of membrane modifying agents, known to promote non-lamellar structures (HII), such as linolenic acid the conversion increased, and the maximal level of violaxanthin de-epoxidation obtained was close to 100%. In contrast, substances promoting lamellar phases (Lα), such as α-tocopherol and 8-cetylether (C16EO8), only 55% and 35% of the violaxanthin was converted at 25 °C, respectively. The results are interpreted in light of the lipid composition of the thylakoid membrane, and we propose a model where a negative curvature elastic stress in the thylakoid lipid bilayer is required for violaxanthin de-epoxidase activity. In this model zeaxanthin with its longer hydrophobic stretch is proposed to promote lamellar arrangements of the membrane. As a result, zeaxanthin relieves the curvature elastic stress, which in turn leads to inactivation of violaxanthin de-epoxidase.  相似文献

Hazard identification is based upon the second “law” of toxicology, the specificity of toxic effects caused by a chemical agent. Specificity reflects the differential reactivity inherent in chemical structure and in the biological niches in which chemicals interact. Just as Paracelsus is identified with the first “law” of toxicology, the dose makes the poison, Paré, a century French surgeon, should be credited with an early formulation of the second “law” of toxicology, the specificity of chemical effects. I discuss a number of aspects of hazard identification, including issues related to oxygenated fuels, to routine safety assessment, to the interpretation of hematological neoplasms and to the Precautionary Principle.  相似文献

The effect of glutathione depletion on cellular toxicity of cadmium was investigated in a subpopulation (T27) of human lung carcinoma A549 cells with coordinately high glutathione levels and Cd++-resistance. Cellular glutathione levels were depleted by exposing the cells to diethyl maleate or buthionine sulfoximine. Depletion was dose-dependent. Exposure of the cells to 0.5 mM diethyl maleate for 4 hours or to 10 mM buthionine sulfoximine for 8 hours eliminated the threshold for Cd++ cytotoxic effect and deccreased the LD50S. Cells that were pretreated with 0.5 mM diethyl maleate or 10 mM buthionine sulfoximine and then exposed to these same concentrations of diethyl maleate or buthionine sulfoximine during the subsequent assay for colony forming efficiency produced no colonies, reflecting an enhanced sensitivity to these agents at low cell density. Diethyl maleate was found to be more cytotoxic than buthionine sulfoximine. Synergistic cytotoxic effects were observed in the response of diethyl maleate pretreated cells exposed to Cd++. Thus the results demostrated that depletion of most cellular glutathione in A549-T27 cells prior to Cd++ exposure sensitizes them to the agent's cytotoxic effects. Glutathione thus may be involved in modulating the early cellular Cd++ cytotoxic response. Comparison of reduced glutathione levels and of Cd++ cytotoxic responses in buthionine sulfoximine-treated A549-T27 cells with those levels in other, untreated normal and tumor-derived cells suggests that the higher level of glutathione in A549-T27 is not the sole determinant of its higher level of Cd++ resistance.Abbreviations BSO DL-buthionine-(R,S)-sulfoximine - DEM diethyl maleate - DMSO dimethyl sulfoxide - GSH reduced glutathione - MT metallothionein  相似文献

Maleonitrile-dithiacrown ethers mn-12S2O2 - mn-21S2O5 (mn = maleonitrile) are preorganized S2On+1-coronands (n = 1–4) which force B, AB and A class metal ions into mixed S/O coordination spheres. Moreover, they form chelate complexes with MX2 salts (M = Pd, Pt; X = Cl, Br), which were studied in this paper. The structures of mn-S2On+1 and [PtCl2(mn-S2On+1)] (n = 2, 3) were investigated theoretically by empirical and semiempirical methods using SYBYL (TRIPOS force field) and MSI/DISCOVER97 (ESFF force field). mn-12S2O2 was investigated experimentally by X-ray analysis and 1D and 2D NMR spectroscopy in solution and the complex formation was studied by 1H, 13C and 195Pt NMR titration experiments, respectively. S-inversion was also investigated in order to determine the ring corresponding interconversional barriers. This study was conducted to examine the effects of dietary taurine supplementation on productive performance, nutrient digestibility, antioxidant status, and the gene expression of ileal nutrient transporters in laying quails reared under heat stress (HS). One hundred and eighty laying Japanese quails (Coturnix coturnix japonica) were fed a basal diet or basal diet supplemented with either 2.5 or 5 g of taurine per kg of diet, and reared at either 22 ± 2 °C for 24 h/d (thermoneutral, TN) or 34 ± 2 °C for 8 h/d (HS) for 12 weeks. The quails reared under HS consumed less feed, produced less egg, and had lower dry matter, organic matter and crude protein apparent digestibilities compared with the quails reared under the TN condition (P = 0.001). However, increasing taurine concentrations in the diet improved feed intake and egg production (P = 0.001), but also the apparent digestibilities (P ≤ 0.027) in quails reared under HS. The greater doses (5 g/kg) of taurine resulted in more responses. The quails reared under HS had greater serum and liver MDA concentrations (P = 0.0001) which decreased with dietary taurine supplementations, particularly greater doses. The gene expressions of ileal PEPT1, EAAT3, CAT1, CAT2, SGLT1, SGLT5, GLUT2, and GLUT5 decreased under HS conditions (P = 0.001). However, supplementing taurine, in a dose-dependent fashion, to the diet of quails reared under HS resulted in increases in the gene expressions of the transporters (P < 0.05) except for CAT1. The results of the present work showed that taurine supplementation, particularly with greater doses (5 g/kg), to the diet of laying quails kept under HS acts as alleviating negative effects of HS, resulting in improvements in productive performance and nutrient digestion, and also upregulation of ileal nutrient transporters.  相似文献

Effective, reproducible, and scalable methods for DNA-lipid assembly are important for the success of non-viral vectors in in vivo gene therapy. We hypothesized DNA-lipid assembly would be optimal if started from a liquid monophase where both DNA and lipids separately form molecular or micellar solutions prior to mixing, without preexisting condensed lipid phases, thus allowing DNA-lipid assembly under conditions close to equilibrium. Previously, we found that mixing plasmid DNA, 1-palmitoyl-2-oleoyl-3-sn-phosphatidylcholine (POPC), cholesterol and a cationic lipid, 1, 2-dioleoyl-3-(trimethylammonio) propane (DOTAP) in 50% (v/v) aqueous ethanol spontaneously produced an optically transparent solution. Upon ethanol removal, DNA-lipid nanoparticles (Genospheres™) were formed. For comparison with well-known technologies, different DNA-lipid particles were prepared by interaction of plasmid DNA and stable or ethanol-destabilized lipid vesicles by combining the components in water or 30% (v/v) aqueous ethanol, respectively. Among the three studied DNA-lipid assembly methods, only Genospheres combined the properties of small size (less than or around 100 nm), high incorporation of both lipid and DNA, high degree of DNA protection (dye accessibility 5-12%), a narrow distribution of particle density and when immuno-targeted, the highest transfection efficiency in HER2-overexpressing cells in vitro. We conclude that the Genosphere assembly methodology offers advantages for the development of effective, scalable and targetable non-viral gene delivery vectors.  相似文献

Eleven flavonoids including three new glycosides were isolated from Brickellia chlorolepis and one new and nine known flavonoids were obtained from B. dentata. The new glycosides from B. chlorolepis are 6-methoxykaempferol 3-rhamnoglucoside, spinacetin 3-rhamnogalactoside and veronicafolin 3-rhamnoglucoside. The known compounds identified from B. chlorolepis are patuletin, casticin, artemetin, eupatolitin 3-galactoside, quercetin 3-rhamnogalactoside, rutin, isorhamnetin 3-galactoside and eupatin 3-SO3Ca12. B. dentata contains the new glycoside eupalitin 3-galactoside and nine known compounds: pectolinarigenin, salvigenin, eupafolin, cirsiliol, eupatorin, eupatolitin, eupatolitin 3-glucoside, eupatolitin 3-galactoside and eupatin.  相似文献

A new flavanone glycoside 5-hydroxy-6,7,3′,4′,5′-pentamethoxyflavanone 5-O-α-l-rhamnopyranoside along with quercetagetin 3,6-dimethyl ether and an anthraquinone glycoside have been isolated from the stem bark of Cassia renigera.
