定量方法在民族植物学研究中越来越占有重要的地住,但民族植物学定量研究中有关取样方面还存在着一些不同的看法。笔者针对云南省金平苗族瑶族傣族自治县的拉祜族民间草医所使用的药用植物资源和草医对不同生境中药用植物的利用情况进行了取样,并以此对民族植物学中有关取样中存在的问题进行了讨论。结果表明不同生境中民族植物学的取样面积与植物生态学中的取样面积比较相近。所以在进行民族植物学野外取样时,司以针对具体的生境和植被特点来借用植物生态学中对该类生境或植被所采用的最小取样面积。 相似文献

Free-energy landscape of a chameleon sequence in explicit water and its inherent alpha/beta bifacial property 下载免费PDF全文
A sequence in yeast MATalpha2/MCM1/DNA complex that folds into alpha-helix or beta-hairpin depending on the surroundings has been known as "chameleon" sequence. We obtained the free-energy landscape of this sequence by using a generalized-ensemble method, multicanonical molecular dynamics simulation, to sample the conformational space. The system was expressed with an all-atom model in explicit water, and the initial conformation for the simulation was a random one. The free-energy landscape demonstrated that this sequence inherently has an ability to form either alpha or beta structure: The conformational distribution in the landscape consisted of two alpha-helical clusters with different packing patterns of hydrophobic residues, and four beta-hairpin clusters with different strand-strand interaction patterns. Narrow pathways connecting the clusters were found, and analysis on the pathways showed that a compact structure formed at the N-terminal root of the chameleon sequence controls the cluster-cluster transitions. The free-energy landscape indicates that a small conditional change induces alpha-beta transitions. Additional unfolding simulations done with replacing amino acids showed that the chameleon sequence has an advantage to form an alpha-helix. Current study may be useful to understand the mechanism of diseases resulting from abnormal chain folding, such as amyloid disease. 相似文献

Hydrogen/deuterium exchange mass spectrometry (H/D MS) of monomeric actin (G-actin), polymeric actin (F-actin), phalloidin-bound F-actin and G-actin complexed with DNase I provides new insights into the architecture of F-actin and the effects of phalloidin and DNase I binding. Although the overall pattern of deuteration change supports the gross features of the Holmes F-actin model, two important differences were observed. Most significantly, no change in deuteration was observed in the critical "hydrophobic plug" region, suggesting this feature may not be present. Polymerization also produced deuteration increases for peptide fragments containing the ATP phosphate-binding loops, suggesting G-actin transitions to a more "open" conformation upon polymerization. However, polymerization produced decreases in deuteration mainly localized to the "inner", filament-axis side as predicted by the Holmes model. Mapping the phalloidin-induced decreases in F-actin deuteration onto the Lorenz binding site produced a single common patch straddling two monomers across the 1-start helix contact, again consistent with the Holmes architecture. Finally, both DNase I and phalloidin were able to alter the deuteration of regions distal to their respective binding sites. These results highlight the great opportunities for H/D MS to exploit high-resolution structures for detailed studies of the organization and dynamics of complex molecular assemblies. 相似文献

目的:比较酶切信号放大法(Cervista)与导流杂交基因芯片技术(Hybri Ma)检测高危型人乳头状瘤病毒(HR-HPV)诊断宫颈上皮内瘤变2级或2级以上(CIN2+)的临床价值。方法:随机选择288例2012年3月至2013年1月在哈尔滨医科大学附属第一医院妇科门诊进行新柏氏液基细胞学检查的年龄在20~65岁的宫颈细胞学检测未明确意义的不典型鳞状细胞(ASCUS)的患者,采用Cervista技术与Hybri Ma技术进行高危型HPV检测,并对入组的患者行阴道镜下宫颈活组织检查。以病理学诊断结果为金标准,比较Cervista技术与Hybri Ma技术诊断宫颈上皮内瘤变2级或2级以上(CIN2+)的敏感度、特异度及ROC曲线。结果:在入组的288例患者中,Cervista技术和Hybri Ma技术检出高危型HPV的阳性率分别为49.31%和51.39%(P0.05),其诊断CIN2+的敏感度分别为95.65%和91.30%(P0.05),特异度分别为59.50%和56.20%(P0.05),阳性预计值分别为30.99%和28.38%(P0.05),阴性预计值分别为98.63%和97.14%(P0.05)。两组ROC曲线下面积分别为0.776和0.738(P0.05)。结论:Cervista技术与Hybri Ma技术诊断CIN2+的临床价值相当。 相似文献

黄河故道不同灌水方式刺槐人工林幼树水分利用效率和生长特性 总被引:1,自引:0,他引:1
干旱一直是限制黄河故道沙区树木生长的重要因素之一,而刺槐(Robinia pseudoacacia)以其较强的耐旱性、较高的成活率在造林时备受重视。适宜的灌水方式不但可以提高当地水资源的高效利用,而且可以促进刺槐迅速生长,改善当地环境生态。因此,在2011至2012年间,对黄河故道沙地进行刺槐人工林的建设与培育,开展滴灌(DI)、沟灌(FI)和软管喷灌(HSI)3种灌水方式对刺槐人工林幼树水分利用效率(WUE)和生长特性影响的研究。结果表明,在不同的水文年份,滴灌刺槐的净光合速率(Pn)、蒸腾速率(Tr)高于软管喷灌,软管喷灌高于沟灌。水分利用效率受年份影响较大,但均表现为前期下降,8月份前后降到最低值,随后缓慢回升。不同灌水方式叶绿素含量(CC)差异显著,滴灌条件下刺槐叶绿素含量最高,其次为软管喷灌,沟灌最低。不同树龄,不同灌水方式刺槐株高、地径和胸径差异显著。一年生林,滴灌刺槐的株高、地径和胸径分别比软管喷灌高出23.4、0.27 cm和0.14 cm;二年生林,滴灌刺槐的株高、地径和胸径分别比软管喷灌高53.7 cm、0.61 cm和0.54 cm。滴灌刺槐的株高、地径和胸径的年度生长量分别为软管喷灌的112.1%、107.1%和111.8%,同时分别为沟灌的121.8%、191.8%和343.6%。不同灌水方式刺槐幼树水分利用效率受年际间气象因素影响较大,滴灌可以显著地改善刺槐幼树的光合生理特性,促进其快速生长。 相似文献

目的:构建、表达重组猪源胰蛋白酶原,并对其进行分离纯化和酶活性分析。方法:利用大肠埃希菌表达系统(pET-28a,宿主菌BL21 DE3)优化天然猪源胰蛋白酶原基因,经乳糖诱导表达重组胰蛋白酶原蛋白,采用阴离子交换色谱法分离纯化,以SDS-PAGE 电泳鉴定目标蛋白,以紫外分光光度法检测重组蛋白的酶活力。结果:测序结果表明,重组猪源胰蛋白酶原基因工程菌构建成功,SDS-PAGE检测显示目的蛋白条带位置与标准猪源胰蛋白酶条带位置一致,且酶活力可达513.09 U?mg-1。结论:成功获得了重组猪源胰蛋白酶原,且酶活力较好,为进一步研究生产提供了基础。 相似文献

Laboratory experiments were performed to determine the sampling rates of pesticides for the polar organic chemical integrative samplers (POCIS) used in Japan. The concentrations of pesticides in aquatic environments were estimated from the accumulated amounts of pesticide on POCIS, and the effect of water temperature on the pesticide sampling rates was evaluated. The sampling rates of 48 pesticides at 18, 24, and 30 °C were obtained, and this study confirmed that increasing trend of sampling rates was resulted with increasing water temperature for many pesticides. 相似文献

To measure fatigue in dental implants and in its components, it is necessary to use a probabilistic analysis since the randomness in the output depends on a number of parameters (such as fatigue properties of titanium and applied loads, unknown beforehand as they depend on mastication habits). The purpose is to apply a probabilistic approximation in order to predict fatigue life, taking into account the randomness of variables.
