Optimal production of bispecific antibodies (bsAb) requires efficient and tailored co-expression and assembly of two distinct heavy and two distinct light chains. Here, we describe a novel technology to modulate the translational strength of antibody chains via Kozak sequence variants to produce bsAb in a single cell line. In this study, we designed and screened a large Kozak sequence library to identify 10 independent variants that can modulate protein expression levels from approximately 0.2 to 1.3-fold compared with the wild-type sequence in transient transfection. We used a combination of several of these variants, covering a wide range of translational strength, to develop stable single cell Chinese hamster ovary bispecific cell lines and compared the results with those obtained from the wild-type sequence. A significant increase in bispecific antibody assembly with a concomitant reduction in the level of product-related impurities was observed. Our findings suggest that for production of bsAb it can be advantageous to modify translational strength for selected protein chains to improve overall yield and product quality. By extension, tuning of translational strength can also be applied to improving the production of a wide variety of heterologous proteins.  相似文献

为了设计一种高效酵母菌培养基,以便在需要酵母菌快速生长繁殖时,使用该类培养基,在较短时间内获得大量酵母菌,分别以LB培养基、麦氏培养基作为最基本的培养基加入一定量的大豆提取物,配制成为一定浓度的培养基,并用于酵母菌的培养。根据酵母菌的生长情况,判定大豆提取物对酵母菌的生长繁殖是否有促进作用。实验证明,大豆提取物对酵母菌生长繁殖具有明显的促进作用,尤其是浓度为6%时作用最明显。结果表明,采用LB或麦氏培养基加入6%的大豆提取物对酵母菌的生长促进作用最佳,是一种高效的酵母菌培养基。  相似文献

A new method for real-time monitoring of the oxygen uptake rate (OUR) in bioreactors, based on dissolved oxygen (DO) measurement at two points, has been developed and tested extensively. The method has several distinct advantages over known techniques.It enables the continuous and undisturbed monitoring of OUR, which is conventionally impossible without gas analyzers. The technique does not require knowledge of k(L)a. It provides smooth, robust, and reliable signal. The monitoring scheme is applicable to both microbial and mammalian cell bioprocesses of laboratory or industrial scale. The method was successfully used in the cultivation of NSO-derived murine myeloma cell line producing monoclonal antibody. It was found that while the OUR increased with the cell density, the specific OUR decreased to approximately one-half at cell concentrations of 16 x 10(6) cells/mL, indicating gradual reduction of cell respiration activity. 本文较详细地报道了鳜鱼的耗氧率和窒息点,并对鳜鱼和作其饲料的鱼同池饲养的可能性进行了探讨。结果表明,鳜鱼耗氧量和体重正相关(r=0.99),耗氧率与体重反相关(r=-0.97);在水温20℃,鱼种耗氧率约为0.14mg/g.h.,成鱼约为0.12mg/g.h.;耗氧量和耗氧率均与水温正相关(r=0.90,r=0.94),水温13—30℃时,体重230±11.7g的鳜鱼。耗氧量为14.31—42.13mg/尾。h.,耗氧率为0.059—0.175mg/g.h.;鳜鱼耗氧率昼夜变化与家鱼相反,黄昏至凌晨是高峰期,为0.12—0.15mg/g.h.(T=20℃),白天是低谷期,为0.07—0.10mg/g.h;;鳜鱼的窒息点与家鱼类较接近,变化范围为0.45—0.76mg/L;鳜鱼和作其饲料的鱼在同一池塘中饲养,既可持续不断地提供鳜鱼充足的适口饵料,又简单易行,成本低,效益高,有较大的价值。  相似文献

Aims Spatial distribution of adult trees in a forest community is determined by patterns of both seed dispersal and seedling recruitment. The objectives of our study were to understand the processes of seed dispersal and seedling recruitment of dominant tree species in a temperate forest of northeastern China and to identify the factors constraining seed dispersal and seedling establishment at different stages of forest succession.Methods During three summer and autumn sessions between 2006 and 2008, altogether 113080 seeds from 22 different tree species were collected in three large field plots representing different forest types in the Changbai Mountain region of northeastern China. The spatial distribution of seed abundance was analyzed using a Syrjala test. Regeneration success of nine major tree species was assessed using variables defining 'limitations' in 'seeds' and 'seedling establishment'.Important findings We found that seed production fluctuated between years and varied greatly with forest types. Four tree species, Acer spp., Fraxinus mandshurica, Tilia amurensis and Betula spp., had the greatest seed production and the widest range of seed dispersal, whereas Quercus mongolica showed the most sustained seed production pattern. The spatial patterns of seed abundance differed significantly among forest types and years. The tree species investigated in this study differed in the degree of seed limitation, as well as in limitation of seedling establishment. There were both negative and positive correlations between seed density and seedling density, depending on site and parental tree density. Seeds of 16 tree species were found in the Populus davidiana–Betula platyphylla forest (PBF) plot, 11 in the conifer and broad-leaved mixed forest (CBF) plot but only 8 in the broad-leaved-Korean pine mixed forest (BKF) plot. The number of seed-contributing species was not only greater in the secondary forests (CBF and PBF plots) than in the primary forest (BKF plot) but was also more variable during the 3 years of assessment. Results from the correlations between seed density and seedling occurrence and that between parental tree density or seed weight and dispersal limitation confirm our intuitive expectations, i.e. heavy seeds had greater dispersal limitation but higher establishment success than light seeds.  相似文献

用DME∶Ham sF12(1∶1)培养液,添加3个水平的EGF和2个水平的胰岛素,组合成6种培养体系(CS)分别培养大熊猫皮肤成纤维细胞。通过对细胞生长速度和染色体数目变异率进行测定,测得在添加10μg/ml的胰岛素和40 ng/ml的表皮生长因子的培养体系中,以1.673±0.185×105/ml密度接种细胞,经3.5天,密度达到6.890×105/ml,其生长速度最快;染色体数目为二倍体的百分率为75.77%。综合衡量,CS-5在本研究中更适合大熊猫皮肤成纤维细胞的培养。  相似文献
