Novel insights on the mechanism of action of alpha-amylase inhibitors from the plant defensin family
Plant defensins are small cysteine-rich proteins commonly synthesized in plants, encoded by large multigene families. Most plant defensins that have been characterized to date show potent antifungal and/or bactericidal activities. This report describes VuD1, an unusual defensin that is able to inhibit insect-pest alpha-amylases. VuD1 was cloned from cowpea (Vigna unguiculata) seeds and expressed in a heterologous system. Inhibitory enzyme assays showed that VuD1 efficiently inhibits alpha-amylases from the weevils Acanthoscelides obtectus and Zabrotes subfasciatus, caused low inhibition toward mammalian enzymes and was unable to inhibit the alpha-amylases from Callosobruchus maculatus and Aspergillus fumigatus. To shed some light over the mechanism of action of VuD1, molecular modeling analyses were performed, revealing that the N-terminus of the molecule is responsible for binding with the active site of weevil enzymes. Moreover, models of VuD1 and mammalian enzymes were also generated to elucidate the specificity mechanisms. 对人b防御素3和植物甜蛋白des-pGlu1-Brazzein嵌合基因的工程菌株BL-pET-hBD3-Bra的IPTG诱导表达条件进行了研究, 同时对所表达的目的蛋白进行了纯化和活性分析。IPTG浓度、诱导时间和诱导温度对菌株生长和目的蛋白表达的试验结果显示: 所取的IPTG浓度(0.2~1.0 mmol/L)对菌株生长和目的蛋白的表达无显著影响(P>0.05); 菌株的生物量随着诱导时间的延长而增加, 6 h优于4 h(P<0.01), 但是蛋白的表达量无明显增加(P>0.05); 其中温度是重要的影响因素, 在30°C诱导时, 目的蛋白的表达量占总蛋白的35%左右。进一步的研究表明, 菌株在30°C~32°C生长, 在 30°C诱导最优。对目的蛋白的活性分析表明, 所得到的hBD3-Bra融合蛋白有甜味, 其甜度大约是蔗糖的200倍, 但是其杀菌活性很弱, 经凝血酶切割后, des-pGlu1-Brazzein的甜度大大提高, 大约为蔗糖甜度的600倍, 重组hBD3对大肠杆菌和金黄色葡萄球菌有明显的抑菌活性。 相似文献

目的:分析抗菌肽人β防御素3(humanβdefensin 3,hBD3)对铜绿假单胞菌PAO-1株的抑制作用。方法:合成抗菌肽hBD3,分别通过最低抑菌浓度(minimal inhibitory concentration,MIC)检测、直接杀菌试验、重要功能基因检测分析其对PAO-1的直接抑制作用;并将其与阿奇霉素、四环素、利福平、氯霉素、链霉素、环丙沙星联合施用,观察对抗生素MIC的影响。结果:HBD3对PAO-1的MIC为32μg/mL;在浓度达到8μg/mL时即有明显杀菌作用。HBD3上调PAO-1株的ahpF基因表达,下调aprA和rhlR基因表达。在联用5μg/mL的hBD3后,四环素、利福平、氯霉素、链霉素、环丙沙星的MIC值均有降低。结论:抗菌肽hBD3对铜绿假单胞菌PAO-1株有显著的抑制作用。 相似文献

为了研究抗菌肽β-防御素130的生物学活性和实现大规模制备,通过改良其分子结构,构建表达载体pET28a-3×β-defensin130,利用大肠杆菌BL21 (DE3)作为宿主细胞诱导表达后为水溶性蛋白。对纯化后抗菌肽进行抑菌实验、稳定性实验、MTT实验和溶血性实验确定其生物活性。最终成功制备出25 kDa的重组蛋白,对金黄色葡萄球菌(ATCC25923)(45μg/mL)和单增李斯特菌(ATCC221633)(80μg/mL)等革兰氏阴性和阳性菌都表现出极强的抗菌活性,且其抗菌活性不受温度、pH值和蛋白酶消化等影响,MTT细胞毒性实验显示其对HEK293细胞无毒性且对兔源红细胞具有极低的溶血性。这将为新型抗菌肽的开发提供理论基础并推动抗生素替代产业快速发展。 相似文献

Human alpha defensins are a class of antimicrobial peptides with additional antiviral activity. Such antimicrobial peptides constitute a major part of mammalian innate immunity. Alpha defensins contain six cysteines, which form three well defined disulfide bridges under oxidizing conditions. Residues C3-C31, C5-C20, and C10-C30 form disulfide pairs in the native structure of the peptide. The major tissue in which HD5 is expressed is the crypt of the small intestine, an anaerobic niche that should allow for substantial pools of both oxidized and (partly) reduced HD5. We used ion mobility coupled to mass spectrometry to track the structural changes in HD5 upon disulfide bond reduction. We found evidence of stepwise unfolding of HD5 with sequential reduction of the three disulfide bonds. Alkylation of free cysteines followed by tandem mass spectrometry of the corresponding partially reduced states revealed a dominant pathway of reductive unfolding. The majority of HD5 unfolds by initial reduction of C5-C20, followed by C10-C30 and C3-C31. We find additional evidence for a minor pathway that starts with reduction of C3-C31, followed by C5-C20 and C10-C30. Our results provide insight into the pathway and conformational landscape of disulfide bond reduction in HD5. 相似文献

Human α‐defensin 6 (HD6), unlike other mammalian defensins, does not exhibit bactericidal activity, particularly against aerobic bacteria. Monomeric HD6 has a tertiary structure similar to other α‐defensins in the crystalline state. However, the physico‐chemical reasons behind the lack of antibacterial activity of HD6 are yet to be established unequivocally. In this study, we have investigated the antimicrobial activity of HD6 analogs. A linear analog of HD6, in which the distribution of arginine residues was similar to active α‐defensins, shows broad‐spectrum antimicrobial activity, indicating that atypical distribution of arginine residues contributes to the inactivity of HD6. Peptides spanning the N‐terminal cationic segment were active against a wide range of organisms. Antimicrobial potency of these shorter analogs was further enhanced when myristic acid was conjugated at the N‐terminus. Cytoplasmic localization of the analogs without fatty acylation was observed to be necessary for bacterial killing, while they exhibited fungicidal activity by permeabilizing Candida albicans membranes. Myristoylated analogs and the linear full‐length arginine analog exhibited activity by permeabilizing bacterial and fungal membranes. Site‐specific labeling of synthetic peptides carrying N‐methoxyglycine (MeOGly) by isothiocyanate is demonstrated. A nonapeptide having MeOGly at its N‐terminus was synthesized by the solid‐phase method and reacted with phenylisothiocyanate under various conditions. In acidic solution, the reaction specifically gave a peptide having phenylthiourea structure at its N‐terminus, leaving side chain amino group intact. The synthetic human β‐defensin‐2 carrying MeOGly at its N‐terminus or the side chain amino group of Lys10 reacted with phenylisothiocyanate or fluorescein isothiocyanate also at the N‐methoxyamino group under the same conditions, demonstrating that this method is generally useful for the site‐specific labeling of linear synthetic peptides as well as disulfide‐containing peptides. Among the three extracellular domains of the tetrameric voltage-gated K+ (Kv) channels consisting of six membrane-spanning helical segments named S1–S6, the functional role of the S1-S2 linker still remains unclear because of the lack of a peptide ligand. In this study, the Kv1.3 channel S1-S2 linker was reported as a novel receptor site for human β-defensin 2 (hBD2). hBD2 shifts the conductance-voltage relationship curve of the human Kv1.3 channel in a positive direction by nearly 10.5 mV and increases the activation time constant for the channel. Unlike classical gating modifiers of toxin peptides from animal venoms, which generally bind to the Kv channel S3-S4 linker, hBD2 only targets residues in both the N and C termini of the S1-S2 linker to influence channel gating and inhibit channel currents. The increment and decrement of the basic residue number in a positively charged S4 sensor of Kv1.3 channel yields conductance-voltage relationship curves in the positive direction by ∼31.2 mV and 2–4 mV, which suggests that positively charged hBD2 is anchored in the channel S1-S2 linker and is modulating channel activation through electrostatic repulsion with an adjacent S4 helix. Together, these findings reveal a novel peptide ligand that binds with the Kv channel S1-S2 linker to modulate channel activation. These findings also highlight the functional importance of the Kv channel S1-S2 linker in ligand recognition and modification of channel activation. 相似文献

Defensins constitute a major family of natural antimicrobial peptides that protect the host against microbial invasion. Here, we report on the antibacterial properties and cellular interaction of Human Defensin 5 as a function of its positive charge and hydrophobicity. We find that selective replacement of arginine residues in HD-5 by alanine or charge-neutral lysine residues reduces antibacterial killing as well as host cell interaction. We identify arginines at positions 9 and 28 in the HD-5 sequence as particularly important for its function. Replacement of arginine at position 13 to Histidine, as observed in a Crohn’s disease patient, reduced bacterial killing strain-selectively. Finally, we find that HD-5 interacts with host cells via receptor-mediated mechanisms. 相似文献
