Despite sharing more than 91% sequence identity, the tomato Cf-4 and Cf-9 proteins discriminate between two Cladosporium-encoded avirulence determinants, Avr4 and Avr9. Comparative studies between Cf-4 and Cf-9 are thus of particular interest. To investigate Cf-4 protein function in initiating defence signalling, we established transgenic tobacco lines and derived cell suspension cultures expressing c-myc-tagged Cf-4. Cf-4:myc encodes a membrane-localized glycoprotein of approximately 145 kDa, which confers recognition of Avr4. Elicitation of Cf-4:myc and Cf-9:myc tobacco cell cultures with Avr4 and Avr9, respectively, triggered the synthesis of active oxygen species and MAP kinase activation. Additionally, an Agrobacterium-mediated transient assay was used to express Cf-4:myc and a newly engineered fusion protein Cf-4:TAP. Both transiently expressed proteins were found to be functional in an in vivo assay, conferring a hypersensitive response (HR) to Avr4. Consistent with previous observations that Cf-9 is present in a protein complex, gel filtration analysis of microsomal fractions solubilized with octylglucoside revealed that epitope-tagged Cf-4 proteins migrated at a molecular mass of 350-475 kDa. Using blue native gel electrophoresis, the molecular size was confirmed to be approximately 400 kDa. Significantly, this complex appeared to contain only one Cf-4 molecule, supporting the idea that, as previously described for Cf-9, additional glycoprotein partners participate with Cf-4 in the perception of the Avr4 protein. Intriguingly, Cf proteins and Clavata2 (CLV2) of Arabidopsis are highly similar in structure, and the molecular mass of Cf-4 and CLV complexes is also very similar (400 and 450 kDa, respectively). However, extensive characterization of the Cf-4 complex revealed essentially identical characteristics to the Cf-9 complex and significant differences from the CLV2 complex. 相似文献

SbHRGP3 encodes an HRGP whose expression is correlated with the cessation of root elongation in soybean. The wound-inducible expression of SbHRGP3 interestingly requires sucrose although wounding alone induces the expression of many HRGP genes. To examine whether sugar serves as a specific signal on the wound-inducible expression or whether sugar is required to provide ATP, we examined SbHRGP3::GUS expression in transgenic tobacco plants. Various oligosaccharides including non-metabolizable sugar induced SbHRGP3::GUS expression in transgenic plants. The inhibitors of photosynthesis and of cellular respiration did not affect the wound-inducible expression of SbHRGP3::GUS. However, the induction was significantly affected by PCMBS, an inhibitor of active apoplastic phloem loading of sucrose, suggesting that SbHRGP3::GUS expression in phloem tissues requires translocated sucrose. AIMS: To study the ability of the plasmid-encoded restriction and modification (R/M) system LlaAI to function as a bacteriophage resistance mechanism in Lactococcus lactis during milk fermentations. METHODS AND RESULTS: Plasmid pAIcat4, carrying the R/M system LlaAI and a chloramphenicol resistance cassette, was introduced into the plasmid-free strain L. lactis MG1614 and the industrial strain L. lactis 964. By measuring changes in conductivity the influence of different phage on the growth was determined. CONCLUSIONS: The plasmid-encoded R/M system LlaAI significantly improves the bacteriophage resistance of L. lactis during milk fermentations. SIGNIFICANCE AND IMPACT OF THE STUDY: It is essential to determine the potential of a phage defence mechanism in L. lactis starter culture strains during growth in milk before steps are taken to improve starter cultures. Effectiveness of selenium in different forms like sodium selenite, selenocysteine and selenomethionine has been compared in four different doses, namely 4, 6, 8 and 10 ppm of each, in terms of their bioavailability and prolongation of survival of Dalton's lymphorna (DL) bearing mice. Selenomethionine, at a dose of 8 ppm, was found to be the most bioavailable and least cytotoxic form that was capable of increasing the life span of the tumour bearing hosts maximally (almost two-fold). Benef iciality of selenomethionine has also been studied by observing continuous changes brought about by this compound on the glutathione (GSH) level, glutathione peroxidase (GPx) activity and extent of lipid peroxidation in the hepatic tissue of the tumour bearing hosts, which are indispensable for a cell to function normally and are found to exhibit significantly altered behaviour in neoplastic cells. Selenomethionine caused the maintenance of high steady state GSH level and a normal GPx activity during the fist phase of tumour growth. It also controlled lipid peroxidation during the first 15-20 days following tumour transplantation. These conditions helped in the maintenance of intracellular redox balance, cellular integrity and metabolic rhythms of cells in DL bearing mice receiving selenomethionine.Affiliation 相似文献

Plants successfully use inducible defence mechanisms to combat potential pathogens. Elicitors are signaling compounds that
stimulate any of such defence responses. Recent progress in the isolation of pure elicitors has made possible investigations
on elicitor-binding proteins which might function as receptors in signal transduction pathways that ultimately activate the
defences. The elicitor-binding sites studied so far show a high degree of ligand specificity, as do the candidate binding
proteins identified for some of the ligands. Following elicitor perception, a number of rapid reactions are detectable in
plant cells, including enhanced ion fluxes across the plasma membrane, formation of reactive oxygen intermediates, changes
in protein phosphorylation, and lipid oxidation. Intriguing questions arising from these observations are whether the elicitor-binding
proteins constitute receptors in plant defence signaling and whether any of the rapid events participate in signal transduction
during defence activation.

We present a design to quantify fitness consequences of jasmonate-induced responses in plants that are competing for limited resources with a conspecific. Under both high and low nitrogen supply rates, uninduced (control) Nicotiana attenuata plants growing next to a plant induced with 250 μg methyl jasmonate (MJ) yielded more seed capsules than control plants competing with another control plant. We conclude that there is a opportunity benefit for control plants growing next to an induced plant. Initially, MJ-induced plants grew more slowly, but by senescence they had produced the same number of seed capsules as control plants that had competed with another control plant. Replacement series showed that the fitness of MJ-induced plants is not influenced by the competitive status of their neighbour plant. We argue that competitive designs are useful tools for evaluating the phenotypic costs of ecologically important traits. 相似文献

I analyse and summarize the empirical evidence in mammals supporting alternative benefits that individuals may accrue when committing nonparental infanticide. Nonparental infanticide may provide the perpetrator with nutritional benefits, increased access to limited resources, increased reproductive opportunities, or it may prevent misdirecting parental care to unrelated offspring. The possibility that infanticide is either a neutral or maladaptive behaviour also is considered. I devote the second half of this article to reviewing potential mechanisms that individuals may use to prevent infanticide. These counterstrategies include the early termination of pregnancy, direct aggression by the mother against intruders, the formation of coalitions for group defence, the avoidance of infanticidal conspecifics, female promiscuity, and territoriality. I evaluate the support for each benefit and counterstrategy across different groups of mammals and make suggestions for future research. 相似文献

Sclerites in the gorgonian coral Briareum asbestinum perform the dual role of skeletal support against wave action and structural defence against predators. Local populations of B. asbestinum vary along gradients of decreasing water movement and decreasing predator abundance with increasing depth, such that sclerite length increases and sclerite density decreases with depth. Based on this pattern, I explored a possible trade-off between the sclerite composition that is most resistant to tearing versus most deterrent to predatory gastropods. Feeding assays revealed that artificial foods containing longer sclerites and those containing higher volume fractions of sclerites are less palatable to the gastropod Cyphoma gibbosum. However, real colonies appear constrained, in that they do not contain both long sclerites and high volume fractions at the same time. Given a choice among real colonies, snails prefer shallow-water colonies with shorter sclerites e ven though the sclerite volume fractions are high. Although least deterrent to snails, shallow-water colonies are 56% more resistant to tearing than their deep-water counterparts. Hence, variation in sclerite composition among local populations of B. asbestinum may be maintained by opposing selection for support versus defense. 相似文献
