A comparative molecular genetic study of 37 Kluyveromyces strains of different origin has made it possible to find molecular markers that can differentiate between the dairy yeast Kluyveromyces lactis var. lactis and the genetically close wild Kl. lactis strains from the European “krassilnikovii” population, which are unable to ferment lactose. A restriction fragment length polymorphism analysis of the IGS2 rDNA region reveals two different AluI profiles, one of which corresponds to Kl. lactis var. lactis while the other corresponds to yeasts from the “krassilnikovii” population. The AluI restriction profile of the IGS2 region of the rDNA also makes it possible to differentiate between the physiologically similar species Kl. marxianus and Kl. lactis. The fermentation of lactose (Lac+) in the dairy yeast Kluyveromyces lactis var. lactis is controlled by the LAC4 (β-galactosidase) and LAC12 (lactose permease) genes. The complementation analysis of twelve Kl. lactis var. drosophilarum natural homothallic Lac? strains of different origin was carried out using the genetic heterothallic lines of Kl. lactis var. lactis of the lac4LAC12 and LAC4lac12 genotypes. It was shown that the natural Lac? strains did not possess the LAC4LAC12 gene cluster. Southern hybridization of chromosomal DNA with LAC4 and LAC12 probes, as well as recombination analysis, showed that Kl. lactis var. drosophilarum yeasts do not have even silent copies of these genes. As distinct from this yeast, natural Lac? strains of the yeast Kl. marxianus are mutants impaired in the lactose permease gene (lac12 analogue), but possess an active β-galactosidase gene (LAC4 analogue). In the present study, the relationship between the temperature–humidity index (THI) and the incidence of medical treatments in lactating dairy cows in Lower Saxony, Germany, was investigated. Records of all veterinary-treated cases over 2 years (2003 and 2005) from eight Holstein–Friesian dairy herds raised in loose-housing systems (55 to 170 cows per herd) were evaluated. After exclusion of management-dependent and climate-independent cases, a total of 5547 treatments were analyzed. Treatments were clustered into the following groups: metabolism, fertility, udder and foot/leg. Meteorological data were compiled from the nearest weather station (average distance ± s.d. 39 ± 13 km). Hourly temperatures and relative humidity values were used to calculate the THI, which was divided into classes. Out of the total number of treatments, 37.4%, 32.9%, 21.6% and 8.1% belonged to metabolism, udder, fertility and foot/leg, respectively. Data were analyzed with a mixed model that included THI class, season and year as fixed effects and farm as random effect. In general, incidences were neither affected by the year (P > 0.05) and season (P > 0.05) nor by THI classes (P > 0.05). In tendency, incidences of metabolic treatments increased with increasing THI and incidences of udder treatments increased with decreasing THI. In conclusion, indications of moderate heat stress during summer months in Central Europe were found in the present study, although THI and season did not affect the different disease complexes significantly. 相似文献

Indoor releases of Spalangia cameroni Perkins and Muscidifurax raptor Girauelt & Sanders (Hymenoptera: Pteromalidae) were conducted in five organic dairy cattle farms to evaluate the overall
effect on parasitism and efficiency at different pupal depths of Musca domestica L. (Diptera: Muscidae). Overall, parasitism increased significantly from 5.3 to 28.8–28.7% of the exposed house fly pupae
due to the release of pupal parasitoids. Spalangia cameroni was by far the most dominant species, contributing approximately 71.5–72.3% of the parasitism in the release and post-release
period, whereas 20.9–24.4% could be attributed to Muscidifurax raptor. A naturally occurring ichneumonid, Phygadeuon fumator Gravenhorst (Hymenoptera: Ichneumonidae) parasitized 4.1–6.8% of the exposed fly pupae. The placement of house fly pupae
at two depths of the bedding, 5–10 and 15–20 cm had no significant effect on overall parasitism whereas M. raptor attacked the house fly pupae significantly more when placed in the 5–10 cm stratum (10.0%) compared to the 15–20 cm stratum
(3.2%). The two pupal depths had no significant effect on parasitism by S. cameroni and P. fumator. Albeit S. cameroni contributed significantly to overall parasitism, M. raptor had a significantly higher attack rate when first a female had located bags with sentinel pupae. Based on the above results,
however, S. cameroni seems the most appropriate species for managing house flies in straw bedded dairy cattle farms in Denmark. A biological control

Grazing pastures to low post-grazing sward heights (PGSH) is a strategy to maximise the quantity of grazed grass in the diet of dairy cows within temperate grass-based systems. Within Irish spring-calving systems, it was hypothesised that grazing swards to very low PGSH would increase herbage availability during early lactation but would reduce dairy cow performance, the effect of which would persist in subsequent lactation performance when compared with cows grazing to a higher PGSH. Seventy-two Holstein–Friesian dairy cows (mean calving date, 12 February) were randomly assigned post-calving across two PGSH treatments (n = 36): 2.7 cm (severe; S1) and 3.5 cm (moderate; M1), which were applied from 10 February to 18 April (period 1; P1). This was followed by a carryover period (period 2; P2) during which cows were randomly reassigned within their P1 treatment across two further PGSH (n = 18): 3.5 cm (severe, SS and MS) and 4.5 cm (moderate, SM and MM) until 30 October. Decreasing PGSH from 3.5 to 2.7 cm significantly decreased milk (−2.3 kg/cow per day), protein (−95 g/day), fat (−143 g/day) and lactose (−109 g/day) yields, milk protein (−1.2 g/kg) and fat (−2.2 g/kg) concentrations and grass dry matter intake (GDMI; −1.7 kg dry matter/cow per day). The severe PGSH was associated with a lower bodyweight (BW) at the end of P1. There was no carryover effect of P1 PGSH on subsequent milk or milk solids yields in P2, but PGSH had a significant carryover effect on milk fat and lactose concentrations. Animals severely restricted at pasture in early spring had a higher BW and slightly higher body condition score in later lactation when compared with M1 animals. During P2, increasing PGSH from 3.5 to 4.5 cm increased milk and milk solids yield as a result of greater GDMI and resulted in higher mean BW and end BW. This study indicates that following a 10-week period of feed restriction, subsequent dairy cow cumulative milk production is unaffected. To ensure a sustainable food supply for the growing population, the challenge is to find agricultural systems that can meet production requirements within environmental constraints and demands. This study compares the impacts of winter wheat production on energy use, land use and 100 years Global Warming Potential (GWP100) under different arable farming systems and farming practices. Life cycle assessment was used to simulate the impacts of organic, conventional and integrated farming (IF) systems along the production chain from input production up to the farm gate. The IF system models were designed to combine the best practices from organic and conventional systems to reduce negative environmental impacts without significant yield reductions. An integrated system that used food waste digestate as a fertiliser, and utilised pesticides and no‐tillage had the lowest energy use and GWP per functional unit of 1000 kg wheat output. When the impacts of some specific practices for reducing energy use and GWP were compared, the highest energy use reductions were achieved by replacing synthetic nitrogen fertilisers with anaerobically treated food waste or nitrogen fixing crops, increasing yields through crop breeding and using no‐tillage instead of ploughing. The highest GWP reductions were achieved by using nitrification inhibitors, replacing synthetic nitrogen fertilisers and increasing yields. The major contributors to the uncertainty range of energy use were associated with machinery fuel use and the assumed crop yields. For GWP results, the main source of uncertainty related to the N2O emissions. In conclusion, farming systems that combine the best practices from organic and conventional systems have potential to reduce negative environmental impacts while maintaining yield levels. 相似文献

This study investigates the effects of two feeding systems and two dairy cow breeds on milk yield and composition, physical and sensorial properties of Camembert and Pont-l’Evêque cheeses. The experiment consisted of a 2 × 2 factorial arrangement of treatments. A low energy grass diet with only 15% of concentrate (LowGS) was compared with a high-energy maize silage diet with 30% concentrate (HighMS). Thirty-four Holstein (Ho) and 34 Normande (No) cows in early lactation were assigned to one of two feeding systems for a 6-week period. Cows on the LowGS feeding system had lower milk yield, fat and protein content. In both feeding systems, No cows had lower milk yields but higher milk protein contents than Ho cows. The LowGS feeding system altered milk fatty acid (FA) composition by reducing saturated FA. Breed had only a small impact on milk FA. Concerning milk coagulating properties, only the firmness was reduced by the LowGS feeding and the Ho breed. The effects of breed and feeding system on the protein content of cheeses were more marked in Camembert cheese than in Pont-l’Evêque cheese. However, the Camembert cheese from Ho-LowGS was, in fact, characterized especially by lower protein content. LowGS feeding system and No breed produced more yellow cheeses. Feeding systems had limited effects on the firmness of Camembert and Pont-l’Evêque cheeses measured by penetrometry. In sensory analysis, Ho breed and LowGS feeding produced a Camembert cheese with a more melting texture in the mouth due to the increase of spreadability index and of proteolysis. The type of cheese also had an influence: the effects were more important on Camembert cheese than on Pont-l’Evêque cheese. Only the Ho-LowGS treatment produced a very specific Camembert cheese different from the others. The feeding systems and breed of dairy cow have no determinant effect on PDO (protected designation of origin) Camembert and Pont-l’Evêque cheeses, especially regarding taste. In this kind of trial, despite the effects of feeding systems and breed on milk composition, the role of cheese ripening and microbiology appears to be of considerable importance. 相似文献

Automatic milking systems (AMS), or milking robots, are becoming widely accepted as a milking technology that reduces labour and increases milk yield. However, reported amount of labour saved, changes in milk yield, and milk quality when transitioning to AMS vary widely. The purpose of this study was to document the impact of adopting AMS on farms with regards to reported changes in milking labour management, milk production, milk quality, and participation in dairy herd improvement (DHI) programmes. A survey was conducted across Canada over the phone, online, and in-person. In total, 530 AMS farms were contacted between May 2014 and the end of June 2015. A total of 217 AMS producers participated in the General Survey (Part 1), resulting in a 41% response rate, and 69 of the respondents completed the more detailed follow-up questions (Part 2). On average, after adopting AMS, the number of employees (full- and part-time non-family labour combined) decreased from 2.5 to 2.0, whereas time devoted to milking-related activities decreased by 62% (from 5.2 to 2.0 h/day). Median milking frequency was 3.0 milkings/day and robots were occupied on average 77% of the day. Producers went to fetch cows a median of 2 times/day, with a median of 3 fetch cows or 4% of the herd per robot/day. Farms had a median of 2.5 failed or incomplete milkings/robot per day. Producers reported an increase in milk yield, but little effect on milk quality. Mean milk yield on AMS farms was 32.6 kg/cow day. Median bulk tank somatic cell count was 180 000 cells/ml. Median milk fat on AMS farms was 4.0% and median milk protein was 3.3%. At the time of the survey, 67% of producers were current participants of a DHI programme. Half of the producers who were not DHI participants had stopped participation after adopting AMS. Overall, this study characterized impacts of adopting AMS and may be a useful guide for making this transition. 相似文献
