New candidates for serological markers against leishmaniasis are required to be identified, since the presence of high titers of anti-Leishmania antibodies remain detected in sera of treated and cured patients, when current antigens have being employed. In this study, the diagnostic performance of a conserved Leishmania hypothetical protein was evaluated against a human and canine serological panel. The serological follow-up of the patients was also evaluated, using this recombinant antigen (rLiHyS) in ELISA assays. In the results, high sensitivity and specificity values were found when rLiHyS was used in the serological tests, while when the recombinant A2 (rA2) protein or an antigenic Leishmania preparation were used as controls, low sensitivity and specificity were found. Regarding the serological follow-up of the patients, significant reductions in the anti-rLiHyS antibody levels were found and, one year after the treatments, the anti-protein IgG production was similar to this found in the non-infected groups, reflecting a drop of the anti-rLiHyS antibody production. In conclusion, the present study shows for the first time a new recombinant antigen used to identify tegumentary and visceral leishmaniasis, as well as being able to serologically distinguish treated and cured patients from those developing active disease.  相似文献

The phylogenetic relationships of the genera of Listroderini LeConte, 1876 are analyzed based on 58 morphological characters. The genera are grouped in four clades, which are given subtribal status: Macrostyphlina new subtribe (Adioristidius, Amathynetoides, Andesianellus, Macrostyphlus, Nacodius and Puranius), Palaechthina Brinck, 1948 (Anorthorhinus, Gunodes, Haversiella, Inaccodes, Listronotus, Neopachytychius, Palaechthus, Palaechtodes, Steriphus and Tristanodes), Falklandiina new subtribe (Falklandiellus, Falklandiopsis, Falklandius, Gromilus, Lanteriella, Liparogetus, Nestrius and Telurus), and Listroderina (Acroriellus, Acrorius, Acrostomus, Antarctobius, Germainiellus, Hyperoides, Lamiarhinus, Listroderes, Methypora, Philippius, Rupanius and Trachodema). The subtribes are characterized and keys to identify them and their genera are provided. Listroderini have four main biogeographical patterns: Andean (Macrostyphlina), Andean-New Zealand (Falklandiina), Andean-Neotropical-Australian (Listroderina) and Andean-Neotropical-Australian-New Zealand-Nearctic-Tristan da Cunha-Gough islands (Palaechthina). Geographical paralogy, particularly evident in the Subantarctic subregion of the Andean region, suggests that Listroderini are an ancient Gondwanic group, in which several extinction events might have obscured relationships among the areas.  相似文献

以喀斯特峰丛洼地不同植被恢复阶段灌丛、次生林、原生林为研究对象,采用全挖法分析了各阶段4个优势种的根系构型参数,探讨该特殊生境条件下植物根系构型的差异性、相似性以及资源合理高效的利用方式。结果表明:(1)各个植被恢复阶段优势种拓扑指数TI均趋向于0.5,呈叉状分支结构,并表现为次生林(0.57)原生林(0.49)灌丛(0.46),有利于根系在贫瘠且浅薄的土壤环境中拓展生存空间。(2)3种植被恢复阶段优势种的根系平均连接长度在34.29cm以上,平均为37.01cm,增加连接长度对植物在养分贫瘠的喀斯特土壤环境的生存有利。(3)次生林根系分支率低于灌丛和原生林,这是不同植被恢复阶段优势种对环境所采取的不同适应策略。(4)3种植被恢复阶段优势种根系的横截面积比均符合Leonardo da Vinci法则,且不随直径的变化而变化。(5)3种植被恢复阶段优势种在土壤养分、水分获取及土壤空间拓展方面没有显著差异性。研究认为,在喀斯特峰丛洼地异质性很强的生境下,3种植被恢复阶段优势种的根系均为叉状分支结构,且均以较长的连接长度和较低的分支率策略适应其特殊的生态环境。  相似文献

Four natural compounds were obtained by concentrating, separating and purifying from the Folium isatidis. These natural compounds have been characterized by elemental analysis, IR spectrum, NMR and single‐crystal X‐ray diffraction analysis. The results show that these natural compounds are 4(3H)‐quinazolinone ( I ), 2,4(1H,3H)‐quinazolinedione ( II ), methyl 3,4‐dihydro‐4‐oxoquinazoline‐2‐carboxylate ( III ) and ethyl 3,4‐dihydro‐4‐oxoquinazoline‐2‐carboxylate ( IV ). The antibacterial activity experiment showed that I and II had better activity against Staphylococcus aureus, Escherichia coli, Bacillus subtilis and Salmonella than III , IV and other multiple components, because III and IV have long branches and steric hindrance effect. Compounds I and II have planar structure, which can more easily combine with these bacteria and kill them. The above results have good guiding significance for studying the antibacterial activity for single components or mixtures from natural origin.  相似文献

Extensive tropical forest loss and degradation have stimulated increasing awareness at the international policy level of the need to undertake large‐scale forest landscape restoration (FLR). Natural regeneration offers a cost‐effective way to achieve large‐scale FLR, but is often overlooked in favor of tree plantations. The studies presented in this special issue show how natural regeneration can become an important part of FLR and highlight the ecological, environmental, and social factors that must be considered to effectively do so. They also identify major knowledge gaps and outline a research agenda to support the use of natural regeneration in FLR. Six central questions emerge from these studies: (1) What are the ecological, economic, and livelihood outcomes of active and passive restoration interventions?; (2) What are the tradeoffs and synergies among ecological, economic, and livelihood outcomes of natural regeneration, restoration and productive land uses, and how do they evolve in the face of market and climate shocks?; (3) What diagnostic tools are needed to identify and map target areas for natural regeneration?; (4) How should spatial prioritization frameworks incorporate natural regeneration into FLR?; (5) What legal frameworks and governance structures are best suited to encourage natural regeneration and how do they change across regions and landscapes?; (6) What financial mechanisms can foster low‐cost natural regeneration? Natural regeneration is not a panacea to solve tensions and conflicts over land use, but it can be advantageous under some circumstances. Identifying under what conditions this is the case is an important avenue for future research.  相似文献

Cells of the coccolithophorid Emiliania huxleyi strain CS-57 grown under an atmosphere of air+0.5% CO(2) showed oxidative damage after 10 days growth with concomitant and major changes to the lipid composition. The fatty acid profile was strongly altered and lacked appreciable amounts of the polyunsaturated fatty acids (PUFA: C(18:5), C(18:3) and C(22:6)) typical of healthy cells. Oxidation products of these PUFA could not be detected, but monounsaturated fatty acids proved to be good indicators of oxidative processes. The presence (after NaBH(4)-reduction) of a high proportion of 11-hydroxyoctadec-cis-9-enoic and 8-hydroxyoctadec-cis-9-enoic acids showed that the degradation of oleic acid involved mainly free radical oxidation processes (70-75% autoxidation and 20-25% photooxidation). We also detected large amounts of degradation products of the oxidation product 9,10-epoxyoctadecanoic acid including diols, methoxyhydrins and chlorohydrins. These oxidative effects were found in all the lipid classes examined. Products included significant amounts of chlorophyll side-chain autooxidation products Z- and E-3,7,11,15-tetramethylhexadec-3-ene-1,2-diols and Z-and E-3,7,11,15-tetramethylhexadec-2-ene-1,4-diols, while phytyldiol was present in relatively low proportions. Delta(5)-3beta,7-epimeric unsaturated steroidal diols arising from the autooxidation of the Delta(5) double bond of epi-brassicasterol and minor amounts of Delta(4)-3beta,6-diols were also detected. Long-chain unsaturated ketone (alkenone) content per cell was much higher in the presence of 0.5% CO(2) likely due to carbon storage under these conditions. The proportions of di- and tri-unsaturated alkenones was relatively stable throughout the growth cycle in the absence of additional CO(2), but not when grown with 0.5% CO(2). The detection of characteristic alkenone autoxidation products in cells grown under these latter conditions allowed us to attribute the significant increase in index observed to the involvement of free radical oxidation processes.  相似文献

Abstract. Within different stands of the white-water inundation forest (várzea forest) in the Central Amazon region, composition, abundance, frequency and basal area of tree species were recorded. Determinations of age and radial growth rates were conducted using dendrochronological methods. Results show significant differences in age, history and species composition between stands as well as different growth strategies among dominant species. Assignment of tree species to growth strategies by means of anatomical and morphological features together with quantitative aspects of vegetational analysis permit the further differentiation of successional stages of várzea forests. General features of successional stages were quantitatively described and compared with forest types from outside the várzea. Many tree species of the várzea forests are widespread in South America, and not limited to floodplains. Their occurrence on sites with distinct dry seasons suggests that they are not specifically adapted to flooding but are tolerant to seasonality in general.  相似文献

The potential differences measured on the cell surface and after penetration into the cytoplasm of activated macrophages are described. Linear regressions are made of the measured potential differences as functions of the tip potential of each microelectrode. The surface potential of the macrophage is not significantly different from zero.Mouse macrophages have a transmembrane potential of ?26 mV, whereas in guinea-pig cells this value is ?18 mV. The input resistances of guinea-pig cells are higher than those of mouse macrophages. The cytoplasmic location of the electrode was characterized both by fluorescent dye injection and by electric criteria.Slow membrane hyperpolarizations are directly elicited by mechanical stimulation. Electric responses evoked by current pulses were further characterized.Our results lead to the establishment of objective criteria to validate intracellular recordings from macrophage.  相似文献

Evolutionists have debated whether population-genetic parameters, such as effective population size and migration rate, differ between males and females. In humans, most analyses of this problem have focused on the Y chromosome and the mitochondrial genome, while the X chromosome has largely been omitted from the discussion. Past studies have compared FST values for the Y chromosome and mitochondrion under a model with migration rates that differ between the sexes but with equal male and female population sizes. In this study we investigate rates of coalescence for X-linked and autosomal lineages in an island model with different population sizes and migration rates for males and females, obtaining the mean time to coalescence for pairs of lineages from the same deme and for pairs of lineages from different demes. We apply our results to microsatellite data from the Human Genome Diversity Panel, and we examine the male and female migration rates implied by observed FST values.  相似文献
