探究不同结香方法对土沉香木质部芳香物质形成的影响,为创新和优化土沉香结香技术提供理论参考。设置钻孔、火烧孔和黄绿墨耳菌溶液3个结香处理,12个月后取样检测各处理的土沉香木质部组织变色范围、淀粉和可溶性糖含量,并用95%乙醇(色谱纯)提取个处理土沉香样品的醇溶提取物,测定沉香四醇和主要芳香物质成分。结果表明:(1) 黄绿墨耳菌溶液处理的土沉香木质部组织变色范围最大,纵向变色距离最长达120cm,横向变色距离最宽达3.84cm;钻孔和火烧孔处理,纵向变色距离均超过120cm,但横向变色距离最宽仅为0.58cm和1.06cm。(2) 3种结香处理的土沉香木质部组织淀粉含量均显著低于CK (P<0.05),降幅范围在24.82~55.25%;黄绿墨耳菌溶液和火烧孔处理的土沉香木质部组织可溶性糖含量均显著高于CK (P<0.05),钻孔与CK差异不显著((P>0.05)。(3)钻孔、火烧孔和菌液处理沉香醇溶性提取物含量达到11.84%、13.26%和21.08%,显著高于CK (P<0.05);沉香四醇含量达到0.18%、0.26%和0.42%,对照处理未检测出沉香四醇。(4) 钻孔、火烧孔和黄绿墨耳菌溶液处理的土沉香芳香物质中分别鉴定出29种、33种和36种,主要成分包括色酮及其衍生物、沉香螺醇、愈创木醇、香树烯、苄基丙酮、二氢卡拉酮、枯苏醇、α-檀香醇和壬醛。说明黄绿墨耳菌溶液诱导土沉香木质部组织变色范围最大,淀粉含量消耗和可溶性糖增幅最大,醇溶提取物、沉香四醇含量及其芳香物质积累的效果最优。 相似文献

利用同步辐射(软X射线)辐照玉米自交系H65和H14D种子,研究其M1代的细胞学效应,并以60Co-γ射线作对照。结果表明,软X射线辐照处理后,不仅能够诱发玉米M1代根尖细胞内核畸变和染色体畸变,而且还能够诱发染色体多种类型的变异,其变异频率随辐照剂量的增加而增大,辐照剂量与细胞总畸变率呈正相关。软X射线对玉米根尖细胞的有丝分裂具有明显地抑制作用,辐照剂量与细胞分裂指数呈负相关。软X射线辐照的细胞学效应与γ射线基本相似,但在诱发的细胞畸变率和染色体变异类型上存在一定的差异。两个供试品系对辐射的敏感性为H14D>H65。 相似文献

If asymmetrical free-floating cells or cell elements are mixed with a material which can be extruded as a fiber, a preparation can be made in which the long axes are parallel to one another. For example, a 10% polyvinyl alcohol (Du Pont Elvanol 71-30) solution is mixed with a cell suspension, and extruded through a nozzle (60-300 β in diameter) into an aqueous solution of sodium sulfate (4%) and borax (0.2%). Measurements of the heat capacity and superfluid fraction of confined 4He have been performed near the lambda transition using lithographically patterned and bonded silicon wafers. Unlike confinements in porous materials often used for these types of experiments3, bonded wafers provide predesigned uniform spaces for confinement. The geometry of each cell is well known, which removes a large source of ambiguity in the interpretation of data.Exceptionally flat, 5 cm diameter, 375 µm thick Si wafers with about 1 µm variation over the entire wafer can be obtained commercially (from Semiconductor Processing Company, for example). Thermal oxide is grown on the wafers to define the confinement dimension in the z-direction. A pattern is then etched in the oxide using lithographic techniques so as to create a desired enclosure upon bonding. A hole is drilled in one of the wafers (the top) to allow for the introduction of the liquid to be measured. The wafers are cleaned2 in RCA solutions and then put in a microclean chamber where they are rinsed with deionized water4. The wafers are bonded at RT and then annealed at ~1,100 °C. This forms a strong and permanent bond. This process can be used to make uniform enclosures for measuring thermal and hydrodynamic properties of confined liquids from the nanometer to the micrometer scale. 相似文献

Integrated crop protection tolerates residual weed floras if they are not harmful for crop production. These weeds can host harmful crop pests, among which parasitic plants such as branched broomrape (Phelipanche ramosa). This holoparasite is responsible for large yield losses in French crops such as oilseed rape. To date, there are no herbicides available to control it. To evaluate ex ante the impact of crop management practices on weed-mediated parasite infection of crops, we developed an indicator calculated from outputs of the weed dynamics model FlorSys. It consists of three components assessing weed impact on (1) stimulation of parasite germination during the whole cropping season, i.e. the potential risk reduction for future crops via a reduction of the parasite seed bank, (2) the stimulation of parasite germination in host crops, i.e. the potential risk increase for the current crop, (3) parasite reproduction on weed plants, i.e. the potential risk increase for future crops. This indicator was then used to predict weed-mediated broomrape risk in cropping systems from six regions from France and one from Spain. Antagonisms and synergies with other indicators of weed-harmfulness for crop production and weed contribution to plant and functional biodiversity were investigated with Pearson correlation analyses. For instance, cropping systems with a high parasite risk also had a high functional biodiversity (e.g. weed-based food offer for bees). Effects of crop management practices on the weed-mediated parasite risk indicator were identified with linear models; regression trees were used to identify the combinations of management practices that maximised or minimised weed-mediated broomrape risk. Parasite risk depended on crop rotation, sowing and harvest dates, tillage, herbicides and mechanical weeding. The lowest risk was observed in fields that were last tilled less than 21 days before sowing, with more than 0.6 herbicides per year (i.e. 3 applications in 5 years) with multiple entry modes into the weeds (e.g. leaves and roots) and the last herbicide sprayed no later than 127 days before harvest. RLQ analyses were used to identify correlations between weed species traits (Q matrix) and simulated parasite risk (R matrix), via simulated weed densities (L matrix). Early summer-emerging weed species increased parasite risk. No other notable correlations were found, indicating that parasite risk results from a weed community of interacting species, and not simply from individual weed species. An advice table was built to summarize and explain the effects of crop management practices on weed-mediated parasite risk. 相似文献

Detection of food‐borne pathogens is of great importance in order to minimize the risk of infection for customers. These analyses should be as fast as possible. Any detection method requires enrichment and quantitative analysis of the enriched microbes. Conventional enrichment methods, which take several days, need to be replaced by faster techniques such as immunomagnetic separation (IMS). This technique is based on the use of paramagnetic microspheres coated with antibodies as ligands that have specific affinity to the microbes that have to be detected. In the studies reported here, a rapid method for the detection of Salmonella enterica serovar Typhimurium (Salmonella Typhimurium), combining IMS and Direct Epifluorescence Filter Technique (DEFT), was developed. It was focused on releasing the target cells from the magnetic beads after IMS, because this is a premise for combining IMS, as an alternative pre‐enrichment, with DEFT. Otherwise, the high number of beads form a layer on the filter membrane that makes the following microscopic analysis for the detection of the contaminants impossible. The CELLectionTM Dynabeads® used in this study, are coated with recombinant streptavidin (rSA) via a DNA linker. The rSA binds biotinylated antibodies that are able to capture target cells. The DNA linker provides the cleavable site, so that the beads can be removed from the captured cells after isolation. In this study a releasing procedure was developed. This procedure allows for an average 74 % ± 4 % of the bead‐bound Salmonella Typhimurium cells to be released from the beads after IMS, so that the detection of the separated cells by DEFT will be possible. 相似文献

Aim Understanding the importance of ecological factors in the origin and maintenance of patterns of phenotypic variation among populations, in an explicit geographical context, is one of the main goals of human biology, ecology and evolutionary biology. Here we study the ecological factors responsible for craniofacial variation among human populations from South America. Location South America. Methods We studied a dataset of 718 males from 40 South American populations, coming from groups that inhabited different geographical and ecological regions. Cranial size and shape variation were studied using 30 cranial measurements. We first used spatial correlograms and interpolated maps to address spatial patterns. We then regressed the shape (principal component scores) and size variables against ecology (mean annual temperature and diet) using multiple and multivariate spatial regression. Finally, the expected magnitudes of shape and size divergence under the influence of genetic drift and mutations alone were evaluated using neutral expectation for the divergence rate. Results The spatial correlograms showed a cline affecting the entire South American distribution. Interpolated maps showed that size and allometric shape vary from south‐east to north‐west. Multiple and multivariate regression analyses suggested that diet has the largest and most significant effect on this pattern of size and allometric shape variation. Finally, the results of the divergence rate test suggested that random processes alone cannot account for the morphological divergence exhibited by cranial size and allometric shape scores among southernmost populations. Main conclusions Correlograms, spatial regression and divergence rate analyses showed that although local factors (neutral processes or local environmental conditions) are important to explain spatial interpopulation differentiation in cranial characteristics among these populations, there is significant correlation of cranial size and allometric shape variation with diet. Gene flow among human populations, or local environmental conditions, could explain spatial variation mainly at smaller spatial scales, whereas the large‐scale pattern of the South American dataset is mainly related to the high proportion of carbohydrates and low proportion of proteins consumed. 相似文献

Non-invasive genetic analyses are important for studies of species that are rare, sensitive or at risk of extinction. This study investigates the possibility of using faeces and urine to obtain microsatellite genotypes for individual identification of wolverines (Gulo gulo). The reliability of the employed method was assessed by analysing independent amplifications of non-invasive samples (a multiple-tube approach) and by comparing genotypes obtained from faeces to genotypes obtained from blood or tissue of the same individual. Ten microsatellite markers were successfully amplified in 65% of the faecal samples (n = 32) and 40% of the urine samples (n = 22). Allelic dropout was found in 12 and 14% of the amplifications from extracts of faeces and urine, respectively. Nevertheless, all multi-locus genotypes were correct, as judged from comparison to data from tissue or blood samples, after three replicates. These results suggest that a non-invasive approach based on DNA-analysis of faeces can be a powerful tool in population monitoring of wolverines, potentially providing reliable estimates of population size and immigration rate. A second objective of the study was to develop markers for DNA-based sex identification in wolverines using non-invasive samples. We developed two Y-linked markers, one that was specific to wolverine and one that also successfully identified sex in another mustelid. Cell lines are widely used for various research purposes including cancer and drug research. Recently, there have been studies that pointed to discrepancies in the literature and usage of cell lines. That is why we have prepared a comprehensive overview of the most common gynaecological cancer cell lines, their literature, a list of currently available cell lines, and new findings compared with the original studies. A literature review was conducted via MEDLINE, PubMed and ScienceDirect for reviews in the last 5 years to identify research and other studies related to gynaecological cancer cell lines. We present an overview of the current literature with reference to the original studies and pointed to certain inconsistencies in the literature. The adherence to culturing rulesets and the international guidelines helps in minimizing replication failure between institutions. Evidence from the latest research suggests that despite certain drawbacks, variations of cancer cell lines can also be useful in regard to a more diverse genomic landscape. 相似文献

Seventy cases of invasive adenocarcinoma of the uterine cervix were predicted on cytological grounds and confirmed by histopathology. The cervical smears from these cases were further analysed and classified into well and poorly differentiated adenocarcinoma on the basis of the pattern of the cells in the smear and their nuclear detail. Using this method of analysis it is possible to increase the accuracy of cytology for the diagnosis of adenocarcinoma of the uterine cervix. 相似文献
