The conformations of four BK antagonists, [D-Arg 0, Hyp3, Thi5, D-Phe7, Acc8]BK (1), Aaa[D-Arg 0, Hyp3, Thi5, D-Phe7, Acc8]BK (2), [D-Arg 0, Hyp3, Thi5, 8, Apc7]BK (3), and Aaa[D-Arg(0), Hyp(3), Thi(5, 8), Apc7]BK (4) were studied by using 2D NMR spectroscopy and MD simulations with time-averaged (TAV) restraints. According to the results of the NMR measurements, the BK antagonists contain 7-30% of minor conformation resulting from cis/trans isomerization of the peptide bonds preceding either Pro or Hyp residues. The major conformation of each peptide possesses all peptide bonds in trans configuration. Peptides modified with the Apc residue at position 7 (peptides 3 and 4) possess a higher percentage of minor isomer.Peptide 1 exhibits the strongest vasodepressor potency among the analogs studied and as a single one forms the betaII-turn in the 2-5 fragment, which is believed to be crucial for antagonistic activity. This peptide is also the most compact. The radius of gyration (Rg) amounts to 6.9 A and is by ca 1.5 A lower than that of the remaining analogs. With peptide 4, the ST-turn of type I within the Ser6-Thi8 fragment was found. 相似文献

The control of wine microbial population during and beyond fermentation is of huge importance for wine quality. Lactic acid
bacteria (LAB) in wine are responsible for malolactic fermentation (MLF) which can be desired in some cases and undesirable
in others. Some LAB do not perform MLF and their uncontrolled growth could contribute to severe wine spoilage such as undesired
flavours. Their identification and detection is considered crucial for numerous biotechnological applications in food fermentations,
where, through acidification and secretion of bacteriocins, they contribute to reduce food spoilage and growth of pathogenic
microorganisms. LAB have traditionally been classified using morphological or biochemical features. Primary isolation, biochemical
identification and phenotypic analysis are laborious, time consuming and inaccurate and often lead to misidentification within
some genera such as Pediococcus. Molecular identification based on suitable marker genes could be an attractive alternative to conventional morphological
and biochemical methods. We assessed here the applicability of four housekeeping genes recA, rplB, pyrG and leuS in combination with the mle gene in multi-loci sequence typing (MLST) of Pediococcus parvulus and Pediococcus damnosus. Sequencing and comparative analysis of sequence data were performed on 19 strains collected during wine fermentation. A
combination of these five marker genes allowed for a clear differentiation of the strains analysed, indicating their applicability
in molecular typing. Analysis of the observed nucleotide polymorphisms allowed designing highly discriminative primers for

Increasing concentrations of organic matter (OM) in surface waters have been noted over large parts of the boreal/nemoral zone in Europe and North America. This has raised questions about the causes and the likelihood of further increases. A number of drivers have been proposed, including temperature, hydrology, as well as ‐ and Cl? deposition. The data reported so far, however, have been insufficient to define the relative importance of different drivers in landscapes where they interact. Thirty‐five years of monthly measurements of absorbance and chemical oxygen demand (COD), two common proxies for OM, from 28 large Scandinavian catchments provide an unprecedented opportunity to resolve the importance of hypothesized drivers. For 21 of the catchments, there are 18 years of total organic carbon (TOC) measurements as well. Despite the heterogeneity of the catchments with regards to climate, size and land use, there is a high degree of synchronicity in OM across the entire region. Rivers go from widespread trends of decreasing OM to increasing trends and back again three times in the 35‐year record. This synchronicity in decadal scale oscillations and long‐term trends suggest a common set of dominant OM drivers in these landscapes. Here, we use regression models to test the importance of different potential drivers. We show that flow and together can predict most of the interannual variability in OM proxies, up to 88% for absorbance, up to 78% for COD. Two other candidate drivers, air temperature and Cl?, add little explanatory value. Declines in anthropogenic since the mid‐1970s are thus related to the observed OM increases in Scandinavia, but, in contrast to many recent studies, flow emerges as an even more important driver of OM variability. Stabilizing levels also mean that hydrology is likely to be the major driver of future variability and trends in OM. 相似文献

Question: Which factors determine diversity of native and alien vascular plant species in semi‐natural dry grasslands? Location: Northern limestone Alps to the southern rim of the Bohemian massif in northern Austria. Methods: In 70 randomly chosen dry grassland patches (0.008 ha ‐ 7 ha) we sampled a complete inventory of vascular plant species at each site. We analysed the correlation between species diversity of natives, archaeophytes (pre‐1500 aliens) and neophytes (post‐1500 aliens). We used GLM to study the relationship of species number (natives, neophytes, archaeophytes) to five explanatory variables (altitude, within habitat diversity, habitat diversity of adjacent areas, within land‐use diversity and land‐use in adjacent areas). Orthogonal components of these variables were derived with a PCA and used in the models. We also tested the influence of minimum residence time (MRT) and the covariables origin, mode of introduction and life form on the number of grassland sites with neophytes with analogous GLMs. Results: Native species diversity species was positively correlated with the species diversity of new, but not old invaders. GLM explains 70% of the variance in the number of native species. Patch size explained the largest part of the variation in the number of native species. PCA axes 1 and 3 were significantly related to the number of native species. Axis 1was related to on‐site habitat and land‐use diversity. The GLM of the archaeophyte diversity explains 18% of the variance. Altitude and presence of fields and grassland in the neighbourhood mainly explained archaeophyte species diversity. The GLM of neophyte diversity explains 12% of the variance. The number of neophytes was positively related to that of archaeophytes. Only PCA axis 3, which is mainly influenced by adjacent land‐use types, showed a relationship with neophytes. MRT, mode of introduction and region of origin (but not life form) were significantly related to the number of grassland sites invaded by neophytes, explaining 35% of the variance. Conclusion: Most factors governing native species diversity are not significantly related to alien species diversity. Additional determinants of the local scale diversity of alien species exist such as region of origin and historical factors (MRT, mode of introduction). 相似文献

The annual life cycle of many birds includes breeding, moult and migration. All these processes are time and energy consuming and the extent of investment in any one may compromise the others. The output from breeding is of course the ultimate goal for all birds, while the investment in moult and migration should be selected so that lifetime fitness is maximized. In particular, long-distance migrants breeding at high latitudes face severe time pressures, which is a probable reason why natural selection has evolved efficient behaviours, physiological and morphological adaptations allowing the maximum possible migration speed. Optimal migration theory commonly assumes time minimization as an overall strategy, but the minimization of energy cost and predation risk may also be involved. Based on these assumptions, it is possible to derive adaptive behaviours such as when and at which fuel load a stopover site should be abandoned. I review some core components of optimal migration theory together with some key predictions. A review of accumulated empirical tests of the departure rule indicates that time minimization is an important component of the overall migration strategy, and hence gives support to the assumption about time-selected migration. I also briefly discuss how the optimal policy may be implemented by the bird by applying a set of simple rules. The time constraints on migrants increase with increasing body size. Some consequences of this are discussed. 相似文献

We model the evolution of flowering time using a multilocus quantitative genetic model with non-selective assortative mating and mutation to investigate incipient allochronic speciation in a finite population. For quantitative characters with evolutionary parameters satisfying empirical observations and two approximate inequalities that we derived, disjunct clusters in the population flowering phenology originated within a few thousand generations in the absence of disruptive natural or sexual selection. Our simulations and the conditions we derived showed that cluster formation was promoted by limited population size, high mutational variance of flowering time, short individual flowering phenology and a long flowering season. By contrast, cluster formation was hindered by inbreeding depression, stabilizing selection and pollinator limitation. Our results suggest that incipient allochronic speciation in populations of limited size (satisfying two inequalities) could be a common phenomenon. 相似文献

The developmental threshold is the minimum size or condition that a developing organism must have reached in order for a life-history transition to occur. Although developmental thresholds have been observed for many organisms, inter-population variation among natural populations has not been examined. Since isolated populations can be subjected to strong divergent selection, population divergence in developmental thresholds can be predicted if environmental conditions favour fast or slow developmental time in different populations. Amphibian metamorphosis is a well-studied life-history transition, and using a common garden approach we compared the development time and the developmental threshold of metamorphosis in four island populations of the common frog Rana temporaria: two populations originating from islands with only temporary breeding pools and two from islands with permanent pools. As predicted, tadpoles from time-constrained temporary pools had a genetically shorter development time than those from permanent pools. Furthermore, the variation in development time among females from temporary pools was low, consistent with the action of selection on rapid development in this environment. However, there were no clear differences in the developmental thresholds between the populations, indicating that the main response to life in a temporary pool is to shorten the development time. 相似文献
