Although a centrifugal bioreactor (CCBR) supports high-density mammalian suspension cell cultures by balancing drag, buoyancy, and centrifugal forces, to date anchorage-dependent cultures have not been tried. Also, steady or intermittent hydrostatic pressures of 8 to 500 kPa, and shears of 0.02 to 1.4 N/m(2) can be simultaneously applied in the CCBR. This article demonstrates the use of a CCBR to stimulate chondrogenesis in a high-density culture. At 3 weeks, histological results show even distribution of glycosaminoglycan (GAG) and collagen, with 1,890 ± 270 cells/mm(2) cell densities that exceed those of 1,470 ± 270 in pellet cultures. Analysis of collagen content reveals similar levels for all treatment groups; 6.8 ± 3.5 and 5.0 ± 0.4 μg collagen/μg DNA for 0.07 and 0.26 MPa CCBR cultures, respectively, in contrast to 6.6 ± 1.9 values for control pellet cultures. GAG levels of 5.6 ± 1.5 and 4.1 ± 0.9 μg GAG /μg DNA are present for cultures stressed at 0.07 and 0.26 MPa, respectively, in comparison to control pellet cultures at the 8.4 ± 0.9 level. Although results to date have not revealed mechanical stress combinations that stimulate chondrogenesis over unstressed controls, system advantages include continuous culture at cell densities above those in the pellet, precise medium control, the ability to independently vary multiple mechanical stresses over a broad range, and the flexibility for integration of scaffold features for future chondrogenesis stimulation studies.  相似文献

细菌是维持养殖水体稳定的重要类群,可以反映出水质情况,微生态制剂作为一种可以去除有害物质的生物方法,目前被广泛应用。了解微生态制剂在投入养殖海水后细菌群落结构和多样性变化,探讨不同季节的水质环境因子对细菌群落的影响。通过高通量测序技术对细菌16S rRNA V3、V4区进行扩增,分析冬、春、夏3个季节下使用完商品型菌剂后,养殖海水中细菌群落分布规律及其多样性,采用冗余分析(Redundancy analysis,RDA)探讨不同季节的水质指标与细菌群落之间的关系。养殖海水中细菌多样性丰富,3个季节主要的优势菌门为变形菌门和拟杆菌门,其中Tenacibaculum(38.28%),卓贝尔氏菌属(Zobellella,30.78%)、盐单胞菌属(Halomonas,13.55%)为冬季优势属;Glycocaulis(34.00%)、盐单胞菌属(11.36%)、Planktosalinus(9.67%)为春季优势属;Pseudofulvimonas(28.94%)、Zobellella(17.51%)、Planktosalinus(15.86%)、海杆菌属(Marinobacter,12.3...  相似文献

Bioprinting as a promising but unexplored approach for cartilage tissue engineering has the advantages of high throughput, digital control, and highly accurate placement of cells and biomaterial scaffold to the targeted 3D locations with simultaneous polymerization. This study tested feasibility of using bioprinting for cartilage engineering and examined the influence of cell density, growth, and differentiation factors. Human articular chondrocytes were printed at various densities, stimulated transiently with growth factors and subsequently with chondrogenic factors. Samples were cultured for up to 4 weeks to evaluate cell proliferation and viability, mechanical properties, mass swelling ratio, water content, gene expression, ECM production, DNA content, and histology. Bioprinted samples treated with FGF-2/TGF-β1 had the best chondrogenic properties among all groups apparently due to synergistic stimulation of cell proliferation and chondrogenic phenotype. ECM production per chondrocyte in low cell density was much higher than that in high cell seeding density. This finding was also verified by mechanical testing and histology. In conclusion, cell seeding density that is feasible for bioprinting also appears optimal for human neocartilage formation when combined with appropriate growth and differentiation factors.  相似文献

Abstract Modification of the ‘intracellular concentration of reduced glutathione’ (IC-GSH) affected the response of cultured rose cells (Rosa damascena) to ultraviolet radiation (UV)-induced leakage of K+. High IC-GSH induced by incubation of cells in 10 mol m?3 GSH (IC-GSH increased linearly with time from 20 to about 600 μmol g?1 in 61.2 ks) caused cells to become significantly less sensitive to UV. Low IC-GSH induced by treatment with 1 mol m?3 buthionine sulphoximine (BSO) plus 1 mol m?3 diethylmaleate (DEM) (IC-GSH decreased from 20 to about 3 μg g?1 in 61.2 ks) reduced, rather than increased, the UV-sensitivity of the cells. However, treatment with DEM also induced a large transient K+ leakage; and treatment with BSO induced a slight leakage. The K+ leaked was recovered by 3.24 ks. Following K+ recovery, the DEM-treated cells showed almost complete insensitivity to UV, and BSO-treated cells showed a slightly reduced sensitivity to UV. These results are in agreement with our previous findings that other treatments (heat, cycloheximide, UV), which also cause a transient leakage of K+, also reduce the induction of K+ leakage by a subsequent UV treatment. We conclude that high IC-GSH may play a role in protecting plant cells from UV-induced K+ leakage. Increased UV-sensitivity with low ICGSH was not observed, we believe, because of the transient K+ leakage, though the mechanism of reduced sensitivity to UV induced by transient leakage of K+ is not known at this time. Treatment with UV did not reduce the IC-GSH, showing that this is not the mechanism by which UV induces K+ leakage.  相似文献

Cultures of endothelial cells and cell lines of endothelial origin were maintained at confluence without medium exchange for a period of 72 h. During this time period the concentration of nutrients — amino acids and glucose — and metabolic waste products — lactate and ammonium — was determined as well as cell vitality and cell numbers. Metabolic rates were calculated and compared for the different cell lines. Surprisingly the primary cells showed significantly higher rates of glucose and glutamine consumption, respectively lactate production than the immortalized cell lines. Except for one tumorigenic cell line all cells showed a significant participation of transaminases in glutamine/ammonium metabolism. Summary Four hematopoietic cell lines (CCRF-CEM, HSB-2, MOLT-4, and RPMI-8402), derived from acute lymphoblastic leukemia and expressing T-cell surface markers (T-HCL), were studied with two specific anti-T-cell sera. The sera were raised in rabbits against human thymocytes (anti-HTY) and against T-cells cultured in the presence of conditioned medium derived from lymphocytes stimulated with PHA (anti-CTC). Both sera were absorbed to obtain a T-cell specific pattern of reaction and were further absorbed with normal peripheral blood lymphocytes or with each of the four T-HCL. The anti-HTY sera absorbed with CEM, 8402, and HSB-2 still reacted with MOLT-4. A similar pattern of reactivity was found only with the anti-CTC absorbed with 8402, whereas, after absorptions with the other cell lines, this antiserum was unreactive against MOLT-4. After absorption with normal peripheral blood lymphocytes, anti-HTY still reacted with thymocytes and MOLT-4 but was negative on CTC. In contrast, anti-CTC absorbed with peripheral blood lymphocytes (PBL) was negative on thymocytes and MOLT-4 but still reacted against CTC. Our data confirm the existence of a T-cell antigen (probably an early T-cell differentiation antigen) shared between thymus and MOLT-4. This antigen is not expressed on CTC, although these cells express an antigenic pattern more complex than PBL. Antisera to CTC represents a source of anti-T-cell sera free of contamination with antibodies to early thymus-related antigens but containing other T-cell-related specificities. 通过现场调查和数据分析,首次探讨了中国近海浮游植物的固碳强度与潜力以及近海人工养殖大型藻类的固碳能力.结果表明,渤、黄、东海的浮游植物固碳强度约为2.22亿t a-1,固碳量的季节变化从大到小的顺序依次为春季、夏季、秋季、冬季,其中春夏季的固碳量占全年的65.3%.南黄海1999~2005年10~11月间浮游植物固碳强度有较大的年际变化,10~11月份7a间其浮游植物最低固碳量为3.54万t d-1,最高为16.58万t d-1,平均为10.50万t d-1,没有明显的年际变化趋势,磷对浮游植物固碳强度的影响最为明显,次之的影响因素是Chl a和亚硝氮 (NO2-N)的含量.南海的固碳能力约为渤黄东海的2倍,为4.16亿t a-1,其季节变化和渤黄东海不同,南海浮游植物在冬季的固碳能力最强,在夏季最弱.整个中国近海浮游植物年固碳量达6.38亿t,可占全球近海区域浮游植物年固碳量的5.77%.实际外海龙须菜的养殖发现,龙须菜每年固定的碳为8.18 t,养殖密度与方式对其产量和固碳量影响巨大.近几年,我国大型经济藻类养殖产量每年在120~150万t左右,换算为固碳量为36~45万t,平均每年40万t,如果海藻养殖产量每年增加5%,到2010年,我国大型经济藻类养殖的固碳量可达57万t a-1,海藻养殖是海洋增加碳汇有多重价值的重要措施.  相似文献

培养的卒中型自发性高血压大鼠(SHR_(sp))及其对照 WKY 大鼠主动脉平滑肌细胞(VSMC)上存在心房钠尿肽(ANP)的特异性受体,它们与~(125)I-ANP 的最大结合量(B_(max))是:SHR_(sp)3.65±0.13和 WKY 1.89±0.09 pmol/mg pr(P<0.01);解离平衡常数(Kd)值分别是72.6±10.2和42.0±4.8×10~(-12)mol/L(P<0.01)。 两种细胞内介导舒血管作用的第二信使、环磷酸乌苷(cGMP)的基础浓度无显著差异,对相同剂量 ANP 刺激引起 cGMP 分别增加139(SHRsp)和271(WKY)倍。可见 SHRsp 的 VSMC ANP 受体数量虽比 WKY大鼠增多,但对相同剂量 ANP 引起的 cGMP 增加反应及 ANP 受体的亲和力均显著降低。高盐培养液孵育24h 后,细胞表面 ANP 受体的亲和力改变不明显,但受体数量下调,SHRsp 和 WKY 大鼠分别降至对照的34.8±8.2%和38.6±9.4%,细胞对 ANP 引起的 cGMP增加反应明显降低,且均以 SHR_(sp)较显著。提示后两种变化可能在高盐促进血压升高的机制中起作用。  相似文献
