薏苡 ( Coix lacryma- jobi)授粉后 2 d,游离核胚乳已转变为细胞胚乳。授粉后 3d,中央细胞被胚乳细胞充满。起初 ,全部胚乳细胞均进行分裂 ,一定时期后 ,细胞分裂主要发生在胚乳周边区。授粉后 1 0 d,表皮停止平周分裂变为糊粉层 ,内方的数层形成层状细胞行平周分裂直到颖果接近成熟。胚乳内部生长则依赖于细胞体积扩大。胚乳基部 (颖果基部的胚乳 )形成了数层传递细胞。授粉后 9d,淀粉积累。授粉后 1 0 d,糊粉层及其内方数层细胞产生了脂体 ,后者的脂体以后又消失。授粉后 1 3、1 5 d,糊粉层细胞的液泡积累蛋白质。授粉后 2 0 d,液泡变为糊粉粒。授粉后 1 5 d淀粉胚乳细胞产生蛋白质体 ,营养物质积累持续到颖果成熟。还观察了胚和胚乳发育的对应关系。  相似文献

To survive winters, woody perennials of temperate zones must enter into endodormancy. Resumption of spring growth requires sufficient exposure to low temperature (chill units, CUs) in winter (chilling requirement), which also plays a role in the development of cold hardiness (cold acclimation). Physiological studies on dormancy breaking have focused on identifying markers, such as appearance or disappearance of proteins in response to varying degrees of chill unit accumulation. However, whether these changes are associated with dormancy transitions or cold acclimation is not clear. In the present study, greenhouse-grown blueberry (Vaccinium section Cyanococcus) plants were used to address this question. Three blueberry cultivars, Bluecrop, Tifblue, and Gulfcoast having chilling requirement of approximately 1 200, 900 and 600 CUs, respectively, were first exposed to 4°C for long enough to provide chill units equivalent to one-half of their respective chilling requirement. This treatment was expected to result in cold acclimation. A fraction of plants was then subjected to a 15/12°C (light/dark) regime for 2 weeks, a treatment expected to be “dormancy-neutral” but cause deacclimation. Before and after each treatment, cold hardiness and dormancy status of floral buds were determined; proteins were extracted from the buds collected on the same sampling date, and separated by one-dimensional SDS-PAGE. Dehydrin-like proteins were identified by immunoblotting, using anti-dehydrin antiserum. Results indicate that the chilling treatment resulted in cold acclimation as indicated by increased bud hardiness in all three cultivars. Data also indicate a distinct accumulation of three dehydrin-like proteins of 65, 60, and 14 kDa during cold acclimation. The cold hardiness and levels of dehydrin proteins decreased during the exposure to 15/12°C for 2 weeks. Results also confirmed that this treatment had no negative effect on chill unit accumulation. Densitometric scans of protein gels indicated a close association between the abundance of dehydrins and degree of cold hardiness in these cultivars. In addition, levels of the dehydrin proteins and cold hardiness remained about the same between 100% and >100% satisfaction of chilling requirement. These results suggest that changes in dehydrin expression are more closely associated with cold hardiness than with dormancy transitions.  相似文献

DNA fingerprinting with three repetitive DNA sequences (OPX2, PB6-4 and Sat-121) was carried out on a set of 10 monosomic additions of Beta procumbens and 75 anonymous B. patellaris-derived monosomic additions in B. vulgaris, for characterization of the alien chromosomes at the DNA level. The probes are Procumbentes-specific and distributed over all chromosomes. Morphological characteristics were also used for the classification of B. patellaris monosomic addition families and for comparison with the morphology of the addition families of B. procumbens.DNA fingerprinting revealed unique patterns for almost all individual addition chromosomes of B. procumbens. However, it was concluded that chromosomes 1 and 6 of B. procumbens may be identical with the only difference that the chromosome referred to as 6 carries a susceptible allele for beet cyst nematode (BCN) resistance. In contrast, it was concluded that the two addition types with chromosome 2 are carrying different chromosomes of B. procumbens, so that one of them was renumbered to become the new chromosome 6.DNA fingerprinting of 75 anonymous B. patellaris-derived monosomic additions facilitated the identification and characterization of the alien chromosomes and the grouping of these additions into nine different groups. Several of these groups could be divided in two sub-groups on the basis of small differences in banding patterns. The results of the DNA fingerprinting led to the conclusion that B. patellaris most likely is an allotetraploid. It was also deduced that the BCN gene(s) in this species are homozygous and located on chromosome 1, while the pair of homoeologous chromosomes does not carry such BCN gene(s). Because of the allotetraploid nature of B. patellaris, preferential association occurs between the two homologous chromosomes containing the allele(s) for BCN resistance. Each group of B. patellaris addition families united by DNA fingerprinting had comparable morphological characteristics. Some of these morphological traits appeared to be chromosome-specific and were very useful for primary classification of the addition families. However, the present study showed that these morphological traits are not adequate for the identification of all alien chromosomes without the aid of additional markers. Because of similarities observed between molecular characteristics or the effects on plant morphology of several chromosomes of B. procumbens and B. patellaris it was concluded that B. procumbens could have been involved in the evolutionary history of B. patellaris.  相似文献

Abstract Optimal conditions for studying the elongation response to a 1 mmol m?2, 2-min pulse of red light in subapical coleoptile sections from dark-grown oat (Avena sativa L. ev. Lodi) seedlings have been determined. A technique for obtaining standard-length coleoptile sections without exposing either seedlings or sections to any light has been developed, and is described. The optimal conditions found were: sampling time, 12 h after irradiation; buffer conditions, 5 mol m?3 potassium phosphate with 5% (w/v) sucrose (pH 5.9). The optima were determined by obtaining the time course for light-induced growth under various conditions. The red light-induced growth response is linear until 12 h after irradiation, when it undergoes an interruption. Optimal incubation conditions were determined by varying the buffer contents systematically and measuring the responses at the optimal lime determined. The results indicate a distinct difference between auxin-induced and light-induced growth responses. Even with variations of basal growth rate and several incubation conditions, the red light-induced elongation appears to be of a constant magnitude, to persist for a constant time period. and to exhibit a constant lag period between irradiation and the onset of response. The use of sections that were produced and handled in complete darkness yielded an unusual response to fusicoccin. A linear, high growth rate in response to I mmol m?3 FC was observed for more than 12 h, both in the irradiated sections and in the dark controls.  相似文献

目的:探讨子宫动脉栓塞术在胎盘前置状态中期妊娠引产中的应用。方法:选择2014年1月~2016年1月我院收治的孕14~24周胎盘前置状态需要终止妊娠患者32例,随机分为两组,其中15例先行子宫动脉栓塞术,3小时后给予利凡诺100 mg羊膜腔内注射引产术,联合口服米非司酮150 mg(A组),17例行常规剖宫取胎术(B组),比较两组的出血量及住院时间。结果:A组产后平均出血量为284.53±33.74 m L,平均住院时间为3.8±0.7天;B组平均产后出血量为546.07±40.69 m L,平均住院时间为6.4±0.3天,差异均有统计学意义(P0.05)。结论:子宫动脉栓塞术可用于胎盘前置状态的中孕引产,而且具有出血少、创伤小、可保护女性生殖生理功能的优点,值得临床推广。  相似文献

目的;探讨硬膜外麻醉、蛛网膜下腔-硬膜外麻、气管插管全麻三种不同麻醉方式对剖宫产产妇血流动力学及胎儿的影响。方法:纳入行择期剖宫产产妇152例,采用随机表法将其分为硬膜外组(48例)、腰硬联合麻醉组(52例)、气管内插管全麻组(52例)。观察麻醉前正常状态时(T1)、麻醉成功后5 min(T2)、胎儿娩出时(T3)三个时间点产妇SBP、DBP、HR、SV、CO、SVR及胎儿相关指标,包括出生后1 min、5 min Apgar评分、脐动脉血pH、PCO_2、PO_2。结果:硬膜外麻、腰麻-硬膜外联合麻醉、全麻三组间麻醉起效时间有明显统计学差异(P0.05),气管内插管全麻组起效时间最快。三组间在T1时间点SBP、DBP、HR、SV、CO、SVR差异无统计学意义(P0.05)。在T2时间点,硬膜外麻、联合麻醉、全麻三组SBP、DBP、HR、SVR水平均较T1时间点明显降低,差异有统计学意义(P0.05)。T2时间点,全麻组SBP、DBP、HR、SVR水平高于硬膜外、联合麻醉组(P0.05)。T3时间点三组SBP、DBP、HR、SV、CO水平均较T2时间相比差异有统计学意义(P0.05)。T3时间点,全麻组SBP、DBP、SVR高于其他两组,心率低于其他两组,差异有统计学意义(P0.05)。三组间新生儿出生后1 min Apagr、5 min Apgar评分、脐动脉血学pH、PCO_2无统计学差异(P0.05)。三组间脐动脉血PO_2有明显统计学差异(P0.05),气管内插管全麻组脐动脉血PO_2最高。结论:三种麻醉均可以用于剖宫产术,全麻剖宫产术对产妇血流动力学影响最小,起效快,但对胎儿脐动脉血氧浓度有一定的影响。  相似文献

目的:探讨剖宫产术后产褥感染的相关危险因素,为临床制定感控措施提供参考。方法:回顾性分析我院接受剖宫产术的1760例孕妇的临床资料,采用单因素卡方检验及多因素Logistic回归方法对剖宫产术后发生产褥感染的相关危险因素进行统计学分析。结果:1760例接受剖宫产手术的孕妇,术后发生产褥感染的有68例,发生率为3.86%;发生产褥感染最常见的部位是急性子宫内膜炎、子宫肌炎,其构成比占57.4%;经过单因素卡方检验及多因素Logistic回归分析,最终筛选出手术时间(OR=1.351)、血红蛋白(OR=1.759)、胎膜早破(OR=2.247)、孕期生殖道感染反复发作(OR=2.047)、妊娠糖尿病(OR=1.473)、前置胎盘反复阴道出血(OR=1.584)与宫产术后发生产褥感染呈正相关(P0.05)。结论:手术时间、血红蛋白、胎膜早破、孕期生殖道感染反复发作、妊娠糖尿病、前置胎盘反复阴道出血是剖宫产后发生产褥感染的高危因素,临床应针对这些高危因素制定干预措施以降低感染的发生率。  相似文献

The bacteriophage lambda Xis protein is one of the proteins required for site-specific excisive recombination by which the lambda prophage is excised from the Escherichia coli bacterial chromosome. We cloned the lambda xis gene under the control of several prokaryotic promoters to obtain a sufficient source of the protein for biochemical studies. Our results demonstrate that E. coli lac promoter and lambda pL promoter fusions to the xis gene produce high levels of Xis protein. Induction of the expression vectors results in a 10- to 50-fold increase in Xis activity. In addition, one of these plasmids allows the control of xis expression in vivo.  相似文献
