The ultrastructural localization of B-1,3-glucanase in three species of sea urchin eggs was determined using a monospecific antibody in an electronmicroscopic immunogold procedure. In all three species, Lytechinus variegatus, Strongylocentrotus purpuratus, and Arbacia punctulata, B-1,3-glucanase was localized specifically to the cortical granules. No other organelle within the egg contained significant label. During the fertilization reaction, B-1,3-glucanase was released from cortical granules into the perivitelline space and became associated with the hyaline layer. No significant label was found in association with the fertilization envelope.  相似文献

The ethanol extract of dried flowers Osmanthus fragrans (OFE) was assessed for free radical scavenging effects measured by the bleaching of the 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical, scavenging of the hydroxyl anion, investigation of the ferric reducing/antioxidant power (FRAP) and lipid-peroxidation inhibition in rat tissues. OFE contained a high amount of total flavonoid and polyphenol. OFE presented the effects in the metal reducing power, FRAP assay with IC50 values of 0.23 μg/ml, and 7.74 μg/ml, respectively. OFE presented similar activities toward the DPPH and hydroxyl anion scavenging ability with IC50 values of 10 μg/ml. OFE with IC50 values between 46 and 97 μg/ml inhibited lipid peroxidation initiated by ferrous chloride in rat brain, liver, heart and kidney mitochodrias. Moreover, the neuroprotective activity of OFE was investigated under different insults (glutamate, arachidonic acid, and 6-hydroxydopamine) in Wistar rat primary cortical neurons. OFE with EC50 values between 66 and 165 μg/ml attenuated the neurotoxicity on MTT and LDH assays. In addition, the AKT protein expression of excitotoxicity and oxidative stress was displayed by western blotting analysis. OFE could up-regulate the glutamate and 6-OHDA decreased AKT expression. This is the first demonstration of the neuroprotective, free radical scavenging and anti-oxidative effects of O. fragrans.  相似文献

Human, rat and mouse pituitary tissues have been examined electron microscopically in transmission (TEM), scanning-transmission (STEM) and scanning (SEM) modes for the surface appearance of the secretory granules in tissue sections. Cryofixed and cryosectioned tissue showed only slightly protruding granule profiles which had a smooth surface. Cryofixed, freeze-dried and Epon embedded pituitaries, on the other hand, demonstrated swollen and furrowed surfaces over the granules after contact with water. This topography could also be seen after glutaraldehyde fixation but less after post-fixation in OsO4. The surface alterations in the sections of pituitary secretory granules are thought to be due to differences in the homogeneity of the resin infiltration, leaving resin-free openings where water can enter. It also seems probable that the Epon resin is more influenced by water than has been previously assumed, based on the findings of efficient elimination of osmium from the granules after incubation of tissue sections in water for only 10 min.  相似文献

Morphogenesis of the ciliate cortex has been viewed as an attractive model system for studying the mechanisms behind the ordered assembly of subcellular structure. Based on the assumption that identifying protein components of the cortex would facilitate the study of cortical assembly, I have produced a number of monoclonal antibodies directed against components of the cortex of Euplotes aediculatus. Several of these antibodies react with the proteins comprising the alveolar plates. These thin polygonal scales, each enclosed within a flattened membranous sac (alveolus) just beneath the cell membrane, tightly abut in a confluent monolayer that appears to lend form and rigidity to the Euplotes cell cortex. Reactivity and specificity of these monoclonal antibodies for the alveolar plates was shown by immunofluorescence staining of whole-cell preparations and of cryosections, and by immuno-gold staining of thin sections by electron microscopy. On immunoblots of SDS-PAGE separated whole-cell extracts, the plate proteins are revealed as two to three closely spaced bands centered at an Mr of 97 kDa, and a larger relative at 125 kDa. Comparative peptide mapping reveals that the members of the 97-kDa protein cluster are closely related. However, the 125-kDa polypeptide varies significantly from the 97-kDa members, and hence is not likely a synthetic precursor. Because bands of these Mr values are prominent in Coomassie blue-stained gels of whole-cell extracts, and are greatly enriched in purified cortical preparations, they likely represent the major proteins comprising the alveolar plates of E. aediculatus. I have proposed the name platein for this family of proteins.  相似文献

Using immunofluorescence microscopy, the cortical microtubular net which is regularly present in cells of young, growing tissue is shown to be absent, or largely reduced, in mature mesophyll cells of Nicotiana plumbaginifolia Viv., N. tabacum L., Petunia hybrida Hort. and Brassica napus L. The onset of division in protoplasts isolated from these fully differentiated tissues is preceded by a period of dedifferentiation. One of the early events during dedifferentiation, as shown for N. plumbaginifolia, is the re-establishment of a net of cortical microtubules, prior to spindle formation. These findings indicate that the presence of the cortical microtubular lattice is a prerequisite for protoplast division. Cell-wall regeneration, which also must precede division, occurs simultaneously with the formation of the lattice. However, the cortical microtubules seem to not exert any influence on the orientation of the microfibrils.  相似文献

A diaminobenzidine posttreatment employing osmium tetroxide and potassium fer-rocyanide was successfully used to intensify the diaminobenzidine stain formed by photoconversion of immunofluorescent labelling. Lactoferrin labelled granules became visible following the photoconversion process. Adequate diaminobenzidine staining was obtained after irradiating the cytospin preparations with ultraviolet light for 30-40 min. The diaminobenzidine stain had advantages over the fluorescent stain owing to its greater stability, greater density, and ability to be intensified. The enhancement procedure intensified both the color and density of the diaminobenzidine product. Consequently, shorter irradiation times could be used. Osmium tetrozide solutions of 3-4% increased the intensity with minimal background staining. Ultrastructural immunogold cytochemistry on ultrathin sections confirmed the existence of the lactoferrin labelled structures observed by light microscopy of cytospin preparations indicating that these were secondary grannies. The photomicroscopy procedures used in this study were simple to perform and could be applied to studies of other cellular antigens prior to using Immnnoultramicroscopy.  相似文献

Abstract: The effect of ATP, Mg2+, or MgATP on the release of luteinizing hormone-releasing hormone (LH-RH) from hypothalamic granules was examined under in vitro conditions. Granules, isolated from adult male hypothalami, were incubated at 37°C in a buffered (pH 7.8) medium containing 0.15 m -KCl. The addition of ATP to the incubation mixture did not stimulate the release of LH-RH. In contrast, the addition of MgATP stimulated the release of LH-RH, the release being 62% greater than control. The addition of Mg2+ to the incubated granules also stimulated the release of LH-RH. However, the magnitude of this Mg2+-stimulated release of LH–RH was significantly ( P < 0.01) lower than that of the MgATP-stimulated release, indicating that ATP stimulates LH-RH release in a Mg2+-dependent manner. As both MgATP and Mg2+ alone stimulated LH-RH release, we characterized further these two release processes by incubating the granules under one of the following conditions: incubation at 4°C in a buffered medium containing 0.15 m -KCl or incubation at 37°C in a medium that does not contain KCl. Under these two incubation conditions, the MgATP-stimulated release of LH-RH was not manifested, whereas the Mg2+-stimulated release of LH-RH was manifested. On the basis of these differences, we propose that two different processes can lead to the release of LH-RH from isolated hypothalamic granules: one process involves ATP and Mg2+ (MgATP) and another process involves Mg2+ alone.  相似文献

A range of granular sludges was taken from industrial anaerobic sludge blanket reactors treating a wide variety of wastewaters and a comparison was made between the polymers which were extractable from the granules and their internal structures. The study of the internal structure, using sequential staining of ultra-thin sections, showed the complexity of granular sludges. Much of the area was occupied by Gram-negative cells and the area which stained positive for protein was found to increase nearer the centre of the granules. This was accompanied by a decrease in the carbohydrate positive areas. Positive areas for lipid were widespread throughout the granules. Changes in the internal structure were observed when the type of wastewater treated by the granules was changed and a comparison between sludges treating the same type of wastewater showed that factors other than the nature of the substrate must be considered as parameters which will affect the structure of the granules. Although an appreciable variation in the granule strengths was noted, it was not possible to relate these differences, on an overall basis, to either the internal structure or the chemical composition of the extracted polymers. However, an examination of data for granules produced during the treatment of nominally similar wastes did suggest that there would be a relationship between polymer composition and granule strength in these cases.  相似文献
