Similar to the situation in mammalian cells and yeast, messenger ribonucleo protein (mRNP) homeostasis in plant cells depends on rapid transitions between three functional states, i.e. translated mRNPs in polysomes, stored mRNPs and mRNPs under degradation. Studies in mammalian cells showed that whenever the dynamic exchange of the components between these states is disrupted, stalled mRNPs accumulate in cytoplasmic aggregates, such as stress granules (SGs) or processing bodies (PBs). We identified PBs and SGs in plant cells by detection of DCP1, DCP2 and XRN4, as marker proteins for the 5'-->3' mRNA degradation pathway, and eIF4E, as well as the RNA binding proteins RBP47 and UBP1, as marker proteins for stored mRNPs in SGs. Cycloheximide-inhibited translation, stress treatments and mutants defective in mRNP homeostasis were used to study the dynamic transitions of mRNPs between SGs and PBs. SGs and PBs can be clearly discriminated from the previously described heat stress granules (HSGs), which evidently do not contain mRNPs. Thus, the role of HSGs as putative mRNP storage sites must be revised.  相似文献

以总有机酸和绿原酸的提取率双指标作为提取条件考核对象,来探讨肠安颗粒中所含有机酸成分的中药材的最佳提取工艺.采用L9(34)正交设计,考察提取溶媒所需加水量、浸泡时间、煎煮时间、煎煮次数四个因素,选择进行3个水平试验.得出结论:以加10倍量的水(第一次煎煮多加1.5倍量),浸泡1 h,煎煮2次,每次1h为最佳提取工艺条件,亦即以A2B2C2D2组合为合理.  相似文献

We studied evoked inhibitory postsynaptic currents (eIPSC) using local electrical stimulation of single presynaptic terminals of cultured rat neocortical neurons. According to pharmacological and kinetic properties, these currents were qualified as GABAA-activated. Using autocorrelation analysis of distributions of the eIPSC amplitudes, which were in all cases polymodal, we examined quantal characteristics of the above eIPSC. These results were compared with the values of quantal parameters (N, p, Q, and m) of the current families obtained using approximation by binomial distribution. Amplitude histograms of spontaneous miniature IPSC recorded under conditions of the minimum quantal release of the neurotransmitter were normal (close to Gaussian) with the mode within a 10 pA range, which is very close to analogous parameters calculated using autocorrelation and binomial techniques.  相似文献

Cytotoxic T cells (CTLs) kill target cells by releasing lytic agents via regulated exocytosis. Three signals are known to be required for exocytosis: an increase in intracellular Ca2+, activation of protein kinase C (PKC) and activation of extracellular signal regulated signal kinase (ERK). ERK activation required for exocytosis depends on activity of PKC. The simplest possibility is that the sole effect of PKC required for exocytosis is ERK activation. Testing this requires dissociating ERK and PKC activation. We did this using TCR-independent stimulation of TALL-104 human leukemic CTLs. When cells are stimulated with thapsigargin and PMA, agents that increase intracellular Ca2+ and activate PKC, respectively, PKC-dependent ERK activation is required for lytic granule exocytosis. Expressing a constitutively active mutant MAP kinase kinase activates ERK independent of PKC. However, activating ERK without PKC does not support lytic granule exocytosis, indicating that there are multiple effects of PKC required for granule exocytosis.  相似文献

活体动物全细胞记录技术不仅可以用于研究感觉系统对自然刺激(如视觉系统的光刺激、听觉系统的声音刺激等)反应的特性和规律,还可以较准确地记录细胞的突触电位(包括阈下反应),实现EPSP和IPSP的相对分离,并实现活体细胞内灌流,从而进一步研究感觉信息的处理机制。本文较为详细地介绍了在活体动物上进行全细胞记录的方法,包括一些技术细节和关键仪器设备的选取原则,举例说明了该技术在视觉系统研究和体感系统研究中的应用,并讨论了这一方法在神经科学中的应用前景。  相似文献

用双声刺激和多管电极方法在 6只大棕蝠 (bigbrownbat,Eptesicusfuscus)的 98个神经元上研究了锐化 (sharpening)蝙蝠听皮层 (primaryauditorycortex ,AC)神经元频率调谐的柱特征。结果发现 ,电极直插在 1个电极通道内连续记录到多个神经元时 ,它们锐化频率调谐的抑制性调谐曲线或抑制区基本相似。电极与AC表面呈 45°斜向推入使其跨越多个功能柱时 ,可观察到锐化频率调谐的抑制区构成也随电极进入不同的功能柱而发生相应的改变。两种不同的电极插入方式均证明锐化AC神经元频率调谐的神经抑制呈柱状组构。这些神经元组合起来排列在同一听觉功能柱内 ,构成AC频率分析的基本功能组构单位“微频率处理器”。实验中还观察到多峰频率调谐曲线神经元 ,它们在声通讯和声定位中不同波谱区域的时间匹配中起作用。此外 ,也有理由认为多峰调谐神经元亦被用于作为复杂波谱信息的“高级调谐预处理器” ,从而极大地提高了神经元对频率分析的能力。为研究锐化频率调谐的神经抑制机制 ,用多管电极电泳γ -氨基丁酸 (γ aminobutyricacid ,GA BA)能a受体拮抗剂荷包牡丹碱 (bicuculline ,Bic)至所记录的神经元 ,发现能大部分或几乎全部取消抑制区 ,从而表明在正常情况下GABA能抑制参与构成锐化AC神经元频率调谐的抑制区 ,  相似文献

On cats with pretrigeminal brainstem transection, we studied the properties of visually sensitive neurons of the extrastriate associative cortical area 21b. The dimensions and spatial distribution of the receptive fields (RF) of the neurons within the vision field were determined. It was found that large-sized RF prevailed within the area 21b (10 to 200 deg2, 61%; greater than 200 deg2, 22%), whereas small-sized RF (1 to 10 deg2) constituted 17% of all the studied RF. Stationary visual stimuli evoked onoff, off, and on responses in 43, 30, and 27% neurons of the area 21b, respectively. In the cases where moving stimuli were presented, 35% of the neurons demonstrated directional sensitivity; the rest of the neurons (65%) were directionally insensitive. We also found a group of neurons that were capable of differentiating not only the direction of the stimulus movement along the RF but also the dimension, shape, and orientation of a complicated moving stimulus. Taking into account the data obtained, we discuss the functional role of the neurons, which demonstrated a specific (specialized with respect to a set of the parameters of visual stimulus, and not to a single parameter) response in central processing of the sensory information.  相似文献

Polychlorinated biphenyls (PCBs) can interfere with normal reproductive functions acting as endocrine disruptors. Aroclor-1254 (A-1254), is a pool of more than 60 congeners used for in vitro studies because its composition is representative of PCBs environmental pollution. We previously demonstrated that the exposure of bovine oocytes to A-1254 during in vitro maturation (IVM) was detrimental not only to the maturation process but also induced a significant increase of polyspermy and a reduction of developmental competence. Therefore, we investigated whether A-1254 acts on two processes that occur during IVM and may be related with its negative effects: maternal mRNA polyadenylation and cortical granules (CGs) migration and exocytosis. Bovine cumulus-oocyte complexes (COCs) were exposed to 0.1 microg/ml of A-1254 during IVM, a level of exposure known to affect oocyte maturation, fertilization, and developmental competence. Oocyte exposure to A-1254 altered the poly(A) tail length of 5 out of 10 genes examined. PCBs effect on mRNA polyadenylation was different depending on the gene considered and resulted either in a shorter or in a longer poly(A) tail. At the end of maturation, Aroclor treated oocytes presented clustered CG in a significantly higher percentage than the control group. In addition, CG exocytosis after 8 hr of fertilization occurred at significantly lower extent in zygotes derived from the exposed group compared to control. Our results indicated that the lower developmental competence of oocytes exposed to PCBs during IVM can be related to the interaction of these contaminants with mechanisms regulating maternal mRNA storage in the ooplasm and normal CGs function.  相似文献
