Inheritance traits of a Cry1Ab-resistant strain of the sugarcane borer, Diatraea saccharalis (F.) were analyzed using various genetic crosses. Reciprocal parental crosses between Cry1Ab-susceptible and Cry1Ab-resistant populations, F1 by F1 crosses, and backcrosses of F1 with the Cry1Ab-resistant population were successfully completed. Larval mortality of the parental and cross-populations were assayed on Cry1Ab diet and Bacillus thuringiensis (Bt)-corn leaf tissue. Maternal effects and sex linkage were examined by comparing the larval mortality between the two F1 populations. Dominance levels of resistance were measured by comparing the larval mortality of the Cry1Ab-resistant, -susceptible, and -heterozygous populations. Number of genes associated with the resistance was evaluated by fitting the observed mortality of F2 and backcross populations with a Mendelian monogenic inheritance model. Cry1Ab resistance in D. saccharalis was likely inherited as a single or a few tightly linked autosomal genes. The resistance was incompletely recessive on Bt corn leaf tissue, while the effective dominance levels (DML) of resistance increased as Cry1Ab concentrations decreased with Cry1Ab-treated diet. DML estimated based on larval mortality on intact Bt corn plants reported in a previous study ranged from 0.08 to 0.26. This variability in DML levels of Cry1Ab resistance in D. saccharalis suggests that Bt corn hybrids must express a sufficient dose of Bt proteins to make the resistance genes functionally recessive. Thus, Bt resistant heterozygous individuals can be killed as desired in the “high/dose refuge” resistance management strategy for Bt corn.  相似文献

通过比较观察紫花含笑(♀)和灰岩含笑()及其杂种F1代叶表皮微形态和叶结构发现,紫花含笑和灰岩含笑在叶表皮微形态及叶解剖结构方面有很大差异。杂种F1代个体间在叶表皮微形态及叶解剖结构方面变异很大,为连续的数量遗传;其中57.7%以上的杂种F1代气孔密度高于父母本。杂种F1代矮化型植株叶片气孔密度较小,推测其抗寒性较强,可以通过进一步杂交改良,获得矮化型的盆栽含笑新品种。观察结果可为深入探讨含笑属种间杂种F1代的遗传变异,并从这些杂种F1代中选育观赏新品种积累科学资料。  相似文献

Reproductive features of the valuable medicinal herbs Glycyrrhiza glabra L., G. uralensis Fisch., and G. korshinskyi Grig. were investigated; an assessment was made of the state of the reproductive systems of populations located under introduction conditions in the Central Urals and in the zone contact of the species ranges in the southern cis-Urals. The states of the male and female reproductive systems, seed productivity elements, pollination features, and the viability of seeds produced by different pollination treatments were studied. For the three Glycyrrhiza species, the seed productivity indices were found to decrease considerably starting from the early stages of development of the female reproductive system and continuing to seed maturity. A quantitative analysis of seed productivity elements taking into account the flowering duration of the species showed that the seed production system of the species is very efficient; even in the introduced populations located north of the range, it is able to produce viable seeds in years with favorable growing conditions. Qualitative and quantitative indices of the reproductive system were used to characterize samples from the zone of contact between the ranges. Embryological analysis confirmed the possibility of interspecific hybridization and the prevalence of hybrid forms among the samples. A broad distribution of interspecific hybridization in Glycyrrhiza in various parts of the ranges is assumed. Glycyrrhiza species are characterized by seed and vegetative reproduction. Vegetative reproduction is limited in space but efficiently ensures reproduction of individuals and formation of vegetative clones. Seed reproduction is characterized by indices such as amphimictic seed formation, strict entomophily, and interspecific compatibility. Seed reproduction is a factor that controls the possibility of wide propagation of the genetic material in space and, hence, the formation of mixed and hybrid populations leading to a higher intraspecific and interspecific diversity of the genus.  相似文献

The focus of this study was to alter the xylan content of corn stover and poplar using SO2‐catalyzed steam pretreatment to determine the effect on subsequent hydrolysis by commercial cellulase preparations supplemented with or without xylanases. Steam pretreated solids with xylan contents ranging from ~1 to 19% (w/w) were produced. Higher xylan contents and improved hemicellulose recoveries were obtained with solids pretreated at lower severities or without SO2‐addition prior to pretreatment. The pretreated solids with low xylan content (<4% (w/w)) were characterized by fast and complete cellulose to glucose conversion when utilizing cellulases. Commercial cellulases required xylanase supplementation for effective hydrolysis of pretreated substrates containing higher amounts of xylan. It was apparent that the xylan content influenced both the enzyme requirements for hydrolysis and the recovery of sugars during the pretreatment process. © 2009 American Institute of Chemical Engineers Biotechnol. Enzymatic hydrolysis of hybrid poplar treated by ammonia recycle percolation (ARP) was studied applying cellulase enzyme supplemented with additional xylanase or pectinase. The effect of xylanase addition was much more significant than pectinase addition. Conversion of ARP‐treated hybrid poplar to ethanol was carried out by simultaneous saccharification and fermentation (SSF) and SS and cofermentation (SSCF). The maximum ethanol yield observed from the SSCF experiment was 78% of theoretical maximum based on the total carbohydrate (glucan + xylan). The same feedstock was also treated by soaking in aqueous ammonia (SAA), a batch pretreatment process with lower severity than ARP. The test results indicated that relatively high severity is required to attain acceptable level of digestibility of hybrid poplar. In order to lower the severity of the pretreatment, addition of H2O2 was attempted in the SAA. Addition of H2O2 significantly enhanced delignification of hybrid poplar due to its oxidative degradation of lignin. Several different H2O2 feeding schemes and different temperature profiles were attempted in operation of the SAA to investigate the effects of H2O2 on degradation of lignin and carbohydrates in hybrid poplar. More than 60% of lignin in hybrid poplar was removed with stepwise‐increase of temperature (60–120°C after 4h of reaction). Selective 2′-hydroxyl acylation analyzed by primer extension (SHAPE) has gained popularity as a facile method of examining RNA structure both in vitro and in vivo, exploiting accessibility of the ribose 2′-OH to acylation by N-methylisatoic anhydride (NMIA) in unpaired or flexible configurations. Subsequent primer extension terminates at the site of chemical modification, and these products are fractionated by high-resolution gel electrophoresis. When applying SHAPE to investigate structural features associated with the wild-type and analog-substituted polypurine tract (PPT)–containing RNA/DNA hybrids, their size (20–25 base pairs) rendered primer extension impractical. As an alternative method of detection, we reasoned that chemical modification could be combined with tandem mass spectrometry, relying on the mass increment of RNA fragments containing the NMIA adduct (Mr = 133 Da). Using this approach, we demonstrate both specific modification of the HIV-1 PPT RNA primer and variations in its acylation pattern induced by replacing template nucleotides with a non-hydrogen-bonding thymine isostere. Our selective 2′-hydroxyl acylation analyzed by mass spectrometry strategy (SHAMS) should find utility when examining the structure of small RNA fragments or RNA/DNA hybrids where primer extension cannot be performed.  相似文献

pUL23是人巨细胞病毒(HCMV)UL23基因编码的皮层蛋白. HCMV皮层蛋白与病毒颗粒的形成、病毒转移、免疫调控等病毒生活过程相关.利用GAL4 酵母双杂交系统筛选人胚肾cDNA文库,获得与人巨细胞病毒皮层蛋白pUL23相互作用的宿主蛋白分子DNAJB6 [DnaJ (Hsp40) homolog, subfamily B, member 6].回复酵母双杂交、体外GST-Pull down和免疫共沉淀试验再次确认两者之间的相互作用.该结果为进一步研究pUL23蛋白在HCMV生活周期中的作用机制提供依据.  相似文献

  • 1 Warren root collar weevil Hylobius warreni Wood (Coleoptera: Curculionidae) is a long‐lived, flightless insect native to coniferous forests across northern North America. Girdling by larval feeding causes significant mortality on young trees. The insect poses considerable challenges to reforestation.
  • 2 Adult weevils feed on all life stages of a variety of coniferous hosts prior to oviposition. Their relative feeding preferences, however, have not been quantified. Moreover, it is not known whether host bark influences oviposition behaviour.
  • 3 Feeding preferences of adult weevils were tested in both choice and no‐choice laboratory bioassays using small branches from three conifers (lodgepole pine Pinus contorta var. latifolia, interior hybrid spruce Picea glauca×engelmannii, and Douglas‐fir Pseudotsuga menziesii) and one deciduous tree (trembling aspen Populus tremuloides). Measurements included the surface area of bark consumed, rate of consumption, the number of days of feeding, and, in the no‐choice assay, the number of eggs oviposited.
  • 4 Bark consumption was greatest on pine and Douglas‐fir, followed by spruce. Little to no feeding occurred on aspen. Consumption did not vary between male versus female insects for any of the feeding metrics quantified.
  • 5 The presence of aspen branches did not inhibit feeding on any of the other species in the choice bioassays.
  • 6 The number of eggs laid by female insects did not differ significantly among tree species in the no‐choice assay. Eggs were laid indiscriminately in the presence of all four host types.
  • 7 Results and opportunities for future research are discussed in the context of formulating new integrated pest management strategies for this insect, which is increasingly important in the period of reforestation subsequent to the mountain pine beetle epidemic in western Canada.

An emerging research area in computational biology and biotechnology is devoted to mathematical modeling and prediction of gene‐expression patterns; it nowadays requests mathematics to deeply understand its foundations. This article surveys data mining and machine learning methods for an analysis of complex systems in computational biology. It mathematically deepens recent advances in modeling and prediction by rigorously introducing the environment and aspects of errors and uncertainty into the genetic context within the framework of matrix and interval arithmetics. Given the data from DNA microarray experiments and environmental measurements, we extract nonlinear ordinary differential equations which contain parameters that are to be determined. This is done by a generalized Chebychev approximation and generalized semi‐infinite optimization. Then, time‐discretized dynamical systems are studied. By a combinatorial algorithm which constructs and follows polyhedra sequences, the region of parametric stability is detected. In addition, we analyze the topological landscape of gene‐environment networks in terms of structural stability. As a second strategy, we will review recent model selection and kernel learning methods for binary classification which can be used to classify microarray data for cancerous cells or for discrimination of other kind of diseases. This review is practically motivated and theoretically elaborated; it is devoted to a contribution to better health care, progress in medicine, a better education, and more healthy living conditions. 用部分改造的 BtCry1Ac基因与慈菇蛋白酶抑制剂(API-A)基因构建的双抗虫基因表达载体,通过农杆菌介导法对三倍体毛白杨进行了转化,对转化后植株体内残存农杆菌在继代培养和移栽过程中进行了跟踪检测。结果表明:通过对转化再生植株的分子生物学检测,42个株系中,33个株系为阳性,阳性率达到80%;用Bt毒蛋白抗血清进行ELSA检测结果表明,7个转基因株系都有Bt杀虫蛋白表达;基因转化后,可采用附加50 mg/L卡那霉素,300 mg/L羧青霉素的筛选培养基消除细菌并进行抗性芽筛选。对28个转基因株系叶片、茎段和根段在含有卡那霉素50 mg/L YEB培养基上进行细菌培养,通过在T-DNA区、质粒Vir区和农杆菌基因组设计引物,进行PCR检测,证明有3个株系(33、37、5号)检测到残存工程农杆菌,并在组培瓶中存活24个月。将带菌的3个株系组培苗移栽到花盆中,室内培养1个月后,在33号株系根际土壤中检测到了目的农杆菌。  相似文献
