This research adds to the limited data on coarse and fine root biomass for blue oak (Quercus douglasii Hook and Arn.), a California deciduous oak species found extensively throughout the interior foothills surrounding the Central Valley. Root systems of six blue oak trees were analyzed using three methods — backhoe excavation, quantitative pits, and soil cores. Coarse root biomass ranged from 7 to 177 kg per tree. Rooting depth for the main root system ranged from 0.5 to 1.5 m, with an average of 70% of excavated root biomass located above 0.5 m. Of the total biomass in excavated central root systems, primary roots (including burls) accounted for 56% and large lateral roots (> 20 mm diameter) accounted for 36%. Data from cores indicated that most biomass outside of the root crown was located in fine roots and that fine root biomass decreased with depth. At surface depths (0–20 cm), small-fine (< 0.5 mm diameter) roots accounted for 71%, large-fine (0.5–2.0 mm) for 25%, and coarse (> 2 mm) for 4% of total root biomass collected with cores. Mean fine root biomass density in the top 50 cm was 0.43 kg m−3. Fine root biomass did not change with increasing distance from the trees (up to approximately 5 m). Thus, fine roots were not concentrated under the tree canopies. Our results emphasize the importance of the smallest size class of roots (<0.5 mm), which had both higher N concentration and, in the area outside the central root system, greater biomass than large fine (0.5–2.0 mm) or coarse (> 2.0 mm) roots. 采用金属螯合亲和层析法,纯化了小鼠腹水来源的抗乙肝核心抗原单克隆抗体,对上样缓冲液的pH和离子强度、洗脱液种类和洗脱方式进行优化。结果表明,采用降低pH分步洗脱时,最佳上样缓冲液为pH8.0,20mmol/LPB+0.5mol/LNaCl,抗体在pH5.0被洗脱下来,抗体回收率80%,纯度85%。采用咪唑浓度梯度洗脱时,最佳的上样缓冲液为pH8.0,20mmol/LPB+5mmol/L咪唑,抗体纯度大于95%,回收率65%;在上样缓冲液中不添加NaCl而添加少量的咪唑,更有利于抗体分离。以上洗脱方式都能较好地保持mAb的生物学活性,为该抗体的应用提供了必要的实验基础。  相似文献

利用SSR分子标记技术,对中国不同生态棉区曾经或正在种植的主要来源于岱字棉、斯字棉、福字棉、乌干达棉的30个陆地棉主栽品种进行了DNA指纹分析。从1 803对SSR引物中筛选到重复性好、多态性丰富的20对核心引物。这些引物分属棉花15条染色体,共检测到116个等位基因,平均每对引物5.8个;PIC值范围为0.384~0.900,平均为0.716;MI值范围为1.152~9.000,平均为4.374。30个品种中有4个品种具有各自的特异引物,可将其与其它26个品种区分开,其它26个品种可利用至少2对引物组合进行区分。为更方便、准确地鉴定各品种,构建了30个品种20对核心引物的十进制数字指纹代码。该研究为陆地棉的品种鉴定和纯度检测、新品种权益保护以及标准DNA指纹库构建提供了重要依据。  相似文献

The effects of climate change are difficult to predict for many marine species because little is known of their response to climate variations in the past. However, long‐term chronologies of growth, a variable that integrates multiple physical and biological factors, are now available for several marine taxa. These allow us to search for climate‐driven synchrony in growth across multiple taxa and ecosystems, identifying the key processes driving biological responses at very large spatial scales. We hypothesized that in northwest (NW) Australia, a region that is predicted to be strongly influenced by climate change, the El Niño Southern Oscillation (ENSO) phenomenon would be an important factor influencing the growth patterns of organisms in both marine and terrestrial environments. To test this idea, we analyzed existing growth chronologies of the marine fish Lutjanus argentimaculatus, the coral Porites spp. and the tree Callitris columellaris and developed a new chronology for another marine fish, Lethrinus nebulosus. Principal components analysis and linear model selection showed evidence of ENSO‐driven synchrony in growth among all four taxa at interannual time scales, the first such result for the Southern Hemisphere. Rainfall, sea surface temperatures, and sea surface salinities, which are linked to the ENSO system, influenced the annual growth of fishes, trees, and corals. All four taxa had negative relationships with the Niño‐4 index (a measure of ENSO status), with positive growth patterns occurring during strong La Niña years. This finding implies that future changes in the strength and frequency of ENSO events are likely to have major consequences for both marine and terrestrial taxa. Strong similarities in the growth patterns of fish and trees offer the possibility of using tree‐ring chronologies, which span longer time periods than those of fish, to aid understanding of both historical and future responses of fish populations to climate variation.  相似文献

In an attempt to overcome the problems associated with LiNiO2, the solid solution series of lithium nickel‐metal oxides, Li[Ni1–xMx]O2 (with M = Co, Mn, Al, Ti, Mg, etc.), have been investigated as favorable cathode materials for high‐energy and high‐power lithium‐ion batteries. However, along with the improvement in the electrochemical properties in Ni‐based cathode materials, the thermal stability has been a great concern, and thus violent reaction of the cathode with the electrolyte needs to be avoided. Here, we report a heterostructured Li[Ni0.54Co0.12Mn0.34]O2 cathode material which possesses both high energy and safety. The core of the particle is Li[Ni0.54Co0.12Mn0.34]O2 with a layered phase (R3‐m) and the shell, with a thickness of < 0.5 μm, is a highly stable Li1+x[CoNixMn2–x]2O4 spinel phase (Fd‐3m). The material demonstrates reversible capacity of 200 mAhg‐1 and retains 95% capacity retention under the most severe test condition of 60 °C. In addition, the amount of oxygen evolution from the lattice in the cathode with two heterostructures is reduced by 70%, compared to the reference sample. All these results suggest that the bulk Li[Ni0.54Co0.12Mn0.34]O2 consisting of two heterostructures satisfy the requirements for hybrid electric vehicles, power tools, and mobile electronics.  相似文献

We cultured avian pox (Avipoxvirus spp.) from lesions collected on Hawai‘i, Maui, Moloka‘i, and ‘Oahu in the Hawaiian Islands from 15 native or non-native birds representing three avian orders. Phylogenetic analysis of a 538 bp fragment of the gene encoding the virus 4b core polypeptide revealed two distinct variant clusters, with sequences from chickens (fowlpox) forming a third distinct basal cluster. Pox isolates from one of these two clusters appear closely related to canarypox and other passerine pox viruses, while the second appears more specific to Hawai‘i. There was no evidence that birds were infected simultaneously with multiple pox virus variants based on evaluation of multiples clones from four individuals. No obvious temporal or geographic associations were observed and strict host specificity was not apparent among the 4b-defined field isolates. We amplified a 116 bp 4b core protein gene fragment from an ‘Elepaio (Chasiempis sandwichensis) collected in 1900 on Hawai‘i Island that clustered closely with the second of the two variants, suggesting that this variant has been in Hawai‘i for at least 100 years. The high variation detected between the three 4b clusters provides evidence for multiple, likely independent introductions, and does not support the hypothesis of infection of native species through introduction of infected fowl. Preliminary experimental infections in native Hawai‘i ‘Amakihi (Hemignathus virens) suggest that the 4b-defined variants may be biologically distinct, with one variant appearing more virulent. These pox viruses may interact with avian malaria (Plasmodium relictum), another introduced pathogen in Hawaiian forest bird populations, through modulation of host immune responses.  相似文献

Preliminary dark incubation of etiolated pea and maize plants at 38 °C allowed to observe a new dark reaction of Chl biosynthesis occuring after photoconversion of protochlorophyllide Pchld 655/650 into chlorophyllide Chld 684/676. This reaction was accompanied by chlorophyllide esterification and by the bathochromic shift of pigment spectra: Chld 684/676 Chl 688/680. After completion of the reaction, a rapid (20–30 s at 26 °C) quenching of Chl 688/680 low-temperature fluorescence was observed. The reaction Chld 684/676 Chl 688/680 was inhibited under anaerobic conditions as well as in the presence of KCN; the reaction accompanied by Chl fluorescence quenching was inhibited in the leaves of pea mutants with impaired function of Photosystem II reaction centers. The spectra position of newly formed Chl, effects of Chl fluorescence quenching allowed to assume that the new dark reaction is responsible for biosynthesis of P–680, the key pigment of Photosystem II reaction centres.  相似文献

本文比较研究了等渗NaCl和KCl胁迫下,高粱幼苗生长及叶片离子含量、质膜相对透性和有关气体交换参数的变化。结果表明,在低浓度NaCl和KCl胁迫7天时,高粱生长、含水量和质膜相对透性与对照相比没有明显变化,而净光合速率、蒸腾速率和气孔导度已明显下降,叶肉细胞间隙CO2浓度明显增加。NaCl胁迫下叶片Na+含量成倍增加,而K+和Ca2+含量无明显变化。KCl胁迫时叶片K+含量明显增加,Ca2+含量明显下降,而Na+含量没有明显变化。随着NaCl或KCl浓度的增加,幼苗生长和叶片含水量明显下降,质膜透性和细胞间隙CO2浓度明显增加,净光合速率、蒸腾速率和气孔导度进一步下降。NaCl胁迫下叶片Na+含量进一步增加,K+和Ca2+进一步下降,而KCl胁迫下叶片K+含量进一步增加,Na+和Ca2+含量进一步下降。KCl对高粱生长抑制、质膜透性、Ca2+含量下降及光合气体交换参数的影响均明显大于等渗的NaCl。  相似文献

Ca2+ ions play a pivotal role in a wide array of cellular processes ranging from fertilization to cell death. In skeletal muscle, a mechanical interaction between plasma membrane dihydropyridine receptors (DHPRs, L-type Ca2+ channels) and Ca2+ release channels (ryanodine receptors, RyR1s) of the sarcoplasmic reticulum orchestrates a complex, bi-directional Ca2+ signaling process that converts electrical impulses in the sarcolemma into myoplasmic Ca2+ transients during excitation-contraction coupling. Mutations in the genes that encode the two proteins that coordinate this electrochemical conversion process (the DHPR and RyR1) result in a variety of skeletal muscle disorders including malignant hyperthermia (MH), central core disease (CCD), multiminicore disease, nemaline rod myopathy, and hypokalemic periodic paralysis. Although RyR1 and DHPR disease mutations are thought to alter excitability and Ca2+ homeostasis in skeletal muscle, only recently has research begun to probe the molecular mechanisms by which these genetic defects lead to distinct clinical and histopathological manifestations. This review focuses on recent advances in determining the impact of MH and CCD mutations in RyR1 on muscle Ca2+ signaling and how these effects contribute to disease-specific aspects of these disorders.  相似文献
