Summary Conditions for the primary culture of branching scleractinian coral (Acropora micropthalma and Pocillopora damicornis) cells were established with a calcium-free seawater cell dissociation method. Cells were isolated and cultured in supple-mented Dulbecco’s modified Eagle media with heat-inactivated fetal bovine serum, antibiotics, and sterile seawater. Among the isolated cell types, large (60–100 μm) multicellular endothelial isolates (MEIs) were seen in high numbers. These isolates were observed to continually spin for up to 300 h without media change. The following parameters were optimized: media, serum, light, trace elements, and growth factor supplements. Rotations per minute were calculated to determine MEI motility in relation to size. Finally, analyses of external and internal structures were conducted with scanning electron microscopy, transmission electron microscopy, and fluorescence microscopy. Additional coral species, Montipora digitata, Stylophora pistillata, Seriatopora hystrix and Porites sp. were also cultured to determine the applicability of isolation techniques. The relatively long survival time of MEIs in primary culture makes them ideal candidates for in vitro studies examining coral disease processes (e.g., mode of infection and intracellular effects of disease-causing agents) as well as aspects of general coral growth and health (e.g., trace element requirements and transfer of products between host cell and zooxanthellae).  相似文献

Both bacteria and algal symbionts (genus Symbiodinium), the two major microbial partners in the coral holobiont, respond to fluctuations in the environment, according to current reports; however, little evidence yet indicates that both populations have any direct interaction with each other in seasonal fluctuation. In this study, we present field observations of a compositional change in bacteria and Symbiodinium in the coral Isopora palifera in three separate coral colonies following monthly sampling from February to November in 2008. Using massively parallel pyrosequencing, over 200 000 bacterial V6 sequences were classified to build the bacterial community profile; in addition, the relative composition and quantity of Symbiodinium clades C and D were determined by real-time PCR. The results showed that coral-associated bacterial and Symbiodinium communities were highly dynamic and dissimilar among the tagged coral colonies, suggesting that the effect of host specificity was insignificant. The coral-associated bacterial community was more diverse (Shannon index up to 6.71) than previous estimates in other corals and showed rapid seasonal changes. The population ratios between clade C and D groups of Symbiodinium varied in the tagged coral colonies through the different seasons; clade D dominated in most of the samples. Although significant association between bacteria and symbiont was not detected, this study presents a more detailed picture of changes in these two major microbial associates of the coral at the same time, using the latest molecular approaches.  相似文献

The question of the age of «Hauts de Meuse coral rag is studied again using new faunas of brachiopods and ammonites collected above, below and inside the coral rag. Ammonites fauna of the well known «Calcaires de Creuë is revised. Results obtained agree closely.The «Hauts de Meuse coral rag grew up during Middle Oxfordian time; and extend part of the Plicatilis zone (Antecedens Subzone) and the whole Transversarium Zone up to the basis of the Bifurcatus Zone, for the first time identified in Lorraine. So it span a short depositional time, about 1 MY, with a high deposition rate, the thickness of the coral rag being more than 100 meters, reaching 150 meters in Saint-Mihiel area. Towards SE, on the other side of the Marne Gap, the sudden change into basinal facies go with a reducing of thickness and deposition rate.Upper Oxfordian deposits show a relative uniformity of facies and thickness. At the top of Middle Oxfordian limestones, the age of the discontinuity and the Sequanian renewal of deposition is basal Bifurcatus Zone. Erosional renewal and terrigeneous deposits are a wide spread event by the beginning of Upper Oxfordian.In the paleontological part, species upon which are based new ages and interpretations are only figured, except the species new or with new paleontological data, which are described. Aim To determine the applicability of biogeographical and ecological theory to marine species at two remote island locations. This study examines how biogeography, isolation and species geographic range size influence patterns of species richness, endemism, species composition and the abundance of coral reef fishes. Location Christmas Island and the Cocos (Keeling) Islands in the tropical eastern Indian Ocean. Methods Published species lists and underwater visual surveys were used to determine species richness, endemism, species composition and abundance of reef fishes at the islands. These data were statistically compared with patterns of species composition and abundance from the neighbouring ‘mainland’ Indonesian region. Results The two isolated reef fish communities were species‐poor and contained a distinct taxonomic composition with an overrepresentation of species with high dispersal potential. Despite low species richness, we found no evidence of density compensation, with population densities on the islands similar to those of species‐rich mainland assemblages. The mix of Indian and Pacific Ocean species and the proportional representations of the various regional faunas in the assemblages were not influenced by the relative proximity of the islands to different biogeographical provinces. Moreover, species at the edge of their range did not have a lower abundance than species at the centre of their range, and endemic species had substantially higher abundances than widespread species. At both locations, endemism was low (less than 1.2% of the community); this may be because the locations are not sufficiently isolated or old enough to promote the evolution of endemic species. Main conclusions The patterns observed generally conform to terrestrial biogeographical theory, suggesting that similar processes may be influencing species richness and community composition in reef fish communities at these remote islands. However, species abundances differed from typical terrestrial patterns, and this may be because of the life history of reef fishes and the processes maintaining isolated populations.  相似文献

Various approaches to coral restoration have been developed to help increase rate of reef recovery from perturbations, among the most common of which is coral transplantation. Success is often evaluated based on short‐term observations that capture only the initial phase of space colonization by coral transplants. Here, an individual‐based model is developed to quantify uncertainty in future trajectories in experimental plots given past observations. Empirical data were used to estimate probabilistic growth, survival, and fission rates of Acropora pulchra and A. intermedia (order Scleractinia) in a sandy reef flat (Bolinao, Philippines). Simulations were initialized with different densities (25 or 50 transplants per species per 16 m2) to forecast possible coral cover trajectories over a 5‐year period. Given current conditions, there is risk of local extinction which is higher in low‐density plots for both species, and higher for A. intermedia compared to A. pulchra regardless of density. While total coral cover is projected to increase, species composition in the future is more likely to be highly uneven. The model was used to quantify effect on recovery rate of protection from pulse anthropogenic disturbances, given different initial transplantation densities. When monitoring data are limited in time, stochastic models may be used to assess whether the restoration trajectory is heading toward the desired state and at what rate, and foresee system response to various adaptive interventions.  相似文献

Recruitment limitation models of coral reef fishes assume that interactions between demersal individuals and settling larvae are of little consequence to local population dynamics, as are interactions among reef residents. To test this premise, I examined the effects of interactions on the settlement and persistence of the ocean surgeonfish, Acanthurus bahianus. I also looked at behavioural exchanges among ocean surgeonfish of different sizes and between ocean surgeonfish and other reef residents to determine whether the nature of these exchanges changes ontogenetically. Settlement of ocean surgeonfish to Tague Bay Reef, St. Croix, U.S.V.I. was increased in the presence of conspecifics and decreased in the presence of a common Caribbean damselfish, the beaugregory Stegastes leucostictus. Additionally, post-settlement persistence was negatively related to the level of aggression received from beaugregories. Interactions among ocean surgeonfish and between ocean surgeonfish and damselfish were size-dependent; the largest and smallest size classes had higher levels of association with conspecifics than did the middle size classes, and larger size classes received more aggression from damselfish than did the smaller size classes. These results suggest that behavioural interactions may significantly affect the distribution, abundance and early post-settlement persistence of ocean surgeonfish settlers, and that the nature of these interactions changes over the lifetime of individuals.  相似文献
