The effect of reduced balanced protein (RP) diet in the F0 and F1 generation of broiler breeders on the learning ability and memory retention of the F2 generation was investigated by means of a reward v. no reward discrimination T-maze test. There were two treatments for the F0 generation: control (C) group, reared on standard commercial diets, and reduced balanced protein (RP) group, fed with RP diets (25% reduction in CP and amino acids). The female F0-progeny of each treatment was again separated into the two dietary treatments, resulting in four treatments for the F1 generation: C/C, C/RP, RP/C and RP/RP (breeder feed in F0/F1 generation). The RP diets fed breeders received on average 10% more feed than C diets fed breeders to achieve a similar target BW. The F2 generation was composed of four treatments coming from the female F1-progeny of the four treatments and were all fed with C diet (namely C/C/C, C/RP/C, RP/C/C and RP/RP/C). All four F2 generation groups were able to complete the T-maze learning test with a slight difference in success rate but a significant difference within groups was observed regarding the time needed to complete the test. In general, the RP/RP/C group needed more time for completing the test compared with the other three groups and the shortest time was recorded for the RP/C/C group. At similar ages, breeders with early learning experience spent significantly less time in completing the test compared with unexperienced breeders. Long-term memory retention was observed in all four groups whereas the learning ability in solving the test decreased with age. It took longer for the breeders to complete the test at older ages. In conclusion, under our experimental conditions, the RP dietary treatment in previous generations had no influence on the T-maze learning ability and memory retention of broiler breeders of the third generation, although it might have effects on the working performance in the T-maze learning test of F2 generation breeders. 相似文献

To describe the effects of sodium-glucose co-transporter 2 (SGLT2) inhibitors on serum uric acid (SUA) in patients with type 2 diabetes mellitus (T2DM). PubMed, EMBASE, and CENTRAL were searched for randomized controlled trials of SGLT2 inhibitors in patients with T2DM up to Aug 10, 2017, without language or date restrictions. Thirty-one studies totaling 13,650 patients were included. SGLT2 inhibitors significantly decreased SUA levels compared with placebo, canagliflozin WMD –37.02?μmol/L, 95% CI [–38.41, –35.63], dapagliflozin WMD –38.05?μmol/L, 95% CI [–44.47, –31.62], empagliflozin WMD –42.07?μmol/L, 95% CI [–46.27, –37.86]. The drug class effect of SUA reduction suggesting SGLT2 inhibitors might be beneficial for diabetic patients with hyperuricemia. 相似文献

The production and use of multi-modal imaging agents is on the rise. The vast majority of these imaging agents are limited to a single length scale for the agent (e.g. tissues only), which is typically at the organ or tissue scale. This work explores the synthesis of such an imaging agent and discusses the applications of our vitamin E-inspired multi-modal and multi-length scale imaging agents TB-Toc ((S,E)-5,5-difluoro-7-(2-(5-((6-hydroxy-2,5,7,8-tetramethylchroman-2-yl) methyl) thiophen-2-yl) vinyl)-9-methyl-5H-dipyrrolo-[1,2-c:2’,1’-f][1,3,2]diazaborinin-4-ium-5-uide). We investigate the toxicity of TB-Toc along with the starting materials and lipid based delivery vehicle in mouse myoblasts and fibroblasts. Further we investigate the uptake of TB-Toc delivered to cultured cells in both solvent and liposomes. TB-Toc has low toxicity, and no change in cell viability was observed up to concentrations of 10?mM. TB-Toc shows time-dependent cellular uptake that is complete in about 30?min. This work is the first step in demonstrating our vitamin E derivatives are viable multi-modal and length scale diagnostic tools. 相似文献

Prostate cancer (PC) is the second most commonly occurring cancer in men. Conventional chemotherapy has wide variety of disadvantages such as high systemic toxicity and low selectivity. Targeted drug delivery is a promising approach to decrease side effects of therapy. Prostate specific membrane antigen (PSMA) is overexpressed in prostate cancer cells while low level of expression is observed in normal cells. In this study we describe the development of Glu-urea-Lys based PSMA-targeting conjugates with paclitaxel. A series of new PSMA targeting conjugates with paclitaxel was designed and synthesized. The cytotoxicity of conjugates was evaluated against prostate (LNCaP, 22Rv1 and PC-3) and non-prostate (Hek293T, VA13, A549 and MCF-7) cell lines. The most promising conjugate 21 was examined in vivo using 22Rv1 xenograft mice model. It demonstrated good efficiency comparable with paclitaxel, while reduced toxicity. 3D molecular docking study was also performed to understand underlying mechanism of binding and further optimization of the linker substructure and conjugates structure for improving the target affinity. We synthesized a pair of compounds containing leucine zipper peptides to deliver protein cargo into cells. One is a cell-penetrating peptide (CPP) with Lz(E), a leucine zipper peptide containing negatively charged amino acids, and the other is a Nanog protein with Lz(K), a leucine zipper peptide containing positively charged amino acids. When cells were treated with these equimolar mixtures, Nanog-Lz(K) hybridized with Lz(E)-CPP was successfully delivered into the cells. Recombinant BK virus (rBKV) is able to express polypeptides under control of its native BKV late promoter. This ability helps to use this construct as a good reporter since it can infect human cells. In this study, we generate a BKV construct containing Renilla luciferase (Rluc) sequences under control of the BKV late promoter. The activity of Rluc was strongly detected in Vero-76 and Cos-1 cells transfected with rBKV-Rluc-myc-2A-VP2 construct, indicating the production of a functional enzyme driven by the native late promoter. Furthermore, a construct made of rBKV-IL2SP-Rluc-myc-2A-VP2 by introducing human IL2 secretion peptide (IL2 SP) caused secretion of IL2SP-Rluc-myc into the culture medium. As a concluding remark, a potential infectious rBKV that can express foreign antigens such as Rluc was generated successfully. The proposed strategy would be useful to engineer recombinant forms of rBKV with many potential applications including development of antiviral assay for new drugs, human vaccines and gene delivery systems for immunotherapeutic or cell transduction. 相似文献

Micelles have demonstrated an excellent ability to deliver several different types of therapeutic agents, including chemotherapy drugs, proteins, small‐interfering RNA and DNA, into tumor cells. Cationic micelles, comprising self‐assemblies of amphiphilic cationic polymers, have exhibited tremendous promise with respect to the delivery of therapy genes and gene transfection. To date, research in the field has focused on achieving an enhanced stability of the micellar assembly, prolonged circulation times and controlled release of the gene. This review focuses on the micelles as a nanosized carrier system for gene delivery, the system‐related modifications for cytoplasm release, stability and biocompatibility, and clinic trials. In accordance with the development of synthetic chemistry and self‐assembly technology, the structures and functionalities of micelles can be precisely controlled, and hence the synthetic micelles not only efficiently condense DNA, but also facilitate DNA endocytosis, endosomal escape, DNA uptake and nuclear transport, resulting in a comparable gene transfection of virus. 相似文献

Plant RNA virus-based guide RNA (gRNA) delivery has substantial advantages compared to that of the conventional constitutive promoter-driven expression due to the rapid and robust amplification of gRNAs during virus replication and movement. To date, virus-induced genome editing tools have not been developed for wheat and maize. In this study, we engineered a barley stripe mosaic virus (BSMV)-based gRNA delivery system for clustered regularly interspaced short palindromic repeat (CRISPR)/Cas9-mediated targeted mutagenesis in wheat and maize. BSMV-based delivery of single gRNAs for targeted mutagenesis was first validated in Nicotiana benthamiana. To extend this work, we transformed wheat and maize with the Cas9 nuclease gene and selected the wheat TaGASR7 and maize ZmTMS5 genes as targets to assess the feasibility and efficiency of BSMV-mediated mutagenesis. Positive targeted mutagenesis of the TaGASR7 and ZmTMS5 genes was achieved for wheat and maize with efficiencies of up to 78% and 48%. Our results provide a useful tool for fast and efficient delivery of gRNAs into economically important crops. 相似文献
