Functional tool use requires the selection of appropriate raw materials. New Caledonian crows Corvus moneduloides are known for their extraordinary tool‐making behaviour, including the crafting of hooked stick tools from branched vegetation. We describe a surprisingly strong between‐site difference in the plant materials used by wild crows to manufacture these tools: crows at one study site use branches of the non‐native shrub Desmanthus virgatus, whereas only approximately 7 km away, birds apparently ignore this material in favour of the terminal twigs of an as‐yet‐unidentified tree species. Although it is likely that differences in local plant communities drive this striking pattern, it remains to be determined how and why crows develop such strong site‐specific preferences for certain raw materials. 相似文献

Haemophilus ducreyi is a Gram-negative bacterium which is the causative agent of chancroid, an ulcerative sexually transmitted disease. In order to understand the pathogenesis of H. ducreyi disease, studies designed to identify potential virulence determinants and construct mutants deficient in the elaboration of these determinants have been undertaken in several laboratories. At the present time, construction of isogenic mutants is accomplished by electroporation of linearized DNA containing insertionally inactivated H. ducreyi genes followed by selection for the resistance marker encoded on the inactivated gene. In our experience, certain mutants are difficult to construct using this procedure. In the construction of strains containing lacZ as a reporter gene, we observed that the growth of lacZ expressing H. ducreyi was inhibited in the presence of X-gal. We have exploited this observation to develop a new strategy for the construction of isogenic H. ducreyi mutants. 相似文献

克隆番茄(Solanum lycopersicum) ARF2基因,并分析其分子特性和亚细胞定位,为研究其功能提供基础.通过生物信息学方法分析SlARF2基因编码蛋白的理化性质和分子特性.采用RT-PCR技术从番茄果实cDNA中扩增SlARF2基因全长,并构建与黄色荧光蛋白(YFP)融合的pBA-ARF2-YFP表达载体,进而再通过农杆菌介导的遗传转化方法,将重组质粒转化到野生型番茄中,将得到的T1代转基因种子萌发,然后取根尖通过荧光显微镜观察了融合蛋白在活细胞内分布的特点.生物信息学分析结果表明,SlARF2是富含Ser、Leu、Gly和Pro以及具有ARF家族典型结构域的可溶性蛋白,其氨基酸序列与葡萄、木薯和拟南芥的同源性分别为70.08 %、66.94 %和60.87 %.经酶切和测序分析证实pBA-ARF2-YFP融合表达载体构建成功,此外,PCR分析表明融合蛋白在转基因植株中得到表达.经荧光显微镜观察,ARF2定位在细胞核中.表明转录因子SlARF2定位在细胞核中,对番茄果实发育和成熟起重要作用. 相似文献

目的 了解河北省三级甲等综合医院图书馆网站建设的现状。方法 选取河北省22家三级甲等医院网站,采用百度引擎对医院网站进行搜索,对出现的图书馆网站及相关信息进行记录、归类和整理。结果 被调查的22家医院图书馆网站中,7家(33.33%)医院网站无图书馆网站及其相关信息;6家(28.57%)在医院网站主界面设置图书馆链接;8家(38.10%)医院在主网页中科教栏及其他栏目中涵盖图书馆链接,且多数医院图书馆网站信息更新频率慢。结论 河北省三级甲等综合医院图书馆网站建设较为落后,应采取相应措施,促进医院图书馆网站逐步完善。 相似文献

Aims In Mediterranean-type ecosystem, the Cape Fynbos, legumes may be able to switch between soil N and atmospheric N 2 sources during growth to adjust the carbon costs of N acquisition. This study investigated the utilization of different inorganic N sources by Virgilia divaricata, a native legume from the Mediterranean-type ecosystem of the Cape Floristic Region.Methods Plants were cultivated in sterile quartz sand, supplied with 25% strength Long Ashton nutrient solution, modified to contain 500 μM Phosphate. At the phosphate level (500 μM), plants were treated with 500 μM NH 4 NO 3 (treatment named N), or grown in N-free nutrient solution and inoculated with effective Burkholderia sp. (Bact.) or treated with combined N sources (500 μM NH 4 NO 3) and inoculated with effective Burkholderia sp. (N+Bact.).Important findings The application of NH 4 NO 3 to the legumes resulted in a greater increase in plant dry matter. Carbon construction costs were higher in plants that were supplied with mineral and symbiotic N sources. Maximum photosynthetic rates per leaf area was maintained, irrespective of the N sources. Although the plant roots were nodulated, the plant dependence on N 2 fixation decreased with addition of N. Roots and nodules of the plants solely reliant on N 2 fixation showed an increase in glutamine content. These results show that V. divaricata is highly adapted for growth at the forest margin. Fynbos and possibly anthropic soils by utilizing both atmospheric and soil N sources. 相似文献
