ABSTRACT. Nuclear factor TDP-43 has been shown to play a key role in Amyotrophic Lateral Sclerosis and Frontotemporal Dementia, where TDP-43 aggregates accumulate in patient''s affected neurons and this event can cause neuronal dysfunction. A major focus of today''s research is to discover the critical factors that lead to TDP-43 aggregation and the consequences for neuronal metabolism. From a structural point of view, several lines of evidence point toward TDP-43 C-terminus as a key domain able to mediate this process. Regarding this region, we have recently described a novel cellular TDP-43 aggregation model based on 12 tandem repetitions of its 339-366 Q/N rich prion-like domain. In addition, we have shown and confirmed that a minimal TDP-43 construct constituted by the N and C-terminal regions, but lacking both RRM domains, induce aggregation of endogenous TDP-43 and leads to its total loss of function as seen by changes in the alternative splicing of endogenous genes. In this work, we further characterize this model and show the importance of the N-terminus structure in the loss of function process. In addition, from a biochemical point of view we report that, as shown in a previous version of this model (GFP 12×Q/N), the endogenous TDP-43 trapped in the aggregates undergoes the 2 most important post-translational modifications seen in pathological TDP-43 inclusions: ubiquitination and hyperphosphorylation.  相似文献

目的:探讨三氧化二砷(As2O3)抑制鼻咽癌的作用机制。方法:采用流式细胞仪(FCM)、激光扫描共聚焦显微镜(LSCM)和荧光技术,检测人鼻咽癌细胞株(CNE1)经As2O3诱导后细胞连接蛋白43(Cx43)表达的变化。结果:FCM显示,经过浓度为4μmol L的As2O3处理后,其Cx43阳性细胞计数率明显升高,与未经As2O3处理和经2μmol L的As2O3处理的CNE1比较,其差异具有非常显著性意义(p<0.01);LSCM图像观察到,经4μmol LAs2O3处理后,标记Cx43的绿色荧光显著增强,集中分布于细胞膜。结论:较高剂量浓度的As2O3能提高鼻咽癌细胞Cx43的表达率,升高细胞膜Cx43的含量。As2O3抑制鼻咽癌细胞生长的作用机制之一,是通过恢复细胞间隙连接通讯功能来实现的。  相似文献

Vertebrate mechanosensory hair cells contain a narrow “pericuticular” zone which is densely populated with small vesicles between the cuticular plate and cellular junctions near the apical surface. The presence of many cytoplasmic vesicles suggests that the apical surface of hair cells has a high turnover rate. The significance of intense membrane trafficking at the apical surface is not known. Using a marker of endocytosis, the styryl dye FM1‐43, this report shows that rapid apical endocytosis in zebrafish lateral line sensory hair cells is calcium and calmodulin dependent and is partially blocked by the presence of amiloride and dihydrostreptomycin, known inhibitors of mechanotransduction channels. As seen in lateral line hair cells, sensory hair cells within the larval otic capsule also exhibit rapid apical endocytosis. Defects in internalization of the dye in both lateral line and inner ear hair cells were found in five zebrafish auditory/vestibular mutants: sputnik, mariner, orbiter, mercury, and skylab. In addition, lateral line hair cells in these mutants were not sensitive to prolonged exposure to streptomycin, which is toxic to hair cells. The presence of endocytic defects in the majority of zebrafish mechanosensory mutants points to a important role of apical endocytosis in hair cell function. © 1999 John Wiley & Sons, Inc. Morphologically distinct TDP‐43 aggregates occur in clinically different FTLD‐TDP subtypes, yet the mechanism of their emergence and contribution to clinical heterogeneity are poorly understood. Several lines of evidence suggest that pathological TDP‐43 follows a prion‐like cascade, but the molecular determinants of this process remain unknown. We use advanced microscopy techniques to compare the seeding properties of pathological FTLD‐TDP‐A and FTLD‐TDP‐C aggregates. Upon inoculation of patient‐derived aggregates in cells, FTLD‐TDP‐A seeds amplify in a template‐dependent fashion, triggering neoaggregation more efficiently than those extracted from FTLD‐TDP‐C patients, correlating with the respective disease progression rates. Neoaggregates are sequentially phosphorylated with N‐to‐C directionality and with subtype‐specific timelines. The resulting FTLD‐TDP‐A neoaggregates are large and contain densely packed fibrils, reminiscent of the pure compacted fibrils present within cytoplasmic inclusions in postmortem brains. In contrast, FTLD‐TDP‐C dystrophic neurites show less dense fibrils mixed with cellular components, and their respective neoaggregates are small, amorphous protein accumulations. These cellular seeding models replicate aspects of the patient pathological diversity and will be a useful tool in the quest for subtype‐specific therapeutics.  相似文献

Excitation-emission maps were constructed by measuring emission spectra from tobacco thylakoids and from thylakoids and intact cells of the cyanobacterium Synechocystis 6803. The measurement of such maps is greatly facilitated by the current diode-array detector technology. We show that excitation-emission maps are valuable tools for studies of the structure and energy transfer pathways in photosynthetic systems. 制备大鼠备用根模型 (切断单侧腰骶背根L2, 3, 4, 6, 及S1, 2, 保留L5 背根), 用免疫组织化学和原位杂交方法研究生长相关蛋白GAP43 在相应节段背根神经节和脊髓背角表达的变化及针刺对其表达的影响。结果发现, 切断一侧L2, 3, 4, 6, 及S1, 2 背根后, 它们对应的背根神经节内GAP43 表达与正常对照组和假手术组相比无显著性差别, 而备用根神经节L5 的GAP43 表达较正常对照组和假手术组明显增强; 手术侧L5 水平脊髓背角与正常对照组相比GAP43 阳性信号加强。针刺后可促进手术侧L5 神经节内GAP43 表达增加; L5 水平脊髓背角GAP43 阳性信号也进一步加强这表明在一定条件下, 神经系统损伤可诱发中枢神经系统GAP43 介导的可塑性变化, 针刺可通过GAP43 对神经的可塑性起调节作用。  相似文献

研究了机械刺激对细胞膜的间隙连接半通道的作用。采用了单滴溶液从不同高度自由落下时对细胞表面的冲击作为机械刺激。单滴染料+EGTA从不同高度落下冲击细胞时,细胞的染料负荷随刺激高度的增加而加强。只用染料而无EGTA时,只有很少量细胞摄取染料。说明EGTA的存在有重要意义。然而,从培养皿边缘缓慢加入5(6)-carboxyfluorescein(简称染料)+EGTA溶液,避免机械刺激时,所有细胞均无染  相似文献

Fibroblast growth factors (FGFs) include a large family of growth factors that play a critical role in maintaining bone homeostasis, but the specific role of its members such as FGF7 does not well understand. Osteoblasts are a kind of major cells essential for bone formation. Osteoblasts interact with one another to create the unique structure of osteons. The well-connected osteons constitute the cortical bone. As an early osteocyte marker that triggers actin cytoskeleton dynamics, E11 is essential for osteoblasts'' dendrites formation. However, the upstream which regulates E11 is mainly unknown. The purpose of this study was to examine the influence of FGF7 on the expression and the distribution of E11 in osteoblasts, which mediated osteoblasts'' processes formation and gap junctional intercellular communication (GJIC) partly through connexin43 (Cx43). We first demonstrated that FGF7 increased the expression of E11 in osteoblasts. We then showed that FGF7 promoted osteoblasts'' dendrites elongation and functional gap junctions formation. Furthermore, E11 interacted directly with Cx43 in primary osteoblasts. MAPK pathway and PI3K-AKT pathway were involved in the effect of FGF7. Our results shed light on the unique role of FGF7 on osteoblasts, which may indicate that FGF7 plays a more significant role in the later stages of bone development and homeostasis.  相似文献
