Nematode diversity may seriously be underestimated when taking into account cryptic speciation. Thoracostoma trachygaster is commonly found in kelp holdfasts along the California coastline and was recently shown to consist of at least two distinct molecular clades (I and II). Here, we provide detailed morphological analysis of both clades, based on measurements taken from video vouchers of respectively eight and 16 individuals from the previous study, as well as 80 newly collected specimens from four Californian beaches. The latter were vouchered, measured, and then subjected to molecular analyses of the mitochondrial cytochrome oxidase c subunit I (COI) gene, and the ribosomal D2D3 and internal transcribed spacer (ITS) regions. This integrative approach shows that the three molecular clades are phylogenetically and morphologically distinct species, but a combination of morphological characters is needed to distinguish them. Two new species, Thoracostoma fatimae sp. nov. and Thoracostoma igniferum sp. nov. , are identified and described. The spicule length of T. fatimae sp. nov. is significantly shorter than that of T. trachygaster. Thoracostoma igniferum sp. nov. can be distinguished by the irregular posterior edge of the cephalic capsule and the two internal subdorsal tropis‐like projections in the wall of the cephalic capsule, which are lacking in T. fatimae sp. nov. and T. trachygaster. © 2012 The Linnean Society of London, Zoological Journal of the Linnean Society, 2012, 164 , 18–35.  相似文献

Mouthparts of unfed larvae Piona carnea (Koch, 1836) (Acariformes: Pionidae) were studied on whole-mount preparations, semi-thin sections and with TEM and SEM methods. The mouth apparatus is incorporated within the pseudotagma, gnathosoma, composed of the infracapitulum and of the chelicerae resting on the roof of the latter. The gnathosoma inclines to the long axis of the body and is inserted at its base into the idiosoma by the circumcapitular fold. The basal cheliceral segments are long and fused. An anterior projection, the proposed fused fixed digits, protrudes from the distal end of the basal cheliceral segment forward between the movable digits. The movable digits are always found in protruded position, strongly curved upward and show a groove on their inner sides. The ventral wall of the infracapitulum is made of the mentum posterior and the malapophyses anterior to the palp articulation. The malapophyses are squeezed between the large palps and envelope the distal portion of the chelicerae from the sides. The ventral portion of the fused malapophyses are provided with a characteristic ventral cuticular fork of unknown function. Each malapophysis terminates by a flexible lateral lip provided by several rigid jags looking posterad. The palps face downward and backward, and bear on the tibia the large curved palpal claws turned laterad. The palp femur bears on the ventral aspect a characteristic wide spade-like projection provided with its own muscles originating on the dorsal wall of the femur. The labrum is a thick cuticular arrow-like structure protruding forward into the preoral cavity, whereas the cervix is a thin weakly sclerotized plate. The particular labral valve projects forward from the dorsal basis of the labrum into the preoral cavity. The labrum and the cervix are provided by their own small labral and cervical muscles originating on the cervical apodemes. The pharynx is totally separated from the ventral wall of the infracapitulum and is devoid of ventral dilators. The dorsal pharyngeal dilators originate on the thick and sclerotized capitular apodeme and, posteriorly, on the paired cuticular branches, capitular apodemes, which end freely in the body cavity and are combined with the common salivary duct. The short sigmoid pieces serve for origin of the levator muscles of the chelicerae. Retraction of the gnathosoma and the chelicerae is mediated by several sets of muscles originating on the dorsal plate.  相似文献

Morphological diversification does not proceed evenly across the organism. Some body parts tend to evolve at higher rates than others, and these rate biases are often attributed to sexual and natural selection or to genetic constraints. We hypothesized that variation in the rates of morphological evolution among body parts could also be related to the performance consequences of the functional systems that make up the body. Specifically, we tested the widely held expectation that the rate of evolution for a trait is negatively correlated with the strength of biomechanical trade-offs to which it is exposed. We quantified the magnitude of trade-offs acting on the morphological components of three feeding-related functional systems in four radiations of teleost fishes. After accounting for differences in the rates of morphological evolution between radiations, we found that traits that contribute more to performance trade-offs tend to evolve more rapidly, contrary to the prediction. While ecological and genetic factors are known to have strong effects on rates of phenotypic evolution, this study highlights the role of the biomechanical architecture of functional systems in biasing the rates and direction of trait evolution.  相似文献

The capacity of species to track changing environmental conditions is a key component of population and range changes in response to environmental change. High levels of local adaptation may constrain expansion into new locations, while the relative fitness of dispersing individuals will influence subsequent population growth. However, opportunities to explore such processes are rare, particularly at scales relevant to species-based conservation strategies. Icelandic black-tailed godwits, Limosa limosa islandica, have expanded their range throughout Iceland over the last century. We show that current male morphology varies strongly in relation to the timing of colonization across Iceland, with small males being absent from recently occupied areas. Smaller males are also proportionately more abundant on habitats and sites with higher breeding success and relative abundance of females. This population-wide spatial structuring of male morphology is most likely to result from female preferences for small males and better-quality habitats increasing both small-male fitness and the dispersal probability of larger males into poorer-quality habitats. Such eco-evolutionary feedbacks may be a key driver of rates of population growth and range expansion and contraction.  相似文献

The highest levels of Pb in soils and needles were detected in the Lisbon Botanical Garden, reflecting the intense car circulation since the end of the late nineteenth century, and the presence of tetraethyl lead in petrol. Levels of Pb in Aleppo bark were at or above the threshold values for plant tissues as defined by the UN-ECE/EC (>4–30 μg/g). Nickel was easily washed by distilled water from the bark, indicating a predominant adsorption due to atmospheric deposition, while only Pb was water-extracted from cone scales, but no more than 0.88 μg/g (4.3% of the total). If we take into account the dry weight of each cone, we can assess the amount of extractable lead present in a given time (ranges between 3.0 and 15.5 μg), although its eventual transfer to the ground through rainfall will be hampered by the cone architecture. The average levels of total Pb in the cones (12.7–35.4 μg/g) fall within the range defined by the UN-ECE/EC, generally; only the reference value is below 4 μg/g. Due to the high capabilities of allocating Pb inside the structure and through adsorption, Aleppo bark and cones can be very interesting tools in the monitoring of Pb.  相似文献

Parkinson''s disease (PD) is the second most common movement disorder and affects 1% of people over the age of 60 1. Because ageing is the most important risk factor, cases of PD will increase during the next decades 2. Next to pathological protein folding and impaired protein degradation pathways, alterations of mitochondrial function and morphology were pointed out as further hallmark of neurodegeneration in PD 3-11.After years of research in murine and human cancer cells as in vitro models to dissect molecular pathways of Parkinsonism, the use of human fibroblasts from patients and appropriate controls as ex vivo models has become a valuable research tool, if potential caveats are considered. Other than immortalized, rather artificial cell models, primary fibroblasts from patients carrying disease-associated mutations apparently reflect important pathological features of the human disease.Here we delineate the procedure of taking skin biopsies, culturing human fibroblasts and using detailed protocols for essential microscopic techniques to define mitochondrial phenotypes. These were used to investigate different features associated with PD that are relevant to mitochondrial function and dynamics. Ex vivo, mitochondria can be analyzed in terms of their function, morphology, colocalization with lysosomes (the organelles degrading dysfunctional mitochondria) and degradation via the lysosomal pathway. These phenotypes are highly relevant for the identification of early signs of PD and may precede clinical motor symptoms in human disease-gene carriers. Hence, the assays presented here can be utilized as valuable tools to identify pathological features of neurodegeneration and help to define new therapeutic strategies in PD.  相似文献

Cell wall deterioration throughout enzymatic hydrolysis of cellulosic biomass is greatly affected by the chemical composition and the ultrastructure of the fiber cell wall. The resulting pattern of cell wall deterioration will reveal information on cellulose activity throughout enzymatic hydrolysis. This study investigates the progression and morphological changes in lignocellulose fibers throughout enzymatic hydrolysis, using (transmission electron microscopy) TEM and field emission scanning electron microscopy (FE‐SEM). Softwood thermo‐mechanical pulp (STMP) and softwood bleached kraft pulp (SBKP), lignocellulose substrates containing almost all the original fiber composition, and with lignin and some hemicellulose removed, respectively, was compared for morphology changes throughout hydrolysis. The difference of conversion between STMP and SBKP after 48 h of enzymatic hydrolysis is 11 and 88%, respectively. TEM images revealed an even fiber cell wall cross section density, with uneven middle lamella coverage in STMP fibers. SKBP fibers exhibited some spaces between cell wall and lamella layers due to the removal of lignin and some hemicellulose. After 1 h hydrolysis in SBKP fibers, there were more changes in the fiber cross‐sectional area than after 10 h hydrolysis in STMP fibers. Cell wall degradation was uneven, and originated in accessible cellulose throughout the fiber cell wall. FE‐SEM images illustrated more morphology changes in SBKP fibers than STMP fibers. Enzymatic action of STMP fiber resulted in a smoother fiber surface, along with fiber peeling and the formation of ribbon‐disjunction layers. SBKP fibers exhibited structural changes such as fiber erosion, fiber cutting, and fiber splitting throughout enzymatic hydrolysis. © 2012 American Institute of Chemical Engineers Biotechnol. There has been a growing interest in whether established ecogeographical patterns, such as Bergmann's rule, explain changes in animal morphology related to climate change. Bergmann's rule has often been used to predict that body size will decrease as the climate warms, but the predictions about how body size will change are critically dependent on the mechanistic explanation behind the rule. To investigate change in avian body size in western North America, we used two long‐term banding data sets from central California, USA; the data spanned 40 years (1971–2010) at one site and 27 years (1983–2009) at the other. We found that wing length of birds captured at both sites has been steadily increasing at a rate of 0.024–0.084% per year. Although changes in body mass were not always significant, when they were, the trend was positive and the magnitudes of significant trends were similar to those for wing length (0.040–0.112% per year). There was no clear difference between the rates of change of long‐distance vs. short‐distance migrants or between birds that bred locally compared to those that bred to the north of the sites. Previous studies from other regions of the world have documented decreases in avian body size and have used Bergmann's rule and increases in mean temperature to explain these shifts. Because our results do not support this pattern, we propose that rather than responding to increasing mean temperatures, avian body size in central California may be influenced by changing climatic variability or changes in primary productivity. More information on regional variation in the rates of avian body size change will be needed to test these hypotheses.  相似文献

Numerous coleopteran species express male‐specific “weapon traits” that often show size variations among males, even within a single population. Many empirical studies have demonstrated that environmental conditions during development affect absolute weapon size. However, relatively few studies in horned beetles support the hypothesis that the relationship between weapon size and body size, also referred to as a “scaling relationship” or “static allometry”, is largely determined by genetic factors. In this study, the heritability of absolute mandible length and static allometry between mandible length and body size were estimated in the stag beetle Cyclommatus metallifer. While no significant heritable variation was observed in absolute mandible length, high heritability (h2 = 0.57 ± 0.25) was detected in the static allometry between mandible length and body size. This is the first report on the genetic effect on male mandible size in Lucanidae, suggesting that absolute mandible size is largely determined by environmental conditions while the static allometry between weapon size and body size is primarily determined by genetic factors.  相似文献
