The present study, utilizing thioglycolamido as the reactive group, describes the synthesis and pharmacology of a new opioid antagonist affinity ligand, 6-thioglycolamido-6-desoxynaltrexone (TAN) and compares TAN with a related known compound, 6-bromoacetamido-6-desoxynaltrexone (BAN). Both compounds were tested for their reversible and irreversible inhibition of [3H]naloxone binding to calf brain membranes. Reversible binding of BAN and TAN had Ki values of 1×10–9 and 1×10–10 M, respectively as determined by log probit plots. Irreversible binding was determined after extensive washing to remove all non-covalently bound ligand. At a concentration of 5×10–8 and 1×10–8 M for BAN and TAN irreversible binding was inhibited 50% of the maximum value. A study of the time course of irreversible inhibition of [3H]naloxone binding revealed that maximal inhibition occurred within 5 min with a concentration of 1×10–7 M of either agent. TAN but not BAN when administered systematically to mice produced an antinociceptive effect as measured by the writhing test. When administered intracerebraventricularly BAN did not block morphine-induced analgesia for more than 2 hr; whereas, with a single ED50 dose of 20 nmoles of TAN i.c.v. morphine-induced analgesia was almost completely blocked for a period of over 24 hr, as determined by the tail flick test. Although the SH group of TAN were required for the covalent interaction with opioid receptors, the site of TAN's interaction appears to involve other than protein SH groups.  相似文献

Abstract A number of sediment incubations were set up to reproduce some of the conditions used by Kristensen and Blackburn [1] and to make a comparison with their results. There were three types of microcosm: aerobic (OX), anaerobic (AN) and aerobic with Nephtys (NOX). In addition to other measurements, dissolved organic nitrogen (DON) pools and fluxes, were measured. The sediment in this experiment contained more particulate organic matter (POM). Nephtys (NOX) had the same effect as Nereis in increasing the rate of mineralization of POC and PON, compared with the OX-cores (2.1 and 2.6 times, respectively). Again, the AN-cores had a higher mineralization rate (loss of POM) than that of the OX-cores, but in addition, mineralization in NOX-cores was not significantly different from AN-cores. It was thus confirmed that anoxic mineralization could be as high, or higher, than the oxic process. Both the temporal patterns of O2-and and CO2-fluxes and their magnitudes were very similar to those reported earlier. This contrasts with the higher loss of POM in the present experiment. However, the loss of C in DOC (associated with the measured DON) can account for the extra POM loss. The pore-water profiles of σCO2 and NH4+ were similar to those in the earlier report, and the fluxes of σCO2, O2, NH4+ and NO3 followed the same temporal pattern.  相似文献

In order to understand the control mechanisms of a large, stable bacterial standing stock, enclosure experiments were conducted in a eutrophic lake, where both bacterial productivity and grazing pressure were very high. Total bacterial number in the different enclosures ranged from 1.2 to 2.7×107 cells mL−1 throughout the experiment. The average bacterial cell production rate estimated from a grazer eliminating experiment was 6.3×105 cells mL−1 h−1. Difference in the bacterial cell production rate between shaded and unshaded enclosures was not apparent. Bacteria showed a reduction in standing stock of only about 25–30% even after the supply of light was cut to 1%. Bacteria in the shaded enclosures then recovered their production rate in the first 12 days of perturbation. Grazing pressure in the shaded enclosures was not less than that for the control. Thus, it was considered a control mechanism of bacterial stable standing stock that the bacteria shifted their organic substrate from extracellular dissolved organic carbon freshly released from phytoplankton to that already stocked in the water column, though it is not known whether the dominant bacteria were the same.  相似文献

The products of activities of calcium and sulphate were calculated for solutions of 75 glasshouse soils. The majority of these products was found to be higher than the solubility product of gypsum, thus indicating that these soil solutions were possibly supersaturated. In another investigation, soil solutions were examined to determine whether such high activity products could be really attributed to supersaturation. By means of ultracentrifuging of solutions of glasshouse soils, it could be established that the solutions were practically free of sulphate-bearing colloidal particles. Some solutions contained calcium-bearing colloidal particles, but the quantities of calcium contained in these particles were too small to substantially influence the calcium activity. Addition of gypsum crystals to soil solutions led to crystallization of so much calcium and sulphate that the products of the activities of calcium and sulphate dropped from values that can be listed as high to values approaching the solubility product of gypsum. The results obtained demonstrate the occurrence of supersaturation of soil solutions with respect to gypsum. It is further postulated that the presence of humic substances in the soil solution is responsible for this supersaturation. The possible occurrence of supersaturation with respect to gypsum in soils other than glasshouse soils is discussed.  相似文献

Small-scale spatial heterogeneity of soil organic matter (SOM) associated with patterns of plant cover can strongly influence population and ecosystem dynamics in dry regions but is not well characterized for semiarid grasslands. We evaluated differences in plant and soil N and C between soil from under individual grass plants and from small openings in shortgrass steppe. In samples from 0 to 5 cm depth, root biomass, root N, total and mineralizable soil N, total and respirable organic C, C:N ratio, fraction of organic C respired, and ratio of respiration to N mineralization were significantly greater for soil under plants than soil from openings. These differences, which were consistent for two sites with contrasting soil textures, indicate strong differentiation of surface soil at the scale of individual plants, with relative enrichment of soil under plants in total and active SOM. Between-microsite differences were substantial relative to previously reported differences associated with landscape position and grazing intensity in shortgrass steppe. We conclude that microscale heterogeneity in shortgrass steppe deserves attention in investigation of controls on ecosystem and population processes and when sampling to estimate properties at plot or site scales.  相似文献

Measurements of suspended matter, particulate organic carbon and dissolved organic carbon were made over a three year period at stations spanning 150 km of the tidal freshwater Hudson River. Suspended matter concentrations varied from year-to-year and were not related to freshwater discharge. The increase in suspended matter with depth in vertical profiles suggests that, during medium to low flow conditions, resuspension of bottom sediments was as important a source of sediment as loadings from tributaries. Particulate organic carbon showed significant variability among stations, and both autochthonous primary production and detrital organic matter are contributing to POC standing stocks. Dissolved organic carbon represented over half of the total organic carbon in the water column and showed little variation among stations.Examining downstream changes in transport showed that there was significant production of both suspended matter and POC within the study reach during the ice-free season. Tributary loadings within the study reach do not appear to be the cause of these increases in downstream transport. Dissolved organic carbon behaved conservatively in that there was no evidence for net production or net consumption within the river.The spatial/temporal patterns and analyses of transport suggest that suspended matter and POC, but not DOC, were controlled to a significant extent by processes occurring within the river and were not simply related to loadings from outside.  相似文献

Intact sediment cores were obtained from three New York lakes in May, July, and October 1981. Radioactive S (as 35SO 4 2− ) was added to the overlying water and cores were incubated without atmospheric exchange for one week near lake bottom temperatures. Headspace flux of 02 as an index of sediment respiration rates varied among lakes and seasonally within lakes. Acidic South Lake had the lowest respiration rate at all seasons and also the smallest net incorporation of the 35SO 4 2− . Summer net isotope transformation into ester sulfate and non-HI reducible S (pyrite and C-bonded S) constituents was 88.6%, 89.4%, and 59.7% of total sediment isotope for Oneida, Deer, and South, respectively. Seasonal variation of net isotope incorporation was observed in each lake as were differences in 35SO 4 2− partitioning into major S pools. Of the S constituents analyzed, HCl digestible S (volatile sulfides) was the smallest pool, while ester sulfate and non-HI reducible S together accounted for greater than 50% of S isotope transformation in all lakes. In addition, ester sulfate is the major product of dissolved SO 4 2− transformation and its formation results in less alkalinity generation than the formation of non-HI reducible S constituents. Thus ester sulfate transformation processes must be considered in calculating alkalinity generation by lake sediments. Nitrate depletion in the riparian zone of a small woodland stream   总被引:1,自引:0,他引:1  
Field enrichments with nitrate in two spring-fed drainage lines within the riparian zone of a small woodland stream near Toronto, Ontario showed an absence of nitrate depletion. Laboratory experiments with riparian substrates overlain with nitrate enriched solutions revealed a loss of only 5–8% of the nitrate during 48 h incubation at 12°C. However, 22–24% of the initial nitrate was depleted between 24 and 48 h when a second set of substrate cores was incubated at 20°C. Short-term (3 h) incubations of fresh substrates amended with acetylene were used to estimate in situ denitrification potentials which varied from 0.05–3.19 g N g–1 d–1 for organic and sandy sediments. Denitrification potentials were highly correlated with initial nitrate content of substrate samples implying that low nitrate levels in ground water and riparian substrates may be an important factor in controlling denitrification rates. The efficiency of nitrate removal in spring-fed drainage lines is also limited by short water residence times of < 1 h within the riparian zone. These data suggest that routes of ground water movement and substrate characteristics are important in determining nitrate depletion within stream riparian areas.  相似文献

The compositions of carotenoids, chlorophylls and lipids at four depths in Ace Lake have been determined as a means of studying the vertical zonation of species in the lake and for comparison with the lipids found in the bottom sediments. The four major species of phytoplankton found in the lake were identified by electron microscopy. The most abundant phytoplankter was Pyramimonas gelidicola McFadden (Chlorophyta, Prasinophyceae) which occurred in greatest numbers at 10 m, the base of the oxylimnion. The pigments and lipids at this depth were mainly derived from this alga. At 11 m (the top of the anoxylimnion) only traces of lipids and pigments attributable to P. gelidicola were found, indicating only limited settling of algal cells through to the anoxylimnion, at least in summer. The pigments at 11 m were dominated by bacteriochlorophylls c derived from green photosynthetic bacteria Chlorobium spp. These pigments were also abundant at 23 m suggesting the presence of intact bacterial cells which had settled out from higher in the water column. Major non-polar lipid classes in the sediments included sterols, alcohols, hydrocarbons and an unusual suite of very long-chain unsaturated ketones and esters which have not previously been reported from antarctic environments. Several novel compounds, not found previously in either sediments or organisms, are reported. These include tri- and tetra-unsaturated straight-chain C39 methyl ketones and C40 ethyl ketones and the methyl ester of a tetra-unsaturated straight-chain C36 fatty acid. The distributions of lipids in the sediment were markedly different from those in the water column indicating extensive bacterial degradation and recycling of labile lipids.  相似文献

Salts of transition elements that alter the rate of sperm cell movement act at or near calcium-binding sites. After living bull sperm cells had been preincubated in VO43?, Ni2+, Zn2+, Mn2+, and also La3+, they were then fixed. Crisply defined organelles and the absence of particulate deposits in the morphological controls contrasted sharply with the treated specimens; the latter contained regions of increased electron density, the nature and distribution of which depended on the test substance, reflecting the differential affinities of the specific ions. La3+ formed fine dense areas, mainly at the exocytic surface of the plasma membrane. VO43? marks the cell surface but also left particulate densities within the cell. Ni2+ caused a nearly uniformly dense deposit at the surface and on the satellite fibers and axonemal microtubules. Zn2+ formed less uniform but coarser deposits, while in Mn2+ the distribution was similar to that in Zn2+ but much denser in the axonemal matrix and on the satellite fibers. Verapamil restricted the size and number of the opacities, while procaine permitted a similar distribution of slightly larger size reaction product. The differences in size and distribution of the enhanced densities were consistent and replicable for the individual assay substances. Vanadate, which specifically inhibits Na, K-ATPase, bound to ouabain-sensitive enzyme loci, however, completely disrupting the axonemal complex. This suggests that an important role of dynein in flagellar motion may relate to intracellular transport of Ca2+.  相似文献
