The ubiquitous glyoxalase system, which is composed of two enzymes, removes cellular cytotoxic methylglyoxal (MG). In an effort to identify critical residues conserved in the evolution of the first enzyme in this system, glyoxalase I (GlxI), as well as the structural implications of sequence alterations in this enzyme, a search of the National Center for Biotechnology Information (NCBI) database of unfinished genomes was undertaken. Eleven putative GlxI sequences from pathogenic organisms were identified and analyses of these sequences in relation to the known and previously identified GlxI enzymes were performed. Several of these sequences show a very high similarity to the Escherichia coli GlxI sequence, most notably the 79% identity of the sequence identified from Yersinia pestis, the causative agent of bubonic plague. In addition to the conservation of residues critical to binding the catalytic metal in all of the proposed GlxI enzymes, four regions in the Homo sapiens GlxI enzyme are absent in all of the bacterial GlxI sequences, with the exception of Pseudomonas putida. Removal of these regions may alter the active-site conformation of the bacterial enzymes in relation to that of the H. sapiens. The PPX/PP4 Ser/Thr protein phosphatases belong to the type 2A phosphatase subfamily and are present in most eukaryotic organisms. We have previously isolated two closely related DNAs encoding PPX isoforms (PPX-1 and PPX-2) of Arabidopsis thaliana. Here we report the molecular cloning of the genes encoding these proteins. The genes PPX-1 and PPX-2 are composed of eight exons and seven introns located at equivalent positions related to the coding sequences. Whereas the intron-exon organization of the PPX genes is completely different from that of the PP2A-3/PP2A-4 A. thaliana family, specific intron-exon boundaries are conserved among PPX genes from distantly related organisms. Based on GUS expression, both PPX genes show the same spatial and temporal pattern of expression: they are expressed in all the organs and tissues analyzed, and from the earliest stage of development. When PPX proteins were localized to the root in semi-thin methacrylate sections by immunofluorescence, staining was predominantly confined to small organelles, shown to be plastids by co-localization of PPX and ferredoxin. Interestingly, only some ferredoxin-positive plastids were also PPX-positive, and PPX staining was consistently brighter in the epidermis. The localization was confirmed with immunogold and electron microscopy. Our results suggest that, despite its strong sequence conservation, PPX in plants functions differently than in animals.  相似文献

黑曲霉T21是由黑曲霉3.795经诱变育种获得的糖化酶高产菌株,为阐明其高产的分子机制,由黑曲霉3.795克隆了糖化酶结构基因及其5′旁侧序列,并与黑曲霉T21的相应序列进行了比较.由黑曲霉3.795菌丝体分离染色体DNA,Southern杂交分析表明,糖化酶结构基因位于~2.5kb的EcoRⅠ-EcoRⅤ染色体DNA片段上,在此EcoRⅠ位点上游约1.0kb处有一SalⅠ位点.为构建糖化酶结构基因及其5′旁侧序列的基因组文库,该染色体DNA分别用EcoRⅠ+EcoRⅤ和EcoR+SalⅠ消化,琼脂糖凝胶电泳分离并回收长度在1.0kb左右和2.5kb左右的DNA片段,分别与pUC19载体连接后转化入E.coliDH5.用原位杂交方法筛选到了携带糖化酶基因编码区及其1505bp5′旁侧序列的阳性克隆.对克隆片段的DNA序列进行了测定并与黑曲霉T21的相应序列进行了比较,结果表明,在糖化酶基因编码区及其150bp3′非编码区内,未发现碱基差异,但在-340~-1505的5′上游区内发生了9个位置的碱基变化,包括缺失、插入和替换.这些结果表明,黑曲霉T21与3.795的糖化酶产量的差异与其结构基因无关,但可能与其  相似文献

One common framework for describing the evaluation and assessment of hazards in the workplace includes the four steps of hazard identification, exposure assessment, exposure-response modeling, and risk characterization (NAS, 1983). We discuss hazards for occupational injury and illness in light of this framework, and we contrast the evaluation of injury hazards with the evaluation of illness hazards. In particular, the nature of the hazards, typical exposure patterns, quantification of exposure, and the attribution of outcome to exposure are discussed. Finally, we discuss the management of occupational illness and injury hazards and issues encountered when evaluating efforts designed to mitigate the effects of occupational hazards.  相似文献

Animal models play a crucial role in fundamental and medical research. Progress in the fields of drug discovery, regenerative medicine and cancer research among others are heavily dependent on in vivo models to validate in vitro observations, and develop new therapeutic approaches. However, conventional rodent and large animal experiments face ethical, practical and technical issues that limit their usage. The chick embryo represents an accessible and economical in vivo model, which has long been used in developmental biology, gene expression analysis and loss/gain of function experiments. It is also an established model for tissue/cell transplantation, and because of its lack of immune system in early development, the chick embryo is increasingly recognised as a model of choice for mammalian biology with new applications for stem cell and cancer research. Here, we review novel applications of the chick embryo model, and discuss future developments of this in vivo model for biomedical research.  相似文献

A needs assessment survey of ethics review committees (ERCs) across Africa was conducted in order to establish their major needs and areas of weaknesses in terms of ethical review capacity. The response rate was 84% (31 of 37 targeted committees), and committees surveyed were located in 18 African countries. The majority of the responding committees (61%) have been in existence between 5 and 10 years; approximately 74% of the respondents were institutional committees, with the remainder being either national (6/31) or regional (2/31).
In terms of the ethical review process, nine of the 31 committees that responded did not have standard operating procedures (SOPs), and seven of the 22 that did have SOPs had never revised them after their initial development (an average period of three years). Of the 31 committees, 10 operated without any ethical guidelines. Many of the committees (13/30) met once per month, and the number of proposals reviewed annually varied, ranging from five to over 100. All respondents relied on paper-based data management and archiving systems.
Overall, the survey identified the major constraints on ERCs as lack of office equipment, outdated or lack of SOPs, lack of electronic data management systems, inadequate resources, lack of or insufficient expertise on the committees, and poor recognition of the importance of the role of the committees. Consequently, the authors are addressing the identified needs and weaknesses through the Bill and Melinda Gates Foundation-funded capacity building project. The impact of the intervention project will be assessed during and at the end of the four-year longitudinal project.  相似文献

Flowering phenology is an important and poorly understood plant trait that may possibly be related to the invasiveness potential of alien species. The present work evaluates whether flowering time of invasive alien species is a key trait to overcome the climatic filters operating in continental Mediterranean ecosystems of Spain (characterised by summer drought and low temperatures in winter). We conducted comparisons between the flowering phenology of the invasive species in their native range and in Spain, and between flowering phenology of 91 coexisting invasive–native species pairs. For the alien species, geographical change from the native to the invaded region did not result in shifts in the start and the length of the flowering period. Overall, climatic conditions in the native range of species selected for a flowering pattern is maintained after translocation of the species to another region. Flowering of tropical and temperate invasive alien species peaked in summer, which contrasts with the spring flowering of native and invasive alien species of Mediterranean climate origin. By exploiting this new temporal niche, these invasive alien species native to tropical and temperate regions benefit from reduced competition with natives for abiotic and biotic resources. We suggest that human-mediated actions have reduced the strength of the summer drought filter in particular microhabitats, permitting the invasion of many summer-flowering aliens.  相似文献

Insect–plant interactions are important to understanding the evolutionary mechanisms responsible for most of the diverse plant‐feeding insects. We aggregated data on sawfly–host plant associations and other resource associations from existing sources to address the following questions: (i) Is there a general correlation between host diversity and sawfly species richness? (ii) Is the pattern of host plant use consistent across sawfly lineages? and (iii) Is there a phylogenetically significant shift in species richness among sawflies? Our analysis comprised 8567 sawfly species, including 2087 species with host plant and other records. In total, there were 2126 records of host usage for sawflies, the overwhelming majority of which were sawflies using angiosperms as resources. Rosales are used by most of the species in sawfly families or subfamilies. We found that there was a strong correlation between the number of host plant orders and the species richness of sawfly families and subfamilies. To examine the points at which sawflies have experienced significant shifts in species richness, we compared sister taxon species richness. Several positive and negative shifts in species richness among sawflies were related to their range of host plant usage and specialized niche, respectively. In general, we found that most of the sawfly families and subfamilies used several orders as host plants, but mainly core eudicots, although some families or subfamilies were specialized on pteridophytes or gymnosperms.  相似文献

Terrestrial invasive plants are a global problem and are becoming ubiquitous components of most ecosystems. They are implicated in altering disturbance regimes, reducing biodiversity, and changing ecosystem function, sometimes in profound and irreversible ways. However, the ecological impacts of most invasive plants have not been studied experimentally, and most research to date focuses on few types of impacts, which can vary greatly among studies. Thus, our knowledge of existing ecological impacts ascribed to invasive plants is surprisingly limited in both breadth and depth. Our aim was to propose a standard methodology for quantifying baseline ecological impact that, in theory, is scalable to any terrestrial plant invader (e.g., annual grasses to trees) and any invaded system (e.g., grassland to forest). The Global Invader Impact Network (GIIN) is a coordinated distributed experiment composed of an observational and manipulative methodology. The protocol consists of a series of plots located in (1) an invaded area; (2) an adjacent removal treatment within the invaded area; and (3) a spatially separate uninvaded area thought to be similar to pre-invasion conditions of the invaded area. A standardized and inexpensive suite of community, soil, and ecosystem metrics are collected allowing broad comparisons among measurements, populations, and species. The method allows for one-time comparisons and for long-term monitoring enabling one to derive information about change due to invasion over time. Invader removal plots will also allow for quantification of legacy effects and their return rates, which will be monitored for several years. GIIN uses a nested hierarchical scale approach encompassing multiple sites, regions, and continents. Currently, GIIN has network members in six countries, with new members encouraged. To date, study species include representatives of annual and perennial grasses; annual and perennial forbs; shrubs; and trees. The goal of the GIIN framework is to create a standard yet flexible platform for understanding the ecological impacts of invasive plants, allowing both individual and synthetic analyses across a range of taxa and ecosystems. If broadly adopted, this standard approach will offer unique insight into the ecological impacts of invasive plants at local, regional, and global scales.  相似文献
