Photosysthetic cleavage of water molecules to molecular oxygen is a crucial process for all aerobic life on the Earth. Light-driven oxidation of water occurs in photosystem II (PSII) — a pigment-protein complex embedded in the thylakoid membrane of plants, algae and cyanobacteria. Electron transport across the thylakoid membrane terminated by NADPH and ATP formation is inadvertently coupled with the formation of reactive oxygen species (ROS). Reactive oxygen species are mainly produced by photosystem I; however, under certain circumstances, PSII contributes to the overall formation of ROS in the thylakoid membrane. Under limitation of electron transport reaction between both photosystems, photoreduction of molecular oxygen by the reducing side of PSII generates a superoxide anion radical, its dismutation to hydrogen peroxide and the subsequent formation of a hydroxyl radical terminates the overall process of ROS formation on the PSII electron acceptor side. On the PSII electron donor side, partial or complete inhibition of enzymatic activity of the water-splitting manganese complex is coupled with incomplete oxidation of water to hydrogen peroxide. The review points out the mechanistic aspects in the production of ROS on both the electron acceptor and electron donor side of PSII.  相似文献

Two Haemaphysalis longicornis ticks were found positive in PCR assay of com-1 gene to detect Coxiella burnetii DNA from 100 ticks. The nucleotide sequences of com-1 and 16S rRNA gene were determined from 2 ticks and compared with those of other C. burnetii strains. The results suggest that H. longicornis harbor Coxiella sp. bacteria in Korea. Furthermore, icd, cbhE', and cbbE' genes are C. burnetii specific genes whereas com-1 gene is Coxiella genus specific gene. This study gives the first documentation to prove the existence of Coxiella sp. in tick collected in Korea.  相似文献

Seeds contain the embryo as a new plant in miniature and have two major functions, reproduction and dispersal. Seed formation completes the process of plant reproduction and, with seed germination, the next plant generation starts. Given the ever-increasing environmental pollution with metal(loid)s, it is perhaps surprising that relatively few reports detail the impacts of metals on seed metabolism, viability and germination in comparison to the numerous publications on the effects of metals in vegetative tissues, particularly roots and shoots. This review provides information on metal(loid) homeostasis, detoxification and tolerance in relation to seed metabolism and performance. The delivery of metals from the mother plant into seeds and their implications on seed development are discussed, as are their uptake upon seed imbibition and subsequent effects on seed germination. Implications for seeds and seedlings on the biochemical and molecular level are discussed and finally, applied aspects are considered regarding the use of seeds for soil and water purification, and in phytoremediation programmes. We conclude with a perspective on future metal research in relation to seed biology.  相似文献

Background

Chronic lead (Pb2 +) exposure leads to the reduced lifespan of erythrocytes. Oxidative stress and K+ loss accelerate Fas translocation into lipid raft microdomains inducing Fas mediated death signaling in these erythrocytes. Pathophysiological-based therapeutic strategies to combat against erythrocyte death were evaluated using garlic-derived organosulfur compounds like diallyl disulfide (DADS), S allyl cysteine (SAC) and imidazole based Gardos channel inhibitor clotrimazole (CLT).

Methods

Morphological alterations in erythrocytes were evaluated using scanning electron microscopy. Events associated with erythrocyte death were evaluated using radio labeled probes, flow cytometry and activity gel assay. Mass spectrometry was used for detection of GSH-4-hydroxy-trans-2-nonenal (HNE) adducts. Fas redistribution into the lipid rafts was studied using immunoblotting technique and confocal microscopy.

Results

Combination of SAC and CLT was better than DADS and CLT combination and monotherapy with these agents in prolonging the survival of erythrocytes during chronic Pb2 + exposure. Combination therapy with SAC and CLT prevented redistribution of Fas into the lipid rafts of the plasma membrane and downregulated Fas-dependent death events in erythrocytes of mice exposed to Pb2 +.

Conclusion and general significance

Ceramide generation was a critical component of Fas receptor-induced apoptosis, since inhibition of acid sphingomyelinase (aSMase) interfered with Fas-induced apoptosis during Pb2 + exposure. Combination therapy with SAC and CLT downregulated apoptotic events in erythrocytes by antagonizing oxidative stress and Gardos channel that led to suppression of ceramide-initiated Fas aggregation in lipid rafts. Hence, combination therapy with SAC and CLT may be a potential therapeutic option for enhancing the lifespan of erythrocytes during Pb2 + toxicity.  相似文献

目的:选育产木聚糖酶活力高的黑曲霉菌株,对其产酶条件进行优化,并研究其酶学性质。方法:通过木聚糖酶解木聚糖产生透明圈的方法,筛选产木聚糖酶菌株,测定固体发酵培养基中玉米芯与麸皮的比例、培养温度、培养时间、添加氮源对产酶的影响。进行了作用温度、pH值、金属离子对酶活力的影响试验,以及酶不同温度下的热稳定性的试验。结果:从自然界筛选得到一株产木聚糖酶的黑曲霉菌株,通过对固态发酵培养条件优化,最终产酶水平达到了5500u/g固体干曲。酶的最适作用温度是45℃、最适作用pH值4.8,是一种偏酸性酶。该酶在45℃以上的温度保存会使酶活力迅速丧失,Mg^2+、Zn^2+对该酶有激活作用,而Mn^2+、Cu^2+、Hg^2+则完全抑制酶的活性。结论:选育的黑曲霉菌株产木聚糖酶活力较高,培养条件简单。  相似文献

High-resolution 13C NMR spectroscopy has been used to analyze the positional distribution of fatty acids in model triacylglycerols. A novel method for quantitative determination of the positional distribution of unsaturated chains in triacylglycerols simultaneously with the ratio of saturated/unsaturated acyl chains has been proposed, utilizing the chemical shift differences of the aliphatic atoms C4, C5, and C6. The use of HSQC-TOCSY spectra allows unequivocal proof of the position of the unsaturated chain as well as complete assignment of the 13C NMR signals in tripalmitin.  相似文献

The objective of this study was to investigate the effect of pre-anthesis high-temperature acclimation on leaf physiology of winter wheat in response to post-anthesis heat stress. The results showed that both pre- and post-anthesis heat stresses significantly depressed flag leaf photosynthesis and enhanced cell membrane peroxidation, as exemplified by increased O2−com/sd/entities/rad" class="glyphImg"> production rate and reduction in activities of antioxiditave enzymes. However, under post-anthesis heat stress, plants with pre-anthesis high-temperature acclimation (HH) showed much higher photosynthetic rates than those without pre-anthesis high-temperature acclimation (CH). Leaves of HH plants exhibited a higher Chl a/b ratio and lower chlorophyll/carotenoid ratio and superoxide anion radical release rate compared with those of the CH plants. In addition, antioxidant enzyme activities in HH plants were significantly higher than in CH. Coincidently, expressions of photosythesis-responsive gene encoding Rubisco activase B (RcaB) and antioxidant enzyme-related genes encoding mitochondrial manganese superoxide dismutase (Mn-SOD), chloroplastic Cu/Zn superoxide dismutase (Cu/Zn-SOD), catalase (CAT) and cytosolic glutathione reductase (GR) were all up-regulated under HH, whereas a gene encoding a major chlorophyll a/b-binding protein (Cab) was up-regulated by post-anthesis heat stress at 10 DAA, but was down-regulated at 13 DAA. The changes in the expression levels of the HH plants were more pronounced than those for the CH. Collectively, the results indicated that pre-anthesis high-temperature acclimation could effectively alleviate the photosynthetic and oxidative damage caused by post-anthesis heat stress in wheat flag leaves, which was partially attributable to modifications in the expression of the photosythesis-responsive and antioxidant enzymes-related genes.  相似文献

Gangliosides are complex glycosphingolipids that are important in many biological processes. The present study investigated the role of gangliosides in the organization of lipid rafts in RBL-2H3 mast cells and in the modulation of mast cell degranulation via Fccom/scidirimg/entities/25b.gif" alt="var epsilon" title="var epsilon" border="0">RI. The role of gangliosides was examined using two ganglioside deficient cell lines (B6A4A2III-E5 and B6A4C1III-D1) as well as the parent cell line (RBL-2H3). All three cell lines examined express Fccom/scidirimg/entities/25b.gif" alt="var epsilon" title="var epsilon" border="0">RI, Lyn, Syk and LAT. However, only in RBL-2H3 cells were Fccom/scidirimg/entities/25b.gif" alt="var epsilon" title="var epsilon" border="0">RI, LAT and α-galactosyl derivatives of ganglioside GD1b mobilized to lipid raft domains following Fccom/scidirimg/entities/25b.gif" alt="var epsilon" title="var epsilon" border="0">RI stimulation. The inhibition of glycosphingolipid synthesis in RBL-2H3 cells also resulted in a decrease in the release of β-hexosaminidase activity after Fccom/scidirimg/entities/25b.gif" alt="var epsilon" title="var epsilon" border="0">RI activation. The two mutant cell lines have a reduced release of β-hexosaminidase activity after Fccom/scidirimg/entities/25b.gif" alt="var epsilon" title="var epsilon" border="0">RI stimulation, but not after exposure to calcium ionophore. These results indicate that the α-galactosyl derivatives of ganglioside GD1b are important in the initial events of Fccom/scidirimg/entities/25b.gif" alt="var epsilon" title="var epsilon" border="0">RI signaling upstream of Ca2+ influx. Since the initial signaling events occur in lipid rafts and in the mutant cell lines the rafts are disorganized, these results also suggest that these gangliosides contribute to the correct assembly of lipid rafts and are essential for mast cell activation via Fccom/scidirimg/entities/25b.gif" alt="var epsilon" title="var epsilon" border="0">RI.  相似文献
