We have identified an 11-residue pattern (KR(F/A)KKFFKK(L/P)K), which we have named the ATRA motif, within the sequence of the Chinese cobra (Naja atra) cathelicidin. A series of 11-residue peptides (ATRA-1, -2, -1A and -1P) were designed to probe the significance of the conserved residues within the ATRA motif, and their contributions to antimicrobial performance. The antimicrobial activities of the peptides were assessed against Escherichia coli K12 strain and Aggregatibacter actinomycetemcomitans Y4. ATRA-1 and -1A, demonstrated potencies comparable to that of N. atra cathelicidin. Structural examination by circular dichroism of the four short peptides suggested the significance of specific amino acid positions within the motif by their contribution to helicity. The results of these studies indicate that short peptides derived from the repeated ATRA motif from the N. atra cathelicidin can demonstrate both low toxicity against host cells and high antimicrobial activity against the gram-negative bacteria used in this study. They constitute novel, effective antimicrobial peptides that are much shorter (and thus less expensive to produce) than the natural cathelicidins, and they may represent new templates for therapeutic drug development.  相似文献

Rodent transgenesis and human‐assisted reproductive programs involve multistep handling of preimplantation embryos. The efficacy of production and quality of results from conventionally scheduled programs are limited by temporal constraints other than the quality and quantities of embryos per se. The emergence of vitrification, a water ice‐free cryopreservation technique, as a reliable way to arrest further growth of preimplantation embryos, provides an option to eliminate the time constraint. In this article, current and potential applications of cryopreservation to facilitate laboratory animal experiments, colony management, and human‐assisted reproductive programs are reviewed. Carrier devices developed for vitrification in the last two decades are compared with an emphasis on their physical properties that infer cooling rate of samples and sterility assurance. 1. In brown food webs of the forest floor, necromass (e.g. insect carcasses and frass) falling from the canopy feeds both microbes and ants, with the former decomposing the homes of the latter. In a tropical litter ant community, we added necromass to 1 m2 plots, testing if it added as a net food (increasing ant colony growth and recruitment) or destroyer of habitat (by decomposing leaf litter). 2. Maximum, but not mean, colony growth rates were higher on +food plots. However, neither average colony size, nor density was higher on +food plots. In contrast, +food plots saw diminished availability of leaf litter and higher microbial decomposition of cellulose, a main component of the organic substrate that comprises litter habitat. 3. Furthermore, necromass acted as a limiting resource to the ant community only when nest sites were supplemented on +food plots in a second experiment. Many of these +food +nest plots were colonised by the weedy species Wasmannia auropunctata. 4. Combined, these results support the more food–less habitat hypothesis and highlight the importance of embedding studies of litter ant ecology within broader decomposer food web dynamics.  相似文献

During the past 15 years in the Ina Valley, Japan, mature fifth instar larvae of the yellow hornet, Vespa simillima, have been occasionally ejected from colonies during September. During 2005, this unusual behavior was particularly widespread, and collection of several V. simillima colonies confirmed that very few fifth instar larvae were present. When compared with an average colony, constructed from 41 colonies collected 20 years previously, colonies in 2005 had 80% fewer fifth instar larvae, despite queen egg‐laying rates remaining similar. It may be that the amount of food provided to the larvae by workers is less than the amount of larval secretions received by workers, which is causing the larvae to become emaciated and preventing them from pupating. This phenomenon normally occurs naturally at the end of the colony cycle in November, when prey abundance decreases and larval secretions or other sources of carbohydrates, for example from ripe fruit, tree sap, or aphid secretions, are needed by sexuals to build up fat reserves. However, another possibility is that the unexplained appearance of this phenomenon in September, which is causing colonies to collapse without producing any or very few sexuals, is due to foragers feeding the larvae prey that are contaminated with insect growth regulators, via pesticides, which are known to prevent successful pupation.  相似文献

Colonization of roots by Pseudomonas chlororaphis O6 ( Pc O6) involves root surface coverage through surface motility and biofilm formation. Root colonization and the production of antifungal phenazines are important in the ability of the bacterium to protect plants against pathogens. In this in vitro study we report that both biofilm formation and phenazine production are differentially influenced by nutrition and the presence of polyethylene oxide/polypropylene oxide triblock copolymer surfactants (Pluronics). Such surfactants are used for many purposes including agricultural formulations. Four Pluronics differing in molecular weight and in hydrophobic/hydrophilic proportions had distinct effects on biofilm formation and secondary metabolite production, although each increased surface motility, termed swarming, to a similar extent. These findings show that Pluronics had specific metabolic impacts on the bacterium, where both up- and downregulation was achieved depending on the medium and the Pluronic composition. In environmental and agricultural settings, Pluronics may have unanticipated effects on soil microorganisms, while in bioprocessing these effects may be leveraged to regulate metabolite yield.  相似文献

Extracellular human granulocyte-macrophage colony stimulating factor (hGM-CSF) expression was studied under the control of the GAP promoter in recombinant Pichia pastoris in a series of continuous culture runs (dilution rates from 0.025 to 0.2 h−1). The inlet feed concentration was also varied and the steady state biomass concentration increased proportionally demonstrating efficient substrate utilization and constancy of the biomass yield coefficient (Yx/s) for a given dilution rate. The specific product formation rate (qP) showed a strong correlation with dilution rates demonstrating growth associated product formation of hGM-CSF. The volumetric product concentration achieved at the highest feed concentration (4×) and a dilution rate of 0.2 h−1 was 82 mg l−1 which was 5-fold higher compared to the continuous culture run with 1× feed concentration at the lowest dilution rate thus translating to a 40 fold increase in the volumetric productivity. The specific product yield (YP/X) increased slightly from 2 to 2.5 mg g−1, with increasing dilution rates, while it remained fairly invariant, for all feed concentrations demonstrating negligible product degradation or feed back inhibition. The robust nature of this expression system would make it easily amenable to scale up for industrial production.  相似文献

c-Fms, a member of the Platelet-derived Growth Factor (PDGF) receptor family of receptor tyrosine kinases (RTKs), is the receptor for macrophage colony stimulating factor (CSF-1) that regulates proliferation, differentiation and survival of cells of the mononuclear phagocyte lineage. Abnormal expression of c-fms proto-oncogene is associated with a significant number of human pathologies, including a variety of cancers and rheumatoid arthritis. Accordingly, c-Fms represents an attractive therapeutic target. To further understand the regulation of c-Fms, we determined the 2.7 A resolution crystal structure of the cytosolic domain of c-Fms that comprised the kinase domain and the juxtamembrane domain. The structure reveals the crucial inhibitory role of the juxtamembrane domain (JM) that binds to a hydrophobic site immediately adjacent to the ATP binding pocket. This interaction prevents the activation loop from adopting an active conformation thereby locking the c-Fms kinase into an autoinhibited state. As observed for other members of the PDGF receptor family, namely c-Kit and Flt3, three JM-derived tyrosine residues primarily drive the mechanism for autoinhibition in c-Fms, therefore defining a common autoinhibitory mechanism within this family. Moreover the structure provides an understanding of c-Fms inhibition by Gleevec as well as providing a platform for the development of more selective inhibitors that target the inactive conformation of c-Fms kinase.  相似文献

干细胞移植治疗肿瘤具有重要的临床价值.应用人间充质干细胞条件培养液作用H7402肝癌细胞,拟探讨间充质干细胞对肿瘤细胞的抑制作用,为今后应用人间充质干细胞进行肿瘤细胞治疗奠定理论基础.应用胎儿真皮来源的 Z3 间充质干细胞和胎儿骨髓来源的 BMMS-03 间充质干细胞的条件培养液作用于H7402肝癌细胞,采用软琼脂克隆形成实验、流式细胞仪技术、基因芯片技术和免疫印迹技术观察 H7402 细胞的克隆形成、增殖和基因表达谱变化.结果显示,H7402 细胞在间充质干细胞条件培养液作用下,克隆形成和增殖受到了明显抑制;基因芯片检测结果显示,H7402 细胞在间充质干细胞条件培养液作用下有 23 个基因上调表达,17 个基因下调表达,这些差异表达的基因与细胞的转录调控、新陈代谢、信号转导、细胞周期、应激反应和细胞粘附等功能相关.本实验结果表明,人间充质干细胞对 H7402 肝癌细胞的克隆形成和增殖具有抑制作用,并有多种基因的表达发生改变,这些基因表达的改变可能参与了对上述肿瘤细胞的抑制.  相似文献

DNA colonies formed during PCR in a polyacrylamide gel and RNA colonies grown in an agarose gel containing Qβ replicase can be identified using the procedure of transfer of molecular colonies onto a nylon membrane followed by membrane hybridization with fluorescent oligonucleotide probes. The suggested improvements significantly simplify and shorten the procedure. By the example of a chimeric AML1-ETO sequence, a marker of frequently occurring leukemia, the express hybridization method was shown to allow the rapid identification of single molecules and the determination of titers of DNA and RNA targets. Hybridization with a mixture of two oligonucleotide probes labeled with different fluorophores complementary to components of the chimeric molecule ensures the identification of molecular colonies containing both parts of the chimeric sequence and improves the specificity of diagnostics.  相似文献

Phlebotomine sandflies, vectors of Leishmania (Kinetoplastida: Trypanosomatidae) parasites that affect millions of people worldwide, breed in terrestrial biotopes. As immature stages are rarely accessible, the detection of their natural breeding sites is primarily based on findings of juvenile males with unrotated external genitalia. In males, permanent 180° rotation on the longitudinal body axis occurs soon after eclosion; however, no study has as yet addressed this aspect in detail. The present study describes the timing and duration of the rotation of male external genitalia in eight highly medically important sandfly species belonging to the genera Sergentomyia, Lutzomyia and Phlebotomus (all: Diptera: Psychodidae), kept under controlled laboratory conditions. The average duration of rotation was species‐specific and varied from 12 h in Sergentomyia schwetzi to 33 h in Phlebotomus sergenti. Significant differences in rotation times were found among species, even between two closely related species of the subgenus Larroussius, Phlebotomus orientalis and Phlebotomus tobbi. The rotation of genitalia in all three studied genera was randomly oriented and similar numbers of clockwise and counter‐clockwise events were observed. The study also addresses the effects of some external factors. In all species studied, rotation was not affected by the time of day of eclosion. Similarly, no differences in total rotation time were found between Phlebotomus papatasi males maintained at 25 and 20 °C, respectively. The present findings will assist in the search for natural breeding sites and in studies aimed at elucidating strategies for integrated sandfly and leishmaniasis control.  相似文献
