Xylan structure,microbial xylanases,and their mode of action   总被引:9,自引:2,他引:7  
Xylans, the major portion of the hemicellulose of plant cell walls and grasses, are heteropolymers consisting principally of xylose and arabinose. Microbial xylanases with different multiplicities and properties are reported. Most studies on the mode of action of these xylanases have been carried out with fungi and there is very little information available on bacterial xylanases. Fungal xylanases have three or more substrate binding sites: for exampleAspergillus niger, Ceratocytis paradoxa, Cryptococcus albidus andChainia sp. endoxylanases have four to seven subsites with the catalytic site located at the centre of these sub-sites. The analysis of these sub-sites is either by kinetic or end-product analysis studies. Kinetic studies are used for exo-type enzymes while the end-product analysis studies are more convenient for endo-type enzymes. This review covers microbial xylanases with special emphasis on studies of sub-site mapping. The industrial applications of the microbial xylanases are also discussed.  相似文献

To date, the sequences of 45 Bradyrhizobium japonicum genes are known. This provides sufficient information to determine their codon usage and G+C content. Surprisingly, B. japonicum nodulation and NifA-regulated genes were found to have a less biased codon usage and a lower G+C content than genes not belonging to these two groups. Thus, the coding regions of nodulation genes and NifA-regulated genes could hardly be identified in codon preference plots whereas this was not difficult with other genes. The codon frequency table of the highly biased genes was used in a codon preference plot to analyze the RSRj9 sequence which is an insertion sequence (IS)-like element. The plot helped identify a new open reading frame (ORF355) that escaped previous detection because of two sequencing errors. These were now corrected. The deduced gene product of ORF355 in RSRj9 showed extensive similarity to a putative protein encoded by an ORF in the T-DNA of Agrobacterium rhizogenes. The DNA sequences bordering both ORFs showed inverted repeats and potential target site duplications which supported the assumption that they were IS-like elements.  相似文献

Summary Dissected ampullae of Lorenzini of the skate (Raja clavata) were studied with the aim of determining the synaptic transmitter between electroreceptor cell and afferent fibre. Resting activity and stimulus-evoked activity in response to electrical pulses were recorded in single afferent units at constant perfusion with normal and test solutions containing different putative neurotransmitters. Presynaptic transmitter release was blocked by Mg2+ (up to 50 mM) to investigate the effects of the test substances upon the postsynaptic membrane. l-Glutamate (l-GLU) and l-aspartate (l-ASP), both at concentrations between 10-7 and 10-3 M, enlarged strongly resting and stimulus-evoked discharge frequency in the afferent fibre. If transmission was blocked by high Mg2+, resting discharge frequency could be restored by l-GLU or l-ASP. The glutamate agonists quisqualate (10-8–105 M) and N-methyl-D-aspartate (10-5–10-3 M) enlarged spontaneous activity in the afferent fiber. The same was found for kainic acid (10-9–10-5 M). Taurine at concentrations between 10-5 and 10-3 M caused a concentration-dependent decrease in afferent activity. The same was found for gammaaminobutyric acid (GABA; 10-5–10-4 M), and for the catecholamines adrenaline and noradrenaline, both in concentrations between 10-5 and 10-3 M. Serotonine (10-5–10-3 M) and dopamine (10-5-10-3 M) had no effect on resting or evoked activity in the Lorenzinian ampulla afferents. Acetylcholine (ACh; 10-4 M) enlarged discharge frequency in those units with initial rates lower than 22–25 Hz, but diminished discharge frequency in fibres with initial activity higher than 25 Hz. When synaptic transmission was blocked by high Mg2+ solution, perfusion with additional ACh did not restore resting activity in the afferent fibre. This paper describes the morphology and response characteristics of two types of paired descending neurons (DNs) (classified as DNVII1 and DNIV1) and two lobula neurons (HR1 and HP1) in the honeybee, Apis mellifera.
1.  The terminal arborizations of the lobula neurons are in juxtaposition with the dendritic branches of the DNs (Figs. 2, 3b, 5). Both of the DNs descend into the ipsilateral side of the thoracic ganglia via the dorsal intermediate tract (Fig. 6) and send out many blebbed terminal branches into the surrounding motor neuropil (Figs. 3c, 7).
2.  Both the lobula and descending neurons respond in a directionally selective manner to the motion of widefield, periodic square-wave gratings.
3.  The neurons have broad directional tuning curves (Figs. 10, 11). HR1 is maximally sensitive to regressive (back-to-front) motion and HP1 is maximally sensitive to progressive (front-to-back) motion over the ipsilateral eye (Fig. 11). DNVII1 is maximally sensitive when there is simultaneous regressive motion over the ipsilateral eye and progressive motion over the contralateral eye (Fig. 12a). Conversely, DNIV1 is optimally stimulated when there is simultaneous progressive motion over the ipsilateral eye and regressive motion over the contralateral eye (Fig. 12b).
4.  The response of DNIV1 is shown to depend on the contrast frequency (CF) rather than the angular velocity of the periodic gratings used as stimuli. The peak responses of both regressive and progressive sensitive DNs are shown to occur at CFs of 8–10 Hz (Figs. 13, 14).

The apple maggot fly, Rhagoletis pomonella (Walsh), has only recently been found in Utah infesting sour cherry, Prunus cerasus L. An electrophoretic comparison of flies from Utah cherries with flies from Illinois hawthorns, Crataegus mollis (T. & G.) Scheele (a native host within the native range of the fly), show a marked reduction of genetic variability in the Utah sample. This result is indicative of a genetic bottleneck associated with the establishment of the apple maggot population in Utah cherries.
Résumé R. pomonella (Walsh), est originaire de Crataegus dans l'Amérique du N.E. Il attaque de nombreux autres fruits, y compris les pommes et les cerises aigres (Prunus cerasus). La mouche a été récemment signalée en Utah, à la fois sur cerises et sur Crataegus douglasii. Nous avons comparé les niveaux de variabilité génétique d'une population de l'Utah contaminant les cerises et d'une population de l'Illinois contaminant C. mollis (la population de l'Illinois est représentative des niveaux de variabilité génétique dans l'aire d'origine de la mouche).La variabilité génétique à 17 loci a été évaluée par électrophorèse sur gel d'amidon. 10 de ces loci sont polymorphes dans la population d'Illinois, mais seulement 4 dans la population de l'Utah. Les fréquences alléliques de ces 4 loci de R. pomonella diffèrent significativement en Utah et en Illinois. La population de l'Utah présente nettement moins d'allèles par locus, un plus faible pourcentage de loci polymorphes et une hétérozygotie moyenne plus faible que la population de l'Illinois. Tous ces résultats sont conformes aux conséquences prévisibles d'un goulot d'étranglement.Deux explications existent pour cette perte de variabilité, toutes les deux liées à la combinaison de la faible taille de la population et de la dérive génétique ultérieure. Pour la première, la colonisation du cerisier par les mouches venant de Crataegus peut avoir provoqué un goulot d'étranglement génétique. Au contraire, la réduction de la variabilité peut avoir été la conséquence de la colonisation de l'Utah par R. pomonella. Nous retenons cette dernière comme la cause la plus vraisemblable de la variabilité génétique de la population de R. pomonella contaminant les cerises de l'Utah.

A biological measure of space available within substrates was used as an index to examine substrate selection by the stonefly nymph Paragnetina media (Walker). Physical measures, such as total surface area of substrate, have not worked well in the past in explaining distribution of aquatic invertebrates. Although analysis of habitable space within substrate did not explain selection completely, the technique provided a precise measure and might be a more rigorous means by which substrate selection could be examined.  相似文献

Summary Genotypes of pearl millet (Pennisetum americanum L. Leeke) were examined for differences in vesicular-arbuscular mycorrhizal (VAM) colonization and response to inoculation. For thirty genotypes tested across three field locations there was a range of mycorrhizal colonization intensity between 25 and 56%. In another experiment with two male-sterile lines, restorer lines and their derived crosses, grown in pots filled with non-sterilized soil there were significant differences between genotypes for colonization by mycorrhiza. This showed hostgenotype dependence for mycorrhizal colonization.Root growth rates, mycorrhizal root length, percentage root colonization and plant growth and P uptake were studied in ten genotypes. A set of 3 genotypes with similar root lengths varied significantly with regard to mycorrhizal root length and the percentage colonization. This supports the suggestion that VAM colonization and spread is dependent on the host genotype. The growth responses differed significantly between the genotypes and they also differed in their responses to P uptake and VAM inoculation. The utility of host-genotype dependent differences in VAM symbiosis in plant breeding is discussed.Journal Article No. 453  相似文献

Export of species from sources (epicenters) of differing ages and complexities was examined using laboratory microcosms. Polyurethane foam (PF) artificial substrates were colonized by protozoans for different time periods in a small pond. Substrates were returned to the laboratory and used as epicenters for protozoan colonization of barren PF islands in initially sterile microcosms. Islands were exposed to epicenters for either 24 h or continuously for 28 d. Islands from pairs of microcosms exposed to epicenters of identical ages were sampled on 1, 3, 7, 14, 21, 28 and 46 d after initial epicenter exposure. Colonization parameters were estimated by fitting numbers of colonizing species to the MacArthur-Wilson equilibrium model. Islands exposed continuously to epicenters were colonized by significantly more species than those exposed for only 24 h. Islands exposed to immature, species poor epicenters were colonized by a greater proportion of the source community than those exposed to more mature, species rich epicenters. All islands were depauperate compared to epicenters except those exposed to the most immature (1 d old) epicenter. Colonization continued at a reduced rate in spite of the absence of the epicenter. Results from communities with rapid species turnover and rapidly reproducing species suggest that the continuous presence of a species source is less important for colonization of a new habitat. Dispersal of potential colonists occurs rapidly in these communities. Less mature communities dominated by pioneer forms are more effective at producing colonists than more mature communities.  相似文献
