The unmet clinical need for myocardial repair after irreversible ischemic injury requires a better understanding of the biological properties of cardiac stem cells (CSCs). Using a primary culture of neonatal rat myocardial cells, we describe the formation and maturation of contracting cardiomyocyte colonies stemming from c-kit+, Sca+, or Isl1+ CSCs, which occurs in parallel to the hypertrophy of the major cardiac myocyte population. The contracting cardiomyocyte colonies (~1–2 colonies per 1 × 105 of myocardial cells) were identified starting from eighth day of culturing. At first, spontaneous weak, asynchronous, and arrhythmic contractions of the colonies at a rate of 2–3 beats/min were registered. Over time, the contractions of the colonies became more synchronous and frequent, with a contraction rate of 58–60 beats/min by the 30th day of culturing. The colonies were characterized by the CSCs subtype-specific pattern of growth and structure. The cells of the colonies were capable of spontaneous cardiomyogenic differentiation, demonstrating expression of both sarcomeric α-actinin and α-sarcomeric actin as well as the maturation of contractile machinery and typical Ca2+ responses to caffeine (5 mМ) and K+ (120 mМ). Electromechanical coupling, characterized by cardiac muscle-specific Ca2+-induced Ca2+ release, was evident at 3 weeks of culturing. Thus, the co-cultivation of CSCs with mature cardiac cells resulted in the formation of contracting cardiomyocyte colonies, resembling the characteristics of in vivo cardiomyogenesis. The proposed model can be used for the investigation of fundamental mechanisms underlying cardiomyogenic differentiation of CSCs as well as for drug testing and/or other applications.  相似文献

This paper develops Belozersky’s early idea of the precedence of RNA in the origin of life on the Earth. Based on the current knowledge of the functional omnipotence of RNA, three new mechanisms are considered that could be critical for the origin and evolution of the ancient RNA world: (1) the reaction of spontaneous transesterification of polyribonucleotides in aqueous media, which has been recently discovered by A.B. Chetverin and colleagues and could result in elongation of short initial oligoribonucleotides and generate sequence variants for further selection; (2) compartmentation of functional RNA ensembles in the form of mixed molecular colonies on moist mineral surfaces, in the absence of membranes and other envelopes; and (3) systematic exponential enrichment of an RNA population with “ functionally the best” molecules due to alternating dissolution of the colonies upon flooding and formation of new colonies upon drying in ancient pools (“primordial natural SELEX”).__________Translated from Molekulyarnaya Biologiya, Vol. 39, No. 4, 2005, pp. 550–556.Original Russian Text Copyright © 2005 by Spirin.  相似文献

This article compares the migration processes to, and the socio‐economic conditions of colonial Caribbean migrants in, the metropoles. Specifically, it discusses: 1) the formation of modern colonies in the Caribbean after World War II; 2) the differences between colonial Caribbean migrations and migrations from Caribbean nation‐states; 3) the different socio‐economic characteristics among Caribbean colonial migrants in the metropoles; and 4) the labour market incorporation and public policies affecting colonial Caribbean migrants in each metropolitan society. There are interesting differences among these Caribbean colonial migrants regarding the process of incorporation into the host society. This is related to the different development of the welfare state in each metropole and to the presence or absence of specific public policies addressing the peculiar socio‐economic conditions of these migrants.  相似文献

We studied the process of offspring production in queenless colonies of Acromyrmex subterraneus brunneus, and particularly evaluated the ovary development of workers as a function of their age. For this, subcolonies were set up and evaluated at different periods of isolation from the queen (2, 4 and 6 months), besides individually labeled age groups. The subcolonies were assessed according to offspring production and ovaries containing oocytes or not. The evaluations showed worker oviposition and development of males originating from worker-laid eggs. At 2 months' absence of the queen, eggs and larvae were found, with eggs in a higher proportion than larvae. After 4 months, the proportion of eggs had reduced while larvae had increased, and a pupa was found in one subcolony. At 6 months, besides a higher share of larvae, one pupa and one adult male were found. Dissection of workers revealed ovaries containing oocytes during the periods of evaluation. Only a group of medium-sized and large workers, 23.3%, 20.9% and 37.5% of the population from each period assessed in queenless subcolonies respectively, presented developed oocytes in the ovary. The same was observed in colonies with a queen, with 17.6%, 19.6% and 7.8% of the group of dissected workers from each time period, respectively. With respect to worker age, we observed by dissection of the ovary, that the greatest percentage of individuals with ovarioles containing oocytes occurred at 45 days (6 weeks) up to 90 days (12 weeks). These results probably are associated with the workers reproduction and the laying of trophic and reproductive eggs in colonies with and without a queen; these eggs have distinct functions in each situation.  相似文献

A bloom of loricate choanoflagellates was recorded for the first time in the Ross Sea polynya during the austral summer 2017. Both individual cells and uncommon large‐size colonies (200 μm length) represent the 42–55% of the total plankton community (i.e. specimens from 5 to 150 μm length). Choanoflagellates serve as a link between low and mid trophic levels since they prey on bacteria and in turn are ingested by zooplankton. This twofold role and the unusual abundance recorded in the Antarctic ecosystem may have relevant but still unknown effects on food web structure and dynamics in that area.  相似文献

Microorganisms are present on all inanimate surfaces creating ubiquitous sources of possible contamination in the laboratory. Experimental success relies on the ability of a scientist to sterilize work surfaces and equipment as well as prevent contact of sterile instruments and solutions with non-sterile surfaces. Here we present the steps for several plating methods routinely used in the laboratory to isolate, propagate, or enumerate microorganisms such as bacteria and phage. All five methods incorporate aseptic technique, or procedures that maintain the sterility of experimental materials. Procedures described include (1) streak-plating bacterial cultures to isolate single colonies, (2) pour-plating and (3) spread-plating to enumerate viable bacterial colonies, (4) soft agar overlays to isolate phage and enumerate plaques, and (5) replica-plating to transfer cells from one plate to another in an identical spatial pattern. These procedures can be performed at the laboratory bench, provided they involve non-pathogenic strains of microorganisms (Biosafety Level 1, BSL-1). If working with BSL-2 organisms, then these manipulations must take place in a biosafety cabinet. Consult the most current edition of the Biosafety in Microbiological and Biomedical Laboratories (BMBL) as well as Material Safety Data Sheets (MSDS) for Infectious Substances to determine the biohazard classification as well as the safety precautions and containment facilities required for the microorganism in question. Bacterial strains and phage stocks can be obtained from research investigators, companies, and collections maintained by particular organizations such as the American Type Culture Collection (ATCC). It is recommended that non-pathogenic strains be used when learning the various plating methods. By following the procedures described in this protocol, students should be able to: Perform plating procedures without contaminating media. Isolate single bacterial colonies by the streak-plating method. Use pour-plating and spread-plating methods to determine the concentration of bacteria. Perform soft agar overlays when working with phage. Transfer bacterial cells from one plate to another using the replica-plating procedure. Given an experimental task, select the appropriate plating method.  相似文献

长木蜂属于野生蜂类,后代的生存和繁衍完全依靠雌蜂所提供的蜂粮,蜂粮保鲜时间的长短对蜂群的繁衍具有关键性作用。本文采用营养琼脂培养基、改良"LB"培养基、MRS(4.5)、MRS(6.5)培养基分别对芍药的手采花粉、长木蜂蜂花粉和不同酿制时间的蜂粮样品中的微生物进行菌落计数、分析和分离鉴定。酿制初期的蜂粮样品在营养琼脂培养基生长的细菌菌落数占优势,随酿制时间的延长而迅速下降;7d、8d蜂粮样品在改良"LB"培养基上生长的细菌菌落数占优势;10d之后的蜂粮样品在MRS(6.5)培养基上微生物菌落数占优势;分离的68株菌株,其中细菌43株,酵母17株,放线菌8株。从蜂粮中分离的酵母菌和放线菌菌株均来自长木蜂雌蜂的体表。分离的68株菌株中有12株有较强抑菌活性,尤其是放线菌NF-34菌株对真菌和细菌均有抑菌活性。  相似文献

For direct identification of toxigenic colonies ofClostridium botulinum type E, suspected colonies are uniformly suspended in a phosphate buffer containing 0.5% (w/v) gelatin and 0.05% (w/v) Tween 20. After centrifuging, the supernatant is tested for botulinal toxin by an enzyme-linked immunosorbent assay (ELISA). Roosts are essential for the survival of most animals. Due to homothermic requirements, mammals are particularly dependent on roost quality and availability. Bats select their roosts in a species-specific way, likely related to species´ different physiological and adaptive needs. Unlike species whose individuals roost solitarily, roost selection is critical for bats forming large colonies due to the requirements for maintaining thousands of individuals in a single shelter. This is the case of Pteronotus (Mormoopidae), whose colonies reach hundreds of thousands of bats. Using captures, bioacoustics, and automated censuses, we evaluated how cave size, ceiling characteristics, environmental stability, temperature, and humidity influence the formation of exceptionally large colonies, species richness and composition in caves in north-eastern Brazil. We expected that colonies would be positively related to cave size and stability, internal cave selection would be species-specific, but larger and more environmentally stable caves would have higher richness. Pteronotus colonies were positively related to cave size, stability, and ceiling characteristics, and their presence strongly influenced cave temperature variation. Species richness was positively correlated to a cave stability index. Species other than Pteronotus preferred different climatic and ceiling characteristics. We detected an indirect influence of the large colonies of Pteronotus on the species richness and occupation inside caves. On the other hand, such caves favor species coexistence, as they offer a range of microenvironments, reducing niche overlap in their interior. Pteronotus gymnonotus and Pteronotus personatus are both key- and umbrella-species for cave ecosystems, stressing the need for specific conservation strategies in Brazil.
