Isoelectric points of viruses   总被引:1,自引:0,他引:1  
Viruses as well as other (bio‐)colloids possess a pH‐dependent surface charge in polar media such as water. This electrostatic charge determines the mobility of the soft particle in an electric field and thus governs its colloidal behaviour which plays a major role in virus sorption processes. The pH value at which the net surface charge switches its sign is referred to as the isoelectric point (abbreviations: pI or IEP) and is a characteristic parameter of the virion in equilibrium with its environmental water chemistry. Here, we review the IEP measurements of viruses that replicate in hosts of kingdom plantae, bacteria and animalia. IEPs of viruses are found in pH range from 1·9 to 8·4; most frequently, they are measured in a band of 3·5 < IEP < 7. However, the data appear to be scattered widely within single virus species. This discrepancy is discussed and should be considered when IEP values are used to account for virus sorption processes.  相似文献

The cell membrane of ram spermatozoa is more sensitive to the freezing process than in other species due to its composition. As a result, the quality and viability of frozen thawed ram spermatozoa are often poor, which together with the specific structure of the ewe's cervix are the main reasons for lower fertility in ewes after intracervical insemination. In the present study we investigated the effects of semen centrifugation through a single layer of a species-specific colloid (Androcoll-O) on post-thaw quality of ram spermatozoa. Motility, viability and morphology were analysed 0, 6, 12 and 24 h after thawing. DNA fragmentation index (%DFI) of the samples was assessed 0 h after thawing, by SCSA™. Membrane and acrosome integrity of spermatozoa were analysed by Sybr-14/PI/PNA test 0 h after thawing.The proportion of motile spermatozoa was significantly higher in SLC – selected samples in comparison to control (not SLC – selected) samples at 0, 6, 12 (P < 0.001) and 24 h (P < 0.05). The proportion of viable spermatozoa was also significantly higher in SLC - selected samples in comparison to control samples at all times (P < 0.001). The proportion of abnormal acrosomes and morphologically abnormal spermatozoa (MAS) were significantly lower in SLC – selected samples compared to control samples at all times (P < 0.001). Analysis of chromatin stability revealed significantly lower %DFI values in SLC – selected samples compared to control samples (P < 0.001). The SYBR-14/PI/PNA test also revealed significantly better values in SLC – selected compared to control samples (P < 0.05). In conclusion, single layer colloid centrifugation significantly improved post-thaw quality and longevity of ram spermatozoa, making it suitable for artificial insemination initiatives.  相似文献

ABSTRACT

Functionalisation of colloidal particles with DNA provides a powerful and flexible path towards self-assembly of ordered materials. Given the nearly limitless possibilities for constructing DNA-functionalised particles, and the wide range of conditions under which they can be assembled, it is crucial to gain an understanding of the principles governing self-assembly of these particles and how their properties affect the structures produced. A number of computational models for DNA-functionalised systems have successfully described their properties, and molecular simulation techniques have provided a unique insight into the factors underlying their assembly. Here, we discuss a variety of efforts using simulations to solve an important design problem in DNA-mediated assembly: how the properties of individual DNA-functionalised particles affect their interactions with each other, and ultimately how these interactions determine what structures can be produced.  相似文献

It is a challenging task to prepare well‐dispersed and highly luminescent quantum dots (QDs) powder and a new strategy is reported in this article. Sodium carboxymethyl starch (CMS‐Na) was employed in this work to prepare the QDs–starch composite. Ultraviolet (UV) light shows that the blank starches had no fluorescence, while the QDs‐starches were highly luminescent. Scanning electron microscopy (SEM) observation shows that the QDs–starch composite has the typical particle morphology with the diameter around 200 nm. Energy dispersive X‐ray spectroscopy (EDX) results show that there are intensive tellurium (Te) and cadmium (Cd) element signals. Combined fluorescent lifetime and steady‐state spectrometer show that the QDs–starch quantum yields (QYs) increase when the QDs loading increases from 1 × 10?6 mol/g to 2 × 10?6 mol/g, but when the loadings further increase, the QYs decrease slightly. For the red colour (λem = 660 nm) QDs, the QYs can reach to as high as 28.2%, and for the other colour QDs they can also have the QYs above 22%. Time‐resolved photobleaching experiments show that the fluorescent QDs–starch composite has a half‐decay time of 40.23 s. These results indicate that the CMS‐Na is a promising QDs dispersant to obtain high QY QD composites.  相似文献

Living and fossil megaspores produced by Selaginella (Lycopsida) and its extinct ancestors form distinctive (and occasionally iridescent) exines. Ultrastructural studies of these spores have provided data that demonstrate a colloidal mode of development which in turn implies a degree of self-assembly in the construction of these exines. We present here experimental evidence in support of the theory of selaginellalean megaspore exine construction by depletion flocculation. Iridescent colloidal flocculations of polystyrene latex particles demonstrate an ultrastructural organization virtually indistinguishable from that of the biological system, and clearly demonstrate that self-assembly of complex Selaginella exines by a relatively simple construction process is plausible.  相似文献

Ferritin, the iron storage protein, is an organic-inorganic hybrid colloid consisting of a hollow protein capsule, which is filled with ferrihydride with up to 4500 iron atoms. Owing to the varying iron content and the resulting density differences, as well as the protein oligomerization, a particle size distribution is superimposed with a density distribution, making a precise analysis of ferritin by analytical ultracentrifugation difficult. This study describes how the information of the sedimentation coefficient distribution can be combined with the diffusion coefficient distribution obtained from flow-field flow fractionation to yield the buoyant molar mass of the oligomers in the mixture, extending the information content of each individual analytical method. In addition, the sedimentation and diffusion coefficients are compatible with a simple hard-sphere aggregation model, suggesting that the ferritin oligomers up to the pentamer have a globular solution structure.Presented at the conference for Advances in Analytical Ultracentrifugation and Hydrodynamics, 8–11 June 2002, Grenoble, France  相似文献

INTRODUCTION: Intracytoplasmic lumens (ICL) with or without magenta material and transgressing vessels are features of Hürthle cell neoplasms (HCN). After detection of intracytoplasmic colloid inclusions (CIs) including targetoid (magenta) body-like structures in the Hürthle cells (HC) in a case of Hashimoto's thyroiditis (HT), we reviewed cases of HT, thyroid neoplasms, hyperplastic nodules (HN) and colloid goitres to determine the frequency of these structures. Further, an attempt was made to find out the significance of CIs. METHODS: FNA smears of 120 HT, 101 colloid goitres, 11 HN, and 76 neoplastic goitres were examined. The presence of CIs and empty ICL were noted in epithelial cells in these lesions. An attempt was made to find out the difference between HT with and without CIs in respect of various cytomorphologic features. The groups were compared using the Fisher's exact test of probability. RESULTS: The CIs were present in 36 (30.0%) of HT, 26 (34.2%) of neoplastic goitres, 3 (27.3%) of HN, and 4 (4.0%) colloid goitres. As compared to colloid goitres, CIs were present in a significant higher number of cases in HN (P = 0.0202), neoplastic goitres (P < 0.0001), and HT (P < 0.0001). Among neoplasms the frequency of CIs in HCN (75.0%) was significantly higher than that of papillary thyroid carcinoma (PTC) (33.3%, P = 0.0466), and follicular neoplasm (14.3%, P = 0.0083). The CIs were more frequent in HC in HT and HCN but in follicular cells (FC) in other lesions. The HT cases with CIs differed significantly from those without CIs in respect of HC and their cellularity, cellularity of reactive lymphoid cells, extracellular colloid and empty ICL. CONCLUSION: Care should be taken not to diagnose HT cases with an excessive Hürthle cell component and CIs, and PTC cases with Hürthlization and CIs, as HCN in FNA smears. Based on review of the literature and our findings, it is suggested that the Hürthle cell metaplasia in HT is a survival response of FC and the presence of CIs in Hürthle cell may represent their limited ability to synthesize colloid.  相似文献

Abstract

Remediation of toxic metals by bacteria offers a relatively inexpensive and efficient way for the decontamination of soil and associated environments. The present study was carried out to investigate the surface characteristics, adsorption, and remobilization of Cd and Cu on bacteria and their composites with soil colloidal components, which are the most active constituents in soils. The bacterial strain NTG-01 (Enterobacter aerogenes), which was both Cd- and Cu-resistant, was isolated from a heavily Cu-contaminated soil of the mining area in Daye suburb of Hubei Province, China. Batch laboratory experiments with NTG-01 and soil colloids were performed to quantify adsorption of Cu and Cd. The surface area of kaolinite and the soil colloids from an Alfisol and Ultisol increased by 3.0–8.8% after the introduction of the bacteria. In the presence of bacterial cells, the negative charges of soil colloid systems increased and the positive charges decreased, shifting pH from 4.0 to 6.5. Our results demonstrate that bacteria promote the adsorption of Cd and Cu by kaolinite and soil colloid systems. However, the heavy metals bound by the bacterial composites could also be easily released by NH4NO3 and EDTA. Caution should be taken when using such bacterial strains in bioremediation of heavy metal-contaminated soils.  相似文献

Surface sculpture of spores, pollen and other walled microscopic organisms commonly resembles patterns seen elsewhere in nature. These patterns are often species specific and of significant use in taxonomic study, particularly so in the fossil record where other data may be minimal. It can be argued that patterning, which must be governed to some extent by genotype, could simply reflect other natural patterns as a result of physical and chemical interaction during development. But does this diminish the view that patterning can often perform important biological functions? With examples drawn from fossil and living walled structures, we analyse the complex relationship between genetic constraints, construction mechanism and biological function, and we conclude that similar function may often result in similar pattern, perhaps further enhanced by similar aspects of development. The genetic complement, by way of selection, ‘learns’ to repeat the pattern, but each pattern creation mechanism retains a ‘personal signature’ reflecting its evolutionary history. With this new perspective in mind, we assess the potential implications in the study of Palaeozoic microfossils when many different groups are first developing surface patterning.  相似文献

Absorption, fluorescence emission, and fluorescence excitation spectra of stilbazolium merocyanine (1-(n-butyl)-4[(3,5-dimethoxy-4-oxocyclohexa-2,5-dienylidene)ethylidene]-1,4-dihydropyridyne) dye in water solution without and with colloidal silver addition were measured. In the presence of the colloid, besides the absorption band assigned to the protonated species of the dye (at 391 nm), an absorption band related to the free-base species appears at 490 nm. From the absorption and emission spectra, measured at various dye concentrations, follows that the aggregates are not effectively formed. Therefore, the long-wavelength absorption and fluorescence bands have to be related to some dye forms created by the solvatochromic effects. The fluorescence bands of the protonated and the free-base species are located at 559 nm and at about 630 nm, respectively. The shape of the long-wavelength band suggests the occurrence of more than one free-base form of the dye. At some dye and colloid concentrations, the global emission of the sample is enhanced as a result of silver addition. The increase in the emission yield of the dye could be partially due to not only the change in the concentrations of dye forms exhibiting various emission yields but is also due to the resonance surface plasmon effect. Because of the superposition of several effects, before the practical application of merocyanine as an indicator of metal presence in biological samples, its spectral properties in the system investigated should be established.  相似文献
