The aim of the present investigation is to compare two geographically well separated populations ofC. afra with different mating systems with emphasis on (1) factors controlling the length of the male phase with special attention paid on the pollen collecting hairs, (2) crossing ability among populations with different mating systems; shouldC. afra be regarded as one or several species?, and (3) correlations between pollen germinability and mating system; how does pollen germinability vary with anthesis and stigma development? From the results it can be concluded that the length of the male phase and the development of the stigma depend on the stimulation of the pollen collecting hairs rather than on the amount of pollen removed. Furthermore, the length of the male phase was found to be correlated with the mating system, i.e., in the population with a mixed mating system, the differences between treated and untreated flowers were less pronounced than in the population being obligately outcrossing. I also found correlations between mating system and pollen germinability, where the population with a mixed mating system had a high germinability at stigma receptivity, whereas for the xenogamous population, the germinability approached zero at stigma development. No crossing incompatibility was found between the two populations ofCampanula afra in any of the combinations.  相似文献

The influence of several endogenous factors on micropropagation of mature elite trees of beech (Fagus sylvatica L.) has been investigated. February was the most beneficial month for explantation of dormant buds, since infection with endogenous bacteria was still low and in vitro growth of the plant material was the highest. The genotypes tested gave different results concerning their growth on various tissue culture media. Out of 51 mature grafted genotypes only seven could be established in vitro. Grafting of branches of mature stock plants on juvenile rootstocks resulted in a high increase of multiplication rate compared with corresponding mature material, which could not be subcultured in vitro. Apical buds, larger than 20 mm, from 1-year-old shoots proved to be the most suitable explant source. The mechanisms that underlie the profibrotic effect of interleukin (IL)-1β are complicated and not fully understood. Recent evidence has suggested the involvement of the calcium-sensing receptor (CaSR) in tubular injury. Therefore, the current study aimed to investigate whether CaSR mediates IL-1β-induced collagen expression in cultured mouse inner medullary collecting duct cells (mIMCD3) and to determine the possible downstream signaling effector. The results showed that IL-1β significantly upregulated the expression of type I and III collagens in a concentration- and time-dependent manner. Moreover, CaSR was expressed in mIMCD3 cells, and its expression was increased by increasing the concentrations and times of IL-1β treatment. Selective inhibitors (Calhex231 or NPS2143) or the siRNA of CaSR attenuated the enhanced expression of type I and III collagens. Furthermore, IL-1β increased nuclear β-catenin protein levels and decreased cytoplasmic β-catenin expression in cells. In contrast, blockage of CaSR by the pharmacological antagonists or siRNA could partially attenuate such changes in the IL-1β-induced nuclear translocation of β-catenin. DKK1, an inhibitor of β-catenin nuclear translocation, further inhibited the expression of type I and III collagens in cells treated with IL-1β plus CaSR antagonist. In summary, these data demonstrated that IL-1β-induced collagen I and III expressions in collecting duct cells might be partially mediated by CaSR and the downstream nuclear translocation of β-catenin.  相似文献

Members of the epidermal growth factor (EGF) family bind to ErbB (EGFR) family receptors which play an important role in the regulation of various fundamental cell processes including cell proliferation and differentiation. The normal rodent kidney has been shown to express at least three members of the ErbB receptor family and is a major site of EGF ligand synthesis. Polycystic kidney disease (PKD) is a group of diseases caused by mutations in single genes and is characterized by enlarged kidneys due to the formation of multiple cysts in both kidneys. Tubule cells proliferate, causing segmental dilation, in association with the abnormal deposition of several proteins. One of the first abnormalities described in cell biological studies of PKD pathogenesis was the abnormal mislocalization of the EGFR in cyst lining epithelial cells. The kidney collecting duct (CD) is predominantly an absorptive epithelium where electrogenic Na+ entry is mediated by the epithelial Na+ channel (ENaC). ENaC-mediated sodium absorption represents an important ion transport pathway in the CD that might be involved in the development of PKD. A role for EGF in the regulation of ENaC-mediated sodium absorption has been proposed. However, several investigations have reported contradictory results indicating opposite effects of EGF and its related factors on ENaC activity and sodium transport. Recent advances in understanding how proteins in the EGF family regulate the proliferation and sodium transport in normal and PKD epithelial cells are discussed here. Protein phosphorylation plays a critical role in the signaling pathways regulating water and solute transport in the distal renal tubule (i.e., renal collecting duct). A central mediator in this process is the antidiuretic peptide hormone arginine vasopressin, which regulates a number of transport proteins including water channel aquaporin-2 and urea transporters (UT-A1 and UT-A3). Within the past few years, tandem mass spectrometry-based proteomics has played a pivotal role in revealing global changes in the phosphoproteome in response to vasopressin signaling in the renal collecting duct. This type of large-scale ‘shotgun’ approach has resulted in an exponential increase in the number of phosphoproteins known to be regulated by vasopressin and has expanded on the established signaling mechanisms and kinase pathways regulating collecting duct physiology. This article will provide a brief background on vasopressin action, will highlight a number of recent quantitative phosphoproteomic studies in both native rat kidney and cultured collecting duct cells, and will conclude with a perspective focused on emerging trends in the field of phosphoproteomics.  相似文献

The mature renal medulla, the inner part of the kidney, consists of the medullary collecting ducts, loops of Henle, vasa recta and the interstitium. The unique spatial arrangement of these components is essential for the regulation of urine concentration and other specialized kidney functions. Thus, the proper and timely assembly of medulla constituents is a crucial morphogenetic event leading to the formation of a functioning metanephric kidney. Mechanisms that direct renal medulla formation are poorly understood. This review describes the current understanding of the key molecular and cellular mechanisms underlying morphological aspects of medulla formation. Given that hypoplasia of the renal medulla is a common manifestation of congenital obstructive nephropathy and other types of congenital anomalies of the kidney and urinary tract (CAKUT), better understanding of how disruptions in medulla formation are linked to CAKUT will enable improved diagnosis, treatment and prevention of CAKUT and their associated morbidity.  相似文献

Suction sampling is widely used to estimate arthropod abundance and diversity. To test the reliability of abundance data derived from suction sampling, we examined sampling efficiency across a wide range of arthropod groups and tested for effects of species traits, vegetation density, and differences between sites. Suction sampling efficiency was quantified by vacuuming an enclosed meadow area and subsequent removal of the turf, which was treated with heat extraction to collect the remaining arthropods. We obtained 250 pairs of suction and turf samples from seven grasslands with variable vegetation density. High suction sampling efficiencies between 49 and 86% were obtained for Auchenorrhyncha, Heteroptera, Araneida, Curculionoidea, Hymenoptera, and Diptera. In contrast, efficiencies were below 30% for Aphidae, Thysanoptera, Staphylinidae and other Coleoptera, and for soil arthropods such as Collembola, Isopoda, Diplopoda, and Formicidae. Efficiency varied significantly among habitats (sites) for most groups, often more than two‐fold. Surprisingly, sampling efficiency for Hymenoptera, Diplopoda, and Collembola increased with vegetation density, probably because aboveground activity of these taxa was higher in dense vegetation. Suction sampling was nearly twice as efficient for spiders living in the vegetation than for spiders living near the soil surface, and cursorial and large‐bodied spider species were more efficiently sampled than web‐builders and small species. Depending on the sampling effort, suction sampling missed between 49% (one sample) and 31% (250 samples) of the spider species present. Suction sampling efficiency varied more strongly among sites and among arthropod groups than previously recognized. Abundance data derived from suction sampling are strongly underestimated, especially for arthropods living near the soil surface. Thus, comparisons of abundance and diversity between sites should be restricted to vegetation‐dwelling species of the most efficiently sampled groups. The positive relationship of sampling efficiency with vegetation density demonstrates that variation in efficiency is mediated by arthropod behaviour.  相似文献

Pollination syndromes are defined as suites of floral traits evolved in response to selection imposed by a particular group of pollinators (e.g., butterflies, hummingbirds, bats). Although numerous studies demonstrated their occurrence in plants pollinated by radically different pollinators, it is less known whether it is possible to identify them within species pollinated by one functional pollinator group. In such a framework, we expect floral traits to evolve also in response to pollinator subgroups (e.g., species, genera) within that unique functional group. On this, specialised pollination systems represent appropriate case studies to test such expectations. Calceolaria is a highly diversified plant genus pollinated by oil‐collecting bees in genera Centris and Chalepogenus. Variation in floral traits in Calceolaria has recently been suggested to reflect adaptations to pollinator types. However, to date no study has explicitly tested that observation. In this paper, we quantitatively test that hypothesis by evaluating the presence of pollination syndromes within the specialised pollination system formed by several Calceolaria and their insect pollinators. To do so, we use multivariate approaches and explore the structural matching between the morphology of 10 Calceolaria taxa and that of their principal pollinators. Our results identify morphological matching between floral traits related to access to the reward and insect traits involved in oil collection, confirming the presence of pollinator syndromes in Calceolaria. From a general perspective, our findings indicate that the pollination syndrome concept can be also extended to the intra‐pollinator group level.  相似文献
