High speed video imaging microscopy and the pH-sensitive fluorophore2′,7′,-bis(2-carboxyethyl)-5(6)-carboxyfluorescein (BCECF)
were used to examine acid-base functions of beta-intercalated cells of the rabbit cortical collecting duct. The presence of
intercalated cells was established and the properties of apical and basolateral acid-base transporters assessed by monitoring
cell pH during acid loading and luminal and basolateral ion substitutions. We showed that treatment of beta-intercalated cells
with ammonium chloride (20 mm) induced a profound decrease of their intracellular pH from 6.98 ± 5.93 ± 0.08. pH recovery occurred after different lag
periods ranging between 2 to 15 min (0.22 ± 0.04 dpH/dt). We demonstrated that this pH recovery mechanism was independent
of basolateral Na+ and apical HCO−
3 and K+. It was also not affected by apical and basolateral addition of NEM, by basolateral DIDS and by apical application of the
H-KATPase inhibitor SCH28080. The process of pH recovery was however, critically dependent on basolateral HCO−
3. These results are best explained by acid-induced insertion and/or activation of chloride-bicarbonate exchangers that are
functional properties with their apical analogues.

Members of the epidermal growth factor (EGF) family bind to ErbB (EGFR) family receptors which play an important role in the regulation of various fundamental cell processes including cell proliferation and differentiation. The normal rodent kidney has been shown to express at least three members of the ErbB receptor family and is a major site of EGF ligand synthesis. Polycystic kidney disease (PKD) is a group of diseases caused by mutations in single genes and is characterized by enlarged kidneys due to the formation of multiple cysts in both kidneys. Tubule cells proliferate, causing segmental dilation, in association with the abnormal deposition of several proteins. One of the first abnormalities described in cell biological studies of PKD pathogenesis was the abnormal mislocalization of the EGFR in cyst lining epithelial cells. The kidney collecting duct (CD) is predominantly an absorptive epithelium where electrogenic Na+ entry is mediated by the epithelial Na+ channel (ENaC). ENaC-mediated sodium absorption represents an important ion transport pathway in the CD that might be involved in the development of PKD. A role for EGF in the regulation of ENaC-mediated sodium absorption has been proposed. However, several investigations have reported contradictory results indicating opposite effects of EGF and its related factors on ENaC activity and sodium transport. Recent advances in understanding how proteins in the EGF family regulate the proliferation and sodium transport in normal and PKD epithelial cells are discussed here. This study reports the presence of large mucous cells lining the large renal collecting ducts and opisthonephric ducts of the Australian freshwater catfishes Tandanus tandanus and Tandanus tropicanus. Histologic examination of the renal tissue from four other species of Australian freshwater catfishes suggests that this feature is unique to the two species of Tandanus studied. The potential functions of renal mucous secretion in T. tandanus and T. tropicanus are discussed. 相似文献

Indium‐tin‐oxide (ITO) free polymer solar cells prepared by ink jet printing a composite front electrode comprising silver grid lines and a semitransparent PEDOT:PSS conductor are demonstrated. The effect of grid line density is explored for a large series of devices and a careful modeling study enabling the identification of the most rational grid structure is presented. Both optical and light beam induced current (LBIC) mapping of the devices are used to support the power loss model and to follow the evolution of the performance over time. Current generation is found to be evenly distributed over the active area initially progressing to a larger graduation in areas with different performance. Over time coating defects also become much more apparent in the LBIC images. 相似文献

Water channel aquaporin-2 (AQP2) strictly regulates body water homeostasis in mammals. Trafficking of AQP2 to the apical membrane is critical to the reabsorption of water in renal collecting ducts. Controlled apical positioning of AQP2 suggests the interaction of AQP2 with other proteins. To isolate AQP2-binding proteins, immunoaffinity chromatography of extracts from rat kidney papilla was performed using a column covalently coupled with anti-AQP2 antibody. Using this method 42-kDa protein was purified and subsequently identified as beta- and gamma-isoforms of actin by two-dimensional gel analysis and matrix-assisted laser desorption-ionization time-of-flight mass spectrometry. AQP2 was indeed coimmunoprecipitated with actin from cell lysates of rat kidney papilla. In addition, surface plasmon resonance analyses showed that the C-terminal fragment of AQP2 strongly bound to actin and the K(D) value was 3.18x10(-8)M. In this experiment we have elucidated the direct binding of channel protein AQP2 to cytoskeletal protein actin, providing a novel mechanism for trafficking of not only AQP2 but also recycling channel proteins. 相似文献

Summary. Heavy rain causes major problems with using pitfall traps to collect ants and other surface-active arthropods. A simple design for a rain-resistant pitfall trap is described. The trap is constructed by grinding a hole in the side of a plastic vial and then covering it with fine-gage wire screen. This hole allows excess water to drain out the side of the trap. The distribution of tropical plant and animal diversity is still poorly documented, especially at spatial resolutions of practical use for conservation. In the present study, we evaluated the level to which geographical incomplete data availability of species occurrence affects the perception of biodiversity patterns (species richness and endemism) among pteridophytes in Bolivia. We used a data base of Bolivian pteridophytes (27,501 records), divided it into three time periods (1900–70, up to 1990 and up to 2006), and created grid-files at 15'-resolution for species richness and endemism. For each of these biodiversity properties we estimated the species richness (Chao 2) and the index of sampling completeness (C index) per grid, and then all these variables at both species richness and endemism were correlated. Patterns of richness were fairly consistent along all periods; the richest areas were placed along the humid-montane forest, even though they were strongly influenced by collecting intensity. Endemism had a lower degree of correlation with collecting intensity, but varied much more strongly through time than species richness. According to the C index, which gives the ratio between estimated (by Chao 2) and recorded values of species richness and endemism, both biodiversity properties tended to be undersampled in the richest grid cells. Inter-temporal correlations showed sharper differences of correlations for endemism than species richness. Consequently, already in 1970, botanists had a correct idea of the spatial distribution of pteridophyte richness in Bolivia (even though the magnitude was grossly underestimated). Summary The light and electron microscopic structure of the high-endothelial postcapillary venules of lymph nodes were studied in the mouse, rat, guinea pig, and rabbit. The venules were most frequently found in the mouse and rat. In all species, they reached their highest degree of differentiation in the paracortical area. The morphology in the light microscope was identical in all four species. The venules in the rat and mouse were surrounded by a cuff of concentrically-arranged lymphocytes, which was rarely seen in the guinea pig and rabbit.The ultrastructure of the high-endothelial cells in all four species was very complex; a prominent Golgi apparatus was present which often occupied large parts of the cytoplasm. Coated and uncoated vesicles originating in the Golgi apparatus often permeated the cytoplasm. These vesicles emptied their contents into the extracellular space after fusion with the plasma membranes.Numerous lymphocytes traversed vessel walls. During their passage, they were always located between, not inside the high-endothelial cells. 相似文献
