In the past decade, the availability of genetically modified animals has enabled the discovery of interesting roles for phosphatidylinositol 3‐kinase‐γ (PI3Kγ) and ‐δ (PI3Kδ) in different cell types orchestrating innate and adaptive immune responses. Therefore, these PI3K isoforms appear to be attractive drug targets for the treatment of diseases caused by unrestrained immune reactions. Currently, pharmacological targeting of PI3Kγ and/or PI3Kδ represents one of the most promising challenges for companies interested in the development of novel safe treatments for inflammatory diseases. In this review we provide a general outline of PI3Kγ‐ and PI3Kδ‐specific functions in distinct subsets of inflammatory cells. We also discuss the therapeutic impact of novel compounds targeting PI3Kγ, PI3Kδ or both, in mouse models of autoimmune disorders (systemic lupus erythematosus (SLE) and rheumatoid arthritis), respiratory diseases (allergic asthma and chronic obstructive pulmonary disease) and cardiovascular dysfunctions (atherosclerosis and myocardial infarction). 相似文献

The pathogenesis of psoriasis, an immune-mediated chronic inflammatory skin disease, remains unclear. Studies have shown an association between psoriasis and intestinal inflammation; in this context, the influence of the gut microbiota on the immune response of psoriasis has become a focus of recent research. The present research evaluated the composition and diversity of the gut microbiota of 21 participants with psoriasis from a Brazilian referral dermatology service compared to 24 healthy controls. A stool sample was collected from each participant at the time of inclusion in the study, and the samples were analysed by sequencing the 16S rRNA gene. The recruitment of research participants involved matching between groups by sex, age, body mass index, comorbidities and smoking and the exclusion of several criteria that could potentially influence the gut microbiota and the interpretation of the data. There was an increase in the Dialister genus and Prevotella copri species in patients with psoriasis compared to the control group. A reduction in the Ruminococcus, Lachnospira and Blautia genera, as well as in the Akkermansia muciniphila species, was also verified in the psoriasis group compared to the control group. Furthermore, patients with psoriasis exhibited less gut microbiota diversity than controls. 相似文献

Use of recombinant interferon gamma in pediatric patients with advanced juvenile chronic arthritis 总被引:1,自引:0,他引:1
Recombinant interferon (IFN) gamma was used in 10 patients,6 to 15 years old, with juvenile chronic arthritis (JCA) for5 to 11 years, resistant or with severe side effects toother treatments. Six patients had systemic JCA and 4started as pauciarticular. Three of the latterbecame polyarticular. Treatment schedule was 50000 IU–kg daily for 4 weeks, then 3 times per week for 3 months andtwice a week up to 2 years. Eight cases had favourableclinical response. Prolonged steroid regime could besuspended in 7/8 cases who previously received it. Twopatients with systemic JCA did not respond to IFNtreatment. Side effects were fever (9), headache (8),chills (6), distal cyanosis, hypotension, leukopenia andmyalgia (2), and vomiting (1). All were mild or moderate. IFN gamma was more tolerable than other drugs and seems to be beneficial for patients with JCA resistant to other treatments. 相似文献

Several studies in animals and humans have shown that copper metabolism could be affected by inflammation or by corticosteroids.
The relative importance of these two factors, often imbedded in clinical practice, was assessed by investigating the effects
of acute prednisolone administration (30 mg/kg, ip) on healthy and adjuvant arthritis rats. Plasma copper levels were significantly
higher in arthritic rats compared to healthy animals, whereas there was a slight, but nonsignificant increase in liver copper.
Acute prednisolone administration in healthy rats resulted in a significant increase in plasma copper (10–15%) as early as
4 h after corticosteroid administration, which was maintained for 12 h. In arthritic rats, the response was much higher (25–40%),
but somewhat delayed and shorter. Liver copper was not clearly modified by prednisolone treatment in both groups. This time-controlled
study showed that acute prednisolone administration increased plasma copper in both healthy and arthritic rats, but in different
ways, indicating that inflammation and corticosteroids may act synergistically. 相似文献

Human fibroblast-like synoviocytes (FLSs) play a role in joint synovial inflammation in rheumatoid arthritis (RA). Some evidence indicates that particulate matter (PM) in air pollution could contribute to the progression of RA. However, more research is needed to clarify this relationship. Up-regulation of cyclooxygenase (COX)-2 and its metabolite prostaglandin E2 (PGE2) are implicated in various inflammatory diseases. Resveratrol, a polyphenol found mainly in grapes and red wine, has antioxidant and anti-inflammatory activities. In the present study, we demonstrated that resveratrol reduced PM-induced COX-2/PGE2 expression in human FLSs, and attenuated PM-enhanced NADPH oxidase activity and ROS generation. In addition, PM induced Akt, ERK1/2, or p38 MAPK activation, which was inhibited by resveratrol. Finally, we demonstrated that PM enhanced NF-κB p65 phosphorylation and the NF-κB promoter activity, which were reduced by pretreatment with a ROS inhibitor or resveratrol. Thus, we concluded that resveratrol functions as a suppressor of PM-induced inflammatory signaling pathways by inhibiting COX-2/PGE2 expression. 相似文献

甲氨蝶呤(MTX)是治疗类风湿关节炎(RA)的首选药物,但 20%~40% 的 RA 患者服用 MTX 无效。MTX 的血浆半衰期短, 其浓度与疗效无相关性。MTX 在红细胞、滑膜细胞等细胞内转化为多聚谷氨酸化甲氨蝶呤(MTXPGs),在细胞内的滞留时间较长,且与 叶酸途径中关键酶的亲和力更强、抗炎作用也更强,因此选用红细胞内 MTXPGs 浓度来预测治疗效果及安全性成为目前研究的重点。综述 MTXPGs 与 RA 疗效和不良反应的相关性研究进展,并对影响 MTX 作用的相关因素进行分析,为临床合理用药提供参考。 相似文献
