Aims: Water quality at two Florida beaches was compared using faecal indicator bacteria measurements, microbial source tracking (MST) methods for detecting human source pollution and the assessment of pathogen presence. These values were also compared before and after remediation of wastewater infrastructure at one beach. Methods and Results: Faecal coliforms, Escherichia coli and enterococci were enumerated in estuarine water and sediment samples. PCR assays for the human‐associated esp gene of Enterococcus faecium and human polyomaviruses (HPyVs) were used to detect human sewage. Culturable Salmonella and enteric viruses were also analysed. MST identified human sewage contamination at one beach, leading to repair of a sewer main and relocation of portable restrooms. Exceedances of Florida recreational water regulatory standards were significantly reduced after remediation (by 52% for faecal coliforms and 39% for enterococci), and the frequency of detection of MST markers decreased. Coxsackie virus B4 and HPyVs were codetected following a major sewage spill, but Salmonella was not detected during the study. Conclusions: These data indicate that infrastructure remediation significantly reduced pollution from human sewage at the impacted beach. Significance and Impact of the Study: A comprehensive microbial water quality study that can identify contamination sources through the use of MST markers and close collaboration with local/and state agencies can result in tangible actions to improve recreational water quality and safety.  相似文献

AIMS: To compare survival of Escherichia coli and faecal coliforms (FC) in bovine faeces deposited in a pasture or incubated in a controlled laboratory environment at temperatures within the same range. METHODS AND RESULTS: Faecal samples from three cow herds were deposited as shaded and nonshaded cowpats in a field and incubated in a laboratory for one month at 21.1, 26.7 and 32.2 degrees C. Both FC and E. coli concentrations increased as much as 1.5 orders of magnitude both in the field and in the laboratory during the 1st week and subsequently decreased. In shaded cowpats, the die-off of E. coli and FC was significantly slower, and the proportion of E. coli in FC was significantly larger as compared with nonshaded cowpats. The die-off was faster in the field than in the laboratory at similar temperatures. CONCLUSIONS: FC and E. coli die-off rates were substantially lower in laboratory conditions than in the field within the same range of temperatures. SIGNIFICANCE AND IMPACT OF THE STUDY: This study underscores the importance of field data on survival of manure-borne FC and E. coli, and indicates that laboratory die-off rates have to be corrected to be used for field condition simulations.  相似文献

The incidence of serotypes of Salmonella in three types of environmental water (sea, river and fresh reservoirs) from north-east Spain was investigated. The study was performed at specific sampling locations during the summer for a period of five years (1992–1996). A total of 823 strains were isolated and 55 different serotypes were identified, 42 were recovered from sea water, 32 from river water and 12 from freshwater reservoirs. The most frequently isolated serotypes coincided with those involved in clinical cases in the area studied. Salmonella enteritidis was the most common (111 isolates), it was found in all types of water, although most predominantly in sea water (16.1% of the isolates). This serotype, together with S. hadar, significantly increased in frequency during the five year study period. The most frequent serotypes in river water and freshwater reservoirs were S. virchow (9.5%) and S. mikawasima (23.8%) respectively. Significant differences were assessed in the indicator organism densities between the samples with serotypes of clinical significance (S. enteritidis, S. infantis, S. typhimurium, S. virchow and S. paratyphi B) and those without clinical significance. Therefore their presence in all environmental waters may be of epidemiological significance.  相似文献

The freshwater mussel Anodonta cygnea and four marine shellfish (mussels, Mytilus edulis; cockles, Cerastoderma edule; clams, Mya arenaria; Scrobicularia plana) from a total of six sites were surveyed for Escherichia coli, Clostridium perfringens, faecal streptococci, 25 and 37 °C coliforms, 25 °C and 37 °C total viable numbers and fluorescent pseudomonads. The A. cygnea from an urban lake contained greater numbers of the faecal indicator bacteria than animals from a rural lake. There were also differences in the other bacterial counts and these were discussed with respect to bacterial parameter and animal characteristics. When freshwater mussels were transferred from the city site to the rural site for 24 h the load of faecal indicator bacteria was eliminated or significantly reduced. Other bacterial types took longer to become stabilised. Loss of indicator bacteria from Anodonta was also demonstrated using cleansing in the laboratory. Very high bacterial numbers were found in some marine molluscs notably Scrobicularia plana and most shellfish contained significant numbers of the three faecal indicator bacteria at every sampling occasion. The relationship between bacterial types was discussed and it was concluded that in both freshwater and marine animals the bacterial numbers were determined more by sampling site than by species of shellfish.  相似文献

Aim: A new medium, EC‐Blue‐10, containing chromogenic and fluorogenic substrates, KNO3 and sodium pyruvate has been developed for the rapid simultaneous detection and enumeration of total coliforms and Escherichia coli in water. Methods and Results:  Two evaluations of EC‐Blue‐10 were carried out. Firstly, EC‐Blue‐10 was compared with Colilert‐MPN for 96 water samples using MPN for total coliforms and E. coli. Secondly, the detection of coliforms and E. coli were compared using 2400 tubes of EC‐Blue‐10 and Colilert‐MPN. The regression coefficients between EC‐Blue‐10 and Colilert‐MPN for total coliforms and E. coli were 0·91 and 0·89, respectively. For the detection results, the Cohen’s kappa values between the two media were 0·79 for coliforms and 0·72 for E. coli. Conclusions: EC‐Blue‐10 is almost same as Colilert‐MPN for the detection of coliforms and E. coli in surface waters. Further evaluation for EC‐Blue‐10 is needed to verify in different geographical areas. Significance and Impact of the Study: EC‐Blue‐10 is useful method for the rapid and simultaneous detection of total coliforms and E. coli in water sample.  相似文献

Modelling the faecal coliform concentrations in the Bilbao estuary   总被引:1,自引:0,他引:1  
Since 1984, an ambitious sewerage scheme is being implemented in the metropolitan area of Bilbao City (North Spain) in order to recover the environment of the highly polluted Bilbao estuary. To check the efficiency of this scheme, the performance of the sewer system and the water quality in the Bilbao estuary and beaches have been simulated during a future hypothetical bathing season. The system has been tested under dry and wet weather conditions, using selected historical rainfall records, in order to evaluate the effect of the faecal pollution arising from Combined Sewer Overflows on the water quality of the beaches located in the outer part of the estuary. The results obtained indicate that under dry weather and optimal operational conditions of the sewer and treatment works, the mandatory European standards on bathing waters will be met in all beaches. The discharges caused by storm overflows will increase the faecal contamination in the beaches compromising the compliance with the guideline standards. Therefore, a disinfection of the treatment works effluent, the main faecal discharge to the estuary, may be needed.  相似文献

AIMS: To investigate changes in the bacterial population and the safety of the biofilm at the end of the drinking water distribution system in Seoul (Korea), selective media and bacterial community analyses were applied to a semi-pilot galvanized iron pipe (GIP) model. METHODS AND RESULTS: No total coliforms or faecal streptococci were detected on m-Endo or m-Enterococcus agar. No Salmonella spp. and Shigella spp. were detected on bismuth sulphite agar or Hektoen enteric agar, respectively. The latter two media detected coliforms, where m-Endo was negative. Biofilm formation started within 1 week (ca 104 CFU cm(-2)) and exceeded 105 CFU cm(-2) within 6 weeks. Although the fatty acid methyl ester analysis revealed dynamic changes in bacterial composition, Micrococcus, Bacillus, and Pseudomonas spp. were persistent members of the biofilm community. Micrococcus spp. was detected most frequently and in high numbers. CONCLUSIONS: Coliforms and Enterococcus species can be recovered from biofilms in water distribution systems. SIGNIFICANCE AND IMPACT OF THE STUDY: This study illustrates the role of biofilms in the chronic deterioration of the water-distribution system in Seoul (Korea).  相似文献

We studied the antibiotic sensitivity of injured coliforms isolated from drinking water of La Plata, Argentina. The antibiotic sensitivity test by the agar diffusion method were proved in: Klebsiella oxytoca (14 strains), Enterobacter aerogenes (4 strains) and Enterobacter cloacae genomic group 3 (14 strains). We found that while these impaired total coliforms were sensitive to piperacillin-tazobactam (TAZ), netilmicin (NTL), ofloxacin (OFLX), and norfloxacin (NFLX) (100%), they had resistant to aminopenicillin-sulbactam (AMS) and nitrofurantoin (NIT) (100%). The resistance to antibiotics demonstrated in these strains would point to the need to promote a rational and judicious use of antimicrobial agents while at the same time implementing a program of active vigilance aimed at ensuring the highest quality of drinking water throughout the system.  相似文献

AIM: To determine the survival of Escherichia coli O157:H7 in dairy wastewater from on-site holding lagoons equipped with or without circulating aerators. METHODS AND RESULTS: Survival was monitored in dairy lagoon microcosms equipped with or without scale-size circulators. Both laboratory strains of E. coli O157:H7 and an isolate of E. coli H7 from wastewater had poor survival rates and none proliferated in water from waste lagoons with or without circulators. Furthermore, the decline of E. coli O157:H7 was not enhanced in those microcosms equipped with circulators. Strain variation in survival was observed in both circulated and settling waters. The decline rate of E. coli O157:H7 Odwalla strain increased proportionately with the inoculum load. Escherichia coli failed to establish itself in wastewater even after four sequential inoculations simulating continuous faecal input into the lagoon. The native aerobic bacteria survived longer with a decimal reduction time of 21.3 days vs either introduced or native E. coli, which declined rapidly with decimal reduction time of 0.5-9.4 days. CONCLUSIONS: Escherichia coli O157:H7 failed to establish and proliferate in dairy wastewater microcosms equipped with or without circulating aerators. SIGNIFICANCE AND IMPACT OF THE STUDY: This study furthers our knowledge of pathogen survival in wastewater, and suggests that proper management of wastewater before its use in irrigation is essential to reduce pathogen transfer to crops.  相似文献

AIMS: To evaluate some physiological characteristics of the Enterobacteriaceae isolated from Pecorino cheese. METHODS AND RESULTS: The production of organic acids, secondary volatile compounds, biogenic amines (BA) and the lipolytic and proteolytic activities of Citrobacter braakii, Enterobacter sakazakii, Escherichia coli, Kluyvera spp., Salmonella enterica ssp. arizonae and Serratia odorifera strains were determined in skim milk after 48 h of fermentation at 30 degrees C. The proteolytic activity observed only in Ser. odorifera and Kluyvera spp. was confirmed by the peptide profiles of the pH 4.6-insoluble fraction using RP-HPLC; however, the lipase activity was evidenced in all the isolates of E. coli, Kluyvera spp. and Salm. enterica ssp. arizonae. During fermentation, all the strains utilized citric acid and produced significant quantities of putrescine followed by histamine, spermine and spermidine as well as acetic and lactic acid. Moreover, the major volatile compounds produced were ethanol, 2,3-butanedione, 3-hydroxy-2-butanone, 2-heptanone and acetone. CONCLUSIONS: The Enterobacteriaceae of dairy origin possess many metabolic activities that could affect the sensory quality of the cheese in which they grow during ripening. SIGNIFICANCE AND IMPACT OF THE STUDY: The important physiological characteristics possessed by Enterobacteriaceae confirm the complexity of the microbiota of Pecorino Abruzzese cheese, which influences the typical sensory properties of this product.  相似文献
