This study experimentally determined the turnover rates of δ13C and δ15N as a function of growth and metabolism and isotopic fractionation for different tissues in captive populations of red rock lobster (Jasus edwardsii) and blue cod (Parapercis colias). Isotopic turnover was estimated using the model of Hesslein et al. [Hesslein, R., Hallard, K., Ramlal, P., 1993. Replacement of sulfur, carbon, and nitrogen in tissue of growing broad whitefish (Coregonus nasus) in response to a change in diet traced by δ34S, δ13C, and δ15N. Can. J. Fish. Aquat. Sci. 50, 2071–2076.]. Isotopic fractionations relative to diet differed among tissues and isotopes. Lobster muscle was more enriched than hemolymph and blue cod fin tissue was more enriched than blood for δ13C and δ15N. The metabolic component of turnover accounted for > 90% of the total isotopic turnover in lobster tissues and 30%–60% in blue cod tissues. Lobster muscle (half-life 147 d) and hemolymph (half-life 117 d) turnover rates were not significantly different but were faster than turnover rates of blue cod tissues. Whole blood, blood plasma fraction, and the blood cellular fraction had similar turnover rates; the whole blood half-life was 240 d for blue cod. Measuring turnover in larger, slower growing animals allowed for a more precise estimate of the metabolic component of isotopic turnover than in fast growing animals in which change is predominantly the result of dilution through growth. The differences in fractionation values among tissues observed here demonstrate that using generic trophic fractionation values would introduce error into diet reconstruction or migration studies. We demonstrate that a modified version of Hesslein et al.'s [Hesslein, R., Hallard, K., Ramlal, P., 1993. Intraguild predation--competition and predation by the same antagonist--is widespread, but its evolutionary consequences are unknown. Intraguild prey may evolve antipredator defenses, superior competitive ability on shared resources, or the ability to use an alternative resource, any of which may alter the structure of the food web. We tested for evolutionary responses by threespine stickleback to a benthic intraguild predator, prickly sculpin. We used a comparative morphometric analysis to show that stickleback sympatric with sculpin are more armored and have more limnetic-like body shapes than allopatric stickleback. To test the ecological implications of this shift, we conducted a mesocosm experiment that varied sculpin presence and stickleback population of origin (from one sympatric and one allopatric lake). Predation by sculpin greatly increased the mortality of allopatric stickleback. In contrast, sculpin presence did not affect the mortality of sympatric stickleback, although they did have lower growth rates suggesting increased nonpredatory effects of sculpin. Consistent with their morphology, sympatric stickleback included more pelagic prey in their diets, leading to depletion of zooplankton in the mesocosms. These findings suggest that intraguild prey evolution has altered food web structure by reducing both predation by the intraguild predator and diet overlap between species.  相似文献

Because most clonal vertebrates have hybrid genomic constitutions, tight linkages are assumed among hybridization, clonality, and polyploidy. However, predictions about how these processes mechanistically relate during the switch from sexual to clonal reproduction have not been validated. Therefore, we performed a crossing experiment to test the hypothesis that interspecific hybridization per se initiated clonal diploid and triploid spined loaches (Cobitis) and their gynogenetic reproduction. We reared two F1 families resulting from the crossing of 14 pairs of two sexual species, and found their diploid hybrid constitution and a 1:1 sex ratio. While males were infertile, females produced unreduced nonrecombinant eggs (100%). Synthetic triploid females and males (96.3%) resulted in each of nine backcrossed families from eggs of synthesized diploid F1s fertilized by haploid sperm from sexual males. Five individuals (3.7%) from one backcross family were genetically identical to the somatic cells of the mother and originated via gynogenesis; the sperm of the sexual male only triggered clonal development of the egg. Our reconstruction of the evolutionary route from sexuality to clonality and polyploidy in these fish shows that clonality and gynogenesis may have been directly triggered by interspecific hybridization and that polyploidy is a consequence, not a cause, of clonality.  相似文献

The dynamics of host susceptibility to parasites are often influenced by trade‐offs between the costs and benefits of resistance. We assayed changes in the resistance to three viruses in six lines of Escherichia coli that had been evolving for almost 45,000 generations in their absence. The common ancestor of these lines was completely resistant to T6, partially resistant to T6* (a mutant of T6 with altered host range), and sensitive to λ. None of the populations changed with respect to resistance to T6, whereas all six evolved increased susceptibility to T6*, probably ameliorating a cost of resistance. More surprisingly, however, the majority of lines evolved complete resistance to λ, despite not encountering that virus during this period. By coupling our results with previous work, we infer that resistance to λ evolved as a pleiotropic effect of a beneficial mutation that downregulated an unused metabolic pathway. The strong parallelism between the lines implies that selection had almost deterministic effects on the evolution of these patterns of host resistance. The opposite outcomes for resistance to T6* and λ demonstrate that the evolution of host resistance under relaxed selection cannot be fully predicted by simple trade‐off models.  相似文献

本文对益生菌在动物及人体上的降胆固醇功能及可能机理进行了综述。益生菌是一类能够对人体健康起到促进作用的活体微生物。现已发现某些乳杆菌、双歧杆菌和肠球菌属的一些菌株具有降低血清胆固醇水平的能力。由于实验动物和人体在生理学上存在差异,因此同一菌株在实验动物和人体上获得的结论可能不同。关于益生菌降胆固醇功能的机理,研究者提出了不同的假说。这些假说包括:(1)益生菌将胆固醇吸收至细胞膜或细胞质中;(2)益生菌将胆固醇吸附到细胞表面;(3)胆固醇和游离胆盐在酸性环境下发生共沉淀;(4)结合胆盐被益生菌的胆盐水解酶水解成了游离胆盐,后者具有较低的溶解度,不易被肠道回收;(5)胆酸被益生菌的荚膜胞外多糖黏附到了细胞表面;(6)益生菌发酵肠道食源性未消化的碳水化合物产生丙酸,后者能够抑制肝脏胆固醇的生物合成,从而导致血清胆固醇水平降低;(7)益生菌通过下调NPC1L1蛋白基因表达来降低小肠细胞对胆固醇的吸收;(8)益生菌抑制胆固醇乳化胶束的形成。这些假说将为我们认识益生菌的降胆固醇机制及潜在降胆固醇功能菌株的筛选提供了依据。  相似文献

Optimal experiment design for parameter estimation (OED/PE) has become a popular tool for efficient and accurate estimation of kinetic model parameters. When the kinetic model under study encloses multiple parameters, different optimization strategies can be constructed. The most straightforward approach is to estimate all parameters simultaneously from one optimal experiment (single OED/PE strategy). However, due to the complexity of the optimization problem or the stringent limitations on the system's dynamics, the experimental information can be limited and parameter estimation convergence problems can arise. As an alternative, we propose to reduce the optimization problem to a series of two-parameter estimation problems, i.e., an optimal experiment is designed for a combination of two parameters while presuming the other parameters known. Two different approaches can be followed: (i) all two-parameter optimal experiments are designed based on identical initial parameter estimates and parameters are estimated simultaneously from all resulting experimental data (global OED/PE strategy), and (ii) optimal experiments are calculated and implemented sequentially whereby the parameter values are updated intermediately (sequential OED/PE strategy).This work exploits OED/PE for the identification of the Cardinal Temperature Model with Inflection (CTMI) (Rosso et al., 1993). This kinetic model describes the effect of temperature on the microbial growth rate and encloses four parameters. The three OED/PE strategies are considered and the impact of the OED/PE design strategy on the accuracy of the CTMI parameter estimation is evaluated. Based on a simulation study, it is observed that the parameter values derived from the sequential approach deviate more from the true parameters than the single and global strategy estimates. The single and global OED/PE strategies are further compared based on experimental data obtained from design implementation in a bioreactor. Comparable estimates are obtained, but global OED/PE estimates are, in general, more accurate and reliable.  相似文献

目的:超声波法优选苦豆子中总黄酮的提取工艺。方法:以超声波为提取方法,以总黄酮提取率为考察指标,选取乙醇浓度(体积分数)、加溶剂倍量和超声时间为考察因素,采用均匀实验和正交实验,优选苦豆子中总黄酮的提取工艺。结果:影响总黄酮提取率各因素主次为:ACB,确定最佳提取工艺为A2B3C3,即70%乙醇,加溶剂40倍量,超声提取75 min。结论:采用该工艺提取苦豆子中总黄酮,提取率较高且稳定,说明该工艺合理可行。  相似文献

典型高寒植物生长繁殖特征对模拟气候变化的短期响应   总被引:1,自引:0,他引:1  
高寒植物的生长繁殖策略对气候变化的响应十分敏感但研究较少。在青藏高原东北的祁连山南麓坡地,于2007年沿3200~3800m海拔进行了植被的等距双向移栽实验并研究了典型高寒植物的生长繁殖策略对模拟气候变化的响应。结果表明,移栽样线年平均气温随海拔升高的递减率为0.51℃/100m。高寒植物移栽到高海拔后,其株高、基叶数、最大(小)叶面积等生长性状指标均发生显著变化,呈现出在3400m海拔处最高,其余3海拔处较低的趋势;而生殖枝数、花数和有性繁殖投入等生殖策略的响应则不明显,但具有随海拔升高而降低,最后在3800m处升高的变化。结果印证了气候变化对高寒植物生长性状的影响比生殖策略快速的假说。  相似文献

The degree to which coral reef ecosystems will be impacted by global climate change depends on regional and local differences in corals’ susceptibility and resilience to environmental stressors. Here, we present data from a reciprocal transplant experiment using the common reef building coral Porites lobata between a highly fluctuating back reef environment that reaches stressful daily extremes, and a more stable, neighbouring forereef. Protein biomarker analyses assessing physiological contributions to stress resistance showed evidence for both fixed and environmental influence on biomarker response. Fixed influences were strongest for ubiquitin‐conjugated proteins with consistently higher levels found in back reef source colonies both pre and post‐transplant when compared with their forereef conspecifics. Additionally, genetic comparisons of back reef and forereef populations revealed significant population structure of both the nuclear ribosomal and mitochondrial genomes of the coral host (FST = 0.146 P < 0.0001, FST = 0.335 P < 0.0001 for rDNA and mtDNA, respectively), whereas algal endosymbiont populations were genetically indistinguishable between the two sites. We propose that the genotype of the coral host may drive limitations to the physiological responses of these corals when faced with new environmental conditions. This result is important in understanding genotypic and environmental interactions in the coral algal symbiosis and how corals may respond to future environmental changes.  相似文献

Understanding soil carbon fractions and their responses to the global warming is important for improving soil carbon management of natural altitudinal forest ecosystem. In this study, the contents of soil total organic carbon (SOC), soil labile organic carbon (LOC), and microbial biomass carbon (MBC) in soil upper layers (0–20 cm) were measured along a natural altitudinal transect in the north slope of Changbai Mountain. The results showed that under natural conditions the contents of SOC and LOC were largest in Betula ermanii forest (altitude 1996 m), moderate in spruce-fir forest (altitude 1350 m), and smallest in Korean pine mixed broad-leaf tree forest (altitude 740 m). MBC contents in different forest ecosystems decreased in the order of Betula ermanii forest, Korean pine mixed broad-leaf tree forest, and dark coniferous forest. In addition, the responses of SOC, LOC, and MBC to soil warming were conducted by relocating intact soil cores from high- to low-elevation forests for one year. As expected, the soil core relocation caused significant increase in soil temperature but made no significant effect on soil moisture. After one year incubation, soil relocation significantly decreased SOC contents, whereas the contents of LOC, MBC, and the ratios of LOC to SOC and MBC to SOC increased.  相似文献
