不同砧穗组合葡萄植株对部分根区干旱的生理生化响应   总被引:1,自引:0,他引:1  
利用自制木箱对嫁接在3309C、420A和110R砧木上的玛瓦斯亚葡萄(M)进行双侧根区交替灌溉(AI)和单侧灌溉(UI)两种水分胁迫处理,以探讨不同葡萄砧穗组合对干旱的适应能力.结果表明:水分胁迫导致葡萄叶片ABA浓度大幅度增加,AI和UI的3种组合平均分别提高了267.5%和394.7%,而气孔导度和蒸腾速率显著下降.UI处理的3种组合叶片SOD和CAT活性显著增加,脯氨酸(Pro)含量极显著增加,且均以M/110R增幅较大,M/3309C增幅较小;而AI处理的SOD和CAT活性增加较小,但Pro含量显著增加.干旱胁迫导致葡萄叶片质膜透性、MDA和H2O2含量极显著增加,以M/3309C增加最多,M/110R增加较少.不同砧穗组合适应干旱逆境的能力主要取决于砧木品种,110R适应干旱逆境的能力强于420A和3309C,其对应的嫁接植株的生长量亦呈同样趋势.与根区单侧灌溉相比,双侧根区交替灌溉对植株造成的伤害较小,是可推广的节水灌溉方式.  相似文献

Abstract Identification of aphid species is always difficult due to the shortage of easily distinguishable morphological characters. Aphid genus Toxoptera consists of species with similar morphology and similar to Aphis in most morphological characters except the stridulatory apparatus. DNA barcodes with 1 145 bp sequences of partial mitochondrial cytochrome‐coxidase I (COI) genes were used for accurate identification of Toxoptera. Results indicated mean intraspecific sequence divergences were 1.33%, whereas mean interspecific divergences were greater at 8.29% (0.13% and 7.79% if T. aurantii 3 and T. aurantii 4 are cryptic species). Sixteen samples were distinguished to four species correctly by COI barcodes, which implied that DNA barcoding was successful in discrimination of aphid species with similar morphology. Phylogenetic relationships among species of this genus were tested based on this portion of COI sequences. Four species of Toxoptera assembled a clade with low support in maximum‐parsimony (MP) analysis, maximum‐likelihood (ML) analysis and Bayesian phylogenetic trees, the genus Toxoptera was not monophyletic, and there were two sister groups, such as T. citricidus and T. victoriae, and two clades of T. aurantii which probably presented cryptic species in the genus.  相似文献

In this study, we used real-time quantitative PCR (RT-qPCR) to evaluate the expression of 32 genes of Xanthomonas axonopodis pv. citri related to pathogenicity and virulence that are also involved in copper detoxification. Nearly all of the genes were up-regulated, including copA and copB. Two genes homologous to members of the type II secretion system (xcsH and xcsC) and two involved in the degradation of plant cell wall components (pglA and pel) were the most expressed in response to an elevated copper concentration. The type II secretion system (xcs operon) and a few homologues of proteins putatively secreted by this system showed enhanced expression when the bacteria were exposed to a high concentration of copper sulfate. The enhanced expression of the genes of secretion II system during copper stress suggests that this pathway may have an important role in the adaptative response of X. axonopodis pv. citri to toxic compounds. These findings highlight the potential role of these genes in attenuating the toxicity of certain metals and could represent an important means of bacterial resistance against chemicals used to control diseases.  相似文献

Plant grafting is an important technique for horticultural and silvicultural production. However, many rootstock plants suffer from undesirable lateral bud outgrowth, low grafting success rates or poor rooting. Here, we used a root‐predominant gene promoter (SbUGT) to drive the expression of a tryptophan‐2‐monooxygenase gene (iaaM) from Agrobacterium tumefaciens to increase auxin levels in tobacco. The transgenic plants, when used as a rootstock, displayed inhibited lateral bud outgrowth, enhanced grafting success rate and improved root initiation. However, root elongation and biomass of SbUGT::iaaM transgenic plants were reduced compared to those of wild‐type plants. In contrast, when we used this same promoter to drive CKX (a cytokinin degradation gene) expression, the transgenic tobacco plants displayed enhanced root elongation and biomass. We then made crosses between the SbUGT::CKX and SbUGT::iaaM transgenic plants. We observed that overexpression of the CKX gene neutralized the negative effects of auxin overproduction on root elongation. Also, the simultaneous expression of both the iaaM and CKX genes in rootstock did not disrupt normal growth and developmental patterns in wild‐type scions. Our results demonstrate that expression of both the iaaM and CKX genes predominantly in roots of rootstock inhibits lateral bud release from rootstock, improves grafting success rates and enhances root initiation and biomass.  相似文献

The Asian citrus psyllid, Diaphorina citri Kuwayama (Hemiptera: Liviidae), is a significant citrus pest and the parasitoid Tamarixia radiata (Waterston) (Hymenoptera: Eulophidae) has been released in various citrus‐producing areas in classical biological control programs targeting D. citri. We investigated the effect of host deprivation on the foraging behavior and patch utilization by T. radiata. In the laboratory, females deprived of hosts for 3 days tended to leave patches of 12 nymphs without parasitizing hosts during the ca. 30 min they spent in the patch before leaving. Moreover, half of these females failed to host feed, and those that did host feed, on average, needed more than 15 min to complete feeding. Conversely, non‐host‐deprived females parasitized on average three nymphs before leaving patches without host feeding during the ca. 39 min they spent in the patch. These laboratory observations were compared to mass‐reared female T. radiata that were released onto colonies of D. citri nymphs infesting citrus in the field. Release vials were provisioned with honey and these females had no opportunities to host feed over the 1‐ to 2‐day containment period prior to release. When introduced onto D. citri colonies, 68% T. radiata females abandoned D. citri patches prior to probing hosts, in part, because Argentine ants, Linepithema humile (Mayr) (Hymenoptera: Formicidae), tending colonies disturbed searching parasitoids. These results from laboratory and field studies are discussed in the context of classical biological control, with the aim of understanding how to manipulate host availability and ant activity so establishment rates and impact of T. radiata can be improved.  相似文献

In an attempt to suppress the propagation of citrus greening disease in Indonesia, we explored pathogens ofDiaphorina citri which vectors the disease. At two orange orchards, manyD. citri adults were found to be dead and covered with fungal mycelia. Two fungi,Paecilomyces fumosoroseus andHirsutella citriformis, were consistently isolated from the infected insects. Molecular phylogeny of their 18S rDNA sequences showed that they belong to the ascomycetous clade of the Clavicipitales/Hypocreales, which embraces many entomopathogenic fungi. When healthy adults ofD. citri were inoculated with conidia of theP. fumosoroseus, the insects died within 6 d.  相似文献

嫁接栽培是茄果类蔬菜防治土传病害和提高产量的重要措施之一。茄子野生近缘种托鲁巴姆(Solanums torvum)因综合抗性强,成为茄子和番茄嫁接的常用优良砧木。但是,由于托鲁巴姆种子的发芽率、发芽势和发芽指数较低,苗龄较长,限制了其在工厂化育苗中的大规模应用,因此迫切需要开发其他方法及相应技术体系提高托鲁巴姆的育苗效率,降低育苗成本。为优化托鲁巴姆微扦插技术,该研究探索并优化了试管内微扦插繁殖托鲁巴姆技术,以无菌播种获得初代无菌苗的茎段为外植体,通过在培养基中添加植物生长调节剂,对比不同浓度植物生长调节剂对托鲁巴姆微扦插繁殖过程中的影响。结果表明:(1)托鲁巴姆在不同培养基中,腋芽诱导、继代增殖和生根培养的效果存在显著差异,初代芽诱导的最佳培养基为MS+KT 0.5 mg·L-1+IBA 0.1 mg·L-1,出芽率达90%。(2)继代扦插最佳培养基为MS+IBA 0.4 mg·L-1,培养30 d的增殖系数达6.11,植株长势健壮。(3)最佳生根培养基为1/2MS+IBA 0.2 mg·L-1,生根培养30 d,单株一级根数4.56条,最长根长125.80 mm、根粗0.50 mm,根系发达。采用试管内微扦插技术繁殖托鲁巴姆种苗,操作简单,增殖系数较高,可满足快速繁育种苗的要求。该研究结果为托鲁巴姆的工厂化规模育苗提供了新途径。  相似文献

Citrus canker [caused by Xanthomonas citri subsp. citri (Xcc)] can cause yield loss of susceptible citrus and result in trade restrictions of fresh fruit. For both regulatory purposes and epidemiological studies, accurate detection and quantification of viable inoculum are critical. Two accepted methods used to detect and quantify Xcc are injection–infiltration bioassay and culture, but these two methods have not been directly compared using field‐obtained samples. The two methods were compared using washates of lesions taken from fruit, leaves and shoots in a commercial orchard in Florida in 2009–2010 and 2010–2011, with bioassay being the assumed standard. Despite some misclassifications, true positives (sensitivity) and true negatives (specificity) were the dominant classes using culture. False positives for lesions from shoots ranged from 13.1 to 21.4% in 2009–2010 and 2010–2011, respectively, and false positives for lesions from fruit and leaves ranged from 4.3 to 15.7%, in the two seasons, respectively. The false positive rate for culture compared with injection–infiltration bioassay was highest (0.16–0.55), due to more frequent recovery of Xcc by culture at ≤103 colony‐forming units (CFU) Xcc per ml. The false negative rate was consistently lower (0.02–0.21), confirming that in only a few cases did culture fail to detect Xcc when it was present. The area under the curve for receiver operator characteristic analysis ranged from 0.80 to 0.97, confirming that culture provided an accurate diagnosis in most cases. There was a higher frequency of lesions from shoots with a CFU ≤103 Xcc compared with lesions from fruit or leaves, making culture more effective at detecting these. The data demonstrate that culture is a reliable way to detect and quantify Xcc compared with injection–infiltration bioassay, particularly when the CFU is ≤103 Xcc per ml.  相似文献

为了解重庆三峡库区柑橘叶片硼营养状况及其影响因子,在该区域12个主产县(区)的代表性果园采集叶片样品954份和土壤样品302份,测定叶片硼含量,并分析了土壤有效硼、土壤pH值、品种、砧木和树龄对叶片硼营养的影响.结果表明: 该区域柑橘叶片硼含量不足(<35 mg·kg-1)的果园比例达41.6%,土壤有效硼含量不足(<0.5 mg·kg-1)的果园比例高达89.4%,柑橘叶片硼含量与土壤有效硼含量的相关性未达显著水平.土壤pH值、品种、砧木和树龄均影响叶片硼含量.pH值4.5~6.4土壤上的柑橘叶片硼含量显著高于pH值6.5~8.5土壤上的柑橘;品种间叶片硼含量为:温州蜜柑>柚类>夏橙>普通甜橙>杂柑>脐橙;枳砧和酸柚砧柑橘叶片硼含量显著高于枳橙砧和红橘砧柑橘;3~8年生柑橘树叶片硼含量适宜(35~100 mg·kg-1)样品比例比8年生以上柑橘树高6.6%.  相似文献
