Hongkong kumquat (Fortunella hindsii) is a wild citrus species characterized by dwarf plant height and early flowering. Here, we identified the monoembryonic F. hindsii (designated as ‘Mini‐Citrus’) for the first time and constructed its selfing lines. This germplasm constitutes an ideal model for the genetic and functional genomics studies of citrus, which have been severely hindered by the long juvenility and inherent apomixes of citrus. F. hindsii showed a very short juvenile period (~8 months) and stable monoembryonic phenotype under cultivation. We report the first de novo assembled 373.6 Mb genome sequences (Contig‐N50 2.2 Mb and Scaffold‐N50 5.2 Mb) for F. hindsii. In total, 32 257 protein‐coding genes were annotated, 96.9% of which had homologues in other eight Citrinae species. The phylogenomic analysis revealed a close relationship of F. hindsii with cultivated citrus varieties, especially with mandarin. Furthermore, the CRISPR/Cas9 system was demonstrated to be an efficient strategy to generate target mutagenesis on F. hindsii. The modifications of target genes in the CRISPR‐modified F. hindsii were predominantly 1‐bp insertions or small deletions. This genetic transformation system based on F. hindsii could shorten the whole process from explant to T1 mutant to about 15 months. Overall, due to its short juvenility, monoembryony, close genetic background to cultivated citrus and applicability of CRISPR, F. hindsii shows unprecedented potentials to be used as a model species for citrus research. 相似文献

Recently, CRISPR‐Cas12a (Cpf1) from Prevotella and Francisella was engineered to modify plant genomes. In this report, we employed CRISPR‐LbCas12a (LbCpf1), which is derived from Lachnospiraceae bacterium ND2006, to edit a citrus genome for the first time. First, LbCas12a was used to modify the CsPDS gene successfully in Duncan grapefruit via Xcc‐facilitated agroinfiltration. Next, LbCas12a driven by either the 35S or Yao promoter was used to edit the PthA4 effector binding elements in the promoter (EBEPthA4‐CsLOBP) of CsLOB1. A single crRNA was selected to target a conserved region of both Type I and Type II CsLOBPs, since the protospacer adjacent motif of LbCas12a (TTTV) allows crRNA to act on the conserved region of these two types of CsLOBP. CsLOB1 is the canker susceptibility gene, and it is induced by the corresponding pathogenicity factor PthA4 in Xanthomonas citri by binding to EBEPthA4‐CsLOBP. A total of seven 35S‐LbCas12a‐transformed Duncan plants were generated, and they were designated as #D35s1 to #D35s7, and ten Yao‐LbCas12a‐transformed Duncan plants were created and designated as #Dyao1 to #Dyao10. LbCas12a‐directed EBEPthA4‐CsLOBP modifications were observed in three 35S‐LbCas12a‐transformed Duncan plants (#D35s1, #D35s4 and #D35s7). However, no LbCas12a‐mediated indels were observed in the Yao‐LbCas12a‐transformed plants. Notably, transgenic line #D35s4, which contains the highest mutation rate, alleviates XccΔpthA4:dCsLOB1.4 infection. Finally, no potential off‐targets were observed. [目的] 明确佛冈县不同类型橘园柑橘木虱的种群动态,为当地柑橘木虱及柑橘黄龙病的防治工作提供科学依据。[方法] 通过2015-2016年的系统调查,对广东省佛冈县砂糖橘园的柑橘木虱种群消长动态进行研究,对比分析了不同处理措施对柑橘木虱种群数量的影响。[结果] 1-3月柑橘木虱发生危害较轻,随着嫩梢增多,其种群数量开始上升,6-9月为种群发生高峰期,期间出现多个发生高峰,10月之后种群数量逐渐进入消退期,种群数量维持在较低水平;2015、2016年佛冈县柑橘木虱的季节性消长动态基本一致,但种群发生量存在明显差别;3种不同类型橘园内柑橘木虱种群发生数量也存在显著差异,常年失管橘园柑橘木虱种群发生数量最高,常规水肥管理橘园次之,二者均显著高于常规水肥管理及化学防治橘园。[结论] 化学防治可在一定程度上控制柑橘木虱的种群数量,及时清理失管橘园对控制柑橘木虱和柑橘黄龙病传播至关重要。 相似文献

Résumé Le tube digestif des cochenilles se caractérise par la présence d'un dispositif intestinal appelé chambre filtrante. Celle-ci, formée par l'accolement des deux extrêmités de l'intestin moyen, est supposée intervenir dans le transit alimentaire en éliminant par «voie raccourcie» l'eau ingérée en excès, mais ce rôle est resté jusque là hypothétique.Chez Planococcus citri, l'étude cytologique et histochimique révèle des caractères propres aux parties de l'intestin moyen participant à la réalisation de ce dispositif. Ces caractères suggèrent une fonction particulière qui peut être liée à un mouvement de l'eau. Les cellules sont de taille très réduite. Par endroits, des différences ultrastructurales s'observent entre les deux anses accolées (présence de vacuoles et d'espaces extracellulaires développés). Les cellules non modifiées de l'anse de départ présentent un caractère morphologique particulier, le système d'espaces intercellulaires et un caractère histochimique dominant, l'importante quantité de glycogène. Les cellules non modifiées de l'anse de retour présentent une structure inhabituelle, les «expansions tubulaires» associées aux microvillosités. En absence du «plasmalemma mitochondrial complex» le passage de l'eau se ferait par osmose dans deux directions: d'une part à travers les épithéliums amincis, d'autre part par le réseau d'espaces intercellulaires.
Studies on the filter-chamber of Planococcus citri risso (Insecta, Homoptera)Histochemistry and ultrastructure
Summary The digestive tract of the coccids is characterized by the presence of a filter chamber formed by the apposition of the two extremities of the midgut. It is supposed that the filter chamber plays a role in the elimination of excess water by a short circuit but this assumption is still hypothetical.In Planococcus citri, cytological and histochemical studies have revealed structural peculiarities in those parts of the midgut which participate in the organization of the filter chamber, which suggest a particular function. The cells are highly reduced in size. In certain regions ultrastructural differences exist between the two apposed extremities of the midgut such as the presence of a vacuole and of highly developed extra-cellular spaces. These features may be related to water transport. The conventional cells of the proximal end of the midgut show the presence of a system of intercellular spaces and are very rich in glycogen. The conventional cells of the distal end of the midgut possess extracellular microtubules associated with the microvilli. In absence of a plasmalemma mitochondrial complex the transport of water could occur by osmosis across the thinned epithelium and the network of intercellular spaces.

Plant-soil interactions in weathered soils are so complex that unqualified statements about a suitable pH for plants are risky.
Conventional experimental designs and statistical methods may not be appropriate for investigating such complexities. Lime
experiments using continuous function designs and observation of plant response to indigenous variability in soil pH permit
detailed observations of plant-soil interactions that are frequently not detected. A graphical boundary-line approach to interpreting
data can make good sense out of apparent confusion.
Increasing the pH of variable-charge soils by adding lime or by indigenous means increased CEC and retarded cation leaching,
but Ca solubility changed very little over the range pH 5 to 6. N fixation and yield was closely related to soil pH, soil
Mn and Mn uptake by soybean. This result was clearly demonstrated regardless of numerous other limiting factors. Plant yield
response curves resolved into distinct segments that corresponded with associated soil properties. Excess Al compounded by
Ca deficiency is suspect in the pH range <5. Excess Mn, and Ca deficiency probably limited yields in the pH range 5.0 to 5.7.
Yields were stable, and Ca and P were constant in the pH interval 5.7 to 6.0. Yields abruptly increased in the pH interval
6.0 to 6.3. This was associated with elevated Ca concentrations in soil solutions. 相似文献

A somatic hybrid plant was obtained by protoplast fusion between navel orange and satsuma mandarin. Protoplasts isolated from nucellar calli of navel orange and from leaves of satsuma mandarin were fused by the PEG method. The fusion products were cultured in a Murashige & Tucker medium containing 0.6 M sucrose. In this medium, some colonies developed into whole plants through embryogenesis. One of the regenerated plants was shown to be a hybrid, which was proven by restriction endonuclease analysis of nuclear ribosomal DNA. The chromosome number of the hybrid was 36. Stylet insertion sites on the leaf surface and leaf cell types fed upon by nymphs and adults of the pear psylla, Psylla pyricola Foerster (Homoptera: Psyllidae) were identified using histological techniques and electronic monitoring of probing and feeding activities. Neither the nymphs nor the adult ingested predominantly from the phloem of Pyrus communis cv. Bartlett. We showed that the pear psylla ingests from all leaf cell types, but that xylem, phloem and bundle sheath cells are more acceptable for ingestion than non-vascular tissues. The possible sensory mechanisms underlying selection of stylet insertion sites on the leaf surface and acceptance of various cell types for ingestion are discussed.
Résumé Les lieux d'insertion des stylets sur la surface de la feuille et les types cellulaires consommés par les larves et les adultes de Psylla pyricola Foerster ont été précisés par étude histologique et par enregistrement électronique du comportement de piqûre et d'alimentation. Ni les larves, ni les adultes n'ont absorbé de préférence le phloème de Pyrus communis de la variété Bartlett. Nous avons constaté que le psylle du poirier ingère le contenu de tous les types cellulaires, mais que le xylème, le phloème et autre cellules des faisceaux libéro-ligneux étaient préferés aux tissus non vasculaires. La discussion a porte sur les méchanismes sensoriels éventuellement responsables de la sélection des lieux d'insertion des stylets à la surface de la feuille et l'acceptation des différents types cellulaires lors de l'ingestion.相似文献

The influence of nitrogen concentration and ammonium/nitrate ratio on N-uptake,mineral composition and yield of citrus 总被引:9,自引:0,他引:9
In short-term water culture experiments with different 15N labeled ammonium or nitrate concentrations, citrus seedlings absorbed NH4
+ at a higher rate than NO3
–. Maximum NO3
– uptake by the whole plant occurred at 120 mg L–1 NO3
–-N, whereas NH4
+ absorption was saturated at 240 mg L–1 NH4
+-N. 15NH4
+ accumulated in roots and to a lesser degree in both leaves and stems. However, 15NO3
– was mostly partitioned between leaves and roots.Adding increasing amounts of unlabeled NH4
+ (15–60 mg L–1 N) to nutrient solutions containing 120 mg L–1 N as 15N labeled nitrate reduced 15NO3
– uptake. Maximum inhibition of 15NO3
– uptake was about 55% at 2.14 mM NH4
+ (30 mg L–1 NH4
+-N) and it did not increase any further at higher NH4
+ proportions.In a long-term experiment, the effects of concentration and source of added N (NO3
– or NH4
+) on nutrient concentrations in leaves from plants grown in sand were evaluated. Leaf concentration of N, P, Mg, Fe and Cu were increased by NH4
+ versus NO3
– nutrition, whereas the reverse was true for Ca, K, Zn and Mn.The effects of different NO3
–-N:NH4
+-N ratios (100:0, 75:25, 50:50, 25:75 and 0:100) at 120 mg L–1 total N on leaf nutrient concentrations, fruit yield and fruit characteristics were investigated in another long-term experiment with plants grown in sand cultures. Nitrogen concentrations in leaves were highest when plants were provided with either NO3
– or NH4
+ as a sole source of N. Lowest N concentration in leaves was found with a 75:25 NO3
–-N/NH4
+-N ratio. With increasing proportions of NH4
+ in the N supply, leaf nutrients such as P, Mg, Fe and Cu increased, whereas Ca, K, Mn and Zn decreased. Yield in number of fruits per tree was increased significantly by supplying all N as NH4
+, although fruit weight was reduced. The number of fruits per tree was lowest with the 75:25 NO3
–-N:NH4
+-N ratio, but in this treatment fruits reached their highest weight. Rind thickness, juice acidity, and colour index of fruits decreased with increasing NH4
+ in the N supply, whereas the % pulp and maturity index increased. Percent of juice in fruits and total soluble solids were only slightly affected by NO3
–:NH4

Aims: To investigate the effect of tea polyphenol (TP) and Candida ernobii alone or in combination against postharvest disease (Diplodia natalensis) in citrus fruit and to evaluate the possible mechanisms involved. Methods and Results: TP at concentrations of 0·1%, 0·5% and 1·0% alone, or in combination with C. ernobii (1 × 106 CFU ml?1), showed a lower infection rate of stem‐end rot. TP at the concentration of 0·5% or above significantly inhibited the spore germination of D. natalensis. TP at the concentration of 1·0% showed inhibitary ability on mycelium growth of D. natalensis. The addition of TP did not affect the growth of C. ernobii in vitro and significantly increased the population of C. ernobii in vivo. Conclusions: TP exhibited an inhibitory effect against D. natalensis and improved the biocontrol efficacy of C. ernobii. It was direct because of the inhibitory effects of TP on spore germination and mycelial growth of D. natalensis in vitro and indirect because of the increased populations of C. ernobii in vivo. Significance and Impact of the Study: The results suggested that TP alone or in combination with biocontrol agents has great potential in commercial management of postharvest diseases in fruits. 相似文献
